Comparison of Active Ingredients between Field Grown and In Vitro Cultured Rhizome of Korean Native Ginger (Zingiber officinale Roscoe) (조직배양생강과 한국재래종 생강의 유효성분 비교)

  • This study was carried out to compare and analyze the active ingredients of Korean native ginger and rhizome derived from in vitro shoot-tip culture of Korean native ginger. Proximate compositions, mineral nutrients, free sugars, fatty acids, volatile components, 6-gingerol, and 6-shogaol were analysed and evaluated. Korean native ginger was proved to have a little more contents than in vitro rhizome in proximate compositions (crude ash, crude lipid, crude protein, carbohydrate). Mineral nutrient contents (Cu, Fe, Mn, Zn) of in vitro rhizome were higher than those of Korean native ginger. Among the mineral nutrients, the quantity of K was the highest, followed by P, Mg, Na, and Ca. Free sugar contents (fructose, glucose, sucrose) of in vitro rhizome were higher than those of Korean native ginger. Fatty acids containing less than C14 was the major among the fatty acids in ginger. Citral ingredient of the unique aromatic compound of Korean native ginger was stronger than that of the rhizome derived from in vitro shoot-tip culture. Gingerol concentration was increased by shoot-tip culture.

Suppressive effects of $Schizandra$ $chinensis$ Baillon water extract on allergy-related cytokine generation and degranulation in IgE-antigen complex-stimulated RBL-2H3 cells

  • $Schizandra$ $chinensis$ Baillon is a traditional folk medicine plant that is used to treat and prevent several inflammatory diseases and cancer in Korea, but the underlying mechanisms involved in its anti-allergic activity are not fully understood. This study was designed to investigate mechanisms of anti-allergic activity of a $Schizandra$ $chinensis$ Baillon water extract (SCWE) in immunoglobulin E (IgE)-antigen complex-stimulated RBL2H3 cells and to assess whether gastric and intestinal digestion affects the anti-allergic properties of SCWE. Oxidative stress is an important consequence of the allergic inflammatory response. The antioxidant activities of SCWE increased in a concentration-dependent manner. RBL-2H3 cells were sensitized with monoclonal anti-dinitrophenol (DNP) specific IgE, treated with SCWE, and challenged with the antigen DNP-human serum albumin. SCWE inhibited ${\beta}$-hexosaminidase release and expression of interleukin (IL)-4, IL-13, and tumor necrosis factor-alpha (TNF-${\alpha}$) mRNA and protein in IgE-antigen complex-stimulated RBL2H3 cells. We found that digested SCWE fully maintained its antioxidant activity and anti-allergic activity against the IgE-antigen complex-induced activation of RBL-2H3 cells. SCWE may be useful for preventing allergic diseases, such as asthma. Thus, SCWE could be used as a natural functional ingredient for allergic diseases in the food and/or pharmaceutical industries.

Anti-inflammatory and Anti-bacterial Active Ingredients Derived from the Extract of the Leaves of Hydrangea Petiolaris (등수국 잎 추출물 유래 항염 및 항균 활성 성분)

  • In this study, the anti-inflammatory and anti-bacterial activities of the extracts from the leaves of the Hydrangea petiolaris were identified, and the chemical structure was identified by separating the active ingredient. As the result of the anti-inflammatory activity experiment using RAW 264.7 cells, it was confirmed that the n-hexane (Hex) and ethyl acetate (EtOAc) fractions inhibited the production of nitric oxide (NO) and the expression of iNOS protein in a concentration-dependent manner without cytotoxicity. In addition, the n-Hex and EtOAc fractions reduced the production of pro-inflammatory cytokines (TNF-α, IL-1β and IL-6). Upon the anti-bacterial tests using Staphylococcus epidermidis and Cutibacterium acnes, the extract, n-Hex, EtOAc and n-butanol (BuOH) fractions showed potent activities. In order to isolate the active constituents, the n-Hex and EtOAc fractions were further purified to afford four phytochemicals; phytol (1), corosolic acid (2), asiatic acid (3) and 1-O-p-coumaroyl-β-D-glucopyranoside (4). All of the compounds 1 - 4 were isolated for the first time from this plant. In addition, the contents of isolated compounds were determined by HPLC and the quantity of phytol (1) was 27.8 mg/g for the 70% EtOH extract. Based on the above research results, it is believed that it will be possible to develop a natural cosmetic material that has anti-inflammatory and anti-bacterial effects using the extract of H. petiolaris leaves.

Characteristics of Wet Distillers Grains on In vitro Ruminal Fermentation and Its Effects on Performance and Carcass Characteristics of Finishing Hanwoo Steers

  • Two experiments were conducted to determine the nutrient composition, in vitro ruminal ammonia concentrations and pH of wet distillers grains (WDG, produced from tapioca 70% and rice 30%) and to evaluate dietary effects of fermented total mixed ration (TMR) using WDG on the performance, blood metabolites and carcass characteristics of Hanwoo steers from mid fattening to slaughter. In Exp. I, average dry matter (DM), crude protein, ether extract, crude fiber, ash, neutral detergent fiber, acid detergent fiber, and nitrogen free extract of seven WDG samples from an ethanol plant with different sampling dates were 19.9%, 24.8%, 3.8%, 21.8%, 8.87%, 60.3%, 34.5%, and 40.7% (DM basis), respectively. For in vitro ammonia concentrations and pH, each sample was assigned to 7 incubation times (0, 4, 8, 12, 24, 48, and 72 h). Linear increase was observed between 12 and 48 h for ammonia concentrations, but final ammonia concentrations (72 h) were not significantly different among WDG samples and fermentation patterns of WDG samples showed similar tendency. In vitro pH varied among treatments from 0 to 24 h, but were not different statistically after 48 h. In Exp. II, 45 Hanwoo steers of 23 months ($641{\pm}123kg$) from mid fattening period to slaughter (248 days) were randomly divided into three groups of 15 pens each (five repetitions/each treatment) and assigned to one of three dietary treatments; i) Control (TMR), ii) WDG 15 (TMR containing 15% of WDG, as fed basis) and iii) WDG 28 (TMR containing 28% of WDG, as fed basis). The body weight (BW), ADG, and feed conversion ratio (FCR) of control and WDG 15 and 28 during 248 days were 760.8, 740.1, and 765.5 kg, and 0.50, 0.50, and 0.52 kg/d, and 18.6, 17.6, and 17.1, respectively. The dry matter intake (DMI) (kg/d) of control (9.11) was higher (p<0.05) than WDG treatments (WDG 15%, 8.57; 28%, 8.70). Nevertheless, DMI did not affect BW, ADG, and FCR of Hanwoo finishing steers. Blood metabolites were in normal ranges and were not different among treatments except the albumin concentration. In carcass characteristics, WDG 15 (30%) showed higher frequency of A-carcass yield grade than WDG 28 (15%) and control (7%), and WDG 28 (61%) showed higher frequency of $1^{{+}{+}}$ and $1^+$-carcass quality grade than WDG 15 (40%) and control (60%). In conclusion, using WDG up to 28% in TMR did not show any negative effect on the performance and blood metabolites, and improved carcass quality of Hanwoo steers. Therefore, WDG can be a useful feed ingredient for Hanwoo steers in mid-fattening period to slaughter.

Analysis on the Components and Safety Evaluation of Abeliophyllum distichum Nakai Leaves and Stems (미선나무 잎과 줄기의 성분 분석 및 안전성 평가)

  • Objectives: This study was carried out in order to analyze the composition of the leaves and stems of Abeliophyllum distichum Nakai, with the aim of obtaining basic data for utilizing the plant as a food ingredient, as well as for processing. Methods: Leaves and stems from Abeliophyllum distichum Nakai were harvested at Cheongcheon-myeon, Geosan-gun, Chungcheongbuk-do, and were subsequently freeze-dried and ground to a fine powder for chemical component analysis and safety evaluation. Results: The moisture contents of Abeliophyllum distichum Nakai leaves and stems were respectively 65.07% and 40.97%, and the crude ash contents were 1.32% and 0.91%. In addition, the crude protein contents were 11.97% and 3.77%, and the crude fat contents were 2.52% and 0.36%, respectively. The fructose and glucose contents were 32.13 mg/g and 56.17 mg/g for leaves, and 11.38 mg/g and 10.59 mg/g for stems. The major fatty acids of the leaves were palmitic acid (31.79%) and stearic acid (14.79%), and those for stems were linolenic acid (32.78%) and palmitic acid (26.75%). The ascorbic acid contents of leaves and stems were 1.32 mg/g and 0.30 mg/g respectively. The calcium content was found to be the highest among the minerals tested, both in the leaves and stems, with the levels being 166.17 mg/100 g for leaves and 592.34 mg/100 g for stems. The content of organic acid was greater in leaves than in stems, with that of malic acid accounting for more than 75% of total organic acids for both samples. The total phenolic compounds and flavonoid contents of Abeliophyllum distichum Nakai were 50.64 mg/g and 13.53 mg/g in leaves and 96.47 mg/g and 18.53 mg/g in stems, respectively. No changes were shown in the number of micronucleated polychromatic erythrocytes (MNPCE) among 2,000 polychromatic erythrocytes compared to the negative control. Abeliophyllum distichum Nakai was administered orally to rats in order to investigate acute toxicity. The $LD_{50}$ values in rats were above 2,000 mg/kg. Conclusion: These results indicate that the leaves and stems of Abeliophyllum distichum Nakai can be used as natural ingredients in the development of nutritional and functional materials.

MMP-2 and MMP-9 Inhibitory Effects of Different Solvent Fractions from Corydalis heterocarpa (염주괴불주머니 분획물의 MMP-2, MMP-9 발현 억제 효과)

  • Natural products have always been an attractive source in terms of novel anti-metastatic compounds which can hinder MMP expression and activity. Corydalis heterocarpa is a salt marsh plant found in the seashores throughout Korea. Its yellow flowers and spikes have been an ingredient in folk medicine to treat spasm and contractions. The present study assessed the potential of different solvent-based fractions from the crude extract of Corydalis heterocarpa (CHE), a halophyte with reported bioactivities, to suppress the PMA-induced MMP expression in human fibrosarcoma HT-1080 cells. The solvent fractions which were named after the solvent used for fractionation (n-hexane, 85% aqueous (aq.) methanol (MeOH), n-butanol (BuOH), and H2O were shown to inhibit the both elevated mRNA and protein expression levels of MMP-2 and MMP-9 and simultaneously relieved the suppression on the expression of the endogenous MMP inhibitors TIMP-1 and TIMP-2. Results indicated that the CHE fractions might intervene with the PMA-induced activation of the MAPK signaling which is the upstream activator of MMP overexpression. Among tested samples, 85% aq. MeOH and n-hexane fractions of CHE was determined to be the most active and future studies to isolate the bioactive substances responsible for the regulation of the MMP expression are, therefore, urged. In conclusion, C. heterocarpa was shown to be a potential source of anti-metastatic compounds and n-Hexane and MeOH fractions might yield lead molecules to develop novel MMP inhibitors.

With fertilizer ingredients in pig sludge chemical qualities, the content of the nitrogen and the phosphoric acid comes 4.4% to be 6.29%, pH 7.02 and content of the copper and the zinc which is a heavy metal which contains in pig sludge with 805 mg/kg and 1,704 mg/kg, it is a restrictive standard of the fertilizer, 300 mg/kg and 900 mg/kg it sees to be high, it manufactures citric acid 1 hydrate with the organic acid solution, heavy metals of pig sludge where it is a mixture ratio of the organic acid solution, it divides to 25%, 50%, 75% and 100% 4 kind levels, the result which measures the heavy metal exclusion ratio of the copper and the zinc, the mixture ratio of the organic acid solution to be many exclusion ratio of the copper and the zinc is showing a just interrelation, from organic acid solution 100% level content of pig sludge remains copper and zinc 330.03 mg/kg and 41.28 mg/kg, it shows the exclusion ratio of copper 59% zinc 97%. 'Cheonganok' growth characteristics with citric acid 1 hydrate, Treatment 2 and control growth characteristics etc, it exclusion the copper and the zinc it doesn't appear on significant difference statistically but, treatment 3 after only pig sludge in resources disposal where it seeding, growth characteristics of leaf area etc. is badness, it compares in control and treatment 2 the growth characteristics badness, it is appearing, it is caused by with disease and insects occurrence of $Ostrinia$ $furnacalis$ and brown spot, the damage was many. From forage value, Treatment 2 where it exclusion the heavy metal with the citric acid 1 hydrate with control it compares and there are not significant difference from crude protein and ADF and NDF contents etc., seeding only Pig Sludge in resources disposal treatment 3, it is caused by with $Ostrinia$ $furnacalis$ etc., trunk and aging of the leaf to be high ADF content is low. but from crude protein, the nitrogen ingredient which pig sludge has and interrelation it seemed and high numerical value were confirmed.

Studies on the Constituents of Gynostemma pentaphyllum Makino (덩굴차(Gynostemma pentaphylum Makino)의 성분에 관한 연구)

  • Gynostemma pentaphyllum Makino is a kind of perennial liana plant belonging to the Cucurbitaceae family. Since it was first discovered and reported to the Japanese academy in 1977, it came to be widely known to China and Southeast Asia areas as well as Japan. In Korea its name began to appear in the first half of 1980's, and it is now being nationwidely cultivated and popularized as tea stuffs. Gynostemma pentaphyllum Makino has remarkably strong propagation power. In addition. since it contains a great quantity of beneficial components, especially saponin it is believed to have excellent medical effects, too. This study was undertaken to examine the value of Gynostemma pentaphyllum Makino as a health-promotint tea, by investigating the components creating the flaver of tea and the saponin ingredient making medical effects as well as by inspecting whether Gynostemma pentaphyllum Makino products show different component arrangements according to their growing districts. As raw materials of the experiment, three kinds of Gynostemma pentaphyllum Makino samples produced in Suwon, Geochang and Uleungdo were taken. To accomplish the formerly stated goals, the contained quantity of the proximate composition, free sugar, reducing sugar, free amino acid, minerals, tannin, caffeine and vitamin C were measured respectively and compared between the samples, while the saponin content was compared with heat extracted from panax ginseng. The results of the experiment are as follows: 1. In the case of the proximate composition, the crude fat content was the highest in the Gymostemma pentaphyllum Makino material from Geochang (1.62%), the second in that from Suwon (1.56%), and the lowest in that from Uleungdo (1.0%). In addition the Geochang: produced sample had the greatest quantity of the crude protein and ash contents: the order of the crude Protein content was the Geochang produced (17.83%), the Suwon-produced (15.87%), and the Uleungdo-produced(12.28%), while that of the ash content was the Geochang-produced (14.80%), the Uleungdo-produced(10.17%), and the Suwon-produced(9.34%). 2. As for the reducing sugar contents, the Suwon-produced scored the highest of the three (3.3%), while the Geochang-produced was 1.3% and the Uleungdo-produced 0.5%. The total content of free sugar was 1.07% (the Suwon-produced), and 0.49% (the Geochang-produced) respectively but the sample from Uleungdo showed almost no free sugar content. The contained quantity of fructose and glucose was the highest among the kinds of free sugar both in the Suwon-produced and in the Geochang-produced. 3. The content of amino acid was the highest in the Suwon-produced(1.41%), the second in the Geochang-produced(1.37%), and the lowest in the Uleungdo-produced(0.53%). In the experiment, sixteen kinds of amino acid were extracted-Asp. Thr. Ser. Glu. Gly. Ala. Val. Het. Ileu. Leu. Tyr. Phe. Lys. His. Arg. Try. All of them except glutamic acid and methionine showed the highest quantity score in the Suwon-produced, while the glutamic acid content was the higest in the Uleungdo-produced and the methionine content in the Geochang-produced. The sequential arrangement of the sixteen contents according to their magnitude ranged from glycine, aspartic acid, and glutamic acid (the highest) to tryptophan, serine and lysine (the lowest). 4. Ten kinds of mineral were detected-Ca, Mn, Cd, K, Na, Pb, Mg, Fe, Zn, Cu. Among them, the content of Cd, Na, Mg, Zn and Fe was the highest in the Geochang-produced and that of K in the Suwon-produced. 5. The Geochang-produced materials contained much a larger quantity of tannin (6.3%) than the Suwon-produced (2.6%). Neither caffeine nor vitamin C was detected in the three kinds of materials. 6. In the case of the saponin content the Geochang-produced showd 2.39%, the Uleungdo-produced 1.77% and the Suwon-produced 1.49% respectively. However, it was found also that the saponin content of Gymostemma pentaphyllum Makino was the same kind as that of panax ginseng.
