닭의 대장을 돼지의 대장이나 소의 제 1위 (Rumen)와 비교 연구 (Comparison of the Chicken Large Intestine to the Large Intestine of Pigs and the Rumen of Cows)

  • 닭의 대장은 해부학적, 생리학적으로 돼지의 대장이나 소의 men과 다르다. 닭의 대장은 소의 men이나 돼지의 대장에 비교하여 잘 발달되어 있지 않다. 그러나 닭의 대장속에 있는 미생물군은 rumen 속의 것과 비슷하다. 특히 닭의 맹장속에는 많은 미생물이 있으며 이 미생물군은 나이, 사료, 성성숙의 정도, 항생제 사용 유무 등에 따라서 다양하게 나타난다. 단백질은 장내의 미생물 형성에 중요한 영양소이다. 닭에서 소장의 길이는 전장 길이의 65%를 차지하며 닭의 맹장은 8.1 %그리고 직장과 공장은 4.6 %를 차지한다. 소장 내에 분포하는 미생물 군은 현재까지 약 10%가 구명된 상태로서 나머지 90%에 대한 정보는 전무하다. 최근의 연구에 따르면 에너지, 저급지방산 (VFA)과 그리고 전해물질이 대장에서 약간씩 흡수되는 것으로 알려져 있다. 닭의 소장은 각종 효소등이 분비되어 소화가 일어나는 중추적인 역할을 하는 것으로 알려져 있다. 앞으로도 많은 연구가 과당류 즉 sucrose thermal oligosaccharide caramel (STOP), fructooligosaccharides (FOS), mannanoligosaccharide (MOS), galactooligosaccharide (GOS) 그리고 isomaltooligosaccharides (IMO)등이 맹장과 대장내에서 어떻게 소화가 일어나는지에 관해서 집중되어야 한다. 과량의 섬유질 함량은 닭의 소화에 치명적일 수 있지만 적당량을 사료에 혼합하여 급여하면 소장의 길이와 함량을 증가시킬 수 있는 효과가 있다.

5-Hydroxytryptamine의 장억제작용(腸抑制作用) (The Inhibitory Effects of 5-Hydroxytryptamine on the Intestine)

  • The inhibitory effect of 5-hydroxytryptamine (5-HT) on the isolated intestinal strips of the tortoise (Amyda japonica), rat, rabbit and guinea pig was investigated. 1) The strips from the middle or lower part of the tortoise intestine responded with relaxation to 5-HT $(10^{-9}{\sim}10^{-5}g/ml)$, and the magnitude of the relaxation was proportional to the dose of 5-HT. The rectal part of the tortoise intestine, in contrast, showed contraction, the magnitude of which also was proportional to the dose of 5-HT. 2) Various blocking agents such as methysergide, morphine, tetracaine, nethalide, bretylium, hexamethonium, mecamylamine and chlorisondamine, showed no selective blocking activity on the relaxant effect of 5-HT on the tortoise intestine. The inhibitory effect of isoproterenol on the tortoise intestine, however, was selectively blocked by nethalide, and the stimulatory effect of 5-HT on the rectal part of the tortoise was blocked by methysergide. 3) In the presence of 5-HT, the stimulatory effect of DMPP on the tortoise intestine was remarkably attenuated, whereas that of acetylcholine and $BaCl_2$ was little affected. In the presence of isoproterenol, the stimulatory effect of acetylcholine and $BaCl_2$ were affected, but that of DMPP was little affected. 4) Large dose of 5-HT($10^{-4}$g/ml) produced inhibitory effect on the strips from the distal part of the isolated colon of the rat, rabbit and guinea pig, when the strips had been exposed to 5-HT($10^{-4}$g/ml), methysergide or phen`oxybenzamine. 5) Bretylium, as well as nethalide, abolished or remarkably reduced, in a few cases of the experiments, the inhibitory effect of the large dose of 5-HT on the distal part of the colon, whereas morphine did not affect it. 6) The ileal strips of the guinea pig also showed relaxation, as in the colonic strips, having been exposed to the large dose of 5-HT or phenoxybenzamine. This effect, however, was not obsered in the case of the rabbit ileum. 7) The property of the action-site of 5-HT in the tortoise intestine seemd to be different from the 5-HT receptors which have been revealed by several investigators. 8) Adrenergic component seemed to be participated in the inhibitory effect of 5-HT on the colon of the rat and rabbit.

Physiology of Small and Large Intestine of Swine - Review -

  • The small and the large intestine of swine represent the organs that extract nutrients from feedstuffs through digestion and fermentation and that allow their absorption and incorporation into the blood circulation. Special attention is directed towards the small intestine of young pigs since the transition to a solid diet at weaning exerts major impacts on the structural and functional integrity of the small intestine. Dietary factors involved in postweaning changes of gut morphology and biochemistry such as removal of bioactive compounds in sows milk at weaning, anti-nutritional factors in weaner diets, dietary fiber and the role of voluntary feed intake will be elucidated. The microbial function of the large intestine which is carried out by a diverse population of microorganisms is dependent on substrate availability. Short chain fatty acids as main fermentation products contribute to the energy supply of the host but they are also important for the maintenance of the morphological and functional integrity of the epithelium in the colon. As a result of bacterial nitrogen assimilation in the large intestine, nitrogen is shifted from the urinary to the fecal excretion route thus saving metabolic energy to the pig because less ammonia would become available for conversion to urea.

도축돈 장관에서 분리한 Enterococcus faeciurnab 특성 (Properties of Enterococcus faecium isolated from the intestineof slaughting pigs)

  • The growth characteristics and the properties of the Enterococcus faecium (E. faecium) isolated from intestine of slaughting pig were examined in order to confirm whether it will be used practically as probiotics or not. E. faecium was identified by morphological and biochemical tests including of final confirming by API strep kit. Among 264 samples, 44 isolates (16.7%) of E. faecium were observed as an exellent growth as a range of $1{\times}10^7CFU/ml{\sim}3.3{\times}10^9CFU/ml$ in viable bacterial count on MRI medium. All isolates were shown non-haemolysis on the blood agar exception of 28 isolates shown ${\alpha}$-haemolysis and were identified as E. faecium by biochemical test using API strep enzyme kits. Eight strains (18.2%) were finally selected from which they were excellent sensitivity in showing of the acid-tolerance and the bile-tolerance in compare with reference strain.

캡슐형 내시경 개발을 위한 소장 내에서의 캡슐저항력 특성 (Tribological Characteristics of Endoscope Capsule Inside of Small Intestine)

  • In order to develop a self-propelled microendoscope, the frictional resistance of the capsule-type endoscope inside the intestine should be understood. In this work the frictional resistance behaviors of capsules with different designs were experimentally investigated using a pig intestine. It was found that cylindrical capsule design had the least frictional resistance. Also, the resistance increased as the speed of the capsule motion was decreased. It is expected that the results of this work will be used to design the optimum propulsion system for the microendoscope.

A Review of Interactions between Dietary Fiber and the Gastrointestinal Microbiota and Their Consequences on Intestinal Phosphorus Metabolism in Growing Pigs

  • Dietary fiber is an inevitable component in pig diets. In non-ruminants, it may influence many physiological processes in the gastrointestinal tract (GIT) such as transit time as well as nutrient digestion and absorption. Moreover, dietary fiber is also the main substrate of intestinal bacteria. The bacterial community structure is largely susceptible to changes in the fiber content of a pig's diet. Indeed, bacterial composition in the lower GIT will adapt to the supply of high levels of dietary fiber by increased growth of bacteria with cellulolytic, pectinolytic and hemicellulolytic activities such as Ruminococcus spp., Bacteroides spp. and Clostridium spp. Furthermore, there is growing evidence for growth promotion of beneficial bacteria, such as lactobacilli and bifidobacteria, by certain types of dietary fiber in the small intestine of pigs. Studies in rats have shown that both phosphorus (P) and calcium (Ca) play an important role in the fermentative activity and growth of the intestinal microbiota. This can be attributed to the significance of P for the bacterial cell metabolism and to the buffering functions of Ca-phosphate in intestinal digesta. Moreover, under P deficient conditions, ruminal NDF degradation as well as VFA and bacterial ATP production are reduced. Similar studies in pigs are scarce but there is some evidence that dietary fiber may influence the ileal and fecal P digestibility as well as P disappearance in the large intestine, probably due to microbial P requirement for fermentation. On the other hand, fermentation of dietary fiber may improve the availability of minerals such as P and Ca which can be subsequently absorbed and/or utilized by the microbiota of the pig's large intestine.

Bowfin-과 Shark-neuropeptide $\gamma$의 구조 및 수축효과 (Structure and Contractile Activity of the Bowfin- and Shark-neuropeptide $\gamma$)

  • Tachykinin peptide의 구조와 생리활성간의 상관관계를 조사하기 위해 고상법으로 합성한 어류 유래의 neuropeptide $\gamma$(Mammalian-, Bowfin- 그리고 Shark-neuropeptide $\gamma$)를 사용하였다. 이들의 이차 구조를 알아보기 위해 circular dichroism (CD)을 이용하였다. CD 연구 결과에 따르면, mammalian-neuropeptide $\gamma$, bowfin-neuropeptide $\gamma$와 shark-neuropeptide $\gamma$는 완충액과 인공지질막 조건하에서 random구조를 나타내었다. 또한 이들 neuropeptide $\gamma$의 장관에 대한 수축 활성을 조사하기 위해 guinea-pig의 회장, rat의 십이지장과 carp intestine을 사용하였다. Carp intestine에 서 bowfin-neuropeptide $\gamma$>shark- neuropeptide $\gamma$>mammalian-neuropeptide $\gamma$순으로 활성 이 나타났다. 그러나, guinea-pig 회장과 rat 십이지장에 대해서 mammalian-NP$\gamma$의 활성이 어류 유래의 neuropeptide $\gamma$들보다 높게 나타났다. 이러한 결과들은 neuropeptide $\gamma$가 종-특이적인 활성을 나타냄을 제시한다.

Genome-wide DNA Methylation Profiles of Small Intestine and Liver in Fast-growing and Slow-growing Weaning Piglets

  • Although growth rate is one of the main economic traits of concern in pig production, there is limited knowledge on its epigenetic regulation, such as DNA methylation. In this study, we conducted methyl-CpG binding domain protein-enriched genome sequencing (MBD-seq) to compare genome-wide DNA methylation profile of small intestine and liver tissue between fast- and slow-growing weaning piglets. The genome-wide methylation pattern between the two different growing groups showed similar proportion of CpG (regions of DNA where a cytosine nucleotide occurs next to a guanine nucleotide in the linear sequence) coverage, genomic regions, and gene regions. Differentially methylated regions and genes were also identified for downstream analysis. In canonical pathway analysis using differentially methylated genes, pathways (triacylglycerol pathway, some cell cycle related pathways, and insulin receptor signaling pathway) expected to be related to growth rate were enriched in the two organ tissues. Differentially methylated genes were also organized in gene networks related to the cellular development, growth, and carbohydrate metabolism. Even though further study is required, the result of this study may contribute to the understanding of epigenetic regulation in pig growth.

돼지분변에서 PCR에 의한 Lawsonia intracellularis 검색 (Detection of Lawsonia intracellularis in swine feces by polymerase chain reaction)

  • Swine proliferative enteritis(SPE) caused by inかsoma intracellularis is a common enteric disaese of grower and finisher pig. Swine affected with SPE show variable clinical signs including diarrhea, weight loss, aberrant growth and death. The characteristic lesion of ileitis at necropsy is marked thickening of the last section of the small intestine. The inner lining of the thickened intestine proliferates almost like a cancer and curved rod bacteria(L intracellularis) are always seen inside the intestinal wall. Infected swine shed the organism in the feces. Isolation and growth of pure L intracellularis in vitro requires a suitable cell culture. This procedure is difficult and not a practical means of diagnosis, thus the polymerase chain reaction(PCR) test of feces can be used to determine whether a pig is shedding the infective organism. A sensitive assay based on amplification of a 319bp DffA fragment of the L intracellularis of Swine proliferative enteritis was attempted for the detection of the organism in the 62 feces of swine. L intracellularis was identified on three herds and detected in 6 fecal samples, representing a infection rate of 9.7%. The PCR was very sensitive and specific on the individual level. The PCR technique could be very useful for the diagnosis of this disease.

Effects of exogenous enzymes from invertebrate gut-associated bacteria on volatile organic compound emissions and microbiota in an in vitro pig intestine continuous fermentation model

  • This study aims to assess the efficacies of exogenous enzymes, derived from invertebrate gut-associated microbes, as feed additives, in reducing volatile organic compound (VOC) emissions using an in vitro pig intestine continuous fermentation system. An in vitro continuous fermentation model was used to simulate a comparable bionic digestion system by co-reacting feed, enzymatic additives (arazyme, mannanase, and xylanase, derived from the gut bacteria of Nephila clavata, Eisenia fetida, and Moechotypa diphysis, respectively), and gastrointestinal microbes, followed by an analysis of their correlations. A significant correlation was observed between exogenous enzyme supplementation and reduced VOC emissions in the fecal phase of continuous fermentation (p < 0.05). The concentration of VOCs decreased by 3.75 and 2.75 ppm in the treatment group following arazyme and multi-enzyme supplementation, respectively, compared to that in the control group (7.83 ppm). In addition, supplementation with arazyme and multiple enzymes significantly affected the microbial composition of each fermentation phase (p < 0.05). In particular, Lactiplantibacillus pentosus and Pediococcus pentosaceus, which changed in abundance according to arazyme or multi-enzyme supplementation, exhibited a positive relationship with VOC emissions. These results suggest that exogenous enzymes derived from invertebrate gut-associated bacteria can be efficiently applied as feed additives, leading to a reduction in VOC emissions.
