Elucidation of Antifungal Metabolites Produced by Pseudomonas aurantiaca IB5-10 with Broad-Spectrum Antifungal Activity

  • Antifungal metabolites were isolated from a culture of Pseudomonas aurantiaca IB5-10. Chemical structures of the metabolites were elucidated as phenazine-1-carboxylic acid (PCA; 1), 2-hydroxyphenazine (2-OH-PHZ; 2), and cyclo-(L-Pro-L-Val; 3), respectively, based on spectroscopic methods. Among them, 3 was isolated for the first time from this strain. The antifungal activities of 1-3 were evaluated against a variety of plant pathogens. To the best of our knowledge, the antifungal activities of 3 against plant fungal pathogens have been evaluated for the first time in this work. PCA (1) showed the most potent antifungal activities against Phytophthora capsici, Rhizoctonia solani AG-1(IA), and Pythium ultimum with MICs (${\mu}g/ml$) of less than 1.0, 1.3, and 2.0, respectively. On the other hand, 2-OH-PHZ (2) showed potent antifungal activity against R. solani AG-1(IA) with the MIC (${\mu}g/ml$) of 2.0, whereas it showed moderate antifungal activity against P. ultimum with the MIC (${\mu}g/ml$) of 50.0. In addition, 3 showed antifungal activity against only R. solani AG-1(IA).

Screening and Evaluation of Yeast Antagonists for Biological Control of Botrytis cinerea on Strawberry Fruits

  • Gray mold (Botrytis cinerea) is one of the most common diseases of strawberries (Fragaria${\times}$ananassa Duchesne) worldwide. Although many chemical fungicides are used for controlling the growth of B. cinerea, the risk of the fungus developing chemical resistance together with consumer demand for reducing the use of chemical fungicides have necessitated an alternative method to control this pathogen. Various naturally occurring microbes aggressively attack plant pathogens and benefit plants by suppressing diseases; these microbes are referred to as biocontrol agents. However, screening of potent biocontrol agents is essential for their further development and commercialization. In this study, 24 strains of yeast with antagonistic ability against gray mold were isolated, and the antifungal activity of the volatile and diffusible metabolites was evaluated. Putative mechanisms of action associated with the biocontrol capacity of yeast strains against B. cinerea were studied through in vitro and in vivo assays. The volatile organic compounds produced by the Galactomyces candidum JYC1146 could be useful in the biological control of plant pathogens and therefore are potential alternative fungicides with low environmental impact.

Occurrence and pathogenicity of Pythium (Oomycota) on Ulva species (Chlorophyta) at different salinities

  • Pythium species are ubiquitous organisms known to be pathogens to terrestrial plants and marine algae. While several Pythium species (hereafter, Pythium) are described as pathogens to marine red algae, little is known about the pathogenicity of Pythium on marine green algae. A strain of a Pythium was isolated from a taxonomically unresolved filamentous Ulva collected in an intertidal area of Oslo fjord. Its pathogenicity to a euryhaline Ulva intestinalis collected in the same area was subsequently tested under salinities of 0, 15, and 30 parts per thousand (ppt). The Pythium isolate readily infected U. intestinalis and decimated the filaments at 0 ppt. Mycelium survived on U. intestinalis filaments for at least 2 weeks at 15 and 30 ppt, but the infection did not progress. Sporulation was not observed in the infected algal filaments at any salinity. Conversely, Pythium sporulated on infected grass pieces at 0, 15, and 30 ppt. High salinity retarded sporulation, but did not prevent it. Our Pythium isolate produced filamentous non-inflated sporangia. The sexual stage was never observed and phylogenetic analysis using internal transcribed spacer suggest this isolate belongs to the clade B2. We conclude that the Pythium found in the Oslo fjord was a pathogen of U. intestinalis under low salinity.

Dual Biocontrol Potential of the Entomopathogenic Fungus, Isaria javanica, for Both Aphids and Plant Fungal Pathogens

  • Dual biocontrol of both insects and plant pathogens has been reported for certain fungal entomopathogens, including Beauveria bassiana and Lecanicillum spp. In this study, we demonstrate, for the first time, the dual biocontrol potential of two fungal isolates identified by morphological and phylogenetic analyses as Isaria javanica. Both these isolates caused mortality in the greater wax moth, and hence can be considered entomopathogens. Spores of the isolates were also pathogenic to nymphs of the green peach aphid (Myzus persicae), with an $LC_{50}$ value of $10^7spores/mL$ 4 days after inoculation and an $LT_{50}$ of 4.2 days with a dose of $10^8spores/mL$. In vitro antifungal assays also demonstrated a strong inhibitory effect on the growth of two fungi that are pathogenic to peppers, Colletotrichum gloeosporioides and Phytophthora capsici. These results indicate that I. javanica isolates could be used as novel biocontrol agents for the simultaneous control of aphids and fungal diseases, such as anthracnose and Phytophthora blight, in an integrated pest management framework for red pepper.

Evidence of Greater Competitive Fitness of Erwinia amylovora over E. pyrifoliae in Korean Isolates

  • Erwinia amylovora and E. pyrifoliae are the causative agents of destructive diseases in both apple and pear trees viz. fire blight and black shoot blight, respectively. Since the introduction of fire blight in Korea in 2015, the occurrence of both pathogens has been independently reported. The co-incidence of these diseases is highly probable given the co-existence of their pathogenic bacteria in the same trees or orchards in a city/district. Hence, this study evaluated whether both diseases occurred in neighboring orchards and whether they occurred together in a single orchard. The competition and virulence of the two pathogens was compared using growth rates in vitro and in planta. Importantly, E amylovora showed significantly higher colony numbers than E. pyrifoliae when they were co-cultured in liquid media and co-inoculated into immature apple fruits and seedlings. In a comparison of the usage of major carbon sources, which are abundant in immature apple fruits and seedlings, E. amylovora also showed better growth rates than E. pyrifoliae. In virulence assays, including motility and a hypersensitive response (HR), E. amylovora demonstrated a larger diameter of travel from the inoculation site than E. pyrifoliae in both swarming and swimming motilities. E. amylovora elicited a HR in tobacco leaves when diluted from 1:1 to 1:16 but E. pyrifoliae does not elicit a HR when diluted at 1:16. Therefore, E. amylovora was concluded to have a greater competitive fitness than E. pyrifoliae.

  • Integrated pest management is essential for controlling plant diseases that reduce crop yields. Rapid diagnosis is crucial for effective management in the event of an outbreak to identify the cause and minimize damage. Diagnosis methods range from indirect visual observation, which can be subjective and inaccurate, to machine learning and deep learning predictions that may suffer from biased data. Direct molecular-based methods, while accurate, are complex and time-consuming. However, the development of large multimodal models, like GPT-4, combines image recognition with natural language processing for more accurate diagnostic information. This study introduces GPT-4-based system for diagnosing plant diseases utilizing a detailed knowledge base with 1,420 host plants, 2,462 pathogens, and 37,467 pesticide instances from the official plant disease and pesticide registries of Korea. The AI plant doctor offers interactive advice on diagnosis, control methods, and pesticide use for diseases in Korea and is accessible at https://pdoc.scnu.ac.kr/.

식물 병원성진균에 대한 도꼬마리 추출물의 항 진균활성 (Antifungal Activity of Extract from Xmthitim strumarium L. Against Plant Pathogenous fungi.)

  • 도꼬마리의 ether 및ethyl acetate 추출물이 식물병원성 진균에 대한 항 진균활송을 측정한 결과, ether와 ethyl acetate 추출물은 P. capsici, S. sclerotiorum과 A. niger에 높은 항 진균활성을 나타내었다. 또한 S. sclerotionum에 대한 ether 추출한 도꼬마리의 $IC_{50}$$335{\mu}g/ml$로 계산되었다. TCL plate상에서 Rf 값이 0.87밴드에서 강한 항 진균활성을 나타냈다.

  • 4품종(남도마늘, 고흥마늘, 단양마늘, 중국산 수입마늘)의 마늘추출액의 식물병원세균과 진균에 대한 항균효과를 조사하였다. 실내실험에서 마늘추출액은 그람양성세균, 그람음성세균, 진균 모두에 좋은 항균효과를 나타내었다. 본 연구에 사용된 4품종의 마늘추출액 중 남도마늘과 중국산 수입마늘이 보다 좋은 항균효과를 나타내었다. 그람양성세균인 Clavibacter michiganensis가 마늘추출액에 가장 민감하였으며, Burkholderia cepacia가 가장 저항성이 컸다. 마늘추출액을 이용한 오이 흰가루병에 대한 항균효과 포장실험에서는 59.7${\sim}$61.9%의 방제가를 보여주었다.

Isolation, Screening, and Identification of Actinomycetes with Antifungal and Enzyme Activity Assays against Colletotrichum dematium of Sarcandra glabra

  • A serious leaf disease caused by Colletotrichum dematium was found during the cultivation of Sarcandra glabra in Jingxi, Rong'an, and Donglan Counties in Guangxi Province, which inflicted huge losses to plant productivity. Biological control gradually became an effective control method for plant pathogens. Many studies showed that the application of actinomycetes in biological control has been effective. Therefore, it may be of great significance to study the application of actinomycetes on controlling the diseases caused by S. glabra. Strains of antifungal actinomycetes capable of inhibiting C. dematium were identified, isolated and screened from healthy plants tissues and the rhizospheres in soils containing S. glabra. In this study, 15 actinomycetes strains were isolated and among these, strains JT-2F, DT-3F, and JJ-3F, appeared to show antagonistic effects against anthracnose of S. glabra. The strains JT-2F and DT-3F were isolated from soil, while JJ-3F was isolated from plant stems. The antagonism rate of strain JT-2F was 86.75%, which was the highest value among the three strains. Additionally, the JT-2F strain also had the strongest antagonistic activity when the antagonistic activities were tested against seven plant pathogens. Strain JT-2F is able to produce proteases and cellulase to degrade the protein and cellulose components of cell walls of C. dematium, respectively. This results in mycelia damage which leads to inhibition of the growth of C. dematium. Strain JT-2F was identified as Streptomyces tsukiyonensis based on morphological traits and 16S rDNA sequence analysis.

Infection and Immune Response in the Nematode Caenorhabditis elegans Elicited by the Phytopathogen Xanthomonas

  • Xanthomonas oryzae pv. oryzae (Xoo) strains are plant pathogenic bacteria that can cause serious blight of rice, and their virulence towards plant host is complex, making it difficult to be elucidated. Caenorhabditis elegans has been used as a powerful model organism to simplify the host and pathogen system. However, whether the C. elegans is feasible for studying plant pathogens such as Xoo has not been explored. In the present work, we report that Xoo strains PXO99 and JXOIII reduce the lifespan of worms not through acute toxicity, but in an infectious manner; pathogens proliferate and persist in the intestinal lumen to cause marked anterior intestine distension. In addition, Xoo triggers (i) the p38 MAPK signal pathway to upregulate its downstream C17H12.8 expression, and (ii) the DAF-2/DAF-16 pathway to upregulate its downstream gene expressions of mtl-1 and sod-3 under the condition of daf-2 mutation. Our findings suggest that C. elegans can be used as a model to evaluate the virulence of Xoo phytopathogens to host.
