형태적 특성이 다른 들깻잎, 상추, 케일 중 농약 잔류량 비교 (Comparison of Pesticide Residues in Perilla Leaf, Lettuce and Kale by Morphological Characteristics of Plant)

  • 본 실험은 엽채류 중 들깻잎, 상추, 케일에 살포된 농약의 잔류량을 비교하기 위해 수행하였다. 농약 9종을 대상으로 2구 선형노즐을 부착한 배부식 동력분무기로 7일 간격 2회 살포 후 0, 2, 5일의 잔류양상을 조사하였다. 무게 대비 엽면적 비율($cm^2/g$)은 들깻잎 58, 상추 27, 케일 23 으로 나타나 들깻잎이 상추 보다 2.1배 높았다. 살포 직후 잔류량은 들깻잎이 상추 보다 1.3~2.3배 높게 나타났으며 케일은 상추보다 2.4~7.3배 높았다. 살포 후 5일에는 들깻잎이 상추 보다 1.3~3.3배 많았으며 상추는 케일보다 5.6~28.1배 높았다. 케일의 잔류량이 적은 것은 잎 표면이 매끄러워 살포된 농약입자가 쉽게 흘러내리는 것이 원인으로 보인다. 상추와 들깻잎의 잔류량 차이는 무게 대비 엽면적 비율의 차이에 따른 영향이 크다고 판단되었다.

Allelopathic and Autotoxic Effects of Alfalfa Plant and Soil Extracts

  • Alfalfa (Medicago sativa L.) plants have been reported to be autotoxic as well as allelopathic. Laboratory and greenhouse experiments through petri-dish and pot test were conducted to determine autotoxic effects of alfalfa leaf and soil extracts on the germination or early seedling growth of alfalfa, and to evaluate allelopathic effects of alfalfa leaf residues on alfalfa, barnyard grass, com, eclipta and soybean. Alfalfa seed germination was delayed depending on aqueous extract concentration, with no difference in final germination after 48 hours. Alfalfa root length was more sensitive to the autotoxic chemicals from leaf extracts than was germination or shoot length. Root growth of alfalfa was significantly inhibited at extract concentration of more than 1 g dry tissue/L (g $\textrm{L}^{-1}$). Hypocotyl growth, however, was not affected by all the concentrations of leaf extracts. Soil extracts from 4-yr-old alfalfa stand significantly reduced alfalfa root length by 66%, while soil extracts from 0,1, and 3yr-old stand stimulated root length up to 14-32% over the control. Residue incorporation with dry matters of alfalfa leaf at 100 g $\textrm{kg}^{-1}$ reduced seedling length of several crop and weed species, ranging from 53 to 87% inhibition. Addition of nutrient solution into alfalfa leaf extracts alleviated alfalfa autotoxic effect. This result indicates alfalfa leaf and soil extracts or residues could exert autotoxic as well as allelopathic substances into soil environments during and after establishment.

토마토 잎에 집적된 Triflumizole 잔류량의 온도에 따른 경시적 동태와 잎곰팡이병균에 대한 약호 (Temporal Dynamics of Triflumizole Residue in Tomato Leaves as Affected by Temperature and Its Effects on Cladosporium fulvum)

  • 토마토 잎에 집적된 triflumizole 잔류량 동태에 미치는 온도 영향과 Cladosporium fulvum에 대한 triflumizole의 살균효과를 구명하였다. 8주간 키운 토마토 식물체에 triflumizole을 분무 살포한 후 식물생장용 항온실에서 키우면서 4가지 온도 조건을 처리하였다. 주기적으로 토마토 잎을 채취하여 HPLC 방법을 이용하여 triflumizole의 잔류량을 조사하였다. Triflumizole 잔류량의 경시적 동태는 약제살포후 적산온도를 독립변수로 사용하는 지수적 감소 몸형으로 나타낼수 있었으며, 이는 triflumizole의 잔류성이 온도 영향을 크게 받는다는 것을 뜻한다. 경기도 농촌진흥원에 위치한 토마토 비닐하우스에서 수행한 실험에서 얻은 자료를 이용하여 평가한 결과 이 모형의 타당성이 인정되었다. Triflumizole의 약효에 대한 in vitro 실험 결과, 이 살균제는 C. fulvum의 균사 생장과 포자형성을 뚜렷이 억제하였으며, 잎곰팡이병 방제를 위해서는 토마토 잎에 집적된 triflumizole 잔류량이 최소한 10 ppm 이상이 되어야 할 것으로 추측된다. 본 연구에서 연구된 지수적 감소 모형은 토마토 잎에 집적된 triflumizole 잔류량을 추정하는데 활용될 수 있으며, 실용적인 측면에서 볼 때, 토마토 잎곰팡이병을 효과적으로 방제하는데 필요한 triflumizole의 최소 잔류량을 식물체에 유지하기 위하여 triflumizole을 살포해야 할 시기를 결정하는데 활용될 수 있다.

  • This experiment was conducted at the forage experimental field, College of Agriculture and Life Sciences, Seoul National University, Suweon in 1992 and 1993 to investigate the effects of tillage method-conventional and rotary-till, rye (Secde cereale L.) harvest date-early (April 14) and late (April 26), and rye residue treatmentno paraquat(1, 1-dinethyl-4,4'-bipyridinium dichloride) and paraquat in minimizing the adverse effects of the rye residue on growth and yield of succeeding corn(Zea mays L.). Corn plant height during the growing season was slighly taller with conventional tillage relative to rotary-till when rye was harvested in early and treated by paraquat. Corn LA1 during the growing season was slighly increased when rye was harvested in early and where conventional tillage was used with paraquat treatment. There were no differences in the leaf number and silking dates of corn among the tillage methods, harvest dates of rye and paraquat treatments. The dry matter yield of corn was significantly increased by paraquat treatment when rye was harvest in early, but no differences were found in the dry matter percentage, ear percent to total dry matter, and stover, ear and estimated TDN yields of corn among the treatments.

물오리나무 유래 고함량 Oregonin 추출방법 및 생리활성 연구 (Extraction Method and Physiological Activity of High Content Oregonin Derived from Plant of Alnus sibirica Fisch. ex Turcz)

  • Oregonin is known as a representative active substance of plants of the Alnus species. In this study, we will show the results of establishing a method for extracting trace amounts of useful substances in plants. Additionally, a variety of physiological activity studies were conducted with extracts containing high content of useful substances. In this study, anti-oxidant, anti-inflammatory and anti-allergic activities of supercritical fluid residue 60% EtOH extracts of Alnus sibirica (ASCFR) were investigated in vitro. To explore anti-oxidant and anti-inflammatory characteristics of supercritical extract, DPPH radical scavenging, and ABTS radical scavenging activity, inhibition of nitric oxide production in LPS-stimulated RAW 264.7 cells and ${\beta}$-hexosaminidase release in IgE-stimulated RBL-2H3 cells were examined, respectively. Results showed a concentration-dependent increase in DPPH and ABTS radical scavenging activity. Cells stimulated with LPS produced more nitric oxide than normal control cells. However, cells treated with supercritical fluid residue 60% EtOH extract decreased production in a concentration-dependent manner. In anti-allergic activity studies, ASCFR showed concentration-dependentanti-allergic activity. Based on results and strong antioxidant activity, ASCFR has potential as basic research materials for development of therapeutic supplements-based medicines or functional cosmetics related to chronic inflammatory skin immunity diseases.

당근의 알콜불용성 잔사와 셀룰로오스 분획의, p-Hydroxybenzoic Acid 추출에 미치는 시판 식물세포벽분해효소의 영향 (Effect of Commercial Plant Cell Wall Degrading Enzymes on Extraction of p-Hydroxybenzoic Acid from Carrot Alcohol Insoluble Residue (AIR) and Cellulose Fraction)

  • Driselase, Cellulase, Macerozyme R-200, Macerozyme R-10및 Sumyzyme MC 등 5종의 시판 식물세포벽분해효소가 당근의 세포벽물질(CWM)로부터 제조한 알콜불용성 잔사(AIR)와 cellulose fraction의 f-hydroxybenzoic acid 추출에 미치는 영향을 조사하였다. 당근AIR의 주된 페놀화합물은 P-hydroxybenzoic acid로 1,977 $\mu$g/g AIR였으며, vanillin, ferulic acid, p-hydroxybenzaldehyde가 각각 55.9, 13.6, 10.6 $\mu$g/g AIR인 것으로 나타났다. 효소에 함유되어있는 ferulic acid의 함량은 Driselase, Cellulase, Macerozyme R-200, Macerozyme R-10, Sumyzyme MC에서 각각 2,319, 2,060, 391, 95.2, 34.1 $\mu$g/g인 것으로 나타났다. 이들 효소와 세포벽물질과의 반응을 조사한 결과, Driselase와 AIR의 반응 후 유리된 p-hydroxybenzoic acid의 함량은 56 $\mu$g/g AIR로 알칼리 (4 M NaOH) 추출한 총량의 2.8$\%$에 해당한다. 따라서 p-hydroxybenzoic acid는 AIR과 셀룰로오스 분획으로부터 이들 효소에 의해 분리되기 어려운 것으로 나타났다.

케일 중 살충제 Chromafenozide와 Pyridalyl의 잔류 특성 (The Residual Characteristic of Chromafenozide and Pyridalyl in Kale)

  • BACKGROUND: It is a very important task to block distribution of the agricultural products contaminated with pesticides in advance to protect consumers from residual pesticides among the agricultural products. Therefore, this study was performed to determine residual characteristics of pesticides in time-dependent manner and present scientific evidences for pre-harvest residue limit (PHRL) setting. METHODS AND RESULTS: The tested pesticides, chromafenozide and pyridalyl were sprayed onto the kale twice (seven day intervals) and then the plant samples were collected at 0 (after 3 hours), 1, 2, 3, 5, 7, and 10 days after the last spraying. Residual concentration of chromafenozide in kale decreased with 74.9% (of decreased fraction, field 1) and 85.3% (field 2) and pyridalyl decreased with 81.2% (field 1) and 85.8% (field 2), calculated by comparisons of the concentrations at 0 day and 10 days. Also biological half-lives of chromafenozide in kale were 5.6 day (field 1) and 3.4 day (field 2), and those of pyridalyl were 4.3 day (field 1) and 3.5 day (field 2). CONCLUSION(S): If the residues of chromafenozide and pyridalyl in kale from 10 days before harvest are less than 37.6 mg/kg and 58.9 mg/kg, respectively, it is expected that safe kale below MRL can be supplied on the pre-harvest day.

The development and validation of a novel liquid chromatography tandem mass spectrometry (LC-MS/MS) procedure for the determination of fluoroquinolones residues in chicken muscle using modified QuEChERS (quick, easy, cheap, effective, rugged and safe) method

  • A novel rapid procedure with liquid chromatography tandem mass spectrometry (LC-MS/MS) detection has been developed by changing various conditions including sample preparation such as QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) methodology. This work has been involved the optimization and validation of detection method for fluoroquinolones which are widespread used in livestock especially in the chicken. Five grams of homogenized chicken muscle were extracted with QuEChERS EN and acetonitrile containing 5% formic acid and cleaned with anhydrous magnesium sulfate and C18 sorbent. The separation was performed on Acquity UPLC HSS T3 (2.1 mm×100 mm, 1.8 ㎛) column. The mobile phase A and B were composed of water containing 0.1% formic acid and acetonitrile containing 0.1% formic acid, respectively. Flow rate was 0.25 mL/min and column temperate was 40℃. LC-MS/MS with multiple reaction monitoring has been optimized for ten fluoroquinolones (ciprofloxacin, danofloxacin, difloxacin, enrofloxacin, marbofloxacin, norfloxacin, ofloxacin, orbifloxacin, pefloxacin and sarafloxacin). The method developed in this study has been presented good linearity with correlation coefficient (R2) of 0.9971~0.9998. LOD and LOQ values ranged from 0.09 to 0.76 ppb and from 0.26 to 2.29 ppb, respectively. The average recoveries were from 77.46 to 111.83% at spiked levels of 10.0 and 20.0 ㎍/kg. Relative standard deviation (%) ranged 1.28~11.90% on intra-day and 3.10~8.38 % on inter-day, respectively. This analysis method was applicable to the livestock residue laboratories and was expected to be satisfactory for the residue surveillance system.

파프리카 재배기간 중 담배나방 방제에 사용되는 살충제의 잔류특성 (Residual Characteristics of Insecticides Used for Oriental Tobacco Budworm Control of Paprika)

  • 본 연구는 파프리카 재배 중 담배나방 방제에 사용되는 살충제의 잔류특성을 알아보고 추천 생산단계 잔류허용기준(Pre-Harvest Residue Limit, PHRL) 설정을 통하여 안전한 파프리카 생산에 기여하고자 수행되었다. 파프리카에 각각의 농약을 기준량 처리와 배량 처리로 살포한 후 1, 3, 5, 7, 10, 12, 15, 18 그리고 21일에 일정한 간격에 맞추어 파프리카 시료를 수확하여 잔류농약을 분석하고 생물학적 반감기를 산출한 다음 추천 생산단계 잔류허용기준(Pre-Harvest Residue Limit, PHRL)을 설정하였다. 시료들은 QuEChERS법으로 추출한 후 $NH_2$ SPE cartridge와 PSA을 이용하여 정제하고 HPLC/DAD와 GLC/ECD를 이용하여 기기분석을 하였다. 5개 살충제의 검출한계는 모두 0.01 mg/kg이며 회수율은 검출한계의 10배, 50배 농도인 0.1, 0.5 mg/kg가 되도록 3반복 처리하여 $81.3{\pm}1.62%$에서 $98.3{\pm}1.58%$였다. 시험 살충제의 생물학적 반감기는 bifenthrin에서 11.8일, chlorantraniliprole은 8.5일, chlorfenapyr는 16.8일, lamda-cyhalothrin은 7.1일, methoxyfenozide에서 31.3일로 나타났다. 생산단계 농약 잔류허용기준은 수확 10일 전 bifenthrin, chlorantraniliprole, chlorfenapyr, lamda-cyhalothrin 그리고 에서 methoxyfenozide에서 각각 1.05 mg/kg, 1.41 mg/kg, 0.93 mg/kg, 2.06 mg/kg 그리고 1.08 mg/kg으로 제안하였다.

단감 재배기간 중 살균제 pyrimethanil과 trifloxystrobin의 생산단계 잔류허용기준 설정 (Establishment of Pre-Harvest Residue Limit of Fungicides Pyrimethanil and Trifloxystrobin during Cultivation of Persimmon)

  • 본 연구는 단감 재배 중 사용되는 살균제인 pyrimethanil과 trifloxystrobin의 생산단계 잔류허용기준 설정을 통하여 안전한 단감 생산에 기여하고자 수행되었다. 먼저 pyrimethanil과 trifloxystrobin을 살포하고, 살포 후 0, 1, 2, 3, 5, 7, 10, 12, 15, 18일에 단감 시료를 채취하여 각각의 농약을 분석하고 생물학적 반감기를 산출한 다음 생산단계 잔류허용기준(PHRL; Pre-Harvest Residue Limit)을 설정하였다. 단감 중 pyrimethanil과 trifloxystrobin은 Acetonitrile을 이용하여 추출하고 $NH_2$ cartridge와 PSA를 이용하여 정제하여 HPLC/DAD를 이용하여 분석하였다. 두 농약에서 검출한계는 모두 0.01 mg/kg이었다. Pyrimethanil의 회수율은 0.1과 0.5 mg/kg 두 수준에서 각각 $81{\pm}1.62%$, $98{\pm}1.58%$ 이었으며, trifloxystrobin의 회수율은 0.1과 0.5 mg/kg 두 수준에서 각각 $91{\pm}2.94%$, $98{\pm}1.25%$이었다. 단감에서 pyrimethanil의 생물학적 반감기는 기준량 살포 시 15.6일, 배량 살포 시 11.6일이었고, trifloxystrobin의 생물학적 반감기는 기준량 살포 시 10.4일, 배량 살포 시 10.3일이었다. 잔류회귀 감소식을 이용한 생산단계 잔류허용기준은 pyrimethanil과 trifloxystrobin은 각각 수확 10일 전 2.69 mg/kg과 0.83 mg/kg으로 제안하였다.
