Comparison of inflammatory cytokine-inducing activity of lipopolysaccharides from major periodontal bacteria

  • Porphyromonas gingivalis (Pg), Aggregatibacter actinomycetemcomitans (Aa), Tannerella forsythia (Tf), Prevotella intermedia (Pi), and Fusobacterium nucleatum (Fn) are major periodontal pathogens. Lipopolysaccharides (LPSs) from periodontal bacteria play an important role in periodontal pathogenesis by stimulating host cells to produce inflammatory cytokines. In this study, highly pure LPSs from the five major periodontopathogens were prepared, and their monocyte chemoattractant protein-1 (MCP-1) and tumor necrosis factor-α (TNF-α)-inducing activities were compared in human umbilical vein endothelial cells (HUVECs) and THP-1 macrophagic cells, respectively. In HUVECs, LPSs from Aa and Fn were potent stimulators for MCP-1 induction; however, LPSs from Pg, Pi, and Tf were much weaker MCP-1 inducers. In THP-1 cells, LPSs from Pg, Aa, and Fn were relatively strong inducers of TNF-α, whereas LPSs from Pi and Tf produced little activity. The Toll-like receptor (TLR)2/TLR4 dependency of various LPSs was also determined by measuring NF-κB reporter activity in TLR2- or TLR4-expressing 293 cells. LPSs from Aa, Fn, and Tf stimulated only TLR4; however, LPSs from Pg and Pi stimulated both TLR2 and TLR4. These results suggest that LPSs from major periodontal bacteria differ considerably in their cell-stimulating activity.

  • The conventional self-tuning methods have the speed control problem of nonlinear BLDC motor which can't adapt against any kinds of noise or operation circumstances. In this paper, supposed to solve these problem to PI parameters controller algorithm using ANN. In the proposed algorithm, the parameters of the controller were adjusted to reduce by on-line system the error of the speed of BLDC motor. In this process, EBPA NN was constituted to an output error value of a BLDC motor and conspired an input and output. The performance of the self-tuning controller is compared with that of the PI controller tuned by conventional method(Z&N). The effectiveness of the proposed control method IS verified thought the Matlab Simulink.

Synthesis and Characterization of Colorless Polyimide Nanocomposite Films Containing Pendant Trifluoromethyl Groups

  • A series of colorless polyimide (PI) nanocomposite films were synthesized from 2,2'-bis(3,4-dicarboxyphenyl)hexafluoropropane dianhydride (6FDA) and 2,2'-bis(trifluoromethyl)-4,4'-diaminobiphenyl (TFDB) with various organoclay contents by solution intercalation polymerization to poly(amic acid)s, followed by thermal imidization. The variation with the organoclay content of the thermomechanical properties, morphology, and optical transparency of the hybrids was examined at organoclay loadings ranging from 0 to 1.0 wt%. The hybrid films showed high optical transparency and almost no color, with cut-off wavelengths ranging from 352 and 356 nm and very low $b^*$ values of 1.19-1.77. The hybrid PI films showed good thermal properties with a glass transition temperature of $280-287^{\circ}C$. Most films did not show any significant thermal decomposition below $490^{\circ}C$. The addition of only a small amount of organoclay was sufficient to improve the tensile properties of the PI films with maximum enhancement being observed at 0.25 wt% organoclay. Moreover, these PI hybrids also had low coefficients of thermal expansion (CTE).

  • Transmission of substituent effects through 5-membered heteroaromaticrings isinvestigated theoretically at the RHF/6-31+G* and B3LYP/6-31+G* levelsusing the deprotonation equilibria of phenol analogues with heteroatoms Y = NH, O, PHand S. The incr ease in the resonance delocalization of the $\pilone-pair$ on the phe-nolic oxygen atom, $n\pi(O)$, accompanied with the deprotonation depends on the heteroatom Y,in the order NH < O < PH < S. This represents the $\pielectron$ accepting ability, or conversely reverse order of the $\pielectron$ donating ability of the $\pilone-pair$ on Y, $n\pi(Y).$ The transmission efficiency of substituenteffects is, however,in the reverse order NH > O > S, which represents the order of delocalizability of $n\pi(Y).$ A better correlation is obtained with ${\sigma}p$ - than with ${\sigma}p$ for the Hammett type plots with the positive slope, $\rho-$ > 0, of the magnitude in the same order as that for the delocalizability of $n\pi(Y).$ Thedeprotonation energy, ΔG = [G(PA) + G(H+)] -G(P), decreases with the increasein the extence delocalization in the order NH > O > PH > S.

Application of the Central Composite Design and Response Surface Methodology to the Treatment of Dye using Electrocoagulation/flotation Process (전기응집/부상 공정을 이용한 염료 처리에 중심합성설계와 반응표면분석법의 적용)

  • This experimental design and response surface methodology (RSM) have been applied to the investigation of the electrocoagulation/flotation of dye wastewater. The electrocoagulation/flotation reactions were mathematically described as a function of parameters current (A), NaCl concentration (B), initial RhB concentration (C) and time (D) being modeled by use of the central composite design (CCD). The application of RSM using the CCD yielded the following regression equation, which is an empirical relationship between the RhB removal (%) and test variables in RhB removal (%) = $-300.42+129.21{\cdot}Current+46.99{\cdot}NaCl-0.11{\cdot}RhB-+43.71{\cdot}Time-5.67{\cdot}Current{\cdot}NaCl-3.18{\cdot}Current{\cdot}Time-2.41{\cdot}NaCl{\cdot}Time-19.79{\cdot}Current^2-2.27{\cdot}NaCl^2-1.59{\cdot}Time^2$. the model predictions agreed well with the experimentally observed result ($R^{2}=0.9728$). The estimated ridge of maximum response and optimal conditions for RhB removal (%) using canonical analysis was 99.4% (A: 1,77 A, NaCl concentration: 2.23 g/L, RhB concentration: 56.12 mg/L, Time: 9.98 min). To confirm this optimum condition, three additional experiments were performed and RhB removal (%) were within range of 86.87% (95% PI low)~111.93% (95% PI high) obtained.

  • Generally, the method of the policy analysis has been used for making a policy decision of the development project of the road through the expert interview based on AHP. According to introducing PI, the study of the method of the integrating process for various opinion of the related groups is required. On this study I evaluate what is a difference between the results of integrated process and the AHP analysis on what is major considering factor in decision-making process. From the analysis, each evaluate factor is in level of significance by ANOVA. First, there is a significance level difference between Group A, C and Group B Second, there is also a significance level difference on the result of AHP analysis for each group in order to evaluate by integrated method. Finally, the method of AHP analysis which considers priority factor is expected as to be useful tool on decision-making of various group.

Development of a Data Acquisition System for the Long-term Monitoring of Plum (Japanese apricot) Farm Environment and Soil

  • Purpose: To continuously monitor soil and climatic properties, a data acquisition system (DAQ) was developed and tested in plum farms (Gyewol-ri and Haechang-ri, Suncheon, Korea). Methods: The DAQ consisted of a Raspberry-Pi processor, a modem, and an ADC board with multiple sensors (soil moisture content (SEN0193), soil temperature (DS18B20), climatic temperature and humidity (DHT22), and rainfall gauge (TR-525M)). In the laboratory, various tests were conducted to calibrate SEN0193 at different soil moistures, soil temperatures, depths, and bulk densities. For performance comparison of the SEN0193 sensor, two commercial moisture sensors (SMS-BTA and WT-1000B) were tested in the field. The collected field data in Raspberry-Pi were transmitted and stored on a web server database through a commercial communications wireless network. Results: In laboratory tests, it was found that the SEN0193 sensor voltage reading increased significantly with an increase in soil bulk density. A linear calibration equation was developed between voltage and soil moisture content depending on the farm soil bulk density. In field tests, the SEN0193 sensor showed linearity (R = 0.76 and 0.73) between output voltage and moisture content; however, the other two sensors showed no linearity, indicating that site-specific calibration is important for accurate sensing. In the long-term monitoring results, it was observed that the measured climate temperature was almost the same as website information. Soil temperature information was higher than the values measured by DS18B20 during spring and summer. However, the local rainfall measured using TR 525M was significantly different from the values on the website. Conclusion: Based on the test results obtained using the developed monitoring system, it is thought that the measurement of various parameters using one device would be helpful in monitoring plum growth. Field data from the local farm monitoring system can be coupled with website information from the weather station and used more efficiently.

Combination Therapy with a PI3K/mTOR Dual Inhibitor and Chloroquine Enhances Synergistic Apoptotic Cell Death in Epstein-Barr Virus-Infected Gastric Cancer Cells

  • The phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin (PI3K/AKT/mTOR) signaling pathway is a promising target for gastric cancer (GC) treatment; however the efficacy of PI3K/mTOR dual inhibitors in GC has not yet been maximized. Additionally, the effect of autophagy regulation by PI3K/mTOR dual inhibitors has not been clearly elucidated in GC treatment. We aimed to show that our newly developed PI3K/mTOR dual inhibitor, CMG002, when combined with an autophagy inhibitor, chloroquine (CQ), potently induces effective cancer cell death in Epstein-Barr virus (EBV)-associated gastric cancer (EBVaGC) cells, where both the PI3K/AKT/mTOR and autophagy pathways play important roles in disease pathogenesis. EBV- and mock-infected AGS and NUGC3 GC cell lines were treated with CMG002 +/- CQ. PI3K/AKT/mTOR signaling pathway mediators, cellular apoptosis and autophagy markers were confirmed by Western blot assay. Cell viability was assessed using the Cell Counting Kit-8 (CCK-8) assay. CMG002 effectively blocked the PI3K/AKT/mTOR pathway by markedly decreasing phosphorylation of AKT and its downstream mediator S6. CMG002 induced G0/G1 cell cycle arrest and enhanced apoptotic cell death in AGS and NUGC3 cells, particularly EBV-infected cells compared with mock-infected cells, as confirmed by flow cytometric analyses and TUNEL (terminal deoxynucleotidyl transferase dUTP nick end labeling) assays. The combination of CMG002 plus CQ synergistically increased apoptotic cell death in EBV-infected GC cell lines when compared with CMG002 alone (P < 0.05). Our results suggest that the new PI3K/mTOR dual inhibitor, CMG002, when used in combination with the autophagy inhibitor, CQ, provides enhanced therapeutic efficacy against EBVaGC.

Anticarcinogenic Effect of S-allylcysteine (SAC) (S-allylcysteine의 항암효과)

  • S-allylcysteine (SAC) is an aged garlic derived water soluble organosulfur compound and has been suggested to have anticarcinogenic activity against diverse types of cancer cells. This review summarizes the cellular signaling pathways and molecular mechanisms whereby SAC exerts its effects on cellular proliferation, apoptosis, cell cycle progression and metastasis based on the results from both in vitro and in vivo studies. SAC activates proapoptotic proteins including Bax and caspase-3, but suppresses antiapoptotic Bcl-2 family proteins to bring about cancer cell death through mitochondria-mediated intrinsic pathway. SAC also inhibits cellular proliferation by inducing cell cycle arrest in which SAC reduces expression and activation of NF-κB, cyclins, Cdks, PCNA and c-Jun, but elevates expression of cell cycle inhibitor proteins p16 and p21 through suppression of both PI3K/Akt/mTOR and MAPK/ERK signaling pathways. And, SAC inhibits invasion and metastasis of cancer cells by inducing suppression of both angiogenesis and epithelial-mesenchymal transition (EMT) through decreased cyclooxygenase (COX)-2 expression and increased E-cadherin expression which were then caused by suppression of inhibitory transcription factors Id-1 and SLUG from SAC-mediated inactivation of both MAPK/ERK and PI3K/Akt/mTOR/NF-κB signaling pathways. Furthermore, SAC prevents toxic compound-induced carcinogenesis by inducing antioxidant enzymes such as glutathione-s-transferase (GST). Thus, SAC can be considered as a potential chemotherapeutic agent for the prevention and treatment of cancer.
