The Environmental Impact Quotient on Fruit and Vegetables Pesticides in Korea (국내 과채류에 등록된 농약에 대한 환경영향지수)

  • The Environmental Impact Quotient(EIQ) has been used to organize the extensive toxicological and soil residue data available on some fruit vegetables pesticides into a usable form for field use. It addresses a majority of the environmental concerns that are encountered in agricultural systems including farm worker, consumer, ecological and environmental safety. The EIQ made use of the rating system by using the EIQ equation. The EIQ of pesticides registered for oriental melon was higher than that of other in terms of farm worker, consumer and environmental safety. Pesticides registered for red pepper showed highest EIQ in ecosystem. The EIQ decreased in order of insecticide> fungicide> herbicide> plant growth regulator. The environment Impact Quotient for the pesticides registered in fruit and vegetables decrease gradually.

Studies on the Propagation of Korean Tea-plant by Tissue Culture (조직배양(組織培養)에 의(依)한 국산다(國産茶)(다수(茶樹))의 증식(増殖)에 관(關)한 연구(研究))

  • In order to contribute to the Korean tea-plant culture and tea industry by means of increasing the production of tea-plants, I have performed the tissue culture of the organs of the anther, leaf and stem. As for the culture-material, I have used the anther of tea (Thea sinensis) at the tetrad uninucleate microspore stage and used medium of modified Murashige and Skoog as the basal medium supplemented with the growth regulators of NAA and 2, 4-D, yeast, kinetin and others at various concentrations. As for the handling of material, I have followed the common methods of sterilization and microtoming and paraffine imbedding method and observed systematically periodic changes of the microspores in culture. I have divided the leaf, stem and root into segments and sterilized them and used the modified Murashige and Skoog as the basal medium and observed the differentiation of roots and callus and the results are as follows. 1. In case of anther, I have found 2n callus was found in 30 out of 100 segments in M2 medium. 2. The differentiation of roots appeared in 24.5% of total leaf segments cultured and in 50.5% of stem and in 43.9% of root. 3. When the differentiation of stem in different parts was observed, the most frequent differentiation was found in the second part of all the 4 parts. 4. The most frequent formation of callus was noticed from the anther-walls in case of anther culture and from the veins in case of leaf culture. It is concluded that the seedlings of tea-plant could be multiplied most by means of tissue culture of the second part of the tea-plant stem and reduction in the expenditures of tea-plant propagation was possible through tissue culture.

Effects of Some Plant Growth Regulators on Protein Biosynthesis of Carrot Cells (당근 세포의 단백질 생합성에 대한 몇가지 식물생장조절제의 영향)

  • Electrophoretic studies of protein extracts from carrot calluses suspension-cultured on the media containing kinetin, BA, IAA, NAA or $GA_3$ at the levels of $10^{-6},\;10^{-5},\;10^{-4}M$, respectively, were performed to identify polypeptides and proteins regulated by auxin, cytokinin or GA. Fifteen bands of polypeptide(s) were observed in the callus cultured in the control medium devoid of growth regulators, and their molecular weights were $18._4,\;20._2,\;20._0,\;34._9,\;35._7,\;37._4,\;40._3,\;42._2,\;44._1,\;44._4,\;49._3,\;55._0,\;56._6,\;58._1,\;and\;59._9\;KD$, respectively. The synthesis of polypeptide appeared to be promoted in two bands by kinetin, in six bands by BA, in one band by IAA, in two bands by NAA, and in four bands by $GA_3$, while inhibited in five bands by kinetin, in three bands by BA, in four bands by IAA, in three bands by NAA and in three bands by $GA_3$. The polypeptides of $40._3\;KD\;42._2\;KD$ seemed to be regulated by cytokinins, and those of $44._1\;KD,37._4\;KD,\;and\;56._6\;KD$ by auxins. The proteins of three bands with relative mobilities of 0.56, 0.84, and 0.92, respectively, increased in the calluses cultured on the media containing kinetin, IAA, $GA_3$, NAA or BA, compared to the control, but it was difficult to identify the proteins specific for each growth regulator.

Effects of Plant Growth Regulator Treatments During the Grain Filling Period on Seed and Viviparous Germination in Rice (벼 등숙기의 생장조절제처리가 종실 및 수발아에 미치는 영향)

  • Experiments were conducted to find out the method of reducing viviparous germination of rice seeds using growth regulators during grain filling period. Among the recommended varieties, early Japonica type showed higher rate of viviparous germination, while late Japonica type and Tongil type varieties did not show viviparous germination in field condition. Grain showed high germination ability from 30 days of grain filling period in Daeseongbyeo of Japonica type and 35 days in Taebackbyeo of Tongil type. Growth regulators, NAA, CMH and Reglone were effective for reducing viviparous germination to the two rice varieties and three treatment stages in this experiment. NAA and CMH were most effective at 100 ppm and Reglone was at 0.4-0.6% concentration. Germination rates of rice seed were not different from non-treatment when NAA CMH were treated, while Reglone lowered germination rate. NAA and CMH had no effects on grain filling rate for both varieties, which showed high grain filling rate as the non-treatment, but Reglone results low grain filling rate. Grain color was not changes when NAA was applied, while CMH and Reglone made a lot of grain to dark brown as the concentration became higher. When grain were hulled, NAA had no difference with the non-treatment whereas CMH and Reglone had lower rate of perfect brown rice when they were applied at 20-30 days after heading.

Effects of Shading Rate, Rooting Media and Plant Growth Regulators on Rooting of Veronoca L. by Cuttings (삽목 번식 시 차광정도, 삽목용토 및 생장조절제 처리가 꼬리풀(Veronica L.)의 발근에 미치는 영향)

  • This study was conducted to develop a mass production for a commercial use by cutting of 4 kinds of V. glabrifolia Kitag., V. pusanensis Y.N.Lee, V. glabrifolia Kitag. × V. Spicata 'Alba' and V. spicata 'Ulster Blue Dwarf' × V. longifolia. Veronica L. Effects of shade, media and concentrations of plant growth regulators on the rooting of Veronica L. were examined. The rooting rate of V. glabrifolia Kitag. was higher as a 60% than the V. pusanensis Y.N.Lee as a 20% in the control of commercial media. In shading treatment, the rooting rate was highest at 30% or 60% shading, and the 30% shading was the best in number of root and root length, but the 90% shading was lower than no shading treatment. For cutting media, the rock wool and 100% perlite increased the rooting rate by more than 10% compared to commercial media, and increased the number of roots by 2 or 3 times. However the Cocopeat of media was lower of rooting rate, root number, and root length compared to another treatment. In the plant growth regulator treatment, the rate of rooting increased in all treatment compared to control, and was highest at IBA 1,000mg·L-1 as a 82.2% and NAA 200 or 400mg·L-1 as a 82.2% or 84.4% respectively, in the V. glabrifolia Kitag. × V. Spicata 'Alba'. However, the root number and root length was decreased as the concentration of growth regulators increased.

Development of a Simultaneous Analytical Method for Determination of Trinexapac-ethyl and Trinexapac in Agricultural Products Using LC-MS/MS (LC-MS/MS를 이용한 농산물 중 식물생장조절제 Trinexapac-ethyl과 대사산물 Trinexapac의 동시분석법 개발)

  • BACKGROUND: Trinexapac-ethyl is a plant growth regulator (PGR) that inhibits the biosynthesis of plant growth hormone (gibberellin). It is used for the prevention of lodging, increasing yields of cereals, and reducing mowing of turf. The experiment was conducted to establish a determination method for trinexapac-ethyl and its metabolites trinexapac in agricultural products using LC-MS/MS.METHODS AND RESULTS: Trinexapac-ethyl and trinexapac were extracted from agricultural products with methanol/ distilled water and the extract was partitioned with dichloromethane and then detected by LC-MS/MS. Limit of detection(LOD) was 0.003 mg/kg and limit of quantification(LOQ) was 0.01 mg/kg, respectively. Matrix matched calibration curves were linear over the calibration ranges (0.01-1.0 mg/L) for all the analytes into blank extract withr2> 0.997. For validation purposes, recovery studies were carried out at three different concentration levels (LOQ, 10LOQ, 50LOQ,n=5). Recoveries of trinexapacethyl and trinexapac were within the range of 73.6-106.9%, 72.7-99.2%, respectively. The relative standard deviations (RSDs) were less than 9.0%. All values were consistent with the criteria ranges requested in the CODEX guideline(CAC/GL 40, 2003).CONCLUSION: The proposed analytical method was accurate, effective and sensitive for trinexapac-ethyl and trinexapac determination and it can be used to as an official method in Korea.

Characterization of In Vitro Totipotency by Armoracia rusticana (서양고추냉이의 기내 전형성능에 관한 특성)

  • Plantlets were regenerated from various explants (shoot tip, leaf blade, petiole and root segments) via organogenesis and/or somatic embryogenesis from Armoracia rusticana(Lam) Gaerth., Mey, et Scherb.. Shoot regeneration rate from callus was highest on the MS mediums supplemented with 0.5 ㎎/L IAA, 5.0㎎/L BA and 10.0㎎/L spermine. A Low frequency of regeneration occurred on hormone-free MS medium. Multiple shooks were regenerated at a pH of 4.0 to 8.0 on MS medium supplemented with 1.0 ㎎/L BA and 0.1 ㎎/L NAA. Polyamines promoted shoot- and root-formation by 2 to 4 times normal, Specific proteins associated with organogenesis were identified. Somatic embryogenesis occurred directly from the leaf blade, petiole and root segments cultured on MS medium with 2.0 ㎎/L BA and 2.0 ㎎/L BA and 2.0 ㎎/L NAA. Three types of regeneration in A, rusticana were clearly established, which could be applied to the study of morphogenesis and genetics at cell, tissue and organ levels.

The Effect of Nitrogen Supply on Tomato Plants by NH4-Beaker-Deposits (토마토에 대한 NH4-Beaker Deposit 비료의 질소공급 효과)

  • Long term cultivation crops like tomato, capsicum, melon etc. demand much amount of continuous supplying of nutrition during the whole growing periods. It is not easy to cover satisfactorily the nutritional demands for them by splitted top dressings, slow release fertilizer applications and fertigation systems. To overcome these problems, the "CULTAN" (Controlled Uptake Long Term Ammonium Nutrition) Beaker Deposit techniques have been developed and it was put into PVC beaker with the combined nitrogen fertilizer type mixed with the ratio of one-third of ammonium sulfate-N and two-thirds of urea-N, in which nitrogen was loaded on the demanding amount of a tomato plant during the growing period. Gypsum was mixed as a binder, and loamy soil and compost were used as a diffusion regulator. It was placed upside down into root zone of tomato at the transplanting. Tomato roots were spreaded into the Deposit beaker by ammonium ions which attract root growth. The tomato fruit yield and nitrogen uptake by plant were increased by application of $NH_4$-Beaker deposit fertilizer rather than those of common fertilizer treatment. In conclusion, it was able to improve economic and ecological benefits through CULTAN system compared with common fertilization systems. CULTAN system was estimated as a prospective alternative to enhance productivity and minimize nutrient lose. In addition, it shows further developing possibility of CULTAN system by the supplement of micro-nutrients and pesticides in the macro-nutrient beaker deposits.

Cutting Efficiency Using Phragmites australis Culms According to Content and Timing of Indole-acetic Acid Treatment (옥신 처리 농도 및 시기에 따른 갈대 지상경 삽목 효율)

  • In this study, we tried to overcome a limit in cutting timing of reed culm by applying a plant hormone, indole-acetic acid (IAA) as a growth regulator with various contents ($10^{-3}$ M, $10^{-6}$ M, $10^{-9}$ M, $10^{-12}$ M). 19 shoots emerged from 240 segments of hardened reed culm from montane fen and eight out of the 19 shoots emerged by $10^{-6}$ M IAA treatment as the most in $5^{th}$ Sep. 2012. 50 shoots emerged from 60 segments of non-hardened reed culm from a population in Seoul National University transplanted from Mt. Odae by $10^{-6}$ M IAA treatment despite the cutting was performed about two weeks later ($19^{th}$ Sep.). Via third cutting experiment performed about 40 days later ($29^{th}$ Oct.), only two shoots out of 60 segments were observed by the same experimental condition except atmospheric temperature. It seemed likely that it was too low temperature in third experiment ($10^{\circ}C$) than the former experiments (about $20^{\circ}C$) to form adventitious buds from culm segment. We recommend to utilize the thick reed culm in culm cutting as possible because the thicker culm segment we used, the thicker emerged shoot we could observe (i.e., diameters of emerged shoots were about 20% of the planted segment's diameters).

Difference in Rooting in the Scion from Different Node of Ligusticum chuanxiong Hort (토천궁(土川芎)의 삽수(揷穗) 채가(採歌) 부위(部位)에 따른 발근차이(發根差異))

  • Scions from the one- and two-years old and the nodes of Ligusticum chuanxiong stem were used to study the rooting ability with the treatment of plant growth regulators planted to different bed soils as the alternative propagation for the present method by rhizome. The rooting ratio in the scion from the one- and the two-years old plants showed any difference, but that was ranged from 94% to 100% in the first node and from 56% to 64% in the second node from the bottom of the stem. The scion of first node from both the plants showed more than 90% in the root ratio and 16 in the number of rooting and growed more than 31cm in the root length. The Rootone-F and 250ppm of NAA were most effective for the promotion of rooting. Rooting ratios in the sand and in the mixture of vermiculite and perlite as bed soils were about 18%, but that in the cultured soil considerably decreased.
