약용식물 추출물의 숙취 해소 효과에 관한 연구 (Effect of medicinal plant extract for hangover relief)

  • The present study was performed to evaluate the effect of medicinal plant extract on relieving hangovers in mice administered alcohol. The animals were divided into three groups. Each group was treated with fermented plant extract, non-fermented plant extract, or water 30 min after consuming ethanol (2 mL/kg). A locomotor activity test showed that all groups had decreased motor activity until 40 min after plant extract administration. The mice treated with water had lower motor activity until 100 min post-administration. However, the group treated with non-fermented plant extract showed increased motor activity 40 min post-administration, and the higher activity level was maintained until 120 min post-administration. The animals treated with fermented plant extract had a level of motor activity between those of the groups treated with water or non-fermented plant extract. Blood was collected from each mouse 120 min post-administration and aldehyde concentration was measured. The group treated with non-fermented plant extract had a significantly higher (p < 0.05) aldehyde concentration than the other groups. These results demonstrate that the non-fermented medicinal plant extract helped alleviate hangovers 40 min after administration by reducing aldehyde concentrations in the blood.

Ethyl Acetate Extract of Bacillus pumilus SH122 Induces Resistance Against Phytophthora Blight in Pepper Plant

  • In order to obtain bacterial metabolites inducing disease resistance in pepper plant, two hundred bacterial isolates were isolated from the rhizosphere soil of tobacco, cucumber, and pepper plant. Ethyl acetate extract of each bacterial culture was used to screening for induction of resistance against phytophthora blight of pepper plant. Application of ethyl acetate extract of an isolate SH122 culture to pepper plant conferred resistance against phytophthora blight consistently and significantly. According to cellular fatty acid analysis and other characteristics, the SH122 culture were significantly lower than those on control plants treated with ethyl acetate extract of nutrient broth. The B. pumilus SH122 itself of ethyl acetate extract of its culture did not show antifungal activity against phytophthora blight in pepper plants.

The Effect of Wood Extract as a Water-Soluble Fertilizer in the Growth of Lactuca sativa

  • Recently, due to environmental and toxicity issues, there has been increasing attention on research regarding natural products that can reduce the use of chemical fertilizers. Wood extracts derived from the biorefining process contain various fertilizer ingredients. HPLC analysis revealed that wood extract contains approximately 5.2% hemicellulosic sugar. The growth of lettuce (Lactuca sativa) upon treatment with wood extract (extract obtained from steam-exploded pine) or water-soluble fertilizers containing different nutrients was analyzed in this study. After two weeks, the growth characteristics of lettuce as affected by wood extract or water-soluble fertilizers were significantly different. The effect of water-soluble fertilizers containing ascorbic acid, magnesium sulfate, citric acid, potassium nitrate, amino acids, or seaweed extract was less desirable than that of wood extracts regarding plant height (18.6 cm), number of leaves (10), leaf length (14.1 cm), shoot fresh wight (9.8 g/plant), root fresh weight (0.8 g/plant) and shoot dry weight (0.6 g/plant). The plant height, number of leaves, leaf length, shoot fresh wight, root fresh weight, shoot dry weight of water-soluble fertilizers containing wood extract were significantly different compared to the control (plant height :13.5 cm, number of leaves : 7, leaf length : 9.4 cm, shoot fresh wight : 5.3 g/plant, root fresh weight : 0.7 g/plant, shoot dry weight : 0.4 g/plant, root dry weight : 0.07 g/plant). From these results, it was concluded that wood extract can be used as a potential water-soluble fertilizer to increase the yield of leafy vegetables.

Lysimachia foenum-graecum Herba Extract, a Novel Biopesticide, Inhibits ABC Transporter Genes and Mycelial Growth of Magnaporthe oryzae

  • To identify a novel biopesticide controlling rice blast disease caused by Magnaporthe oryzae, 700 plant extracts were evaluated for their inhibitory effects on mycelial growth of M. oryzae. The L. foenum-graecum Herba extract showed the lowest inhibition concentration ($IC_{50}$) of $39.28{\mu}g/ml$, which is lower than the $IC_{50}$ of blasticidin S ($63.06{\mu}g/ml$), a conventional fungicide for rice blast disease. When treatments were combined, the $IC_{50}$ of blasticidin S was dramatically reduced to $10.67{\mu}g/ml$. Since ABC transporter genes are involved in fungicide resistance of many organisms, we performed RT-PCR to investigate the transcriptional changes of 40 ABC transporter family genes of M. oryzae treated with the plant extract, blasticidin S, and tetrandrine, a recognized ABC transporter inhibitor. Four ABC transporter genes were prominently activated by blasticidin S treatment, but were suppressed by combinational treatment of blasticidin S with the plant extract, or with tetrandrine that didn't show cellular toxicity by itself in this study. Mycelial death was detected via confocal microscopy at 24 h after plant extract treatment. Finally, subsequent rice field study revealed that the plant extract had high control efficacy of 63.3% and should be considered a biopesticide for rice blast disease. These results showed that extract of L. foenum graecum Herba suppresses M. oryzae ABC transporter genes inducing mycelial death and therefore may be a potent novel biopesticide.

  • 육계 사료에 허브와 식물체의 추출물의 첨가가 생산능력, 영양소 소화율, 장내 미생물, IgG, 혈액의 조성에 미치는 영향을 구명하고자 2차례의 실험을 수행하였다. 시험 1은 대조구, 항생제($Aviilamix^{(R)}premix-avilliamycin$ 0.02 %) 0.03%구, Herb M (Herb $mix^{(R)}$-지황 10%, 산약 10%, 당귀 20 %, 오미자 5%, 감초 5%, 천궁 10%) 0.2%구, Plant extract B(BIOSTONG $510^{(R)}-Anise$ Extract 20%, Thyme Extract 17%, Quillaja 30%) 0.02%구, Plant Extract A($APEX^{(R)}$ - garlic, anise, cinnamon rosemary, thyme, Talin등 50%) 0.02%구 등 5개 처리구를 두었다. 시험 2는 대조구, 항생제($Aviilamix^{(R)}$premix- avilliamycin 0.02%) 0.03% 구, Plant Extract D($Digestarom^{(R)}$-peppermint oil, gentian, horse-tail herb, saponins, oak bark 50 %) 0.02 %구, Plant extract P($Phellozyme^{(R)}$ -Kilmora lavan extract, bromelain extract, bioflavonoid, propolis 15%) 0.1%구, Plant Extract G($Galicin^{(R)}$-allicin 10%) 0.05%구, Plant Extract C(CRINA $POULTRY^{(R)}$-cinnamon, lemongrase, savory, rosewood, spearmint, tea) 0.05%구 등 6개 처리구를 두었다. 시험 1은 1일령 육계($Ross^{(R)}$종) 1,000수(암 수 각각 500수)를 공시하여 5처리 4반복 반복당 50수(암 수 동수) 씩을 floor pen(가로: 2.0 m, 세로: 2.4 m)에 완전 임의 배치하였다. 시험 2는 1일령 육계($Ross^{(R)}$종) 240수(암 수 각각 120수)를 공시하여 6처리 4반복, 반복당 10수(암 수 동수) 씩을 케이지(가로: 35.5 cm, 세로: 45 cm, 높이: 55 cm)에 완전 임의배치하여 각각 35일간사양 시험을 실시하였다. 시험 1에서 생산지수는 대조구에 비해 첨가구들이 높은 경향이 있었고 herbs M구가 가장 높았다. 시험 2에서 $4{\sim}5$주 사료 섭취량은 대조구에 비해 첨가구들이 유의적으로 높았다(P<0.05) 사료 요구율은 항생제 처리구가 다른 처리구보다 낮았다. 시험 1의 RBC와 적혈구 용적 (HCT 또는 PCV), Hb는 첨가구들이 대조구보다 유의적으로 높았다(P<0.05). 시험 2의 BA는 대조구보다 첨가구들이 유의적으로 낮았다(P<0.05). 시험 1과 시험 2의 혈청 IgG 농도는 대조구에 비해 첨가구들이 유의적으로 높았다(P<0.05). 시험 1과 시험 2의 장내 미생물 균총과 영양소 이용율은 처리간에 통계적 차이가 없었다. 결론적으로 일부 한방제와 생약제제는 육계에서 항생제를 대체하여 사용이 가능하며 특히 혈액의 성분에 유의한 영향을 미치는 것으로 사료된다.

몇 가지 식물추출물이 배양 NIH3T3 섬유모세포의 세포생존율과 세포부착률에 미치는 세포독성에 관한 연구 (A Study on the Cytotoxic Effects of Several Plant Extracts on the Cell viability and Cell Adhesion Activity in Cultured NIH3T3 Fibroblast)

  • This study was aimed to clerify the cytotoxicity of some plant extracts such as Hosta longissima HONDA (HL), Hemerocallis fulva var. Kwanso REGL (HFVK), Hemerocallis fulva L (HF), Macrocapium officinale NAKAI (MO) and Mentha canadensis var. piperascens HARA (MCVP), the cultured NIH3T3 fibroblasts were treated with 25, 50, 100, 150 and $200{\mu}g/mL$ of five kinds of plant extracts for 48 hours, respectively. The cytotoxicity of plant extracts was measured by MTT and NR assays for the cell viability, and XTT assay for the cell adhesion activity. In this study, HL, MO and FHVK extracts showed the range of midtoxic-non toxic by the criteria of chemical cytotoxicity. While, the HF and MCVP extracts showed midtoxic. In the extract cytotoxicity, HL, MO and FHVK extracts showed non-toxic by the criteria of extract cytotoxicity. While, HF extract was determined as lower-toxic. In the responsive sensitivity of each plant extract on colorimetric assays, HF extract was sensitive to mitochondrial enzyme by MTT assay, lysosomal enzyme by NR assay and mitochondrial nucleus by XTT assay. While, MCVP extract was sensitive to mitochondrial enzyme by MTT assay and lysosomal enzyme by NR assay than other assays. While, HL, HFVK and MO extracts were most sensitive to NR assay. Cell culture is one of useful materials in the screening of cytotoxic and recovary effect on the putative chemical agents or plant extract. And also, colorimetric assay is regarded as very useful tools for quantitative measurement of cytotoxic effect on plant extracts in vitro.

7종의 주요 해충에 대한 식물추출물의 살란 효과 (Ovicidal Effect of Plant Extract Mixture Against Seven Major Insect Pests)

  • 몇 종의 주요 농작물 해충에 대한 식물추출물(cinnamon extract 5% + citronella oil 10% + citrus oil 30% + derris extract 10% + neem extract 20% + penetrating surfactant 25%)의 살란 효과를 분무법으로 검정하였다. 노린재류의 경우 알이 부화하는 과정에서 치사하는 경향을 보였다. 식물추출물(500배액) 처리시 썩덩나무노린재(Halyomorpha halys), 톱다리개미허리노린재(Riptortus clavatus), 홍비단노린재(Eurydema dominulus), 온실가루이(Trialeurodes vaprarorium), 담배가루이(Bemisia tabaci), 파밤나방(Spodoptera exigua), 검거세미밤나방(Agrotis ipsilon)에 대한 최종 살란 효과는 100%로 높게 나타나, 향후 산업화가 가능할 것으로 생각된다.

Cinnamon Plant Extract as Corrosion Inhibitor for Steel Used in Waste Water Treatment Plants and Its Biological Effect on Escherichia coli

  • The inhibition effect of cinnamon plant extract as a green corrosion inhibitor for steel in sulfide polluted salt water was studied by potentiodynamic polarization, electrochemical impedance spectroscopy (EIS), and electrochemical frequency modulation (EFM). The results showed that cinnamon plant extract in sulfide polluted salt water is a good corrosion inhibitor with inhibition efficiency reached to 80% at 250 ppm of the plant extract. The adsorption of cinnamon obeys Temkin adsorption isotherm, and acts as a mixed-type of inhibitor but dominantly as a cathodic inhibitor in sulfide polluted salt water.

  • The antimicrobial activity of seventy five ethanol extracts obtained from 67 different kinds of plant species of the Mongolian flora were evaluated by means of the disc diffusion method against five species of microorganisms, Escherichia coli, Enterococcus faecalis, Staphylococcus aureus, Micrococcus luteus and Pseudomonas aeruginosa. Among the plant extracts examined, 34 kinds of extracts demonstrated significant antibacterial activity against one or more species of microorganisms, respectively. Especially, the root extract of Paeonia anomala, the whole herb extract of Myricaria alopecuroides, the whole herb extract of comarum zalesovianum, the whole herb extract of Agrimonia pilosa and some other plant extracts demonstrated a particularly potent antimicrobial activity. The ethylacetate fractions obtained from the whole herb extract of Myricaria alopecuroides and from those of Sedum aizoon, Paeonia anomala, Sedum hybridum and Dasiphora fruticosa exhibited a particularly potent antibacterial activity especially against Staphylococcus aureus and Micrococcus luteus.

Anemarrhena asphodeloides Extract Inhibits the Mycelial Growth of Magnaporthe oryzae and Controls the Rice Blast Disease

  • Previously, we have reported a plant extract isolated from Lysimachia foenum gracum Herba as a new environment friendly biopesticide that has the mycelial growth inhibition effect on Magnaporthe oryzae, the pathogenic fungus of the rice blast disease. For the finding of additional biopesticide candidate, we tested the mycelial growth inhibitory effects about 700 species of plant extracts on PDA media. Among them, the extract of Anemarrhena asphodeloides showed prominent inhibitory effect of which $IC_{50}$ was $139.7{\mu}g/ml$. Mycelial radii of M. oryzae were measured on PDA medium containing the four organic solvent fractions isolated from total extract from A. asphodeloides. Ethyl acetate fraction showed the impressive inhibitory effect of $IC_{50}$, $54.12{\mu}g/ml$. In the subsequent rice field test for the total extract of A. asphodeloides, we obtained encouraging 62.0% control rate of rice blast disease without any phytotoxicity. It is almost equivalent to that of chemical pesticides implying the applicability of the extract as a new biopesticide. In further study, the analysis of active ingredients of the extract would be necessary for the development of a new biopesticide and for the verification of cellular mechanism by which the mycelial growth of M. oryzae inhibited.
