Screening of Insecticide to Control Rhophalosiphum nymphaeae for Alisma plantago as Second Crop in Paddy Field

  • The purpose of this study was to evaluate the effect of pesticides on insect pest control of Rhophalosiphum nymphaeae, growth characteristies and dry root yield from the cultivated after early maturing rice cropping. All pesticides treated had no effect on the growh period, flowering rate of Alisma plantago. The major pesticides were methomyl-Lf, 24.1 %, imidacloprid-Wp, 10%, carbosulfan-Wp, 20%, and methomyl-Wp, 45%. Dry yield of root were increased largely with imidacloprid-Wp, 10% (10g/20$\ell$), pesticide than the other pesticides and control. All pesticides were had no injury with recommended concentration. On the other hand all pesticides were slightly harmful in the double dosage level for the Alisma plantago.

Selecting of Useful Chemicals Reducing of Leaf Blight on Job's Tears (Coix lachry-ma-jobi L. Var.)

  • This study was conducted evaluate the control effect of fungicides on control of Leaf blight, growth characteristics, and dry yield in the cultivation of Job's tears. All fungicides treated had no effect on the growth of Job's tears. The major fungicides were mancozeb Wp, $75\%$, chlorothalonil Wp, $75\%$, dithianon Wp, $43\%$, difenoconazole Wp, $10\%$, benomyl Wp, $50\%$, and propineb Wp, $70\%$. Dry seed yield were increased largely with chlorothalonil Wp, $75\%\;(33g/20\ell)$, fungicide than the other fungicides and control. All fungicides had no injury with standard dosage. On the other hand all fungicides had slight injury in the double dosage level for the Job's tears.

Symptom of Leaf Injury and Varietal Difference to Ozone in Rice and Soybean Plant (벼와 콩의 오존 피해증상과 품종간 차이)

  • This study was carried out to elucidate the symptom of leaf injury to ozone and to determine varietal difference to ozone injury in rice and soybean plant. Ozone was produced by electrostatic discharge in oxygen and was monitored by UV absorption ozone analyzer. The rice leaves were spotted red and rolling leaf edge, discolored to reddish brown or yellowish white in response to ozone, and the leaves that were severely stressed were withered from the tip of leaf. The soybean ones were also discolored to lemon yellow, yellow or dark brown. The leaf injury in both rice and soybean was clearly appeared at the reverse side of leaf and in lower leaves. Milyang 23 and Nonganbyeo among rice cultivars tested were resistant to ozone, but Chucheongbyeo was resistant to it. The ratio of leaf injury was increased and chlorophyll content was decreased as the extension of ozone exposure from 2 to 8 hours in rice. Keunolkong and Danyeopkong among soybean cultivars tested showed resistant reaction to ozone, but Kwangankong and Muhankong were susceptible to it. It was observed that the soybean plants grown for 45 days after seeding were severely damaged by ozone than those of other growth stages.

Stachys sieboldii M iq. Protects SH-SY5Y Cells Against Oxygen-Glucose Deprivation/Reoxygenation-Induced Injury by Inhibition of Mitochondrion-Mediated Apoptosis Pathway (허혈-재관류 유도 SH-SY5Y 모델에서 미토콘드리아 매개 Apoptosis 기전 제어를 통한 초석잠 추출물의 세포보호 효과)

  • Oxygen glucose deprivation/re-oxygenation (OGD/R) induces neuronal injury via mechanisms that are believed to mimic the pathways associated with brain ischemia. Stachys sieboldii Miq. (Chinese artichoke), which has been extensively used in oriental traditional medicine to treat of ischemic stroke; however, the role of S. sieboldii Miq. (SSM) in OGD/R induced neuronal injury is not yet fully understood. The present research is aimed to investigate the protective effect and possible mechanisms of SSM extract treatment in an in vitro model of OGD/R to simulate ischemia/reperfusion Injury. Pretreatment of these cells with SSM significantly attenuated OGD/R-induced production of reactive oxygen species (ROS) by increasing GPx, SOD, and decreasing MDA. SSM decreased mitochondrial damage caused by OGD/R injury and inhibited the release of cyt-c from mitochondrion to cytoplasm in SH-SY5Y cells. Furthermore, neuronal cell apoptosis caused by OGD/R injury was inhibited by SSM, and SSM could decrease apoptosis by increasing ratio of Bcl-2/Bax and inhibiting caspase signaling pathway in SH-SY5Y cells. SSM demonstrated a neuroprotective effect on the simulated cerebral ischemia in vitro model, and this effect was the inhibition of mitochondria-mediated apoptosis pathway by scavenging of ROS generation. Therefore, SSM may be a promising neuroprotective strategy against ischemic stroke.

Effect of Uniconazole and Free Radical Scavenger Treatments on Reduction of $SO_2$ Injury in Platanus occidentalis (Uniconazole 및 Free Radical Scavenger처리가 양버즘나무의 $SO_2$ 피해경감에 미치는 효과)

  • The objective of this research was to increase phytoprotective effects by combined treatment of uniconazole and free radical scavengers such as ascorbic acid or sodium benzoate on $SO_2$ injury in P. occidentalis. The plant injury, chlorophyll content and enzyme activity of superoxide dismutase(SOD) and peroxidase(POD) affected by combined treatment were also investigated. The phytoprotective role of uniconazole was nullified by spray of Diethyldithiocarbamate(DDTC) resulting in the decrease of SOD and POD activities. Free radical scavengers, sodium benzoate and ascorbic acid, did not affect SOD and POD activity, but significantly inhibited the development of visible injury, degradation of chlorophyll, and SOD and POD activity in leaves exposed to $SO_2$. The spray of ascorbic acid decreased plant susceptibility to $SO_2$ induced by DDTC application. These results indicate that uniconazole application increase SOD activity that play a role of antioxidant in plant body, but sodium benzoate and ascorbic acid do not affect enzyme activities of SOD or POD.

Risk Priority Number of Chemical Facilities by the Risk Assessment of Injury Analysis in the Chemical Plant (재해분석을 통한 화학공장의 위험성 평가에 따른 화학설비의 위험도 우선순위)

  • There have been large explosions at some chemical plants recently. Accidents in chemical plants have been caused mainly by servicing for maintenance. There is a need to find a key target for effective injury prevention in maintenance. In this paper, facilities were selected as a key target and the risk priority numbers of the facilities were calculated in order to prioritize preventative measures. The research method was based on the followings; the list of the facilities is found through injury analysis. Then, the risk of each facility was calculated by the frequency of accidents and the working day loss through injuries. In addition, the risk of the facilities was calculated again by the frequency and the severity based on knowledge and experience of experts. As a result, the facilities in chemical plant maintenance are ranked in order of high to low risk priority number; reactor, dryer, tank lorry, etc. In the future, special measures and attention should be directed according to the risk priority number during the maintenance of chemical plants.

Rapid Diagnosis of Resistance to Glufosinate-ammonium in Transgenic Sweet Potato (형질전환 고구마에 대한 Glufosinate-ammonium 저항성 간이진단법)

  • Transgenic herbicide-resistant sweet potato plants [Ipomoea batatas (L.) Lam.] produced through a biolistic transformation were used in this study. The objective of this research was to find out a rapid and reliable assay method for confirming glufosinate-ammonium resistance. The techniques tested include whole-plant bioassay, one leaf bioassay, and leaf disk bioassay. Parameters investigated in this study were leaf injury and ammonium accumulation at 1 and 5 days after treatment of glufosinate-ammonium. In the leaf disk bioassay, leaf injury of the transgenic line 7171 was 1.9-fold less affected by glufosinate-ammonium than the wild type. The leaf injury of 7171 in one leaf and whole-plant bioassays was 59- and 92-fold less affected by glufosinate-ammonium, respectively, compared with that of the wild type. Leaf disk, one leaf, and whole-plant bioassays showed that ammonium accumulation of the 7171 was 2 to 20-, 4 to 43-, and 6 to 115-fold less affected by 0.5-5 mM glufosinate-ammonium than that of the wild type. All three bioassays successfully distinguished the resistance from the transgenic lines, but one leaf bioassay is the simplest and quickest. Leaf injury and ammonium accumulation were the same in leaves 1, 3, 5, 7, and 10 of 3 mM glufosinate-ammonium treated plants or nontreated plants. The one leaf bioassay was chosen as the standard procedure for future confirmation of resistance in transgenic sweet potato because it is a rapid and reliable assay.

Effect of safener fluxofenim on crop injury of chloroacetanilides and enzyme activity of glutathione S-transferase in grain sorghum seedlings (수수유묘에 있어서 fluxofenim의 약해경감효과와 glutathione S-transferase 효소활성)

  • Effects of safener fluxofenim was investigated for crop injury of acetanilide's upland herbicides and for enzyme activity of glutathione S-transferase (GST) in grain sorghum. Bioassay with etiolated grain sorghum [Sorghum bicolor (L.) Moench. cv. 'G522DR'] seedlings grown in agar containing metolachlor or alachlor showed that they are strong inhibitors on root growth of grain sorghum ($GI_{50}=4.5{\mu}M$ for metolachlor and $6.2{\mu}M$ for alachlor). The safener fluxofenim applied by seed soaking protected growth of grain sorghum from crop injury of metolachlor or alachlor at the concentrations of 1 to 10 ${\mu}M$. There was a significant increase in glutathione-herbicide conjugates in root tissues of fluxofenim-treated seedlings. Activities of $GST_{-metolachlor}$ and $GST_{-CDNB}$ were increased by 82% and 70%, respectively, in the cytosolic fraction of roots with fluxofenim treatment.

Effect of Flooding and Soil Salinity on the Growth of Yam (Dioscorea batatas) Transplanted by Seedling of Aerial Bulblet in Saemangeum Reclaimed Tidal Land

  • The effect of flooding and soil salinity on the growth of yam (Dioscorea batatas) were studied on the experimantal site temporally established in the south-eastern part of Saemangeum Reclaimed Tidal Land (near Gwanghwal myun, Gimjae-gun, Jellabukdo, Korea). Yam seedlings planted by using aerial bulblet as alternative of sliced tubers, were grown for 20-days and transplanted in black-vinyl mulched ridges (about 20 cm in height) at 70cm interval by $20{\times}60cm$ spacing in the $4^{th}$ of May, 2010. Soil salinity was maintained at lower than 1.2 ds $m^{-1}$ during the growing period and did not result to salt injury in all plants. However, flooding injury very seriously led to plant death and plant mortality rates at $67{\pm}21$ and $82{\pm}9%$ of yam plants in the compost and no compost treatment, respectively, died by heavy flooding during the rainy summer season. The main reasons of the flooding injury included the decreased rainfall acceptable capacity (RAC) after the rising of water table and a slowdown of water infiltration rate after the formation of an impermeable soil crust in the furrow bottom with continuous and heavy downpour during the rainy summer season. The effect of compost treatment was not statistically observed because of the severe spatial difference caused by wet injury, although yam tuber yield was higher at 30 kg $10^{-1}$ in the compost treatment than in the no-compost treatment at 20 kg $10^{-1}$. However, the size of tuber ranged at 1.23 to 1.60 cm in diameter and 3.7 to 5.0 cm in length in all both treatment, which means they are still reproducible for the next cropping season. Conclusively, proper counter-flooding measure and soil salinity control critically important for successful yam production in Saemangeum Reclaimed Tidal Land.

Ethane Evolution in Cucumber Plants by Air Pollutants in Relation to Plants Injury (대기오염 물질 처리에 의한 오이 장해와 에탄 생성)

  • Ethane was measured to know whether active oxyzens may induce phytoxicity in stressedcucumber plant.The time course of the increase in ethane evolution was the same as that of the increase of visible injury in all treatments except $SO_2$ treatment.This result showed that air pollutants-induced plant damages were closely related to ethane evolution.And evolution of ethane was more increased in combined stress than singly one,suggesting that phytotoxicity was more severe in complex sterss.Also, evolution of ethane was enhanced in the light condition and scavengers of active oxygen were inhibited,showing that plant damage that plant damage were cause by active oxygens.
