Isolation and Structural Analysis of Acetyl Soyasaponin $A_1$ from Hypocotyl of Soybean (콩 Hypocotyl에서 Acetyl Soyasaponin $A_1$의 분리 및 구조 분석)

  • Soyasaponins are phytochemicals of major interest fur their health benefits. Chemical investigation of a soybean phytochemical concentrate resulted in the isolation and identification of triterpenoid saponins. The MeOH extraction of defatted hypocotyl separated from soybeans was peformed by the automated solvent extractor (ASE). Fractionation was performed on a flash column ($150mm{\times}40mm$ i.d.) packed with a preparative $C_{18}$ reverse phase bulk packing material $(125\AA,\;55-105{\mu}m)$ and monitored at 210 nm, and collected 14 fractions. Consequent Fsat preparative column liquid chromatography (Fast PCLC) was performed for the purification of Fraction-I (Fr-I) collected from the fraction 8 and 9 of flash chromatography. Fsat PCLC was performed on a Luna $C_{18}\;10{\mu}m,\;100{\AA}$, semipreparative reverse phase column ($250cm{\times}50mm$ i.d.) for the purification of isolated unknown compound (Fr-I-2). Chemical structure of acetyl soyasaponin $A_1\;(MW:1436.6,\;C_{67}H_{104}O_{33})$ was identified and determined by a combination of extensive NMR ($^1H-NMR$, 400 MHz; $^{13}C-NMR$, 100 MHz; DEPT), IR, UV, and ESI-MS analysis.

  • This study aims to develop knitted ware design to meet desire to express diversity in the modern fashion design so that we designed knitted ware by applying african geometric pattern and color to suggest new knitted ware design. We collect data about african texture pattern through technical books, publications, internet, and preceding research and visit and investigate the African museum. We investigate knitted Jacquard texture through preceding research and collect sample and data which is insufficient in the data source. The conclusions in this study are summarized as follows: First, African textile pattern is formulated with animism based on their religious view of art for a basis and African regards nature like animal and plant as a motive and interprets nature in the so that they can create symbolized geometric features that constitute African texture pattern. Those patterns is composed of extremely geometric figures so that they we fit to apply for color jacquad knit design. Second, color knitted jacquad can be distinguished by knitting method and status of knitting as 7 kinds of techniques such as Nomal, Bird'eye, Floating, Tubular, Ladder's back, Blister, Transfer Jacquard, and as a result of preceding research and knitting texture directly, jacquard technique makes different texture under same condition like consistent spinning rate and same crochet hook. Third, Bird'eye Jacquard used generally to make knitted ware and Ladder's back Jacquard, Tubular Jacquard used to make knitted ware light are fit to apply them to 7GG and 12GG machines. We design a cloak as a outer garment, a coat shaped like one-piece dress and a coat with hood by using Tubular Jacquard which can make thick texture and design a jacket, a skirt and a one-piece dress by using Bird'eye Jacquard. we make a light and flimsy one-piece dress by using Ladder's back Jacquard. Fourth, we apply the contrast of $4{\sim}6$ color and line and the contrast of texture and raw material to jacquard in order to emphasize texture property and visual property.

Redox Characteristic and Evolution of a Fragipan of Gangreung Series Commonly Developed in Coastal Terraces (해성단구지에서 발달된 강릉통의 이쇄경반층(Btx) 토양의 산화.환원적 특성에 관한 연구)

  • Soil pan typically presents the problems in soil water movement or in aeration which is not appropriate for a plant root growth, In this study physico-chemical characteristics of soils and micromorphological characteristic of clay accumulated zone were investigated to identify redox characteristic and evolution of a fragipan of Gangreung series commonly developed in coastal terraces. Gangreung series is classified as Aquic Fragiudalfs according to the USDA soil taxonomy. It is known that sedimentary ocean floor results in soil pan having parallel liner soil structure due to landscape evolution around 200 to 250 million years ago. it is considered that illite, kaolinite, and vermiculite are major clay minerals contained in a fragipan of Gangreung series. Mixed gray and reddish brown colored band around soil pores was found and would be the redoxmorphic features of fragipan. It is possibly due to accumulated illuvial clay and ferriargillans in soil pores and aggregates in reducing conditions eluding ferrous material. Therefore, mixed colored band around pores in soils of Gangreung series would be developed from the eluted ferrous materials which were accumulated in fragipan during the emerged land formation.

Fermentation of wheat bran through lactic acid bacteria: Changes in flavor components and free amino acids and potential applications in baking (밀기울의 유산균 발효: 향기성분 및 유리아미노산 변화를 통한 제빵 소재로서의 가능성)

  • The aim of this study was to enhance the use of wheat bran in lactic acid bacteria (LAB) fermentation. LAB fermentation of wheat bran and the flavor components and amino acids of fermentation products were analyzed using gas chromatography-mass spectrometry (GC/MS) and high-performance liquid chromatography (HPLC). The results showed that total flavor components increased by 93% and 73% in the animal-based LAB mixture (T2) and plant-based LAB mixture (T3), respectively, after fermentation. Among these components, 2,3-butanedione (diacetyl), known for its buttery flavor, was detected at concentrations of 18.44 ng/g (T2) and 16.95 ng/g (T3). Levels of hexanal and nonanal, which causes off-flavor components in wheat bran, dramatically decreased after T2 fermentation; similarly, levels of total free amino acids decreased by 37.6% (T2) and 36.7% (T3) after fermentation. This may explain why some components were bound to volatile compounds during LAB fermentation. These results suggest that LAB-fermented wheat bran is a potential value-added food material.

Selection of Binder and Solid Materials for Pelleting Welsh Onion(Allium fistulosum L.) Seeds (파 펠렛종자 생산을 위한 접착제 및 피복재료 선발)

  • This study was carried out to select new pelleting binder and material for Welsh onion seeds. The optimum treatments of the various plant growth regulators to improve seed germination of the Welsh onion was also estimated. There were no significant effects of growth regulators on the germination percentage, but germination was faster according to the number of days to 50% of the final germination ($T_{50}$) and the mean number of days to germination (MDG) than those of the control. Germinability was increased when the seeds were soaked in gibberellic acid ($GA_3$) solution for 24hrs, even though there was no synergy effect on the germinability when $GA_3$ was mixed with 6-benzylaminopurine (BAP). The optimum treatment for improving germination of Welsh onion was observed when the tested seeds was soaked in 500 $\mu$M of $GA_3$ at $20^{\circ}C$ for 24hrs. Also, when the seeds soaked in the aforementioned treatment, the rate of germination was increased at lower temperature than at $20^{\circ}C$, the optimal temperature. The percentage and the speed of seed germination depended on the kinds of pelleting binder and their concentration. It showed that the higher the concentration of the binder for seeds pelleting, the lower the percentage of seed germination. Among the pelleting binder, polyvinyl alcohol (PVA) and polyvinyl pyrrolidone (PVP) were the best binders for seed pelleting, because seed germination using those binder did not affect on the concentration of binder. On the other hand, Carboxymethy cellulose (CMC) and methyl cellulose (MC) severely inhibited the seed germination The germinability was also different arcording to the pelleting materials. Among the different 58 pelleting materials, kaoline alone, the mixture of bentonite and kaoline, the mixture of bentonite, calcium carbonate and diatomaceous earth #300 were found as the best pelleting materials for welsh onion seeds.

  • A greening phenomenon has been observed in some plant foods such as chestnut, sweet potato, burdock, and others during processing. The formation of the pigments was postulated as reactions of primary amino compounds with chi orogenic acid or caffeic acid ester, yielding acridine derivatives. Acridine derivatives have been regarded as mutagenetic agents. For the reason, the bacterial reverse mutation test was carried out to evaluate the genotoxicity of green pigment using Salmonella typhimurium TA98 and TA100. Alanine, arginine, aspartic acid, glycine, lysine, and phenylalanine were reacted repectively with chlorogenic acid to synthesize model compound. Green pigment was extracted from sweet potato. Maximum concentration of 2 and 50 mg/plate was tested for the synthetic green pigments and extracted green pigment respectively, taking bacterial survival, solubility, and color intensity into consideration. There was no signigicant increase in the reverse mutation either with or without S9 activation system by any test material. Though further studies with other genotoxicity test system are necessary, both synthetic and sweet potato green pigments seemed not to cause mutation despite the acridine moiety in their structures.

Changes in Freshness of Endive (Cichorium endivia L.) by Different Packaging Types (엔다이브 포장방법에 따른 저장 중 선도 변화)

  • The effect of packaging material in enhancing the shelf life and maintaining the postharvest quality of endive (Cichorium endivia L.) was studied. Endive were packed in three packaging materials: paper box (control); paper box with endive wrapped in high density polyethylene (HDPE) film; and plastic box container covered with high density polyethylene (HDPE) film. The quality characteristics, such as fresh weight loss, soil plant analysis development (SPAD) value, and appearance of endive were investigated during 20 days of storage at $2^{\circ}C$. The endive wrapped with HDPE film inside the paper box showed the lowest weight loss, highest SPAD value and the best appearance compared to those wrapped in other packaging types during storage. The results indicate that the marketability of endive can be optimized with proper packaging and storage.

Effect of Drying Temperature on High Quality Functional Processed Products of Chinese Matrimony Vine (가공용 소재로서 구기자나무 잎의 건조온도 조건)

  • Background : Chinese matrimony vine (Lycium chinense Mill.) is a deciduous shrub belonging to the Solanaceae. The leaves are used as an alternative raw material replacing dried fruits associated with high production costs in many industires. The aim of this experiment was to determine the effect of drying temperature on the leaves used in the manufacture of functional products. Methods and Results : The leaves of Chinese matrimony vine were harvested when the plant height reached 60 - 70 cm in the spring and treated at four different temperatures ($40^{\circ}C$, $50^{\circ}C$, $60^{\circ}C$ and $70^{\circ}C$). The time to reach the desiccation state of 20% was 63 h at $40^{\circ}C$, 37 h at $50^{\circ}C$, 17 h at $60^{\circ}C$ and 11 h at $70^{\circ}C$. The drying rate per hour was 1.28% at $40^{\circ}C$, 2.25% at $50^{\circ}C$, 4.94% at $60^{\circ}C$ and 7.60% at $70^{\circ}C$. No significant difference were observed in ash, crude fat, polyphenol or rutin content of treated samples. Crude protein and betaine content decreased with higher dry temperatures. Nitrogen free extract of the treated samples increased with higher drying temperatures. Conclusions : Taking into consideration drying time, drying rate, color value, energy consumption and functional ingredients is advantageous to dry the leaves of the Chinese matrimony vine at $60^{\circ}C$ using a hot air agricultural dryer.

Authentication and classification of strawberry varieties by analysis of their leaves using near infrared spectroscopy.

  • It is well known now that near infrared spectroscopy (NIRS) is a fast, no destructive, and inexpensive analytical technique that could be used to classify, identify, and authenticate a wide range of foods and food items. Therefore, the main aims of this study were to provide a new insight into the authentication of five strawberry (Fragaria x ananassa) varieties and to correlate them with geographical zones and the propagating methods used. Three weeks plants of five different strawberry varieties (F. x ananassa Duch. cv Camarosa, Seascape, Chandler, F. Chiloensis, and F. Virginiana) were cultivated in vitro first and then transferred to pots with special soil, and grown in a greenhouse at CINVESTAV, all varieties were acquired from California (USA). After 18 months, ten leaves from each variety were collected. Transmission spectra from each leave were recorded over a range of 10, 000-4, 000 cm$-^{1}$, 32 scans of each strawberry leave were collected using a resolution of 4 cm$-^{1}$ with a Paragon IdentiCheck FT-NIR System Spectrometer. Triplicates of each strawberry leave were used. All spectra were analyzed using principal component analysis (PCA) and soft independent modeling class analogy (SIMCA). The optimum number of components to be used in the regression was automatically determined by the software. Camarosa was the only variety grown from the same shoot but propagated by a different method (direct or in vitro). Five different classes (varieties) or clusters were observed among samples, however, larger inter class distances were presented by the two wildtype samples (F. Chiloensis and F. Virginiana). Camarosa direct and Camarosa in vitro displayed a small overlapping region between them. On the other hand, Seascape variety presented the smallest rejection percentage among all varieties (more similarities with the rest of the samples). Therefore, it can be concluded that the application of NIRS technique allowed the authentication of all strawberry varieties and geographical origin as well. It was also possible to form subclasses of the same materials. The results presented here demonstrate that NIRS is a very powerful and promising analytical tool since all materials were authenticated and classified based on their variety, origin, and treatment. This is of a tremendous relevance since the variety and origin of a plant material can be established even before it gives its typical fruit or flower.

The Correlation between the Gall-forming Process by Aphids and the Various Amino Acids in Zelkova Serrata Makino (느티나무에서 면충에 의한 충영형성과 아미노산종과의 연관성)

  • After investigating the developmental process of galls forming on leaf of Zelkova serrata and analyzing the composition and contents of the free amino acids in leaves, galls and haemolymph of aphids, we have come to the following results. Galls induced were mostly distributed on the middle of lower part of leaf to the petiole along vein and most of investigated leaves had one gall only. The growth of galls continued in length and width until Paracolopha morrisoni habitants which transformed into alate adult left the gall. These results also indicate that galls are closely connected with a portion of plant tissues as well as foods and habitats of aphids. 26${\sim}$29 kinds of free amino acids such as proline, alanine and valine and so on, detected in leaves, gall and haemolymph of aphids. The composition of free amino acids were similar in three materials examined, but proline and asparatic acid showed the highest quantity in the leaves and falls, respectively. In case of aphids, alanine was extremely higher as 18.4% of total contents of free amino acids, in 1st instar of fundatrix stage. Total contents of free amino acids in the apteral adult stage of aphid were also increased than in the 1st instar stage. This investigation implies that nitrogen source such as free amino acids is apparently a major growth-material like carbohydrates during development of galls in Zelkova serrata.
