${\beta}$-sitosterol, one of phytosterols, exhibited numerous pharmacological effect including anti-inflammatory, anti-cancer and immune-modulating properties. This study attempted to determine the pharmacological effects of ${\beta}$-sitosterol on atopic dermatitis (AD). We investigated to ascertain the pharmacological effects of ${\beta}$-sitosterol on 2, 4-dinitrochlrobenzene (DNCB)-induced AD symptom and histamine-induced scratching behaviors in mice. Additionally, we evaluated the effects of ${\beta}$-sitosterol on the interleukin (IL)-6 levels in HaCaT cells and skin tissue of AD. The findings of this study demonstrated that ${\beta}$-sitosterol reduced AD clinical symptoms such as eczematous, erythema and dryness and serum histamine and IgE levels in DNCB-induced AD model and histamine-induced scratching behaviors in mice. Additionally, ${\beta}$-sitosterol inhibited the IL-6 expression in AD-like skin lesion and HaCaT cells. Collectively, these findings provide that ${\beta}$-sitosterol could be a therapeutic agent for skin inflammation including AD.
The scope of the research is investigation of immune-modulating activities of A. rugosa (Baechohyang) extract was preformed through the screening active constituents using in vitro assays and evaluating anti-inflammatory activity and anti-atherosclerotic activity of the extract and active compound (tilianin) in vivo. In addition, various functional foods using the extract and whole plant was developed. The extract showed strong anti-inflammatory activity in carrageenan-induced acute edema mouse model and anti-atherogenic lesion activity in LDLR (low density lipoprotein receptor) deficient mouse model. These activities were thought to be resulted from modulation activity of several pathways of inflammation process. Among the main constituents of Baechohyang, polyunsaturated fatty acids (PUFA), Phytosterols, oleanolic acid and rosmarinic acid showed anticomplement activity, and PUFA, acacetin and tilianin newly showed potent ICAM-1 expression inhibition activity. The processes of extraction, mixing ratio of additives and storage conditions were established for drinks, granule tea, leaf tea, mixed tea and furigake.
Lipid-based functional components present in foods undergo chemical changes during cooking. Useful n-3 and n-6 fatty acids, phytosterols, tocopherols, and carotenoids are degraded by thermal cooking such as frying, resulting in loss of their physiological functions. However, conjugated linoleic acid and diacylglycerols can be formed during heating, which would be beneficial to the health. Degree of degradation and formation of the functional components depends on the cooking method, cooking temperature and time, lipid matrix containing the components, and the presence of other materials. Cholesterol is a vital component of the human body. It stabilizes cell membranes and is the precursor of bile acids, vitamin D and steroid hormones. However, cholesterol accumulation in the bloodstream (hypercholesterolemia) can cause atherosclerotic plaques within artery walls, leading to heart attacks and strokes. The efficiency of cholesterol absorption in the small intestine is of great interest because human and animal studies have linked cholesterol absorption with plasma concentration of total and low density lipoprotein cholesterol. Cholesterol absorption is highly regulated and influenced by particular compounds in the food supply. Therefore, it is desirable to learn more about natural food components that inhibit cholesterol absorption so that food ingredients and dietary supplements can be developed for consumers who wish to manage their plasma cholesterol levels by non-pharmacological means. Food components thus far identified as inhibitors of cholesterol absorption include phytosterols, soluble fibers, phospholipids, and stearic acid.
To clarify chemical constituents of Clematis heracleifolia, isolation and structure elucidation of the underground parts of C. heracleifolia were performed. Five compounds were isolated from $CHCl_3$ and n-BuOH soluble fraction of this plant. On the basis of spectral and physico-chemical data, the structure of isolated compounds were identified as coniferyl alcohol (1), scoparone (2), (+)-lariciresinol (3), phytosterols (4), and daucosterol (5), respectively. All compounds are isolated from this plant for the first time. To evaluate anti-acetylcholinesterase activity of the isolated compounds, compounds 1, 2, 3, and 5 were tested inhibition activity against acetylcholinesterase. Among tested compounds, daucosterol (5) showed acetylcholinesterase inhibitory activity with $IC_{50}$ value of $6.1{\mu}M$.
Three coumarins, one chromone, and a phytosterol were isolated from the stem of Ostericum koreanum. Basis on the spectral data, we determined to be the structures of isolated compounds as bergapten, xanthotoxin, auraptenol, hamaudol, and mixture of phytosterols. Auraptenol was isolated for the first time from this plant.
The rate of growth and production of bioactive substances from Rehmannia glutinosa Liboschitz (Scrophulariaceae) were studied with the variation on the constituents of the culture media. The best growth was observed from MS basal medium containing 3.0 ppm NAA and 2.0 ppm kinetin. Carbohydrates (fructose, glucose and sucrose), phytosterols(${\beta}-sitosterol$, campesterol and stigmasterol) and carotenoid like substances were identified by GC-MS and TLC from the callus mass. However, catalpol was not detected from both solid and cell suspension cultures containing geraniol. Callus cultured Rehmannia glutinosa in the MS basal medium containing 0.1 ppm NAA and 0.1 ppm kinetin become differentiated to root.
Saururus chinensis has been reported to contain compounds such as lignans, alkaloids, diterpenes, flavonoids, tannins, steroids, and lipids. Fermentation is commonly used to break down certain undesirable compounds, to induce effective microbial conversion, and to improve the potential nutraceutical values. Previous studies have reported that the fermentation process could modify naturally occurring constituents, including isoflavons, saponins, phytosterols, and phenols, and could enhance biological activities, specifically antioxidant and antimicrobial properties. The probiotic strains used for fermentation exert beneficial effects and are safe. In this study, the antioxidative effects of the Bacillus subtilis fermentation of Saururus chinensis were investigated in a rat model with Triton WR-1339-induced hyperlipidemia by comparing the measured antioxidative biological parameters of fermented Saururus chinensis extract to those of nonfermented Saururus chinensis extract. Fermentation played a more excellent role than nonfermentation in ultimately protecting the body from oxidative stress in the liver of the experimental rats with Triton WR-1339-induced hyperlipidemia.
미강유(米糠油)의 부(不)검화물(化物)을 분석(分析)하여 sterol조성(組成)을 살펴본 결과(結果)는 다음과 같았다. 4-Desmethylsterol로서는 10개가 동정(同定)되었으며 이 중(中) sitosterol이 주성분(主成分)을 이루고 있었다. 2개의 소량성분(少量成分)인 ${\Delta}^7$-stigmastenol(3%)과 ${\Delta}^7$-avenasterol(1%)를 제외(除外)하면 모두가 ${\Delta}^5$-sterol였으며 소위(所謂) ${\Delta}^5$형(型) sterol분포(分布)를 보이고 있었다. 4-monomethylsterol fraction에서는 9개가 확인(確認)되었으나 cycloeucalenol, citrostadienol, gramisterol가 주성분(主成分)이었다. 환계(環系)의 구조가 다양(多樣)하여 sterol의 생합성경로(生合成經路)의 중간단계(中間段階)임을 보여주고 있다. 4,4-Dimethylsterol(triterpene alcohol)로서는 4개가 동정(同定)되었으나 이들은 모두 9,19-cyclopropanering을 가지고 있다. 이들중(中) sterol생합성(生合成)의 초기단계(初期段階)의 물질(物質)인 cycloartenol와 24-methylenecyclo-artenol가 이 fraction의 96%를 점(占)하고 있는 것으로 봐서 미(米)강의 4,4-dimethylsterol는 짧은 경로(經路)를 거쳐 쉽게 4-monomethylsterol로 전환(轉換)되는 것 같다. Side chain에서 E-, Z-관계(關係)인 이성체(異性體)가 4-desmethylsterol로서는 fucosterol와 ${\Delta}^5$-avenasterol가 그리고 4-monomethylsterol로서는 28-isocitrostadienol와 citrostadienol로서 각각 1쌍식이 검출(檢出)된 것으로 미루어 볼때 sterol의 생합성과정(生合成過程)에서 side chain에서 보다 환계(環系)에서 먼저 변환(變換)이 일어나는 것으로 보아진다.
