Establishment of propagation system for in vitro calla plants(Zantedeschia spp.) by using auxin and cytokinin hormones treatments (오옥신과 사이토키닌 호르몬 처리를 이용한 칼라 기내 식물체 대량증식체계 확립)

  • There are two kinds of calla belonging to the Zantedeschia spp. depending on the growing condition. Between them, various colored calla with flower shape and flower color are very popular as cut flowers. However, colored calla is very low in natural reproduction rate, and it takes a long time to obtain a flowering bulb. So it is urgent to establish a large breeding system of good varieties. In this study, various plant growth hormone treatments were carried out to improve the growth of the plants in calla. Root formation was the highest at 81.5 % in basal MS medium, and the formation of shoot and multiple shoots were the highest at 100.0 % and 36.4 % in the combined treatment of BA 2.0 mg/L and IBA 1.0 mg/L, respectively. In fresh weight, the highest growth rate was observed with 11.2 times increase in BA 2.0 mg/L single treatment. Auxin and cytokinin mixed treatment were widely used in previous studies and positive effects on the growth of calla plants will help to establish the micro-propagation system.

Plant growth promotion effect of Arthrobacter enclensis Yangsong-1 isolated from a button mushroom bed (양송이배지로부터 분리한 Arthrobacter enclensis Yangsong-1의 식물생장촉진효과)

  • An auxin-producing bacterium Yangsong-1 was isolated from a button mushroom bed in Chung cheongnam-do. The strain Yangsong-1 was classified as a novel strain of Arthrobacter enclensis based on a chemotaxonomic and phylogenetic analysis. The isolated A. enclensis Yangsong-1 was confirmed to produce indole-3-acetic acid (IAA), which is one of the auxin hormones. When the concentration of IAA was assessed by HPLC quantity analysis, the maximum concentration of IAA, $152.903mg\;L^{-1}$, was detected from the culture broth incubated in R2A medium containing 0.2% L-tryptophan for 48 h at $35^{\circ}C$. A negative relationship between IAA production and pH was estimated to show that the increase in IAA caused pH acidification of the culture. The effect of the supplement on L-tryptophan, a known precursor of IAA production, appeared to be at maximal production at 0.2% concentration and was rather reduced at concentration above 0.4%. To investigate the growth-promoting effects on the crops, the culture broth of A. enclensis Yangsong-1 was placed in water cultures and seed pots of mung beans and lettuce. In consequence, the adventitious root induction and root growth of mung beans and lettuce were 1.5 and 1.9 times higher, respectively, than those of the control.

  • As one of the fundamental studies for the breeding of marine algae, this paper deals wit_the effects of plant hormone on the growth of microscopic filamentous gametophyte of Undaria pinnatifida. The results obtained are summarized as follows: (1) All zoospores were settled on slide glass, germinated and developed into gametophytes, without the growth of germination tubes when treated with 0.1 mg/l of 2.4-D, or 0.1, 1.0, or 5.0 mg/1 of Keinetin. (2) The best growth effect in total average was observed at 0.1 mg/1 of Keinetin, when the growth-rate was $248.9\%$ in contrast with control, and was followed by 1.0 mg/1 of IAA ($243.3\%$), and 0.05 mg/1 of 2.4-D ($205.6\%$). (3) It was certain that the growth-effect by each plant hormone had some differences between male and female gametophytes. IAA was very effective in the growth of male gameto-phytes but Keinetin in that of females. Especially in females, the efficiency of Keinetin was recorded best at $239.0\%$ at 5.0 mg/l and $222.0\%$ at 0.1 mg/1. On the other hand, it was $195.1\%$ in its best at 0.5 mg/1 of IAA, and $146.6\%$ was recorded best in 2.4-D at 0.05 mg/1.

Hormonal Effect on the Callus Induction from Perennial Weeds (다년생잡초(多年生雜草)로부터 Callus 유도(誘導)와 생장조절제(生長調節劑)의 영향(影響))

  • This experiment was conducted to evaluate effect of various hormones on callus induction, and on plantlet formation on various media, and to detect of Londax [Methyl 2-[[[[[(4,6-dimethoxy pyrimidin-2-yl) amino] carbonyl] amono] sulfonyl) methyl] benzoate] and Basta[Ammonium-(3-amino-3-carboxy-propyl)-methyl phosphinate] on callus growth and reaction of succinate dehydrogenase in callus against TTC, using various species such as Eleocharis kuroguwai, Cyperus serotinus, Oryza sativa (samgangbyeo) and Echinochloa crusgalli P. Beauv. var. caudata Kitagawa. The optimal levels of 2,4-D in MS medium seems to be different among species tested, 2.0 ppm for rice and E. crusgalli, 1.0 ppm for Eleocharis kuroguwai, and 4.0 ppm for C. serotinus derived callus from shoot-tip. In case of combination of 2,4-D with BA, 1.0 plus 0.3 ppm appeared the most appropriate level to induce callus from rice and E. kuroguwai, and I.0 plus 0.1 ppm for C. serotinus and E. crusgalli. When 2,4-D treated with TIBA, 1.0 plus 0.5 ppm appeared the most appropriate rates to induce callus derived from seeds of rice, E. crusgalli, seeds of C. serotinus and E. kuroguwai, 1.0 plus 0.3 ppm for shoot-tip of C. serotinus. Positive reaction of succinate dehydrogenase against TTC was observed regardless of calli and herbicides tested, indicating that they all are alive, and these herbicides were not able to kill the calli tested within the short period of time 20 hrs treatment. Regardless of plant species used, the rate of plantlet formation from callus was very low. However, some plantlet formed from E. crusgalli at 0.8 ppm of 2,4-D plus 8.0 ppm of kinetin, and from E. kuroguwai at 1.6 ppm of 2,4-D plus 16.0 ppm of kinetin, showing effectiveness of 2,4-D with kinetin mixture treatment. No callus was induced from C. serotinus treated with Basta from $10^{-6}M$ to $10^{-3}M$. In general, rice was the least susceptible to Basta among plant species tested, followed by E. crusgalli, and E. kuroguwai. In Londax treatment, rice showed the least inhibition rate in callus growth. Callus was induced from rice even at $10^{-3}M$ of Londax. However, $10^{-3}M$ of Londax completely inhibited callus induction from the test species. Rice showed most tolerant to both herbicides, indicating the existence of different responses among plant species.

Plant Regeneration of Bupleurum spp. through Somatic Tissue Culture (자호(紫胡)의 체세포조직배양(體細胞組織培養)에 의한 식물체재분화(植物體再分化))

  • This study was conducted to determine the optimum conditions of inducing callus, proliferating callus, forming somatic embryos, and regenerating plantlets via somatic embryogenesis, for the purpose of producing artificial seeds and substantially developing plant factory technologies that can be employed to all seasons production of Bupleurum plants. Callus was efficiently induced from leaf tissues at three leaf stage in the MS medium supplemented with 2, 4-D 2mg /1 and thidiazuron(TDZ) 0.lmg /1. Callus induction from leaf tissues at maturity was mostly effective in the mixture of 2,4- D 2mg /1 and TDZ 1.0mg /1 while that from flower bud tissues was fairly good in the MS medium containing 2,4-D 1 or 2mg /1.Callus was formed in 15 to 20 days after culture initiation in the MS media supplemented with 2, 4- D 1-2mg /1 and TDZ 0.l-1.0mg /1. Such hormones as kinetin 3mg /1, GA 1mg /1, and the mixture of GA 1mg /1 and TDZ 1mg /1 effected markedly to proliferate the callus cells.The optimum temperature and light intensity for callus culture were found to be $25^{\circ}C$ and 3000 Lux, respectively. Direct plant regeneration from cultured callus was fairly made on hormone-free MS or half-strength MS medium. Somatic embryogenesis was most frequently observed in hormone-free media:60 somatic embryos per 20ml in MS medium and 28 somatic embryos per 20ml in half -strength MS medium. There were three stages-globular, heart, and torpedo-in development of somatic embryos, among which globular stage was more frequently observed in MS medium rather than in half-strength MS medium. Somatic embryos induced from suspension culture fairly differentiated a number of shoots and roots on hormone-free and half-strength MS solid medium.

Changes in Endogenous Abscisic Acid, Jasmonic Acid and Sucrose Content during Bulb Development in the Cold-type Cultivar of Garlic (Allium sativum L.) of Korea (한지형 마늘의 인경 발육과정에서 식물내생호르몬 Abscisic Acid, Jasmonic Acid 및 당 함량변화)

  • This study was performed to investigate the role of plant growth substances on the bulbing of cold type of garlic (Allium sativum L. cv. Uiseongmaneul) during long and short day conditions. The change in endogenous plant hormones such as abscisic acid (ABA), jasmonic acid (JA) and sugar contents in leaf blade and sheath was examined during the growth stage from bulb differentiation (starting at April 16) to bulbing (April 24 to May 18) in cold type of garlic. In the long day condition, ABA contents were higher than short day condition and ABA contents of leaf sheath were higher than leaf blade. ABA contents of growth stage in garlic were not changed during growth stage from April 16 to May 2, however it rapidly increased during bulbing (May 2 to May 18). On the other hand, endogenous JA contents in short day condition did not change in long day condition, it increased from April 16 to May 2. JA contents in the leaf sheath (33.85-62.04 $ng{\cdot}g^{-1}$ DW) were higher than leaf blade (15.39-30.04 $ng{\cdot}g^{-1}$ DW). These results showed that garlic bulb differentiation and bulbing was induced by JA in leaf sheath. In long day condition, total sugar contents in the leaf blade were increased from bulb differentiation (April 16) to bulbing (May 4) and it was decreased during bulbing (May 4 to May 18) while the total sugar content in leaf sheath were gradually increased from bulb differentiation (April 16) to bulbing (May 18). In conclusion, our results showed that there is a significant correlation between the bulb development of garlic and hormonal content in the leaf sheath.

Characterization of Growth and Auxin Formation in a Bacteria Isolated from Waste bed of Agaricus bisporus (양송이 수확 후 배지로 부터 분리한 옥신생산 세균의 생육특성)

  • An auxin-producing bacteria (2SJ8-02) was isolated from waste mushroom bed of Agaricus bisporus in Buyeo-Gun, Chungnam. The strain 2SJ8-02 was classified as a novel strain of Pantoea rwandensis based on a chemotaxanomic and phylogeneticanalyses. The isolate was confirmed to produce indole-3-acetic acid (IAA), one of auxin hormones, by TLC and HPLC analyses. The maximum concentration of IAA, $122mg\;L^{-1}$ was detected from the culture in R2A broth containing 0.1% tryptophan for 24 h at $35^{\circ}C$. The molecular weight of the main peak obtained by LC-mass analysis was 175 corresponding to that of IAA. To investigate the growth-promoting effects to the crops, the culture broth of Pantoea rwandensis 2SJ8-02 was infected to water cultures and seed pots of mung bean. In consequence, the adventitious root induction and root growth of mung bean were two times higher than those of the control.

Effects of $GA_3$ and ABA Application on After-ripening of Panax quinquefolium Seeds during Stratification ($GA_3$ 및 ABA 사용이 매장처리 중 미국삼 종자의 후숙에 미치는 영향)

  • The effects of gibberilin ($GA_3$) on levels of endogenous indole-3-acetic acid (IAA) and zeatin in both fresh and stratified American ginseng (Panax quinquefolium) seeds were investigated. In our first experiment, the fresh seeds were stratified after soaked in 80 ppd $GA_3$ solution for 24 hours. We found that the IAA concentration in embryo increased by 50.7% and 82.1% respectively at the 120th day and the 188th day of stratification, and the zeatin concentration also increased by 3.8% and 51.6% respectively. In our second experiment, we treated the seeds after 134 days stratification with 80 ppm GA3 for 24 hours and then continued to stratify them. We found that the IAA concentration in embryo increased by 32.9% and 17.7% respectively at the 164th day and the 208th day of stratification while zeatin concentration increased by 22.7% and 30.6% respectively In our another experiment, we studied the effects of $GA_3$, abscislc acid (ABA) and GA, plus ABA on germination rate of seeds treated with these plant hormones during stratification. We found that when the stratified seeds whose ratio of embryo had reached 75% were treated with 80 ppm GA3 for 24 hours and then were allowed to be stratified for another 88 days, the weight and length of embryo (p < 0.05), and germination rate (p < 0.01) increased. In contrast, the 25 ppm ABA treated with for 24 hours was found to Inhibit the growth of embryo (p < 0.05) and reduce the germination rate (p < 0.05) . The experiment of combination treatment of $GA_3$ and ABA showed that $GA_3$ could relieve the inhibitory effects of the ABA on the development of the seeds.

  • Calcium ($Ca^{2+}$) plays pivotal roles as an intracellular second messenger in response to a variety of stimuli, including light, abiotic. and biotic stresses and hormones. $Ca^{2+}$ sensor is $Ca^{2+}$-binding protein known to function in transducing signals by activating specific targets and pathways. Among $Ca^{2+}$-binding proteins, calmodulin (CaM) has been well reported to regulate the activity of down-stream target proteins in plants and animals. Especially plants possess multiple CaM genes and many CaM target proteins, including unique protein kinases and transcription factors. Thus, plants are possible to perceive different signals from their surroundings and adapt to the changing environment. However, the function of most of CaM or CaM-related proteins have been remained uncharacterized and unknown. Hence, a better understanding of the function of these proteins will help in deciphering their roles in plant growth, development and response to environmental stimuli. This review focuses on $Ca^{2+}$-CaM messenger system, CaM-associated proteins and their role in responses to external stimuli of both abiotic and biotic stresses in plants.

Mycelial Growth of Ganoderma lucidum and Grifola frondosa in Milk Whey (유청을 이용한 영지버섯과 잎새버섯의 균사체 배양)

  • For the production of mycelia of mushroom by submerged culture, the experiment was carried out. mushroom. The optimum culture broth for Ganoderma lucidum NG-L were compo­sed of CMC 0.8%(w/v) and $(NH_4)_2SO_4$ 0.2%, with 10%(v/v) of inoculum size and pH 5.5 when the milk whey was used as basal medium. In case of Grifola frondosa ATCC48688, the optimum broth were composed of soluble starch 2%(w/v) and $KNO_3$ 0.l%(w/v), with 8%(v/v) of inoculum size and pH 5.2. Among several plant growth hormones,indole-3-acetic acid and gibberellin $A_3-3-acetate$ stimulated the mycelial growth of Ganoderma lucidum NG-L and Grifola frondosa ATCC 48688 respectively. The culture broth of these mushrooms inhibited the growth of B. subtilis and P. aeruginosa.
