재식밀도 차이가 약용작물 홍화의 수량에 미치는 영향 (Effect of Row-width and Plant-spacing within Row on Yield in Safflower, Carthamus tinctorius L.)

  • 홍화증수를 위한 알맞는 재식밀도를 구명하고 그에 따를 수량 및 농업 형질의 변이를 조사하기 위하여 1980년 3월부터 10월30일까지 강원도 원주상지대학 실습포장에서 실험을 실시하여 얻은 결과를 요약하면 다음과 같다. 1. 초장은 휴폭과 주간이 좁을수록 커지는 것으로 나타났으며 이들 양자에 의해 영향을 받았다. 2. 주당 분지수는 휴폭과 주간이 좁아짐에 따라 감소하였으며 이를 양요인에 의해 영향을 받았다. 3. 지표면에서 제1분지까지의 높이는 주간에 의해 영향을 받으며, 좁을수록 그 위치가 높아졌다. 4. 초장은 휴폭과 부의 상관을 가지며, 초장과 제1분지까지의 높이는 휴폭x주간과 부상관을 갖는다. 또한 주당 분지수는 휴폭, 휴폭x주간과는 정상관, 초장과는 부상관을 보였다. 5. 수량은 주간, 휴폭x주간에 관하여 부상관을 가지며 제 1분지의 높이와는 정상관을 가졌다. 6. 수량은 주간에 의해 크게 영향을 받으며 다음은 주간xs휴폭이 영향하는 것으로 나타났다. 휴폭은 수량에 크게 영향하지 않으나 30cm 또는 40cm구가 가장 증수되었으며 주간은 5cm구가 가장 증수되어 30cm x 5cm구, 또는 40cm x 5cm구가 홍화의 증수를 위한 적철한 재식밀도인 것으로 나타났다.

SSR 마커를 이용한 유럽 양송이 자원의 유전적 다양성 및 집단구조분석 (Genetic diversity and population structure of European button mushroom (Agaricus bisporus) using SSR markers)

  • 본 연구에서는 유럽 양송이 자원들을 SSR marker를 통해 유전적 다양성과 집단 구조, 유전적 분화에 대하여 분석하였다. 본 연구에서 유럽의 양송이 자원들은 유전적 거리기반의 4개의 그룹으로 나뉘었고 집단구조 분석을 통하여 2개의 subpopulation으로 이루어져 있었다. 본 연구에서 사용한 SSR 마커로 유럽의 양송이 자원들은 지리적 그리고 갓색으로 구분되지 않았다. 유전적 다양성은 유전적 거리기반의 그룹에서는 Group 4, 집단구조 분석을 통한 subpopulation에서는 Pop. 2의 다양성이 높았다. 그리고 양송이 자원들은 유전적 분화가 매우 낮았다. 본 연구의 결과는 차후 양송이의 육종 등에 이용 할 수 있을 것이다.

QTL Analysis of Plant Height in Rice Using CNDH Population

  • Rice is a staple crop used by more than 50% of the world's population. However, in rapidly changing climates such as abnormal high temperatures and typhoons, the food security of rice is greatly threatened. Plant factories have the advantage of being able to grow crops regardless of climate change, so they can be a response to climate change. However, in plant factories, crops are grown by placing the culture bed vertically, so shorter crops are more efficient. Therefore, in order to search for genes related to the height of rice, QTL analysis was performed by investigating the plant height of Cheongcheong/Nagdong doubled haploids from 2017 to 2021. Plant height of rice investigated for five years showed a normal distribution, meaning that genes related to rice height are quantitative traits. As a result of QTL analysis, a total of 12 QTLs were detected, and QTLs overlapped for 5 years in RM12285-RM212 on chromosome 1. Also, The QTLs of plant height detected in 2019 has a LOD score of 17.64 in RM12285-RM212 region of chromosome 1. As a result of QTL analysis, 44 height-related genes were searched from the detected chromosomes, and among them, Os01g0757200 in RM 12285-RM212 on chromosome 1 region, named OsGA2ox3q1, were selected as genes related to the height of rice. The relative gene expression level of OsGA2ox3q1 was highly expressed in cultivar with short culm lines, and was low expressed in cultivar with long culm lines. OsGA2ox3q1 can be used to breed semi-dwarf cultivar in rice more efficiently.

Genetic Diversity and Relationship in Soybean MDP (Mutant Diversity Pool) Revealed by TRAP and TE-TRAP Markers

  • Mutation breeding is the useful tool to improve agronomic traits in various crop species. Soybean is most important crop and is rich in protein and oil contents. Despite of the importance as economic value and various genetic resource of soybean, there have been limited studies of genetic relationship among mutant resources through radiation breeding. In this study, the agronomical phenotype for selecting various genetic resources was evaluated in 528 soybean mutant lines. As a result, 210 soybean mutants with their original cultivars were selected with various traits. We named 210 selected lines as Mutant Diversity Pool (MDP). The genetic diversity and the relationship of the MDP were investigated using TRAP and TE-TRAP markers. In TRAP analysis, sixteen primer combination (PC)s were used and a total of 551 fragments were amplified. The highest (84.00%) and the lowest (32.35%) polymorphism levels were showed in PC MIR157B+Ga5 and B14G14B+Ga3, respectively. The mean of PIC values was 0.15 ranging from 0.07 in B14G14B+Sa12 to 0.23 in MIR157B+Sa4. Phylogenetic and population structure analysis indicated that the 210 MDP lines dispersed to four groups among the wild types and their mutants. The highest genetic diversity among populations was observed between lines Paldal and 523-7 (Fst=0.409), whereas the lowest genetic diversity was between population KAS360-22 and 94seori (Fst=0.065). AMOVA showed 11.583 (21.0%) and 43.532 (79.0%) variations in inter and intra mutant population, respectively. Overall, the genetic similarity of each intra mutant populations was closer than that of inter mutant population. A total of 408 fragments were amplified in the 210 MDP using twelve PCs of TE-TRAP markers that were obtained from a combination of three TIR sequence of transposable elements (MITE-stowaway; M-s, MITE-tourist; M-t, PONG). The highest (77.42%) and the lowest (56.00%) polymorphism levels were showed in PONG+Sa4 and PONG+Sa12, respectively. The mean of PIC values was 0.15 ranging from 0.09 in M-s+Sa4 and M-s+Ga5 to 0.21 in M-t+Ga5. AMOVA of M-s showed 2.209 (20%) and 8.957 (80%) variations in inter and intra mutant population, respectively. AMOVA of M-t showed 2.766 (18%) and 12.385 (82%) variations in inter and intra mutant population, respectively. AMOVA of PONG showed 3.151 (29%) and 7.646 (71%) variations in inter and intra mutant population, respectively. According to our study, the PONG had higher inter mutant population and lower intra mutant population. This mean was that for aspect of radiation sensitivity, M-s and M-t showed higher mobility than that of PONG. Our results suggest that the TRAP and the TE-TRAP markers may be useful for assessing the genetic diversity and relationship among soybean MDP and help to improve our knowledge of soybean mutation/radiation breeding.

수종 식물의 밀도-경쟁효과에 관한 연구 (Studies on the Competition-Density Effect of Some Higher Plants)

  • The studies of density effect or the effect of population density on plant growth have been done on basis of dry matter production with Raphanus acanthiformis var. simoodaeguen, Brassica campestris var. Pekinensis f. namsounsokoombecheu, Oryza sativa f. kimmajae and O. sativa f. mangyeng grown in the various spacing. 1. In the early period of plant growth in dry weight was not different each other among varying densities, but as time advanced the plant grown vast space grew sufficiently compared with those of narrow one. 2. Iogarithmic relation between the growth of plant (W) and the density (P), log W-log P in the material plants, were approximated by two straight lines, one was horizontal line and another inclined: the former showed non-competition density and the latter competition density addition to these the point interlinking both lines were implied of the optimum density per unit land area at certain growth period. 3. The values of relatvie growth rate (RGR) and net assimilation rate (NAR) were decreased as increase in the density, while those of leaf area ratio (LAR) were rather increased in the same condition, with minor exception. From these results and relation between the productive structure and due to lack of the recieved light intensity owing to the mutal shading among the plants.

  • Although it has been known though many morphological and physiological studies, its genetic diversity and population structure have not yet been investigated in this species. Therefore, detailed studies, in particular at the DNA level, on genetic diversity of natural populations of wild sea lettuce, and genetic relationships between natural sea lettuce and cultivated sea lettuce are necessary from the viewpoint of plant evolution. 저온 생장성 Pseudomonas fluorescens M45와 MC07을 이용한 오이의 생육촉진과 모잘록병의 방제 (Enhancement of Plant Growth and Suppression of Damping-off of Cucumber by Low Temperature Growing Pseudomonas fluorescens Isolates)

  • Growth rates of the low temperature growing isolates, Pseudomonas fluorescens M45 and MC07, reached maximum stationary phase within 50 hrs at the low temperature, 4$^{\circ}C$. But an ordinary biocontrol agent P. putida Pf3 did not reach logarithmic growth phase until 80 hrs. The culture filtrates of M45 and MC07 significantly inhibited the mycelial growths of Pythium ultimum, Rhizoctonia solani and Phytophthora capsici. Detached cotyledons of cucumber grown on Murashige and Skoog agar medium were much enhanced in their growth, compared to those without the filtrates. Population densities of M45 and MC07 in the rhizosphere at 14$^{\circ}C$ were more stable than at 27$^{\circ}C$. When M45 and MC07 were treated into soil, the population density of MC07 continuously increased up to 9 days after treatment, and sustained the initial inoculum density up to 60 days. Cucumber damping-offs caused by P. ultimum and R. solani were significantly reduced by applying M45 as seed-inoculant and by soil treatment with MC07. The combined treatment of M45 and MC07 provided greater effect in reducing the disease incidence than that obtained by single treatments.

Vegetative Compatibility Groups and Pathogenicity Variation among Isolates of Fusarium oxysporum f.sp. melonis

  • A total of 90 isolates of Fusarium oxysporum f.sp. melonis, the causal agent of oriental melon (Cucumis melo var. makuwa) wilt, was isolated from symptomatic tissues of oriental melon from 4 provinces in Korea. These isolates were grouped into vegetative compatibility groups (VCGs) by demonstrating heterokaryosis through complementation using nitrate nonutilizing (nit) mutants. No self-incompatibility was observed in any of isolates. All isolates were grouped into 3 VCGs ; A, B, and C. iSOLATES BELONGING TO VCG A and VCG B accounted for 87% and 91% of the fungal population collected in 1991 and 1993, respectively. As the increment of cultivation period in the same field, the proportion of isolates belonging to VCG B increased whereas that of isolates belonging to VCG A decreased. Mean virulence of a total population increased as the increment of cultivation period in the same field. Isolates belonging to VCG B showed the highest increment of virulence. These data suggest that replanting of a host plant in the same field may cause increase of virulence in the pathogens. Furthermore, virulence of F. oxysporum f.sp. melonis isolates is related to the VCGs.
