Isolation of the Constituents with Cancer Cell Growth Inhibition and Anti-inflammatory Activity from Persicaria nepalensis (암세포 성장 저해 및 항염증 효능을 나타내는 산여뀌 성분의 분리)

  • This study was initially explored to procure biomaterials capable of inhibiting cancer cell growth from nine Persicaria species (Polygonaceae). The extract of P. nepalensis that was selected from the initial screenings was further fractionated to identify bioactive compounds. The ethyl acetate (EtOAc) fraction was shown to be the most active in the inhibition of cell growth against six cancer cell lines (IC50 value of 3.77-12.87 ㎍/ml). Phytochemical study led to the isolation of two galactolipids of 1,2-di-O-linolenoyl-3-O-β-D-galactospyranosyl-sn-glycerol (1) and 1-O-linolenoyl-3-O-β-D-galactospyranosyl-sn-glycerol (2) from the hexane fraction and three phenylpropanoyl sucroses of lapathoside A (3), vanicoside B (4) and lapathoside C (5) from the EtOAc fraction. These isolated compounds have not been reported from this plant. Compounds 3 and 4 exhibited the effective growth inhibition against a panel of cancer cell lines (IC50 value of 6.90-18.09 μM). In addition, the anti-inflammatory activity was evaluated to determine lipopolysaccharide (LPS)-induced nitric oxide (NO) formation in RAW264.7 mouse macrophage cells. The EtOAc fraction (IC50; 34.14 ㎍/ml) and its constituents, 3 (8.55 μM) and 4 (7.83 μM) were shown to be effective in the inhibition of LPS-induced NO production. Therefore, compounds 3 and 4 were considered to be active constituents for anti-inflammatory and antitumor activity from P. nepalensis.

Purification of Antioxidant substance from the stem bark of Rhus verniciflua

  • The Rhus verniciflua contains alkly(en)-catechol type allergens with a saiurated or unsaturated alkly chain of 15 or 17 carbon atoms. It has been recognized as an extremely active allergen causing skin reactions similar In poison ivy. The allergic contact dermatitis induced by the urushiol is known to be mediated be T lymphocytes whicht specifically recognize the hepten urushiol. Therefore. direct use of this plant as a medicinal purpose might imply a considerable hazard in Korea. In this study, using the established method for the detoxification from the stem bark of Rhus verniciflua, an strong antioxidant substance was isolated and characterized DPPH (diphenypricryl hydrazyl) assay measures hydrogen atom-donating activity and hence provides a measure of free radical scavenging antioxidant activity. DPPH, a purple-colored stable free radical, is reduced to yellow-colored diphenylpicryl hydrazine by antioxidants to deducing agents. Antioxidative effects of the water extract from RV were measured by DPPH assay. Twenty microliters of the extract was added to 1ml of 100mM DPPH solution in ethanol The mixture was shaken and left to stand for 10min at room temperature. The crude water extracts was purified by using HPLC method with a DEAE (anionic type), CN, ODS column. The purified compound remained stable at pH 3.0-6,0, but unstable above pH 6.5. It was stable heat at 10$0^{\circ}C$ for 4 hours, but still had about 80% of residual activity after treatment at 10$0^{\circ}C$ for 5 hours. The elemental composition of the HR-EI mass spectrum at m/z 170.02 was estimated the empirical formula as $C_{7}$ $H_{6}$ $O_{5}$. $C_{10}$ $H_4$ $O_2$N$_1$, $C_{5}$ $H_4$ $O_4$N$_3$, $C_{8}$$H_2O$$_1$N$_4$. In antimicrobial test, no inhibition was observed against Gram-positive and negative bacteria. This compound was stronger than that of commercial antioxidant by DPPH test, such as BHT, BHC at the same concentration (20$\mu$g/ml).ml).

Vegetation Structure and Management Proposal of Hwangsong Park in Kyongju (경주 황성공원의 식생구조 및 관리방안)

  • Vegetation structure of $Hwangs\v{o}ng$ Park in $Ky\v{o}ugju$ was investigated to study ecological value and to suggest a desirable management proposal. To do this, twenty plots were set up and surveyed. The $Hwangs\v{o}ng$ Park forest was classified into five plant communities by the DCA technique, which were two types of Pinus densiflora communities, Quercus spp.-P. densiflora, Robinia pseudoacacia and Q. mongolica community. In the diversity analysis, 29 woody species were observed, but species composition was simple. No. of average species is $5.4\pm2.9$ per a plot($100m^2$), and Pinus densiflora is a dominant species in DBH $27\sim47cm$ class. From the results of tree ring and growth analysis, it was found out that pinus densiflora community of $Hwangs\v{o}ng$ Park has a historical and ecological value, but recently the growth rate was decreasing. The management proposal was suggested upon to the above results.

Flavonoid Compounds from the Leaves of Kalanchoe prolifera and Their Cytotoxic Activity against P-388 Murine Leukimia Cells

  • Seven flavonoid compounds, kaempferol (1), quercetin (2), quercetin-3-O-${\beta}$-D-glucopyranoside (3), kaempferol-3-O-${\beta}$-D-glucopyranoside (4), kaempferol-3-O-${\alpha}$-L-rhamnoside (5), quercetin-3-O-sophoroside (6) and quercetin-3-O-rutinoside (7), were isolated from the methanolic extract of leaves of Kalanchoe prolifera. Compounds 1-7 were isolated for first time from this plant. These compounds were evaluated their cytotoxic activity against P-388 murine leukimia cells in vitro. Among those compounds kaempferol (1) and quercetin (2) showed strongest cytotoxic activity with $IC_{50}$ values of $4.45{\pm}0.05$ and $6.28{\pm}0.02{\mu}g/mL$, respectively.

  • Photoinhibition of photosystem (PS) n was induced in primary leaves of 25 day-old peppers grown $100\;{\mu}mol{\cdot}m^{-2}{\cdot}s^{-1},\;at\;25^{\circ}C$. The modulation of PSII functionality in vivo was induced by varying both irradiance ($0-3000\;{\mu}molm^{-2}{\cdot}s^{-1}$) and duration (0-70 min) of light treatment. The functionality of PSII was investigated in terms of photochemical efficiency of PSII (Fv/Fm) and quantum yield of $O_2$ evolution, and expressed as a function of photon exposure [$mol\;photons{\cdot}m^{-2}$, the product of irradiance and duration of light treatment (Bell and Rose, 1981)]. Contrary to the linear decline of Fv/Fm ratio showing 50% decreases by absorption of $10\;mol\;photons{\cdot}m^{-2$, quantum yield of $O_2$ evolution decreased biphasically with increasing photon exposure, showing 50% decreases by absorption of $5.5\;mol\;photons{\cdot}m^{-2}$. Treatment of low temperature at $15^{\circ}C$ for 30 min alone did not affect the functionality of PSII, but high temperature ($45^{\circ}C$) significantly inactivated PSII activity. However, when Jeaves of pepper were subjected to low or high temperature in the presence of light, PSII was substantially photoinactivated. These results suggest the presence of different photoinhibitory mechanisms at low and high temperature.rature.

Recently Augmented Natural Habitats of Forsythia koreana (Rehder) Nakai and Abeliophyllum distichum Nakai in Korea (개나리와 미선나무의 새로운 자생지 보고)

  • This study was performed to report the augmented wild populations of Forsythia koreana (Rehder) Nakai and Abeliophyllum distichum Nakai in Korea. Forsythia koreana was found in Budong-myeon, Cheongsong-gun, Gyeongsangbuk-do. The overall size of the natural habitat of Forsythia koreana is $5m{\times}5m$. A total of 12 individuals were recorded in the wild, and 3 of them were in the seedling stage. The wild habitat is in a secondary forest, which is co-dominated by Pinus densiflora Siebold & Zucc and Robinia pseudoacacia L, with 15% coverage. The coverage of the shrub layer is 40% and is dominated by Spiraea prunifolia for. simpliciflora Nakai. The natural habitat of Abeliophyllum disitichum, which is recorded in Uisong-up, Gyeongsangbuk-do, is distributed in sites characterized by boulders along streams, with a patchwork of 15 scattered areas over a habitat size of $50m{\times}15m$. Quercus variabilis Blume is a dominant species in this habitat, with this species dominating at the sub-tree layer. Celtis aurantiaca Nakai also dominates at the shrub layer. The natural ranges of these two species in Korea were mapped based on the terms of the extent of occupancy (EoO).

Tests on Magnesium Phosphate Composite Mortar Mixtures with Different Molar Ratios of MgO-to-KH2PO4 (MgO-KH2PO4 몰비 변화에 따른 마그네시아-인산염 모르타르의 배합실험)

  • The objective of this study is to seek a reliable mixture proportion for magnesium potassium phosphate composite(MKPC) mortars with a near-neutral pH value (below 9.5) and a relatively good compressive strength exceeding 30MPa. The main parameter selected was the molar ratios($M_{mp}$) of $MgO-to-KH_2PO_4$ which varied from 30.4 to 3.4. The setting time of the MKPC mortars tended to shorten with a decrease in $M_{mp}$ value. With regard to the strength development ratio normalized by the 28-day strength, the ranges measured in the mortars with an $M_{mp}$ below 7.9 were 50~61% at 1 day and 60~73% at 3 days, indicating a highly rapid early-strength development. With a decrease in $M_{mp}$, the formation of struvite-K crystal identified as a primary hydration product increased, which led to the decrease of the macro-capillary pores in micro-structures. For achieving the targeted requirements for pH value and compressive strength, the $M_{mp}$ needs to be selected as below 5.1.

Free Radical Scavenging Activity of Butanol Fraction from the Fruit of Citrus junos

  • In this study, we investigated the free radical [1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical (${\cdot}OH$) and superoxide anion (${O_2}^-$)] scavenging activity of MeOH extract and 3 fractions of Citrus junos. Of the tested fractions, the BuOH fraction showed the strongest DPPH scavenging activity, showing the $IC_{50}$ values of 63.4 mg/mL. Therefore, we continuously carried out DPPH, ${\cdot}OH$ and ${O_2}^-$ scavenging activity tests of BuOH fraction of Citrus junos. The BuOH fraction of Citrus junos inhibited DPPH radical to 97.5% at a concentration of 1000 mg/mL and the scavenging activities were increased concentration-dependently. In addition, BuOH fraction from Citrus junos also scavenged ${\cdot}OH$ in a concentration dependent manner from 5 to 1000 mg/mL. Furthermore, BuOH fraction showed about 56% ${O_2}^-$-scavengimg activity at 25 mg/mL concentration but, the scavenging activities were not enhanced in a dose dependent manner. The present results suggest that BuOH fraction of Citrus junos would have the protective potential from oxidative stress induced by free radicals.

Antimicrobial Effects of Artemisia capillaris Extracts on the Pathogenic Bacteria In Vitro (이진쑥 추출물의 병원성 세균에 대한 시험관내 항균효과)

  • Artemisia capillaris THUNB is a perennial herb that belongs to the family Compositae spp and the most common plant among the various herbal folk remedies used in treatment of abdominal pain, hepatitis, chronic liver disease, jaundice and coughing in Korea. In this study, antimicrobial effects of Artemisia capillaris extracts on the food poisoning bacteria were investigated for further clinical application, which is an alternative for the use of antibiotics and their unexpected resistance. Artemisia capillaris extract using ethyl acetate showed the highest antimicrobial effects on S. enteritidis, E. coli O157 : H7, L. monocytogenes and S. aureus. The chloroform extract showed strong effects on all kinds of bacteria; whereas ethanol and methanol extracts showed weaker effects. Finally, ether and water extracts showed the weakest effects under the same conditions. The minimum inhibitory concentration (MIC) of ethyl acetate extract was 1 mg/mL for E. coli O157 : H7 and L. monocytogenes, and 2 mg/mL for S. enteritidis and S. aureus. The inhibitory effects on all the bacteria continued for 12 hours after incubation using 20 mg/mL and 30 mg/mL of ethyl acetate extract. The inhibitory effects continued maximally for 72 hours. The results of these studies indicate Artemisia capillaris extract exhibited excellent antimicrobial and inhibitory effects on the food poisoning pathogenic bacteria; S. enteritidis, E. coli O157 : H7, L. monocytogenes and S. aureus.

High Temperature Stress of Summer Chinese Cabbage in Alpine Region (고랭지 여름배추의 고온장해 원인 해석)

  • The objective of this study was to assess the regime of heat shock protein and leaf temperature caused by high temperature stress in chinese cabbage grown in alpine region. In monsoon period, high temperature and heavy rain have caused a stress condition for the cultured higher plants. Chinese cabbages were grown in different altitude, i.e. 600 m and 1,100 m. It was demonstrated that heat shock protein (Hsp 90) in alpine chinese cabbage leaf was actively expressed by high temperature and surplus nitrogen application. As a results of thermo-graphically observed leaf temperatures, chinese cabbage grown in high altitude region were ranged from 20.5 to $24.3^{\circ}C$ while in low altitude from 24.0 to $31.5^{\circ}C$. Furthermore, analysis of assimilated nutrients indicated that total nitrogen content was higher in plant grown under high temperature than under low temperature.
