EFFECT OF D-PROPRANOLOL ON TRANSMEMBRANE ACTION POTENTIAL OF CARDIAC PAPILLARY MUSCLE OF CAT (고양이 유두근의 활동전압에 미치는 d-propranolol의 영향)

  • Adult cats were light anesthetized with ethyl ether and heart was removed fastly. cardiac papillary muscle was dissected from heart in organ bath con taining Tyrode solution saturated with 95% O₂+5% CO₂, and prepared papillary muscles were placed in Tyrode solution that was continuously circulated and gassed with 95% O₂+5% CO₂at 32℃. The isolated papillary muscle was stimulated continuously with platinum pin electrode at frequency of 15/min and 90/min by means of electric stimulator and transmembrane action potentials were recorded with microelectrdes on the oscilloscope. The drug used was d-propranolol and its concentration was 0.5, 1.5 and 5.0 mg/L. The results obtained were as follows: 1. D-propranolol increased the threshold voltage of papillary muscle and raised by average of 213.6% of control. 2. D-propranolol had no effect on duration of action potential. 3. Conduction time of isolated papillary muscle was increased by d-propranolol and its effect was prominent at frequency of 90/min. 4. the maximum upstroke velocity was decreased by d-propranolol and its effect was dose-depndent decrease.

Physicochemical Properties of Poly-γ-glutamic Acid Produced by a Novel Bacillus subtilis HA Isolated from Cheonggukjang

  • A novel bacterium isolated from Cheonggukjang was identified as a glutamate-dependent Bacillus subtilis HA with 98.3% similarity to Bacillus subtilis Z99104. Optimization of poly-$\gamma$-glutamic acid ($\gamma$-PGA) production by modulating fermentation factors including carbon sources, nitrogen sources, inorganic salts and fermentation time was investigated. Optimum culture broth for $\gamma$-PGA production consisted of 3% glutamate, 3% glucose and various salts, resulting in the PGA production of 22.5 g/L by shaking culture for 72 hr at $37^{\circ}C$. Average molecular weight of $\gamma$-PGA was determined to be 1,220 kDa through MALLS analysis. The $\gamma$-PGA solution showed a typical pseudoplastic flow behavior, and a great decrease in consistency below pH 6.0 regardless of the same molecular weight of $\gamma$-PGA. The molecular weights of isolated $\gamma$-PGA were drastically decreased by heat treatment in various acidic conditions, resulting in different hydrolysis of $\gamma$-PGA. The consistency of $\gamma$-PGA solution was greatly decreased with increase heating time in acidic conditions.

Optimization of the Production of Fibrinolytic Enzyme from Bacillus firmus NA-1 in Fermented Soybeans

  • Bacillus strains capable of producing fibrinolytic enzyme were isolated from traditional fermented Korean soybean paste and Japanese fermented soybean (Natto). Among the 16 strains, a selected Bacillus sp. was identified as bacillus firmus, with 80.7% homology, by API kit analysis. Seed starter or B. firmus NA-1 was prepared with 5% soymilk prepared from micronized soybean powder. To produce fibrinolytic enzyme by B. firmus NA-1 the liquid culture was performed with NB broth (pH 7.0) fortified with 1% galactose, 0.1% tryptone, and 0.5% $K_2$HPO$_4$, by shaking with 180 rpm at 37$^{\circ}C$. Fibrinolytic enzyme activity reached the highest value at 7.8 unit/mL (plasmin unit) after fermentation for 72 hr. The crude fibrinolytic enzyme showed higher relative activity in the range of pH 7.0∼9.0. The activity of crude fibrinolytic enzyme was well maintained even after concentration by the vacuum evaporation at 5$0^{\circ}C$ for 1 hr.

  • Fretting is a kind of wear which effects on reliability and durability. When machine parts are joined joint in parts such as a bolt or a rivet or a pin, fretting phenomenon is occurred by micro relative movement. When fretting occurs in joint parts, there is wear which is the cause of fatigue crack. Recently, although the ways of assessment of fatigue and damage tolerance are established, there is no way to evaluate fatigue crack initiation life by fretting phenomenon. Consequently, the prediction of life and prevention plan caused by fretting are needed to improve reliability. The objective of this paper is to predict fretting wear by using a experimental method and contact analysis considering wear process. For prediction of fretting wear volume, systematic and controlled experiments with a disc-plate contact under gross slip fretting conditions were carried out. A modified Archard equation is used to calculate wear depths from the contact pressure and stroke using wear coefficients obtained from the disc-plate fretting tests.

A Study on the Current Sensor Using an Optical Modulator with BSO (BSO와 ZnSe를 광 변조기로 이용한 전류센서에 관한 연구)

  • In this paper, a magneto-optic modulator has been designed by using single crystal BSO and polycrystal ZnSe as Faraday cells. And practical core-type optical current sensors using pure iron and permalloy have been prepared and experimented. In order to obtain efficient magnetic field detection, LED(NEC OD08358, 0.87 $\mu$m) was used as optical source, PIN-PD(OD-8454)as optical receiver and multi-mode optical fiber (100/140$\mu$m) as transmission line. The characteristics matrix of the optical element was calculated by Stokes parameter, and optic modulation characteristics equations were derived by Muller matrix. Electromagnetic analysis program (FLUX 2D, micro VAX 3600) by finite element method was used to find the magnetic flux density around the core. The measuring error of the output voltage to input current has been masured below 5% in the range of 50A to 1000A. As the temperature was changed from -20$^{\circ}C$ to 60$^{\circ}C$, the maximum measurement error of the optical output has been found to be 0.5% at 60$^{\circ}C$. These experimental results show good temperature and linearity characteristics. The SNR of the overall system was 47dB in case of 600A (250.2 Oe) conductor current and the system has good noise immunity.

  • The quality of steel plate products is determined by the number of defects and the process problems are estimated by shapes of defects. Therefore pinholes defects of cold rolled steel have to be controlled. In order to improve productivity and quality of products, within each production process, the product is inspected by an adequate inspection system individually in the lines of steelworks. Among a number of inspection systems, we focus on the pinholes detection system. In this paper, we propose an embedded system using FPGA which can detect pinholes defects. The proposed system is smaller and more flexible than a traditional system based on expensive frame grabbers and PC. In order to detect consecutive defects, FPGAs acquire two dimensional image and process the image in real time by using correlation of lines. The proposed pinholes detection algorithm decreases arithmetic operations of image processing and also we designed the hardware to shorten the data path between logics due to decreasing propagation delay. The experimental results show that the proposed embedded system detects the reliable number of pinholes in real time.

Impact of Multi-dimensional Core Thermal-hydraulics on Inherent Safety of Sodium-Cooled Fast Reactor (다차원 노심열수력 현상이 소듐고속로 고유안전성에 미치는 영향)

  • A metal-fueled pool-type liquid metal fast reactor (LMFR) provides large margins to sodium boiling and fuel damage under accident conditions. The favorable passive safety results are obtained by both a reactivity feedback mechanism in the core and a passive decay heat removal system. Among the various reactivity feedbacks, the ones by a thermal expansion of a radial dimension of the core and by the control rod drivelines are strongly dependent on the flow conditions in the core and the hot pool, respectively. The effects of multidimensional thermal hydraulic characteristics on these reactivity feedbacks are investigated by the system-wide safety analysis code SSC-K with advanced thermal hydraulics models. Particularly a detailed three dimensional thermal hydraulics reactor core model is integrated into SSC-K for use in a whole system analysis of the passive safety aspects of LMR designs. The model provides fuel and cladding temperatures for every fuel pin in a reactor and coolant temperatures for every coolant sub-channel in the reactor.

  • L Company is producing the second sept pin, Shadow Mask which is the chief part of CRT in W, Monitor. Inside of the CRT pan, Shadow Mask leads the electronic beam to express the three primary colors; red, green, blue, and it is the core part to embody the colors. In the etching process, it produces this part with manufacturing by eroding the iron chloride. Even though the iron chloride is harmless to human body. it is necessary to diminish the amount of it on the view of preservation of environment. In addition, by studying the method of the dispossed liquid process, cutting down the manufacturing cost is a necessary task on the aspect of reconsideration of competitive business. This study shows the case that through the six sigma innovation activity, it reforms the following the former processing flow, and it reduces the amount of it by improving the efficiency of the iron chloride. By rationalization of the standard requirement cooperative company, it could cut down the manufacturing cost in the cooperative company, so it could promote the common profits.

  • To predict the deformation and fracture during tube expansion using the finite element (FE) method, a material model is considered that incorporates the damage evolution due to the deformation. In the current study, a Rice-Tracey model was used as the damage model with inclusion of the hydrostatic stress term. Since OFHC Cu is not significantly affected by strain rate, a Hollomon flow stress model was used. The material parameters in each model were obtained by using an optimization method. The objective function was defined as the difference between the experimental measurements and FE simulation results. The parameters were determined by minimizing the objective function. To verify the validity of the FE modeling, cross-verification was conducted through a tube expansion test. The simulation results show reasonable agreement with the experiments. The design for a minimum diameter of expansion tube using the FE modeling was verified by a simplified tube expansion test and simulation results.

Sliding Wear Characteristics of the Fe-17Mn Alloy with Various Phases (Fe-17M 합금의 상에 따른 미끄럼 마멸 거동의 변화)

  • It is reported that $\varepsilon$ (HCP) and $\gamma$ (FCC) phases of a Fe-17Mn alloy transform to $\alpha'$ phase, which has BCC structure, under a deformation condition. In this study, we investigated the effect of strain-induced-transformed $\alpha'$ phase on sliding wear of the Fe-17Mn alloy that originally had e and y phases. Wear tests of the materials were carried out using a pin-on-disk wear tester at various loads of 0.5N-50N under a constant sliding speed condition of 0.38m/s against glass $(83\%\;SiO_2)$ beads. The sliding distance and radius were loom and 9 mm, respectively. Wear rate of the Fe-17Mn alloy was calculated by dividing the weight loss, measured to the accuracy of $10^{-5}g$ by the measured specific gravity and sliding distance. Worn surface and wear debris of the specimens were examined using an SEM and XRD. During the wear, $\alpha'$ phase of BCC structure was formed by strain-induced transformation when the applied wear load exceeded critical values. The $\alpha'$ phase formed by the strain induced transformation increased the wear rate of the Fe-17Mn alloy.
