Confirmation of Genetic Homogenecity among Wild Strains of Piptoporus betulinus Collected from a Single Tree (하나의 나무에서 채집된 자작나무버섯의 균주간 유전적 동일성 확인)

  • Four strains of Piptoporus betulinus were isolated from a trunk of the Betula ermani in Seorak mountain. These strains were studied to observe whether they are originated from the same genet or not, by using the method of pairing culture between strains and by comparing part of the nuclear rDNA of ITS regions amplified with NSI1 and ITS4B primers. The demarcation line was not formed on the potato dextrose agar plates in the combinations of two different strains. Partial nuclear rDNA sequences of the four strains were also the same. As a result, it is considered in high probability that they are derived from a single spawn.

Log Cultivation of the Birch Fungus Piptoporus betulinus using Betula davurica (물박달나무를 이용한 자작나무버섯의 원목재배)

  • A birch fungus, Piptoporus betulinus, is judged the mushroom with economical value that produces the fruiting body on Betula trees, but the mushroom rarely occurs in Korean birch forest. So, the fungus was applied to log cultivation for the production of fruiting body on Betula davurica. The fungus produce the fruiting bodies in the early August of the next year after inoculation in April and its mushroom matured in October. The mushroom produced 212 g to 1,298 g fresh weight in one or two mushrooms per a log. Thus, it is considered that log cultivation of the fungus has a potential to mass production of fruiting body and is expected to introduce to more researches to develop the use of this mushroom.

Antibacterial Activity of Macromycetes Mycelia and Culture Liquid

  • The antibacterial activities of thirty mushroom species belonging to Basidiomycetes and Ascomycetes, cultivated on two liquid media, were evaluated against gram-positive (Bacillus subtilis and Staphylococcus aureus) and gram-negative (Escherichia coli) bacteria by the disk diffusion method. All of the mushrooms, except Auriporia aurea, Fomes fomentarius, and Lyophyllum shimeji, showed different antibacterial activity levels—from 9.5 mm in diameter of the inhibition zone to full inhibition of growth of the test bacteria. The antibacterial activities of Crinipellis schevczenkovi, Hohenbuehelia myxotricha, Oxyporus obducens, and Spongipellis litschaueri were observed for the first time. The antibacterial potential of culture liquids of the investigated species was higher than that of their mycelia activity. Dependence of the intensity of antibacterial activity on the culture medium was shown. The antibacterial efficiency of the most active species (Lentinus edodes, Piptoporus betulinus, and Phellinus igniarius) was verified and compared with those of some commercial antibiotics and natural essential oils of Salvia and Eucalyptus. The culture liquid of Piptoporus betulinus, obtained after cultivation on glucose-peptone-yeast culture medium, is a potential substance for further creation of antibacterial products.

Effects of Preservation Period at Low Temperature on the Mycelial Growth and the Lignocellulolytic Enzyme Activities of Basidiomycetes (저온보존기간이 담자균류의 균사생장과 목질분해효소의 활성에 미치는 영향)

  • Subculture is the most common method for preservation fungi, but has a disadvantage of accumulation of spontaneous mutations during the repeated subculture. To reduce the subculture frequency, the effect of preservation period at $4^{\circ}C$ in a slant culture was examined on the mycelia growth and lignocellulolytic enzyme activities of various basidiomycetes. Mushrooms, including Stereum ostrea, Coprinellus micaeus, Trametes versicolor, Hypholoma fasciculare, Wolfiporia extensa, Pleurotus pulmonarius, Piptoporus betulinus and Ganoderma applanatum were not affected by the preservation period more than two years, indicating that they can be maintained by subculture every two years. Some other tested fungal strains showed a significant decrease in both viability and enzyme activity when they were maintained for two years, suggesting that they should be subcultured at least once in a year. A little correlation was found between the recovery of mycelial growth and extracellular enzyme activity. In conclusion, mycelial activity and enzyme activity according to storage period is expected to be a way of deciding on subculture times for fungal preservation.
