영지버섯(Ganoderma lucidum) 생장점으로부터 분리한 단백다당류의 면역활성 (Immunoactivities of the Protein-Polysaccharides of the Tips of the Growing Carpophores of Ganoderma lucidum)

  • Two protein-polysaccharide fractions, GLA and GLB, respectively, were prepared from the pileus of the fully grown carpophores and the tips of the growing carpophores of Ganod erma lucidum. At a dose of 100mg/kg/day ip, GLA and GLB inhibited the growth of sarcoma 180 solid tumor in ICR mice by 56.3% and 81.8%, respectively. In a flow cytometric (FCM) analysis, GLA and GLB enhanced the formation of lymphoblasts of BALB/c, splenic leukocytes at a concentration of 100 ${\mu}$g/ml, by 38.3% and 61, 3%, respectively. When ip injected into ICR mice, GLB exerted anti-leukopenic effect against cyclophospamide (75mg/kg, ip) in that the leukocyte counts of the peripheral blood of the normal and the cyclophosphamide-treated mice. respectively. was (11.1 ${\pm}$ 3.8) ${\times}$ 10$^3$ and (4.0 ${\pm}$ 1.8) ${\times}$ 10$^3$, while the GLB-cyclophosphamide treated mice showed a leukocyte count of (10.8 ${\pm}$ 5.1) ${\times}$ 10$^3$ CELLS/${\mu}$l. These results suggest that GLB is a promising candidate for an effective, cancer immunotherapeutic agent.

Breeding of New Strains of Mushroom by Basidiospore Chemical Mutagenesis

  • Chemical mutagenesis of basidiospores of Hypsizygus marmoreus generated new mushroom strains. The basidospores were treated with methanesulfonate methylester, an alkylating agent, to yield 400 mutant monokaryotic mycelia. Twenty fast-growing mycelia were selected and mated each other by hyphal fusion. Fifty out of the 190 matings were successful (mating rate of 26.3%), judged by the formation of clamp connections. The mutant dikaryons were cultivated to investigate their morphological and cultivation characteristics. Mutant strains No. 3 and No. 5 showed 10% and 6% increase in fruiting body production, respectively. Eight mutant strains showed delayed and reduced primordia formation, resulting in the reduced production yield with prolonged cultivation period. The number of the fruiting bodies of mutant No. 31, which displayed reduced primordial formation, was only 15, compared to the parental number of 65. Another interesting phenotype was a fruiting body with a flattened stipe and pileus. Dikaryons generated by mating with the mutant spore No. 14 produced flat fruiting bodies. Further molecular biological studies will provide details of the mechanism. This work shows that the chemical mutagenesis approach is highly utilizable in the development of mushroom strains as well as in the generation of resources for molecular genetic studies.

Cloning and Expression Analysis of Phenylalanine Ammonia-Lyase Gene in the Mycelium and Fruit Body of the Edible Mushroom Flammulina velutipes

  • Phenylalanine ammonia-lyase (PAL) gene is known to be expressed in plants, and is involved in the differentiation, growth and synthesis of secondary metabolites. However, its expression in fungi remains to be explored. To understand its expression in mushroom fungi, the PAL gene of the edible mushroom Flammulina velutipes (Fvpal) was cloned and characterized. The cloned Fvpal consists of 2,175 bp, coding for a polypeptide containing 724 amino acids and having 11 introns. The translated amino acid sequence of Fvpal shares a high identity (66%) with that of ectomycorrhizal fungus Tricholoma matsutake. Distinctively, the Fvpal expression in the mycelium was higher in minimal medium supplemented with L-tyrosine than with other aromatic amino acids. During cultivation of the mushroom on sawdust medium, Fvpal expression in the fruit body correspondingly increased as the mushroom grew. In the fruiting body, Fvpal was expressed more in the stipe than in the pileus. These results suggest that F. velutipes PAL activity differs in the different organs of the mushroom. Overall, this is first report to show that the PAL gene expression is associated with mushroom growth in fungi.

  • Di-mon 교잡에 의해 만들어진 표고품종 산림7호를 원목 재배한 결과, 1년 중 가을발생이 가장 많았으며, 생표고용으로 적합하였다. 발생된 버섯은 평반구형이며, 갓의 직경은 47mm 정도 되었다. 주름살 측면은 평활형이며, 폭은 좁았다. 버섯발 이온도는 평균 $11{\sim}23^{\circ}C$였고, 산발발생하는 경향이 있었다. 그리고 4년간 생산된 버섯량은 골목 1본당 약 1.4 kg이었다.

  • 2006년 7월25일 국립공원 계룡산 상신리 지역의 활엽수림에서 채집한 무당버섯인 R. eburneoareolata Hongo의 형태학적 특징 및 생태학적 특성을 기초로 하여 상아무당버섯으로 명명하였다. 본 종의 자실체는 중형이며, 갓은 4-9 cm의 크기이며 성숙하면 표면이 갈라져 상아색 돌기 모양이 나타난다. 주름살은 조밀하며 짙은 크림색이며 자루는 길이가 3-6 cm, 두께가 12-20mm 정도이다. 포자는 타원형이며 표면에 사마귀모양의 돌기가 가는 띠로 연결되어 분포한다. 크기는 $7-8{\times}6-7\;{\mu}m$이다. 담자기는 크기가 $35-40{\times}10-11\;{\mu}m$ 정도이며 4개의 담자포자가 달린다. 낭상체는 반원통형 또는 방망이 모양으로 끝부분이 약간 뾰족하다.

  • As a result of survey the dealings amounts of agricultural products in the public management wholesale market from 1997 to 2009 for 13 years, that makes composted of 54,706 tons in 1997, 130,345 tons in 2003 by increasing 238%, that is resulted of about 20% increase every year. As a result of survey the dealings money of agricultural products in the public management wholesale market from 1997 to 2009 for 13 years, that makes composted of 168,074 million won in 1997, 323,232 million won in 2003 by increasing 192%, that is resulted of about 16% increase every year. It has corelation with dealings amounts and money each other, also appeals to transition without discrimination, but making high amounts than money. It is divided into excellent, good and poor products according to freshness, pileus size, color and stem length to forwarding method field survey in farm produce wholesale market.

느타리버섯 신품종 육성 연구(I) -병재배용 신품종 『진미』 느타리버섯의 특성- (Characteristics of a new oyster mushroom variety 『Jinmi』 for the bottle cultivation)

  • 병재배 느타리버섯의 품종 다양화를 위해 단핵균주교잡에 의해 육성된 진미느타리버섯의 균사생장적온은 $25{\sim}30^{\circ}C$, 버섯발생 및 생육온도는 $15{\pm}1^{\circ}C$이고 갓색은 회색이며 얕은깔대기형으로 발이수가 많았다. 배양일수는 $20^{\circ}C$에서 22일, 초발이 소요일수는 4일이며 가늘고 긴형으로 농가실증시험에서 병당수량은 145.9g으로 높은 편이었다. 버섯의 갓색과 갓형태, 이형개체 발생, 균사생장량, 종균배양기간 등에 대한 균일성도 양호하였고, 세균성 갈변병에 대한 저항성은 소정도로 나타났으나 푸른곰팡이병은 발생되지 않았다. 또한 균사활력이 강하고 15 이하에서는 진한회색을 나타내며 육질이 부드럽고 대가 백색으로 깨끗한 직립형의 특성을 보유하고 있다.

신령버섯의 기형으로부터 배양불능세균의 PCR 검정 (PCR-based identification of uncultural bacterium from malformed Agaricus blazei)

  • 신령버섯은 항암작용과 더불어 면역강화작용 등 약용으로서 이용되고 있습니다. 최근에는 신령버섯 재배농가에서 새로운 병징 현상이 보고되어 내생세균에 대한 분자생물학적 방법으로 조사하였다. 병징을 나타내는 수집된 기형버섯을 eubacterial 16S rDNA 영역을 이용하여 PCR한 결과 기형버섯만 증폭되었다. 이 영역을 부분적으로 염기서열을 결정한 결과 CFB bacterium과 유사성이 가장 높았고, 이 염기서열을 이용하여 프라이머를 디자인한 후 nested PCR를 부위별로 확인한 결과 기형을 일으킨 갓의 주름살 부위에서 가장 강하게 증폭되었고 포자수확도 되지 않았으며 배양불능세균 group인 CFB bacterium임을 확인하였다.

Cultural and Morphological Characteristics of a New White Button Mushroom Cultivar 'Saedo'

  • Since development of the first Korean button mushroom cultivar 'Sae-Ah' in 2010, mushroom farmers have demanded an improved cultivar. Although a new cultivar, 'Saejeong', was developed in 2011, this cultivar has thus far been difficult to cultivate in typical mushroom farms. To solve this problem, another new cultivar, 'Saedo', was developed in 2012. This new cultivar was selected by genetic analysis from crossing the heterokaryon line A175 and the homokaryon line ASI1346-15. The heterokaryon A175 was a crossed line between strains ASI1038-211 and ASI1346-20. The mycelium of 'Saedo' showed good growth on compost dextrose agar at $25^{\circ}C$. The morphological traits such as the pileus (cap) and stipe of the 'Saedo' cultivar were thicker than those of the reference control 'Saejeong', and the production yield was increased by 1.89-fold compared to that of the reference control. A field experiment of the 'Saedo' cultivar has been ongoing since 2013. The yield of the 'Saedo' cultivar was found to be substantially higher than that of the reference control during the $2^{nd}$ flush.

  • 우수한 약리기능성을 보이는 검은비늘버섯의 병재배에 적합한 새로운 품종 진황을 교잡육종법으로 육성하였다. 진황은 포플라톱밥을 이용한 병제배와 폐면을 이용한 상자재배가 모두 가능하여 재배농가에서 생산이 용이하나, 약용 또는 기능성버섯으로서의 인지도가 낮으므로 소비 확대를 위한 약리효과의 홍보가 필요하다. 진황 검은비늘버섯의 주요특성은 다음과 같다. 가. 균사배양 최적용도는 $25^{\circ}C$ 자실체발생 및 생육온도는 $15 {\sim}18^{\circ}C$이다. 나. 자실체의 색깔은 황갈색이며 갓표면에 흰색~갈색의 인피가 덮혀 있으며 점성이 있다. 다. 병당 유효개채수가 평균 26개로 많고 대도 굵어 품질이 양호하다.
