Days to Germination and Effect of Growth Regulator on Rhizome Growth in Cymbidium goeringii Hybrid

  • Germination efficiency, such as days to germination and conditions for the hybrid seeds, was tested after hybrid seeds were sown on the medium. Seeds were germinated from 67 to 126 days after seeding in all cross combinations, and germination condition was different among hybrid combinations. There was big difference on days to germination based on the different media, that is, days to germination in the hyponex medium was shorter than that in the Knudson C medium. Mutants such as MMS and NaN$_3$ were used to cause mutation. Germination occurred with 0.1 % NaN$_3$ and MMS. However, even though germination occurred in other treatments (0.01 % and 0.2% of NaN$_3$ and MMS), brownish phenomenon was intense, or did not proceed and got worse after 4-5 months of seeding. In addition, it was performed to choose appropriate medium for the growth of Cymbidium rhizome whose media adaptability is more different than other orchids. Different concentration of NAA and Kinetin was used. As a result, C. goeringii hybrid showed difference for the concentration of NAA and Kinetin combination.

Evaluation of Fungicides for Dollar Spot Control on Creeping Bentgrass and Annual Bluegrass Putting Green (크리핑벤트그래스와 애뉴얼블루그래스 그린에서 동전마름병에 대한 살균제 효과 평가)

  • Dollar spot, caused by Sclerotinia homoeocarpa F.T. Bennett, is the most economically important turfgrass disease on golf courses in North America due to its persistent nature. Fungicides, plant growth regulators, and other products were evaluated for preventative control of dollar spot and the secondary effects on brown patch and Pythium in a mixed stand of creeping bentgrass and annual bluegrass golf course putting green. Most fungicides provided excellent control of dollar spot throughout the trial but Rhapsody alone did not provide adequate control. Civitas applied alone and tank-mixed with Banner MAXX(R) and Daconil Ultrex(R) provided acceptable control throughout the study. No phytotoxicity was observed on any of the treatments applied but a darker green color and reduction in turfgrass growth was observed in a treatment with plant growth regulator, Trimmit. Treatments that contained Civitas and the Civitas Harmonizer also exhibited green pigmentation.

Enhancement of centelloside production from cultured plants of Centella asiatica by combination of thidiazuron and methyl jasmonate

  • In order to produce centellosides from whole plant cultures of Centella asiatica (L.) Urban, we evaluated the synergistic effects of thidiazuron (TDZ) and methyl jasmonate (MJ) on whole plant growth and centelloside production. After 4 weeks of treatment with 0.025 mg/L of TDZ coupled with 0.1 mM MJ, the production of made-cassoside and asiaticoside from whole plant cultures was estimated to be 2.40- and 2.44-fold, respectively, above that of MJ elicitation alone. When whole plants were treated with a growth regulator and an elicitor, the growth of whole plants, as compared to the controls, did not differ. Additionally, total phytosyterol content in the leaves of whole plants co-treated with MJ and TDZ was 1.08-fold greater than those of MJ alone. These results demonstrate that combined treatments not only stimulate the accumulation of centellosides in the leaves but also inhibit the reduction of phytosterol levels caused by MJ elicitation.

  • In order to investigate the possibility of in vitro mass propagation via somatic embryogenesis from Sicyos angulatus L., effects of plant growth regulators and carbon sources on callus induction and somatic embryogenesis were evaluated. Optimal combinations of plant growth regulator for callus induction from cotyledon and inflorescence explants were 2,4-D 2.0 mg/L + BA 0.1 mg/L and 2,4-D 1.0 mg/L + BA 0.1 mg/L in MS basal medium supplemented with sucrose 30 g/L,, respectively. Somatic embryogenesis was observed from cultured inflorescence explants, but it could not be achieved from leaf or cotyledon explants. The most effective plant growth regulators for somatic embryogenesis from callus was NAA 1.0 mg/L + kinetin 10 mg/L in the half strength of MS basal medium supplemented with 20 g/L sucrose.

  • This experiments were carried out to determine the optimum concentrations of plant growth regulators for the direct embryogenesis from the cotyledon culture of Paeonia lactiflora Pall. Zygotic embryos rescued from true seeds were developed abnormally in the medium containing ABA. But somatic embryogenesis from cotyledons of abnormal seedlings was induced more effectively. Also the somatic embryogenesis from cotyledons was promoted in the medium containing ABA. The frequency of embryogenesis was maximum(59.9%) from the cotyledons cultured on MS medium containing 0.5 mg/L ABA on which the frequency of somatic embryos with two cotyledons was 22.6%.

  • This study was conducted to enhance the propagation of Acanthopanax divaricatus and Acanthopanax koreanum through different cutting practices. For propagation by cuttings, scions were taken on September 1. This already has hard skin and showed much higher values in terms of root length, root number, rooting ratio and root weight than those taken on June 30 and August 1. Regardless of cutting date, 50% shading resulted to the highest root length, root number, root ratio and root weight. Meanwhile, 95% shading significantly reduced these parameters compared with no shading. These results suggest that over-shading may inhibit root growth. Two growth regulators, IBA (concentration of 1,000, 2,000, 3,000 ppm) and Rootone-F (0.4%) were also tested of its effect to the cuttings. Rootone-F was found to be more effective than IBA. Cutting treated with Rootone-F had slightly higher root length and root number. A. koreanum which grows well in hot climatic condition showed better rooting ability than A. divaricatus.

Effects of Growth Regulators and Organic Nitrogen Sources on the Production of Heavy Chain Immunoglobulin G in Suspension Cultures of Transgenic Tobacco Cells

  • To enhance the production of heavy chain immunoglobulin G (HC IgG) in the suspension cultures of transgenic tobacco cells (Nicotiana tabacum), the effects of adding various cytokinins (i.e., growth regulators) and organic nitrogen sources to culture media were investigated. Four different cytokinins including kinetin, isopentenyladenine (IPA), 6-benzylaminepurine (BA), and zeatin were tested with or without dichlorophenoxyacetic acid (2,4-D), which is a typical growth regulator supplemented in the standard Murashige and Skoog (MS) medium. The productivity of intracellular HC IgG was increased by 36 and $42\%$, compared to the control, especially when IPA (2 mg/l) or BA (0.2 mg/l) was added to the media in the presence of 2,4-D, respectively. In the study of organic nitrogen sources, addition of each casein hydrolysate and tryptone to the culture media at a final concentration of 0.01 and 1 g/l, respectively. increased the productivity or he IgG as much as 68 and $67\%$, respectively, in comparison with the control, which was is MS medium without supplementation of any organic nitrogen sources. This study shows that the optimization of media composition could offer significant improvements in the production of foreign proteins in the suspension cultures of transgenic plants.

Effect of Seed Treatment on Improved Germinability of Agastache rugosa O. Kuntze (배초향 종자처리가 발아성 향상에 미치는 영향)

  • This study was carried out to establish optimal conditions for breaking dormancy of Agastache rugosa O. Kuntze seeds. A series of experiments according to seed maturity and treatment with plant growth regulators were performed to improve germination percentage and synchronize germination of the seeds. In addition, it was conducted to test whether the useful effect of seed treatment before sowing leads to healthy seedling and early vigorous growth. The average seed size was 1.85 mm (length) × 0.82 mm(width). The seed size was much smaller than other vegetable seeds. Seeds colorappeared dark brown, the shape of the seeds was oval and the weight of 1,000seeds was 352.8 mg. The optimum germination temperature was 22℃. Light exposure during germination did not affect germination promotion, suggesting that A. rugosa seeds are a kind of dark germinating seeds. Seed dormancy lasted for 40 days after harvesting, and GA3 treatment of dormant seeds could break dormancy. There were significant differences in germination percentage and rate according to the maturity of seeds. The germination percentage of mature seeds was 10 - 18% higher than that of immature seeds, and germination rate was 2 days faster. GA3 treatment during growth regulator treatment improved germinability, but BAP or ethephone treatment did not. The optimal growth regulator concentration of for germination was the combination treatment of 100 mM GA3 + 100 mM BAP.

Shoot Induction and Genetic Stability of in vitro Cultured Pea

  • Pea (Pisum sativum) is one of important legume crops in the world. It is commonly used as a protein source for animal and human diet, and also used as a natural nitrogen source which is produced by a symbiotic bacterium in their root nodule and helpful for terrestrial ecosystem. The successful in vitro manipulation is depended on three main factors including physiology of plant donor, in vitro manipulation approach, and stress physiology during plant cultivation. Moreover, genotype is an important for plant manipulation; different genotype gives the different response to regeneration efficiency. An efficient condition of shoot induction for pea (Pisum sativum cv. 'Sparkle') was developed by using optimum explant, plant growth regulator concentrations, and pretreatment of BA onto explant. The average shoot number per explant showed the highest on two kinds of shoot induction media (MSB5 media containing 2 mg/L BA and a combination of 2 mg/L BA and 1 mg/L TDZ) with cotyledonary node explants culture. Moreover, the pretreatment of explant in 200 mg/L BA solution was found to be more effective in shoot induction than that of non-pretreatment. The analysis of genetic stability of regenerants by using 13 ISSR markers presented that in vitro regenerated plants showed polymorphism with 8.3% compared with their mother plants.

Effects of Media Composition on Plant Regeneration and Callus Formation of Abeliophyllum distichum Nakai

  • This experiments were carried out to find out the effects of different explant materials, kinds and concentration of plant growth regulators, and total nitrogen and sucrose contents on the in vitro regeneration of Abeliophyllum distichum Nakai. The effects of growth regulators on regeneration from 3 explant sources (leaf, internode and node) were more or less same. Leaf explants produced only callus with 2ip (Isopentenyladenine) and NAA (Naphthaleneacetic acid) treatment and other regulators had no effects. Test with internode explants yielded about same results but callus was obtained with 2,4-D (2,4-Dichlorophenoxyacetic acid). Node explants resulted in shoot regeneration by all regulator treatment except NAA and 2,4-D, but control also showed similar results. Callus formation from internode and node explants was vigorous by 2ip, zeatin, and 2,4-D treatments and high NAA concentration resulted in higher callus formation. In this experiment, various mixed treatment of growth regulators were also employed, using node as explant material. Shoot regeneration was obtained with BA (Benzyl adenine) + NAA treatments but the results were comparable with control. Generally shoot and root regeneration was poor with all combined treatment except 2ip + NAA and 2,4-D + NAA. However, callus was formed readily with all treatments. In this experiment, combined treatments of regulators were applied on the callus derived from singular regulator treatment. The results showed no shoot and root regeneration with any combination of 2,4-D, IAA (Indoleacetic acid) and NAA, but soft milky white callus was formed in all the treatments. No shoot and root regeneration was observed with any combination of 2iP, NAA and IAA, but somewhat hard, light green callus was formed in all the treatments. Callus formation decreased with high kinetin concentration in case of kinetin + NAA treatment. The experiments with total nitrogen content of media showed that low concentrations of 15 and 30mM were effective for the shoot and root regeneration. Sucrose experiment demonstrated shoot regeneration with 1${\sim}$4% concentration, and root and callus formation with 2${\sim}$4%. No root and callus formation was observed with 0 and 1% sucrose.
