Enzymatic characterization and Expression of 1-aminocycloprophane-1-carboxlyate deaminase from the rhizobacterium Pseudomonas flourescens

  • Ethylene, known as a stress hormone regulate wide developmental processes including germination, root hair initiation, root and shoot primordial formation and elongation, leaf and flower senescence and abscission, fruit ripening. The acceleration of ethylene biosynthesis in plant associated with environmental and biological stresses. 1-Aminocycloprophane-1-carboxlyate deaminase(ACCD) is an enzyme that cleaves ACC into and ammonia, a precursor of the plant hormone ethylene. Plant growth-promoting rhizobacteria (PGPR) having ACCD can decrease endogenous ACC level of tissue, resulting in reduced production of ethylene in plants. ACC deaminse was a key enzyme for protect stressed plants from injurious effects of ethylene. ACCD gene was encoded from Pseudomonas flourescens, PGPR and was cloned in Escherichia coli. We expressed the recombinant ACCD(rACCD) containing 357 amino acids with molecular weight 39 kDa that revealed by SDS-PAGE and western blot. The rACCD was purified by Ni-NTA purification system. The active form of rACCD having enzyme activity converted ACC to a-ketobutyrate. The optimal pH for ACC deaminase activity was pH 8.5, but no activity below pH 7.0 and a less severe tapering activity at base condition resulting in loss of activity at over pH 11. The optimal temperature of the enzyme was $30^{\circ}$ and a slightly less severe tapering activity at 15 - 30$^{\circ}$, but no activity over $35^{\circ}$. P. flourescens ACC deaminase has a highly conserved residue that plays in allowing substrate accessibility to the active sites. The enzymatic properties of this rACCD will provide an important reference for analysis of newly isolated ACCD and identification of newly isolated PGPR containing ACCD.

Pteris cretica 'Wilsonii'의 기내 식물체 재생에 미치는 배지구성물질의 영향 및 기외순화 (Medium Composition Affecting In Vitro Plant Regeneration and Acclimation of Pteris cretica 'Wilsonii')

  • 기내배양을 이용하여 Pteris cretica 'Wilsonii'의 효율적인 대량번식법을 개발하기 위한 목적으로 근경, 엽신 및 엽병조직으로부터 식물체 재분화를 유도하였다. 그 결과, 균질화된 근경의 절편에서만 신초가 재생되었다. 식물체 재분화를 유도하는데 적합한 배지는 1%의 sucrose와 $50\;mg{\cdot}mL^{-1}$$NaH_2PO_4$를 첨가한 1/8MS배지였다. 근경 절편으로부터 식물체의 재생을 촉진하기 위하여 생장조절물질이 첨가된 배지에서 2달 동안 절편을 배양한 후, 생장조절물질을 첨가하지 않은 배지로 이식한 결과, $1{\mu}M$의 kinetin과 $5{\mu}M$의 NAA를 혼용첨가한 처리구에서 신초의 재생이 가장 왕성하였다. BA 첨가구에서는 분열조직의 일종인 GGB가 다량 유도되었으나, 형성된 GGB에서 신초 재생율은 낮은 것으로 나타났다. MS배지의 적정 무기물 및 비타민의 농도를 구명하는 실험을 마친 뒤, 형성된 유묘를 이용하여 기외순화 시킨 결과, 1/8MS배지에서 형성된 유묘가 생존율이 가장 높았으며, 생육 또한 왕성하였다.

Brassinosteroids-mediated regulation of ABI3 is involved in high-temperature induced early flowering in plants

  • The interplay of plant hormones is one of the essential mechanisms for plant growth and development. A recent study reported that Brassinosteroids (BR) and ABSCISIC ACID (ABA) interact antagonistically in early seedling developments through the BR-mediated epigenetic repression of ABSCISIC ACID-INSENSITIVE 3 (ABI3). However, the other physiological roles of the BR-mediated regulation of ABI3 and ABA responses beyond early seedling developments remain largely unknown. Here, we showed that the activation of BR signaling by high temperatures promotes flowering time through the suppression of ABI3 expressions. Elevated ambient temperature induced early flowering in wild type Col-0 plants, but not in BR-defective bri1-116 mutant plants. Conversely, a hyper BR biosynthetic dwf4-D mutant displayed more sensitive thermomorphic long shoot elongation and early flowering. Both expression patterns and physiological responses supported the biological roles of ABI3 in the regulation of floral transition and reproduction under high temperature conditions. Finally, we confirmed that the lowered expressions of the transcript and protein levels of ABI3 brought on by elevated temperature were correlated with warmth-induced early flowering phenotypes. In conclusion, our data suggest that the BR- and warmth-mediated regulation of ABI3 are important in thermomorphic reproductive phase transitions in plants.

SMALL AUXIN UP RNA 유전자 집단의 기능과 조절 메커니즘에 대한 최근 연구 동향 (Recent research progress on the functional roles and regulatory mechanisms of SMALL AUXIN UP RNA gene family)

  • The plant hormone auxin regulates the overall metabolic processes essential for plant growth and development. Auxin signaling is mediated by early auxin response genes, which are classified into three major families: AUXIN/INDOLE ACETIC ACID (AUX/IAA), GRETCHEN HAGEN3 (GH3) and SMALL AUIN UP RNA (SAUR). The SAUR gene family is the largest family among early auxin response genes and encodes the small and highly unstable gene products. The functional roles of SAUR genes have remained unclear for many years. The traditional genetic and molecular studies on the SAUR functions have been hampered by their likely genetic redundancy and tandem arrays of highly related genes in the plant genome, together with the molecular characteristics of SAUR. However, recent studies have suggested possible roles of SAUR in a variety of tissues and developmental stages in accordance with the novel approaches such as gain-of-function and RNA silencing techniques. In this review, the recent research progress on the functional roles and regulatory mechanisms of SAUR and a set of possible future works are discussed.

옥신 반응 SMALL AUXIN UP RNA 유전자의 최근 연구 동향 및 작물 개량을 위한 적용 (Auxin-responsive SMALL AUXIN UP RNA genes : recent research progress and its application for crop improvement)

  • Auxin is a key plant hormone which regulates overall plant growth development. A number of researches to investigate auxin signaling identified three major classes of early auxin response genes: AUX/IAA, GH3 and SMALL AUXIN UP RNA (SAUR). Among these genes, in planta functions of SAUR gene family are largely ambiguous, while both AUX/IAA and GH3 genes are analyzed to mediate negative feedback on auxin response. SAUR genes encode small plant-specific proteins. SAUR gene products are highly unstable and transiently expressed in the tissue- and developmental-specific manners in response to auxin and various environmental stimuli. In the decades, molecular and genetic approaches to elucidate in planta functions of SAURs have been hampered by several factors such as the unstable molecular features and functional redundancy among them. However, a series of recent studies focusing on several subgroups of SAUR gene family made significant progress in our understanding of its biochemical and physiological functions. These works suggest that many SAUR proteins mainly regulate auxin-related cell expansion and auxin transport. In this review, the recent progress in SAUR research and prospects for crop improvement through its genetic manipulation are discussed.

자원식물로서 응용을 위한 야광나무 열매의 식물화학적 연구 (Phytochemical Study for Botanical Utilization of the Fruits of Malus baccata)

  • Very little utilization of the fruits of Malus baccata(Rosaceae) has been employed for food and medicinal plants except for preparing fruit beverages. But, it was estimated as valuable to investigate the chemical components for the botanical resource of this plant. In this study, it was found that the fruits of this plant contained primary long chain alcohol, ${\beta}-sitosterol$, campesterol, ursolic acid and ${\beta}-_D-glucosides$ of ${\beta}-sitosterol$ and campesterol. However, phloretin(dihydrochalcone) and its 5-O-glucoside(phloridzin) known as plant growth regulators in many Rosaceae plants were not found in this plant material by co-TLC analysis with authentic specimens. Although plant sex hormone, estrone, was often contained in relates of M. baccata, e.g., Prunus spp., Crataegus spp. and Malus spp., this compound was not detected in this fruit by comparison with an authentic material. By RIC chromatography, it was suggested that the Soxhlet extraction by the solvent of ether was excellently useful to extract ursolic acid efficiently.

Overexpression of human erythropoietin in tobacco does not affect plant fertility or morphology

  • Human erythropoietin (EPO) is a leading product in the biopharmaceutical market, but functional EPO has only been produced in mammalian cells, which limits its application and drives up the production costs. Using plants to produce human proteins may be an alternative way to reduce the cost. However, a recent report demonstrated that overexpression of the human EPO gene (EPO) in tobacco or Arabidopsis rendered males sterile and retarded vegetative growth, which raises concern whether EPO might interfere with hormone levels in transgenic plants. In the present study, we demonstrated that overexpressing EPO with additional 5'-His tag and 3' ER-retention peptides in tobacco did not cause any developmental defect compared to GUS plants. With our method, all 20 transgenic plants grew on selective medium and, further confirmed by PCR, were fertile. Most of them grew similarly compared to GUS plants. Only one transgenic plant (EPO2) was shorter in plant height but had twice the life span compared to other transgenic plants. When 11 randomly selected EPO plants, along with the abnormal plant EPO2, were subjected to RT-PCR analysis, all of them had detectable EPO transcripts. However, their protein levels varied considerably; seven of them had detectable EPO proteins analyzed by western blot. Our results indicate that overexpressing human EPO protein in plants does not have detrimental effects on growth and development. Our transformation systems allow us to further explore the possibility of glycoengineering tobacco plants for producing functional EPO and its derivatives.

Agrobacterium rhizogenes에 의하여 형질전환된 인삼 모상근의 세포 유전학적 및 조직학적 특성 (Cytogenetic and Histological Characteristics of Ginseng Hairy Root Transformed by Agrobacterium rhizogenes)

  • Ginseng (Panax ginseng C.A. Meyer) hairy root transformed with Agrobacterium rhizogenes (strain A4) was examined cytogenetically and histologically to assess its characteristics. The optimum growth of hairy root obtained in hormone-free MS medium (sucrose 30 g/L, pH5-6) for long period cultures. All hairy root strains (HB1, HB2, HB3) had the 2n diploid number (2n=48) of chromosomes in root tip cells. There were no alteration in chromosome structure except in one cell of HB3 strain. Results of SDS-PAGE showed a few difference in pattern and number of bands between normal and hairy root of ginseng. The root anatomy of normal root and hairy root differed each other. The hairy root had a clearly defined vascular strand, and the morphology of cortical cells were disorganised with large intercellular spaces.

Microprogation And Environment Conditions Affecting On Growth Of In Vitro And Ex Vitro Of A. Formosanus Hay

  • The goal of this research was to develop the effectiveness of in vitro culture method for A. formosanus and study the environment in vitro conditions affecting on growth. The first series of experiments were examined to investigate the response of three different basal media, MS (Murashige and Skoog, 1962), Knudson (KC; Knudson, 1946) and modified hyponex on growth and multiplication during in vitro culture. Multiple shoot proliferation was induced in shoot tip explants on Hyponex (H3) media supplemented with BA (1 mg1$\^$-1/) or TDZ (1-2 mg1$\^$-1/). Addition of activated charcoal (1%) to the TDZ containing medium promoted rapid shoot tip proliferation (11.1 shoots per explant) but the same medium had an opposite effect resulting in poor proliferation in the nodal explants. However, the regenerated shoots had slow growth rate and failed to elongate. This problem was overcome by transferring the shoot clumps to a hormone free H3 media supplemented with 2% sucrose and 0.5% activated charcoal. Using bioreactor culture for scaling up was also shown the best way for multiple shoot induction and growth of this plant. The second series of experiments was studied to investigate the effect of physical environment factors on growth of in vitro plantlets. The Anoectochilus formosanus plantlets were cultured under different air exchange rate (0.1, 0.9, 1.2h$\^$-1/), without sucrose or supplement 20g.1$\^$-1/ (photoautotrophic or photomixotrophic, respectively), and different photosynthesis photon flux (40, 80, 120 ,${\mu}$mol.m$^2$.s$\^$-1/- PPF). Under non-enrichment CO$_2$ treatment, slow growth was observed in photoautotrophical condition as compared with photomixotrophical condition on shoot height, fresh weigh and dry weight parameters; High air exchange (1.2.h-l) was found to be inadequate for plant growth in photomixotrophical condition. On the contrary, under CO$_2$, enrichment treatment, the plant growth parameters were sharply (visibly) improved on photoautotrophic treatments, especially on the treatment with air exchange rate of 0.9.h-1. The growth of plant in photoautotrophic condition was not inferior compared with photomixotrophic, and the best growth of plantlet was observed in treatment with low air exchange rate (0.9.h-1). Raising the PPF level from 80 to 120${\mu}$mol.m$\^$-2/.s$\^$-1/ decreased the plant height, particularly at 120${\mu}$mol.m$\^$-2/.s$\^$-1/ in photoautotrophic condition, fresh weight and dry weight declined noticeably. At the PPF of 120${\mu}$mol.m$\^$-2/,s$\^$-1/, chlorophyll contents lowed compared to those grown under low PPF but time courses of net photosynthesis rate was decreased noticeably. Light quality mainly affected morphological variables, changes of light quality also positively affected biomass production via changes in leaf area, stem elongation, chlorophyll content. Plant biomass was reduced when A. formosanus were grown under red LEDs in the absence of blue wavelengths compare to plants grown under supplemental blue light or under fluorescent light. Stem elongation was observed under red and blue light in the present experiment. Smaller leaf area has found under blue light than with other lighting treatments. Chlorophyll degradation was more pronounced in red and blue light compared with white light or red plus blue light which consequent affected the photosynthetic capacity of the plant. The third series of experiment were studied to investigate the effect of physical environment factors on growth of ex vitro plants including photosynthesis photon flux (PPF), light quality, growing substrates, electrical conductivity (EC) and humidity conditions. In the present experiments, response of plant on PPF and light quality was similar in vitro plants under photosynthesis photon flux 40${\mu}$mol.m,$\^$-2/.s$\^$-1/ and white light or blue plus red lights were the best growth. Substrates testing results were indicated cocopeat or peat moss were good substrates for A. formosanus growth under the greenhouse conditions. In case of A. formosanus plants, EC is generally maintained in the range 0.7 to 1.5 dS.m-1 was shown best results in growth of this plant. Keeping high humidity over 70% under low radiation enhanced growth rate and mass production.

1-Aminocyclopropane-1-Carboxylate Deaminase from Pseudomonas fluorescens Promoting the Growth of Chinese Cabbage and Its Polyclonal Antibody

  • Bacterial 1-aminocyclopropane-1-carboxlyate (ACC) deaminase (AcdS) is an enzyme that cleaves ACC, a precursor of the plant hormone ethylene, into ${\alpha}$-ketobutyrate and ammonia. The acdS gene was cloned from Pseudomonas fluorescens, which was capable of improving the seedling of Chinese cabbage under salinity condition. The recombinant AcdS (rAcdS) exhibited optimal activity at pH 8.5 and $30^{\circ}C$. Strong activity was sustained at up to 100 mM NaCl. The polyclonal anti-P. fluorescens AcdS antibody was produced in a rabbit that had been immunized with the purified rAcdS. This antibody successfully recognized the homologous antigens derived from the total proteins of isolated plant growth-promoting microorganisms. A statistically significant correlation was observed between the intensity of hybridization signal and AcdS activity measured by a biochemical method, suggesting its application as a useful indicator for active deaminases.
