Livestock grazing and trampling effects on plant functional composition at three wells in the desert steppe of Mongolia

  • Backgrounds: In arid grasslands, wells are subject to heavy trampling and grazing pressure, which can increase vulnerability to local land degradation. To investigate trampling and grazing, we surveyed plant communities at three well sites in the desert steppe of Mongolia, using 1600-m line transects from the wells. The sites (Bshrub, Sshrub, and shrubL) differed by concomitant shrub type (big shrub, small shrub, and shrub-limited) and livestock pressure (light, medium, and heavy). A plant classification scheme based on edibility and morphology (rosette or creeping type) was used to separate grazing and trampling effects on plant communities. Results: Edible plants were dominant at all sites but a fraction of grazing- and trampling-tolerant plants increased in the order Bshrub, Sshrub, and shrubL, following livestock pressure. Clear transition zones from inedible to edible plant groups were recognized but at different locations and ranges among the sites. Trampling-tolerant plants explained 90% of inedible plants at Sshrub with camels and horses, but grazing-tolerant plants prevailed (60%) at shrubL with the largest livestock number. Plant coverage increased significantly along the transects at Bshrub and Sshrub but showed no meaningful change at shrubL. Herbaceous plant biomass showed significant positive and negative trends at Bshrub and shrubL, respectively. Conclusions: Both grazing and trampling can produce larger fractions of inedible plants; in this, camel and horses can have considerable effects on desert-steppe plant communities through trampling.

Direct Multiple Shooting Induction of Taraxacum

  • Plants of the genus Taraxacum are well-known as a traditional herbal remedy with a long history, and they have also been extensively used as food, chemicals and cosmetics. In this study, four Taraxacum species distributed in Korea (T. mongolicum, T. officinale, T. mongolicum variation and T. officinale variation) were utilized for an efficient method for direct multiple shooting induction and regeneration, using leaf blade, transition zone, petiole and root as explants in MS media with various hormone concentration and combination. MS medium containing IAA 0.2 mg/L and TDZ 1.0 mg/L showed the highest induction frequency of all the hormone combinations. Besides, the induction of T. mongolicum variation was most effective comparing with the other three species by the average induction frequency of four explants. While the induction effect of leaf blade explant was more obvious than the other three explants. This system exhibited a rapid propagation of shoots from the leaf blade explants and makes it convenient to make use of these Taraxacum species to develop their diverse applications in the future.

Do Physiognomically Designated Protected Areas Match Well with Ecological Data based upon Diversity Indices and Ordination? Implications for Urban Forest Conservation

  • We surveyed the vegetation of an ecological landscape preservation area (legally protected conservation areas or national parks) and the surrounding areas of Mt. Cheonggye, Republic of Korea, to explore the conservation implications for preservation areas and surrounding transition areas. We calculated diversity indices to identify the properties of the preservation and surrounding areas that are relevant to conservation efforts. We then compared the plant community composition between the areas using field and quadrat surveys in the preservation and surrounding areas. The cover of the dominant species in all tree and herb layers was markedly higher in the preservation area than in the peripheral zones. The species richness indices were significantly higher in the preservation area than in the peripheral zones. Ordination using detrended canonical correspondence analyses showed that the cover of the dominant tree species and rocks could explain the distribution of plant species in the Cartesian space of the ordination. Our results demonstrate that physiognomically designated protected areas match well with ecological data based on diversity indices and ordination analyses and that disturbances in the areas surrounding the ecological landscape of preservation areas can have considerable impacts on plant diversity indices. Hence, the preservation and management of surrounding areas are essential conservation elements for protecting the entire ecological landscape of preservation areas.

Studies of Vegetation Structure Analysis and Vegetation Transition over 25 years of Evergreen Broad-leaved Forest in Hong-Do Island

  • This study aims at classifying and interpreting on the vegetation structure and the vegetation transition over 25 years (between 1986 and 2010), and the correlation with the change of some conditions (the vegetation height and coverage on each layer and the climate factors as WI, CI, mean annual temperature, mean annual total precipitation etc.) in the Evergreen Broad-Leaved Forest,, Hong-Do island. The EBLF is classified into five units of vegetation (Hedera rhombea-Machilus thunbergii community (M-M comm.), Castanopsis sieboldii forest (Machilus japonica-Castanopsis sieboldii community; Raphiolepis indica var. umbellata-C. sieboldii community), community (Qa comm.), Carpinus turczaninovii community (Ct comm.), Camellia japonica stand (Cj stand)). The vegetation transition by CCA had high correlation with the height and coverage on each layer and the climate factors, and it did the succession (transition) that the M-M comm. (2010) from Mallotus japonicus community Machilus thunbergii community Carpinus coreana community (Cc comm.) Aucuba japonica community (Aj comm.) Trachelos permum asiaticum var. intermedium-Quercus acuta community (TQ comm.) (1986), the communities of C. sieboldii forest (2010) from Aj comm. TQ comm. Raphiolepis umbellata-Camellia japonica community (RC comm.) (1986), the Qa comm. (2010) from Ardisia japonica-Castanopsis sieboldii community (AC comm.) and TQ comm. (1986), the Ct comm. (2010) from Cc comm. RC comm. Aj comm. Quercus serrata community and the Cj stand (2010) from AC comm. (1986). the height and coverage on each layer are also changed.

연강(Mild Steel)의 극저온 파괴 거동에 대한 실험적 연구 (Study on Fracture Behavior of Mild Steel Under Cryogenic Condition)

  • LNG선이나 해양플랜트 같은 설비나 구조물은 작동 조건을 고려할 때, 특히 부재마다 가지는 고유한 연성 취성천이온도(Ductile to Brittle Transition Temperature, DBTT)를 고려하여 설계되어야 한다. 본 연구에서는 해양플랜트 및 LNG선에 hull plate로 쓰이는 A-grade 연강(mild steel)에 대해 DBTT를 샤르피 V-노치(CVN) 실험을 통해 알아보았고 파괴형상을 통한 파괴거동을 살펴보았다. 그 결과 온도가 감소함에 따라 충격 흡수에너지는 감소함을 보였다. Upper shelf energy region과 lower shelf energy region이 나타나고 그 사이 구간의 천이점을 통해 DBTT가 결정되었다. 파괴형상에서는 upper shelf energy region에서 수많은 딤플이 연성파괴 형상으로 관찰되고 lower shelf energy region에서는 전형적인 취성파단형상이 관찰되었다. 이를 통해 BCC 구조를 가지는 A-grade 연강은 upper shelf energy 구역과 lower shelf energy구역을 보이고 그 사이 구간의 천이점에서 급격하게 온도가 떨어지는 DBTT구간을 뚜렷하게 보이는 특성을 알 수 있었다.

최적전원차성을 위한 절감 시뮬레이션 방법의 개발 (The Development of the Simplified Simulation Technique for the Best Generation Mix)

  • The simplified simulation technique for the best generation mix is developed and the studied results are described. The best generation mix over study time from the economic point of view can be easily constructed by this technique. Generator maintenance, the operation of pumpgenerator and LNG thermal generator with limited energy are simulated variously, so a role of each generator is also easily evaluated. Through parametric analysis, useful planning guide points are obtained for the best generation mix transition, nuclear power plant construction cost, ruanium cost , oil cost, coal cost and midnight factor in the study case corresponding to real power system size model.

Metabolic engineering of Lilium ${\times}$ formolongi using multiple genes of the carotenoid biosynthesis pathway

  • Lilium ${\times}$ formolongi was genetically engineered by Agrobacterium-mediated transformation with the plasmid pCrtZW-N8idi-crtEBIY, which contains seven enzyme genes under the regulation of the CaMV 35S promoter. In the transformants, ketocarotenoids were detected in both calli and leaves, which showed a strong orange color. In transgenic calli, the total amount of carotenoids [133.3 ${\mu}g/g$ fresh weight (FW)] was 26.1-fold higher than in wild-type calli. The chlorophyll content and photosynthetic efficiency in transgenic orange plantlets were significantly lowered; however, after several months of subculture, they had turned into plantlets with green leaves that showed significant increases in chlorophyll and photosynthetic efficiency. The total carotenoid contents in leaves of transgenic orange and green plantlets were quantified at 102.9 and 135.2 ${\mu}g/g$ FW, respectively, corresponding to 5.6- and 7.4-fold increases over the levels in the wild-type. Ketocarotenoids such as echinenone, canthaxanthin, 3'-hydroxyechinenone, 3-hydroxyechinenone, and astaxanthin were detected in both transgenic calli and orange leaves. A significant change in the type and composition of ketocarotenoids was observed during the transition from orange transgenic plantlets to green plantlets. Although 3'-hydroxyechinenone, 3-hydroxyechinenone, astaxanthin, and adonirubin were absent, and echinenone and canthaxanthin were present at lower levels, interestingly, the upregulation of carotenoid biosynthesis led to an increase in the total carotenoid concentration (+31.4%) in leaves of the transgenic green plantlets.

Development of a new vitrification solution, VSL, and its application to the cryopreservation of gentian axillary buds

  • Vitrification methods are convenient for cryopreserving plant specimens, as the specimens are plunged directly into liquid nitrogen (LN) from ambient temperatures. However, tissues and species with poor survival are still not uncommon. The development of vitrification solutions with high survival that cover a range of materials is important. We attempted to develop new vitrification solutions using bromegrass cells and found that VSL, comprising 20% (w/v) glycerol, 30% (w/v) ethylene glycol, 5% (w/v) sucrose, 10% (w/v) DMSO and 10 mM $CaCl_2$, gave the highest survival following cryopreservation, as determined by fluorescein diacetate staining. However, the cryopreserved cells showed little regrowth, for unknown reasons. To check its applicability, VSL was used to cryopreserve gentian axillary buds and the performance was compared with those of conventional vitrification solutions. Excised gentian stem segments with axillary buds (shoot apices) were two-step precultured with sucrose to induce osmotic tolerance prior to cryopreservation. Gentian axillary buds cryopreserved using VSL following the appropriate preculturing approach exhibited 78% survival (determined by the regrowth capacity), which was comparable to PVS2 and PVS1 and far better than PVS3. VSL had a wider optimal incubation time (20-45 min) than PVS2 and was more suitable for cryopreserving gentian buds. The optimal duration of the first step of the preculture was 7-11 days, and preculturing with sucrose and glucose gave a much higher survival than fructose and maltose. VSL was able to vitrify during cooling to LN temperatures, as glass transition and devitrification points were detected in the warming profiles from differential scanning calorimetry. VSL and its derivative, VSL+, seem to have the potential to be good alternatives to PVS2 for the cryopreservation of some materials, as exemplified by gentian buds.

Trace elements analysis in some medicinal plants using graphite furnace-atomic absorption spectroscopy

  • Ten trace elements (Mg, Ca, V, Cr, Mn, Fe, Cu, Zn, Mo, and Cd) were determined in seven traditionally used wild medicinal plants of the Nagas in the North-East India viz Cynoglossum furcatum Wallich, Elsholtzia blanda Bentham, Lycopodium cernuum Linnaeus, Potentilla fulgens Wallich ex Hooker, Swertia macrosperma C.B. Clarke, Thalictrum foliolosum DC and Valeriana jatamansi Jones. Plant samples were dried, weighed, digested and analyzed for their mineral distribution level ranging from trace to major elements by graphite furnace-atomic absorption spectroscopy. All the medicinal herbs studied were found to contain the ten elements analyzed. The elemental concentration is given in mg/kg. Mg and Ca are present in high concentrations in both the roots and leaves of the plant samples. Among trace transition metals Fe had the highest concentration, followed by Mn, Cr, Cu, V, Zn, and Mo. Analysis showed that the toxic Cd element was at a lower concentration and is within the permissible limit of FAO/WHO, California standards and United States Pharmacopeia Limit for Nutritional Supplements.

Firewood Plantation as an Alternative Source of Energy in the Philippines

  • The increasing price of liquefied petroleum gas (LPG) has forced many households to shift to firewood/charcoal in the Philippines, causing tremendous pressure on the dwindling forest. This transition is more obvious in the rural area where firewood price is almost half of that in the cities. Both problems on big demand for firewood and the pressure on our forest can be partly solved by going into firewood plantation. After all, the country has vast track of idle lands, both private and government lands, waiting to be developed. What the government can do are: a) aggressive promotion of firewood plantation, b) simplify further the old technology on establishing firewood plantation to encourage more farmers, c) improve charcoal processing and d) promote the use of efficient stoves. This write up discusses the need for firewood plantation, areas available for planting, where to plant in farms, what species to plant in a particular soil type/location for optimum growth, planting methods, harvesting, marketing, and the side lights on the ecological benefits derived from plantation aside from wood.
