This study was carried out to identify the causal pathogens and investigated the characteristics of Penicillium spp. isolated from postharvest decay of pear. One hundred and ninety eight Penicillium spp. were isolated from infected pear fruits. The lesions were formed when the isolated pathogen were inoculated into the wounds and unwounds of pear fruit. Total isolates were classified into 15 groups by the size, color, pigment of colony and shape of conidia. These isolates were identified to be P.expansum, P.solitum, and P.crustosum according to the types of morphological, cultural and physiological characteristics. The pathogenicity was higher in wound inoculation at low temperature than unwound one. This result confirmed that wound promoted the disease appearance. P.expansum was appeared to have the most strong virulence, whereas P.solitum and P.crustosum were classified as weak virulent species by pathogenicity test on pear fruits.

A fungal isolate was newly collected from Zoysiagrass 'Anyang-Joongji' in small circular patches on a fairway ofa golf course in Korea, which seriously occurred during the early summer period of 2005. The isolate presented on PDAmedium, named Scz1, was closely identical to Sclerotinia homoeocarpa, a casual fungus of dollar spot disease, in cool season turf grasses such as creeping bentgrass. Hereby, this study was accomplished to characterize the isolate and compare it with the fungus, named Scb1, isolated from dollar spot-infected creeping bentgrass (Agrostis palustris Huds. cv Penncross). On PDAmedium, individual mycelial appearance of three isolates was very similar except for the pigment. Mycelial pigments of Scz1 and Scz2 (another analogous isolate collected) were light pinkish on the reverse side of PDA medium but that of Scb1 was dark brownish. In a microscopic study, three isolates were barely distinguishable in the appearance of mycelia. As expected, in the temperaturesensitivity assay, all pathogens were very delicate to $32^{circ}C$ above but not to $30^{circ}C$ below, in which was explained to be one of typical characteristics in S. homoeocarpa. In an artificial inoculation assay, disease symptoms including leaf spots in Zoysiagrass were appeared within 6-7 days after inoculation through the hand inoculation method with the isolate-infested soil. Then the fungus was re-identified from the infected leaf tissues. Interestingly, inoculation of isolate Scz1 gave rise to distinct symptoms in only Zoysiagrass but not in creeping bentgrass 'Penncross' and Kentucky bluegrass 'Midnight'. The observation might be involved in host specific pathogenecity of S. homoeocarpa Scz1 to Zoysiagrass. In a chemical sensitivity assay for the isolate, Scz1, showed a high mycelial inhibition against two fungicides, iprodione and propiconazole. All results described above suggest that S. homoeocarpa Scz1 is a primary pathogen of Zoysia dollar spot disease.

Some species of the Trichoderma genus are reported as the major problem in oak wood mushroom production in Korea. In spite of economic loss by the fungi, scientific information on airborne Trichoderma species is not much available. To generate information for disease management development we analyzed airborne Trichoderma. A total of 1,063 fungal isolates were purely obtained from indoor air sampling of cultivation houses used for oak wood mushroom using sawdust media. Among the obtained isolates, 248 isolates were identified as Trichoderma fungi including T. harzianum, T. atroviride, T. citrinoviride, and T. pseudokoningii, by morphological and molecular analysis. T. harzianum was dominant among the four identified species. All the four Trichoderma species grew fast on solid nutrient media tested (potato dextrose agar [PDA], malt extract agar [MEA], Czapek's Dox + yeast extract agar [CYA] and cornmeal dextrose agar). Compact mycelia growth and mass spore production were better on PDA and CYA. In addition, T. harzianum and T. citrinoviride formed greenish and yellowish mycelium and spores on PDA and CYA. Greenish and yellowish pigment was saturated into PDA only by T. pseudokoningii. These four Trichoderma species could produce extracellular enzymes of sawdust substrate degradation such as β-glucosidase, avicelase, CM-cellulase, amylase, pectinase, xylanase, and protease. Their mycelia inhibited the growth of oak wood mushroom mycelia of two tested cultivars on dual culture assay. Among of eleven antifungal agents tested, benomyl was the best to inhibit the growth of the four Trichoderma species. Our results demonstrate that the airborne Trichoderma fungi need to be properly managed in the cultivation houses for safe mushroom production.
This study investigated the effects of high light intensity (100 KLw) and high temperature (45 ℃, dark) on enzyme (glucose-6-phosphate dehydrogenase, acid phosphatase, catalase, peroxidase, and proteinase) activities and characteristics of Panax ginseng C.A. Meyer leaves. Enzyme activity and protein content decreased rapidly under treatment with high light intensity In P ginseng the thermal stabilities of catalase and peroxidase were high (above 70%), and the coagulation rates of soluble proteins were low (below 17%). Therefore, the decrease in enzyme activity and protein content was not caused by increase in leaf temperature due to the high light intensity, but by increase in proteolytic activities. The photochemical formation rate of superoxide radical (O-2) was higher in the P ginseng leaf extracts than in Solanum nigmm, and was accelerated by addition of crude saponin to the buffer extracts.
This study was investigated and analyzed the physiological reaction mechanisms and the factors of the chlorophyll bleaching phenomenon on leaf burning-disease of the Ginseng (Panax ginseng C.A. Meyer). Chlorophyll bleaching phenomenon was mainly caused by the photooxidation of singlet oxygen and the autooxidation of hydrogen peroxide($H_2O_2$) accumulation resulted from inactivation of catalase and peroxidase. Chlorophyll bleaching phenomenon was remarkably accelerated by addition of saponin.
Endothelin-1 (ET-1) plays an indispensable role in epidermal pigmentation in hyperpigmentary disorders due to a central role in melanogenesis. Nevertheless, precise mechanism involved in ET-1-induced hyperpigmentation is still undefined. Glycoprotein (transmembrane) non-metastatic melanoma protein b (GPNMB) is a key element in melanosome formation. Therefore, we speculated that GPNMB was correlated with ET-1-induced pigmentation. After culturing with ET-1, melanin synthesis was significantly up-regulated, accompanying with increased expression of GPNMB and microphthalmia-associated transcription factor (MITF). Total number of melanosomes and melanin synthesis were sharply reduced via GPNMB-siRNA transfection, indicating ET-1-induced pigmentation by GPNMB-dependent manner. Furthermore, MITF-siRNA transfection strikingly inhibited GPNMB expression and the melanogenesis, and this suppression failed to be alleviated by ET-1 stimulation. All of these results demonstrated that ET-1 can trigger melanogenesis via the MITF-regulated GPNMB pathway. Taken together, these findings will provide a new explanation of how ET-1 induces hyperpigmentation, and possibly supply a new strategy for cosmetic studies.
A Pseudomonas fluorescens was selected from mushrooms and studied in both batch and fed-batch cultures in order to get maximal biomass concentration. P. fluorescens is an aerobic bacterium and antagonistic to Pseudomonas tolaasii which causes blotch disease on the mushroom cap. P fluarescens and P. tolaasii were identified by Gram staining, gelatin liquefaction, oxidase test, etc. and were characterized by pigment production, temperature sensitivity, salt tolerance and rapid pitting test, etc., Celts of P. fluorescens well in medium containing 30g/L of glucose, whereas the growth was inhibited at the glucose levels at higher than 30g/L. The highest values of specific growth rate and productivity were obtained when using 10g/1 of yeast extract. Optimum concentrations of $NH_4Cl$ and ${(NH_4Cl)}_2SO_4$ for culture were found to be 1.0g/L and 0.1g/L respectively. Optimum concentration of $MgSO_4{\cdot}7H_2O$ used as a sulfursource was 1.0g/L. It was also found that the cell concentrations reached the maximum level when grown on the medium containing 1.0g/L of $KH_2PO_4$ and 0.1g/L of $CaCl_2$. Also, the optimum culture conditions were $30^{\circ}C$ and pH 6.0. Cultivation of P. fluarescens at high dissolved oxygen (DO) concentration led to a decrease of bacterial productivity in batch culture. Maximum productivity was achieved at 40% DO concentration.
We have studied the mycelia growth of four soil-borne fungal pathogens under light qualities and two lighting types (continuous and intermittent) provided by light-emitting diodes (LEDs). As a result, each mycelia growth on Phytophthora cactorum KACC40166, Athelia rolfsii KACC40170, and Helicobasidium mompa KACC40836 strain showed the similar growth rates within 10% or less difference among treatments compared to dark control, regardless of lighting types. However, the mycelia growth on Rosellinia necatrix KACC40168 strain was significantly suppressed by blue, blue+green and blue+red LED as well as fluorescent lamp compared to a dark control, in common with lighting types. The melanin pigment on R. necatrix KACC40168 strain showed relatively to induce more strongly under green LED and fluorescent lamp, whereas no induction under red LED and a control, regardless of lighting types. Thus, the hypha width on R. necatrix KACC40168 was significantly thinned by blue and blue+green LED compared to a control, in common with lighting types.
Creeping bentgrass (Agrosis stolonifera L.) is regarded to be the most widely used cool-season turf grass species grown on golf greens and fairways in temperate climates of North America. Creeping bentgrass is highly susceptible to the fungal disease 'dollar spot' caused by Sclerotinia homoeocarpa. Dollar spot is a foliar disease favored by conditions of high humidity, warm days, and cool nights. Studies using Aliette Signature (Fosetyl Aluminum formulated with a green pigment) mixed with another broad-spectrum fungicide do not always provide additional visual quality benefits compared to the fungicide alone. The exact mechanism for improved summer visual quality, when it occurs, is not known. Fertility management and environment likely contribute. The object of this study is to evaluate fungicide strategies for control of dollar spot and effect on visual quality during summer and on an L-93/G-2 creeping bentgrass green. Nine fungicide combinations were used for this study. Disease control and visual quality by fungicides was evaluated on an established G-2/L-93 creeping bentgrass green at 3-hole Sunshine Golf Course in Lemont, IL. All fungicide combination showed excellent dollar spot suppression except Fore throughout the study. Visual quality of greens by addition of Aliette Signature is enhanced when bentgrass growth is compromised and slow. Dollar spot levels in Fore plots increased to 30% on 14 August, and was no different than untreated plots. Unacceptable quality by Fore was due to lack of dollar spot control.

Onion (Allium cepa L.) production in Korea has increased gradually over the past 15 years, placing second in food consumption survey with 20.6 g daily intake in 2006. Onions, used as an ingredient in many dishes and accepted by almost all traditions and cultures, have been reported to have a range of health benefits which include anticarcinogenic, antiasthmatic, antibiotic, and antioxidative effects. These effects may be attributable to a powerful flavonoid pigment-containing compounds, such as quercetin and alk(en)yl cysteine sulphoxides (ACSOs). Although antiplatelet and antithrombotic activities of onion have been confirmed by many of in vitro or animal studies, only a few human intervention studies have been examined. The majority of human studies identified that onion improves some cardiovascular markers such as lipid profile and platelet coagulant. With regard to antioxidative effects, somewhat positive effects are confirmed through strengthening the resistance of oxidative DNA damage in lymphocyte and urine, while most studies failed to find inhibitory effects on LDL oxidation. The discrepancies among studies might be ascribed to producing area, processing methods of onion, dosage, subject characteristics, study duration, and measurement methods. In this review, we focused on the preventive effect of cardiovascular disease through onion consumption in human intervention studies.
