Effect of Gossypol from Cottonseed Meal Diets on Some Clinico-biochemical Parameters and Humoral Immune Response of Crossbred Calves Fed Barley or Sorghum

  • Effects of feeding of 9.95 mg free gossypol/kg live weight through cottonseed meal (CSM) were studied in 20 intact male calves fed barley or sorghum as source of cereal during the experimental duration of 210 days. Serum concentration of total protein, albumin, globulin and their ratio did not vary because of protein (gossypol) or cereal sources. Serum level of cholesterol and urea were lower (p<0.05) in sorghum than barley fed calves. Feeding of gossypol through CSM enhanced (p<0.05) serum cholesterol. An interaction between protein and period was observed with respect to serum concentrations of urea, creatinine and alanine transferase. The levels of serum creatinine and alanine transferase increased (p<0.05) following 120 days of experimental feeding in calves fed CSM diets compared to the control animals fed groundnut meal diets. No effect of feeding gossypol was, however, evident on the serum enzymes viz. alanine and aspartate transferases and alkaline phosphatase. Moreover, the source of cereal and protein did not appear to influence the metabolic profile of the calves. Humoral immune response, measured through antibody titre against Brucella abortus S99 innoculation, revealed a delayed and depressed seroreactivity indicative of immunocompromisation because of the phytotoxin gossypol. In conclusion, the feeding of gossypol at the designated levels, although had no deleterious clinico-biochemical manifestations, affected the humoral immune response of the calves.

Partial Characterization of Allelopathic Substances in Sorghum Stem by Different Organic Solvents and pH (수수 줄기에 함유(含有)된 타감물질(他感物質)의 용매(溶媒)와 pH에 따른 특성구명(特性究明))

  • The toxic compounds of sorghum stem extracts were isolated by different organic solvents and pH, and characterized and quantified in terms of their inhibition of seed germination and seedling growth in Echinochloa colona(L.) Link and radish(Raphanus sativus L.). Sequential partitioning of stem extract with various organic solvents with increasing polarity showed that all fractions of hexane, ethyl ether, methylene chloride, ethyl acetate, and the aqueous remainder inhibited germination and seedling growth in E. colona. Of the five fractions, the ethyl ether fraction had the greatest inhibitory effect on E. colona. Further separation of the ethyl ether fraction at different pH(pH 2-11) showed that phytotoxic compounds were acidic. The result indicates that the phytotoxin present in the stem extract may be nonpolar and acidic.

Allelopathic Effects of Medicinal Plants on Echinoclhoa crus-galli Beauv. (약용작물의 Allelopathy 효과에 관한 연구)

  • Allelopathic activity of methanol extract from medicinal plants was determined based on the inhibition of germination, shoot and root length of Echinochloa crus-galli. The response was depended on the source of extracts and concentrations. Of the 20 test plants tested, Cnidium officinale showed most inhibition according to germination, shoot and root length of E. crus-galli. Based on the plant parts, the root extract of C. officinale had more inhibitory effect on E. crus-galli than the stem+leaf extract. Phytotoxic effects of the root extract on E. crus-galli showed 75% inhibition of seed germination, 95% inhibition of shoot growth, and 100% inhibition of root growth at the concentration of 1g/petridish. Sequential partitioning of root extract with organic solvents with increasing polarity showed that the ethylacetate fraction had the greatest inhibitory effect on E. crus-galli. Germination was not inhibited at any rate of ethylacetate fraction but shoot and root growth, were significantly inhibited even at 2mg/petridish. This indicates that the most toxic phytotoxin present in the root extract may be moderate polar compound.

  • A zearalenone derivative, methyldihydroxyzearalenone has been isolated from a plant pathogenic fungus, Drechslera portulacae which causes necrosis on the leaves and stem of Portulaca oleracea. It was characterized as 3, 4, 5, 6, 9, 10-hexahydro-8, 9, 16-trihydroxy-14-methoxy-3-methyl-1H-2-benzoxacyclotetradecine-1, 7(8H)-dione on the basis of spectral analysis. Methyldihydroxyzearalenone treatment at $3{\times}10^{-5}M$ inhibited root length of Echinochloa crus-galli and Abutilon avicennae by 35.7% and 72.6% as compared with the control, respectively.

  • Tabtoxin produced in Pseudomonas syringae pv. tabace irreversibly inhibits its known physiological target, glutamine synthetase so that causes wildfire disease on leaves of host plant. In this study, we examined a rapid and sensitive microbiological method for tabtoxin assay in several media. In minimal A agar medium nd minimal glucose agar medium, growth inhibition zone of Agrobacterium tumefaciens was larger than that of other indicator strain. However, mostly, growth inhibition zone of indicator strains on the minimal glucose agar medium was smaller than that of on the miniaml A agar medium. In complex agar medium, growth inhibithiton zone was not observed in all the tested indicator strains. Pseudomonas syringae pv. tabaci produced more tabtoxin according to the incubation time. When glutamine was added to the minimal glucose agar medium, growth inhkbition zone of Agrobacterium tumefaciens was reduced.

Characterization of Streptomyces Species Causing Potato Scab in Korea: Distribution, Taxonomy, and Pathogenicity

  • From 1996 to 1999, potato-growing areas in Korea were surveyed for identification and distribution of potato scab pathogens. Potato scab was widely distributed in the mass cultivation areas, especially in Jriu island, southern areas of Chonnam and Gyounggi provinces, and the alpine area of Gangwon province. Jeju island was the most affected area by this disease. A total of 55 Streptomyces strains were isolated from potato scab lesions, among which 40 strains were pathogenic on progeny tubers. Among the pathogenic strain, 21 strains were identified as previously described S. scabies, 7 Strains as S. turgidiscabies, and 5 Strains as S. acidiscabies, while 7 strains were observed as having distinct phenotypic properties. These strains were classified into six distinct clusters based on phenotypic characteristics and selected representative strains for each cluster. S. scabies (S33) had grey spores in a spiral chain. Mean-while, S. turgidiscabies (S27) had grey spores, S. acidiscabies (S71) had white spores, S. luridiscabiei (S63) had yellow-white spores, S. puniciscabiei (S77) had purple-red spores, and S. niveiscabiei (S78) had thin and compact white spores, all in a rectiflexuous chain. Pathogenicity was determined by the production of thaxtomin A and homologs of necl and ORFtnp genes. In TLC, representative strains S27, S71, S63, S77, and S78 produced a yellow band that co-migrated with the authentic thaxtomin A. However, thaxtomin A was not detected in chloroform extracts from oatmeal broth culture and Slice tuber tissue of S. luridiscabiei (S63) and S. puniciscabiei (S77) by HPLC analysis. In addition, no homologs of necl and ORFtnp genes in S. acidiscabies (S71), S. luridiscabiei (S63), S. puniciscabiei (S77), and S. niveiscabiei (S78) were detected by PCR and Southern hybridization analysis.

  • A laboratory experiment was conducted to find out the abilities of siderophore formation and competition of $Fe^{3+}$ and $Mn^{2+}$ absorption in synthetic medium with inoculation of fluorescence forming pseudomonas and soil saprophytic fungi as Stachybotrys chatarum, Fusarium solani, and F. oxysporum. The results obtained were summarized as follows; 1. The concentration of siderophore in synthetic medium with inoculation of Pseudomonas putida pt-II was increased (with progress of incubation time). 2. The negative correlation was obtained with the increase of $Fe^{3+}$ ion concentration and siderophore in synthetic medium with inoculation of fluorescence forming pseudomonas and soil saprophytic fungi. 3. The fresh weight of fungal hyphae was decreased with the increase of siderophore in synthetic medium. 4. There was insignificant relationship between the concentration of $Mn^{2+}$ and the concentration of siderophore while the positive correlation was obtained with the increase of fresh weight of fungal hyphae.

Phytotoxicity of Endophytic Fungi and Characterization of a Phytotoxin Isolated from Gliocladium catenulatum from Pinus densiflora (식물내생곰팡이의 제초활성 검정 및 소나무에서 분리한 Gliocladium catenulatum이 생산하는 제초활성 물질의 특성 규명)

  • This study was conducted to discover new phytotoxins which may be used as lead molecules for the development of new herbicides. A total of 187 endophytic fungi were isolated from 11 plant species, which were collected from 8 locations in Korea. Their herbicidal activities were screened in vivo by herbicidal and duckweed bioassays after they were cultured in potato dextrose broth and rice solid media. Both fermentation broth and solid culture extract of Gliocladium catenulatum F0006 isolated from red pine (Pinus densiflora) showed 70% herbicidal activity only against cocklebur (Xanthium strumarium) out of the 10 weeds tested. Solid culture extract of F0034 isolated from arrowroot (Pueraria thunbergiana) exhibited 20 to 100% herbicidal activities against all of the weeds. Especially, shattercane (Sorghum bicolor), barnyardgrass (Echinochloa crus-galli), large crabgrass (Digitaria sanguinalis), and fall pauicum (Panicum dichtomiflorum) were sensitive to the solid culture extract of F0034. In addition, solid culture extract of F0043 isolated from red pine displayed 20% to 70% herbicidal activities only against 5 grass species, but not against 5 broad-leaf plant species. On the other hand, as the results of duckweed assay, 8 fermentation broths showed 100% growth inhibitory activity at concentrations less than 5.0% of culture supernatants and 12 solid cultures had a potent inhibitory activity against duckweed growth. A toxic metabolite was purified from the solid cultures of G. catenulatum F0006 by repeated column chromatography and bioassay. It caused a phytotoxic syndrome only on cocklebur out of the 10 weeds tested; it completely killed cocklebur seedlings at $500\;{\mu}g/ml$ and showed 85% herbicidal activity against cocklebur at $100\;{\mu}g/ml$. The molecular weight of the toxic metabolite is 238 daltons and its structure determination is underway.
