  • Aspergillus을 이용하여 생리적 제 성질을 조사하였던 바 다음과 같은 결론을 얻었다. 1)각 균주들은 각각 그들의 특성을 가지고 있었으며 이로서 균동정의 가능성을 나타내었다. 2)Amylase측정결과에서 보면 비교적 역가가 높은 균주들이 관찰되었으며, 이는 일주간의 배양시간의 경과에 따라 역가가 증가하였으나 protease의 역가는 우수한 균주를 발견키가 어려웠다. Iodine의 착색대와 비착색대의 비율에 의한 역가의 정성적인 측정이 가능하였다.

Evaluation of Two Biologically Active Compounds for Control of Wheat Root Rot and its Causal Pathogens

  • The main aim of this study is to evaluate the efficiency of two biologically active compounds(Strom and F-760) in control of wheat root rot disease and its causal organisms. Fusarium graminearum, F. oxysporum, F. solani and Bipolaris sorokiniana were used as target organisms. In vitro, the two compounds showed fungicidal effect on all investigated pathogens resulted in suppression of radial growth and mycelial dry weight of them. Under greenhouse conditions, treatment of wheat grains with either Strom or F-760 before cultivation significantly reduced the percent of disease distribution as well as the mean disease rating of plants in both seedling and flowering stages. Fresh and dry weights of plants as well as water maintenance capacity were increased as the result of applying these compounds as seed dressing. Also data showed that the membrane stability of plants was injured as a result of infection with all investigated organisms, while this injury was alleviated when F-760 and Strom were applied. The $K^+$ efflux and the leakage of UV absorbing metabolites was stimulated with fungal infection. However, F-760 and Storm treatment partially retarded the stimulatory effect on leakage of $K^+$ and UV-absorbing metabolites of fungal infected plants. On the other side, the fungal infection had inhibitory effects on pigment fractions(chlorophyll a, b, and carotenoids) biosynthesis in wheat leaves. This retarding effect was partially or completely alleviated as the grains were treated with the applied compounds.

  • 망막색소상피(retinal pigment epithelium, RPE)는 시기능을 유지하는데 중요한 역할을 하여 RPE의 퇴화는 여러 망막변성질병을 유발한다. 현재 이에 대한 효과적인 치료법이 부족하여 세포 이식에 적합한 지지체를 제작하기 위해, 생분해성 고분자인 PLGA와 항염증, 항산화 작용 등의 기능이 있는 헤스페리딘을 이용하여 하이브리드 필름을 제조하였다. ARPE-19를 파종한 후, MTT 분석법을 이용하여 세포 증식률을 확인하고, 세포의 부착 및 세포 형태를 SEM을 통하여 확인하였다. 또한 RPE 세포의 특이적 유전자 발현정도를 확인하기 위하여 RT-PCR을 수행하였고, RPE65의 발현을 확인하기 위해 AEC 면역화학적 염색을 실시하였다. 그 결과, 헤스페리딘/PLGA 필름은 PLGA보다 RPE 세포의 부착, 증식 및 표현형 유지가 우수함을 확인하였고, 이를 통해 헤스페리딘/PLGA 필름의 망막재생을 위한 조직공학적 담체로써 응용 가능성을 확인할 수 있었다.

Crosstalk of Zn in Combination with Other Fertilizers Underpins Interactive Effects and Induces Resistance in Tomato Plant against Early Blight Disease

  • The present study was undertaken to evaluate the integrated effect of zinc (Zn) with other nutrients in managing early blight (EB) disease in tomato. A pot experiment was carried out with basal application of the recommended level of macronutrients [nitrogen, phosphorus and potassium (NPK)] and micronutrients [magnesium (Mg) and boron (B)] in bilateral combination with Zn (2.5 and 5.0 mg/kg) in a completely randomized deigned in replicates. Results revealed that interactive effect of Zn with Mg or B was often futile and in some cases synergistic. Zn with NPK yield synergistic outcome, therefore EB disease was managed significantly (disease incidence: 25% and percent severity index: 13%), which resulted in an efficient signaling network that reciprocally controls nutrient acquisition and uses with improved growth and development in a tomato plant. Thus, crosstalk and convergence of mechanisms in metabolic pathways resulted in induction of resistance in tomato plant against a pathogen which significantly improved photosynthetic pigment, total phenolics, total protein content and defense-related enzymes [superoxide dismutase (SOD), catalase (CAT), peroxidase (POX), polyphenol oxidase (PPO) and phenylalanine ammonia-lyase (PAL)]. The tremendous increase in total phenolics and PAL activity suggesting their additive effect on salicylic acid which may help the plant to systemically induce resistance against pathogen attack. It was concluded that interactive effect of Zn (5.0 mg/kg) with NPK significantly managed EB disease and showed positive effect on growth, physiological and biochemical attributes therefor use of Zn + NPK is simple and credible efforts to combat Alternaria stress in tomato plants.

피부과 및 외과 질환으로 한방안이비인후피부과에 의뢰된 한방병원 입원환자에 대한 고찰 (A Study of Korean Medicine Hospital Inpatient Consulted by Korean Medicine Ophthalmology, Otolaryngology & Dermatology Department for Dermatology and Surgical Disease)

  • Objectives : The purpose of this study is to retrospectively analyze 3 years of inpatient dermatologic and surgical consultation to Korean medicine ophthalmology, otolaryngology & dermatology department in Korean medicine hospital. Methods : We classified dermatological and surgical inpatient consultations between September 1st, 2019 and August 31st, 2022, based on electronic medical record by gender, age, hospitalized department, categories of diseases. Results : Total number of consultations were 336. The proportion of female and male were 2.5:1, and the average of age was 65.6 and 60, respectively. The most frequently referred skin disease category was infectious skin disease(39.9%), followed by eczema(20.2%), dermatoses due to physical forces(10.1%), pigment anomalies(7.1%), nevi and tumors(4.8%), diseases of the skin appendages(4.2%), urticaria(3.9%), and others(4.9%). Consultations from department of Korean rehabilitation medicine, and Korean internal medicine were most common, because they have a comparatively large number of inpatients and long hospitalization period. In case of infectious disease and skin ulcer, patients from Korean internal medicine were frequently consulted due to characteristics of stroke and internal diseases. Conclusions : This study identified the frequency of consultation and the disease group with many requests for each department. Identifying the characteristics of diseases with a high frequency of consultation will prevent various dermatological problems that may occur by department. In addition, it was found that a prompt consultation was needed for accurate diagnosis and treatment in dermatological problem.

Pink Root of Onion Caused by Pyrenochaeta terrestris (syn. Phoma terrestris)

  • Pink root of onion occurred in the fields of the Onion Experimental Station and in the main onion cultivation area in Korea in 1998 and 1999, respectively. The casual fungus of pink root was isolated only from apricot agar. Formation of pycnidia and pycnidiospores of the fungus was highest in alternating cycles of 12 hours near ultraviolet light and 12 hours in dark condition. Its morphological characteristics and pigment formation on water agar were identical with that of Pyrenochaeta terrestris. The optimum temperature for the growth of the fungus and disease development was $25-28^{\circ}C$. When onion seeds were inoculated with the spore suspension, incubated in test-tube and sown in potted soil, disease symptoms developed in onion roots 7 and 30 days after inoculation.

  • 인삼(Panax ginseng C.A. Meyer) 엽소병에 대한 고사원인을 현상학적 측면에서 관찰, 분석한 결과 chlorohphyll의 bleaching현상은 적색광(660~700nm)에서 일어났으며, 청색광(400~500nm)에서는 광산화에 의한 bleaching현상이 일어나지 않았다. 엽소병에 의한 고사원인으로 대두되었던 환경요인 "온도"는 chlorohphyll의 bleaching현상에 전혀 영향을 미치지 않았다. 따라서 단순연소(simple burning)설은 본 실험을 통해 배재되었으며, 낮은 pH는 chlorohphyll의 bleaching속도를 가속화시켜 주었다. 엽소병에서 chlorohphyll의 bleaching현상을 유발시키는 1차적요인은 인삼잎의 독특한 구조적 차이에 의한 것이었다.

Comparison of Sudden Death Syndrome in Responses to Fusarium solani f. sp. glycines between Korea and U.S. Soybean Lines

  • In order to identify the responses of Korean soybean cultivars to sudden death syndrome (SDS), forty-two Korean cultivars and three check cultivars (Hartwig and PI 520733 are resistant; Hartz 6686 is susceptible) were tested with sorghum seed inoculum infested with Fusarium solani f. sp. glycines isolate 171 in the greenhouse. This isolate has blue pigment cultural shape on potato dextrose agar (PDA) medium. All Korean cultivars inoculated with F. solani isolate 171 showed the typical SDS symptoms and disease severity on soybean leaves in each cultivar varied at 4 weeks after inoculation. Nine cultivars were included in the most SDS susceptible group and six cultivars were included in the most susceptible group based on Duncan's multiple range tests (P$\leq$0.05). In results of the LSD analysis for SDS the resistant group, a total of twenty-five Korean cultivars were included in the same SDS resistant group as PI 520733 or Hartwig and fourteen Korean cultivars were included in the same SDS susceptible group as Hartz 6686. In the second experiment, ten Korean cultivars, ten U.S. cultivars, and one introduced line were compared in the same way as the first experiment Disease severity ranking of check cultivars, Hartwig, PI 520733, and Hartz 6686, were the same as in the first experiment. Within Korean cultivars, seven cultivars showed the consistent severity proportions of leaf symptoms. Disease rankings of these cultivars in this experiment were the same as those in the first experiment. Three US cultivars: Hartwig, Hartz 5454, and Forrest, three Korean cultivars: Keunolkong, Myeongjunamulkong, and Jinpumkong 2, and one introduced line, PI 520733, were included in the highest SDS resistant group. Shinphaldalkong 2, Milyang 87, and Samnamkong consistently showed the highest SDS susceptibility in both experiments. Average disease severity in the first and the second experiment were 49.56% and 45.39%, respectively.

인삼 틸라코이드 막의 지질과 산화 (Lipid Peroxidation of Ginseng Thylakoid Membrane)

  • In order to elucidate the mechanism of the leaf-burning disease of ginseng (Panax ginseng C.A. Meyer), the relationships between thylakoid membrane peroxidation and chlorophyll bleaching were investigated in comparison with the ones of soybean (Glycine max L). When I measured the rate of lipid peroxidation in the thylakoids of ginseng and soybean by irradiation of light(60 w.m-2), it was identified that, the remarkably lower rate of lipid peroxidation was found in the ginseng thylakoid than the case of soybean. When lipid peroxidation of ginseng thylakoid was induced in the dark, chlorophyll contents of thylakoid was not changed. The results suggest that lipid peroxidation does not affect the chlorophyll bleaching in ginseng thylakoid. Thylakoid membrane peroxidation as well as chlorophyll bleaching was closely related with photosynthetic electron transport. But, according to the quenching experiment active oxygen species induced lipid peroxidation may be different species in the case of chlorophyll bleaching.

소맥근권(小麥根圈)에서 분리(分離)한 형광성(螢光性) Pseudomonas spp. 에 관(關)한 연구(硏究) (Studies on the fluorescent Pseudomoηas isolated from the wheat rhizosphere)

  • 밀의 근권(根圈)에서 병원성(病原性) 사상균(絲狀菌)에 대해 길항성(拮抗性)이 높은 균주(菌株)를 분리(分離), 동정(同定)하여 연작장해(連作障害)의 피해경감과 생물방제(生物防除)의 기초자료(基礎資料)를 삼고자 그 균이 생성(生成)하는 siderophore에 의한 사상균(絲狀菌)의 생육(生育) 저해력(沮害力)을 조사한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 밀의 근권(根圈)에서 형광성(螢光性) 세균(細菌)을 선별하여 R. solani, F. oxysperum, F. solani에 대한 생육저해(生育沮害)가 큰 (10mm이상) 균주(菌株)를 P. fluerescens Biovar I(Ps-1, Ps-2, Ps-5)과 Biovar II(Ps-3)로 동정(同定)되었다. 2. 선발균주(選拔菌株)가 생성(生成)하는 색소(色素)는 green-yellow 로 410nm에서 최대의 흡광도(吸光度)를 나타내었다. 3. Siderophore는 $Fe^{+3}$의 첨가시 생성량(生成量)은 현저히 감소하였으나 $Fe^{+2}$의 첨가시에는 생성량(生成量)의 감소는 보이지 않았다. 4. 선발균주(選拔菌株)중 Ps-5가 생성(生成)한 색소(色素)는 R. solani의 생육(生育)에 영향을 미치지 않았으며 $FeSO_4$의 처리시 생육(生育)이 증가되었다. 그러나 균주(菌株) Ps-1, Ps-2, Ps-3가 생성(生成)한 색소(色素)는 R. solani, F. oxysperum, F. solani에 대한 생육저해(生育沮害)는 컷으며 $Fe^{+2}$의 첨가시 저해력(沮害力)은 현저히 감소되었다. 5. 암소재배법(暗所栽培法)을 이용한 오이의 연작지토양(連作地土壤)에 오이를 파종하여 접종효과(接種效果)를 본 결과(結果) Ps-2>Ps-1>Ps-5>Ps-3의 순(順)으로 접종효과(接種效果)가 있었으며 Ps-1과 Ps-2는 captan 50ppm의 처리구 보다도 효과(效果)가 더 좋았다.
