A Study on the Inactivation of Phytophthora Blight Pathogen (Phytophthora capsici) using Plasma Process (플라즈마 공정을 이용한 고추역병균(Phytophthora capsici) 불활성화에 관한 연구)

  • Plasma reactor was used for the inactivation of Phytophthora capsici which is phytophthora blight pathogen in aquiculture. Effects of first voltage, second voltage, air flow rate, pH, incubation water concentration were examined. At the low $1^{st}$ voltage, under 80 V, the lag phase was noticed within 30 sec, however, it was not shown over 100 V. The variation of optimum operation condition was not shown by the variation of microorganisms. However, the inactivation rate was different by the variation of species of microorganisms. The inactivation rate and efficiency were increased by the increase of $2^{nd}$ voltage. The highest initial inactivation rate was shown at pH 3 and the rate was decreased by the increase of pH. The inactivation rate increased by the increase of air flow rate, however, it was shown as similar at the rate of 4 L/min and 5 L/min. The inactivation rate was distinctly decreased at the three times concentration of incubation solution comparing at the distilled water and basic incubation solution.

First Report of Phytophthora Leaf Blight and Vine Rot of Kudzu (Pueraria lobata) in Korea

  • A disease causing leaf blight and vine rot was recognized on kudzu plants (Pueraria lobata) in Korea since 1991. A species of Phytophthora has been repeatedly isolated from the infected leaves. Identification in species level of the Phytophthora sp. remained unsolved. An isolate, KACC 47616 originally collected from Manchon Park in Daegu, has been kept in our laboratory. In 2013, three new isolates, KACC 47617 and KACC 47618 from Yeongyang and KACC 47619 from Gunwi in Gyeongbuk province, were collected and examined to classify up to species level by characterizing morphology, response to temperature and phylogenetic relationship. On the basis of morphological characters such as the nature of hyphal swelling, sporangia and sex organs, absence of chlamydospore production, optimum temperature for mycelial growth, and internal transcribed spacer rDNA and cytochrome oxidase subunit 1 sequence analysis of the pathogen, the causal fungus of kudzu plant was identified as Phytophthora asiatica.

  • This study was conducted in order to find out biological control of sesame wilt and blight caused by Fusarium of oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica by using Streptomyces spp. Two sesame pathogens, Fusarium oxysporum f. sp. vasinfectum and Phytophthora nicotianae var. parasitica were purely isolated from diseased sesame plants of the field. Streptomyces species were isolated from 72 soil samples collected from red pepper and sesame uplands in Chungbuk and selected as antagonists according to the results of dual culture. The selected Streptomyces isolates such as St-11 and St-20 were confirmed their antagonistic effect through mycelial inhibition zone and inhibitory effects on the mycelial growth of the pathogens by culture filterate of the antagonists. Inhibitory effects on the conidial germination of Fusarium oxysporum vasinfectum and Phytophthora nicotianae parasitica by the antagonists were also tested in addition to mycelial Iysis. The antagonists St-11 and St-20 showed inhibitory effect on growth of sesame seedlings after seeds soaked in the suspension. Effect of soil inoculation with antagonist St-11 showed 40 to 78 percent of control effect for two diseases in comparison with control under greenhouse.

Foliage Blight of Vinca (Catharanthus roseus) by Phytophthora nicotianae (Phytophthora nicotianae에 의한 일일초 역병)

  • A disease causing blights on leaves, stems, flowers, and pods of Vinca (Catharanthus roseus L.) was occurred in Aug. 2003 in Chengdo Peach Experiment Station. A species of Phytophthora was isolated from the diseased tissues. The causal fungus was identified as Phytophthora nicotianae on the basis of mycological characteristics and pathogenicity. Sporangia were ovoid to spherical, noncaducous, prominently papillate and averaged 38.0${\times}$31.0 ${\mu}{\textrm}{m}$ in dimension. Chlamydospores were abundantly produced on agar media and sized about 30.5 fm in diameter, The fungus was heterothallic and Al mating type. Oospores were measured 23.1 ${\mu}{\textrm}{m}$ in size. Optimum temperature for growth of the fungus was 25 to 3$0^{\circ}C$. This is the first report of occurrence of foliage blight of Vinca caused by P. nicotianae in Korea.

Biocontrol of Phytophthora Blight and Anthracnose in Pepper by Sequentially Selected Antagonistic Rhizobacteria against Phytophthora capsici

  • We previously developed a sequential screening procedure to select antagonistic bacterial strains against Phytophthora capsici in pepper plants. In this study, we used a modified screening procedure to select effective biocontrol strains against P. capsici; we evaluated the effect of selected strains on Phytophthora blight and anthracnose occurrence and fruit yield in pepper plants under field and plastic house conditions from 2007 to 2009. We selected four potential biocontrol strains (Pseudomonas otitidis YJR27, P. putida YJR92, Tsukamurella tyrosinosolvens YJR102, and Novosphingobium capsulatum YJR107) among 239 bacterial strains. In the 3-year field tests, all the selected strains significantly (P < 0.05) reduced Phytophthora blight without influencing rhizosphere microbial populations; they showed similar or better levels of disease suppressions than in metalaxyl treatment in the 2007 and 2009 tests, but not in the 2008 test. In the 2-year plastic house tests, all the selected strains significantly (P < 0.05) reduced anthracnose incidence in at least one of the test years, but their biocontrol activities were variable. In addition, strains YJR27, YJR92, and YJR102, in certain harvests, increased pepper fruit numbers in field tests and red fruit weights in plastic house tests. Taken together, these results indicate that the screening procedure is rapid and reliable for the selection of potential biocontrol strains against P. capsici in pepper plants. In addition, these selected strains exhibited biocontrol activities against anthracnose, and some of the strains showed plant growth-promotion activities on pepper fruit.

Comparison of Microbial Fungicides in Antagonistic Activities Related to the Biological Control of Phytophthora Blight in Chili Pepper Caused by Phytophthora capsici

  • Two similar microbial fungicides (termed as MA and MB) developed in a Korean biopesticide company were analyzed and compared each other in their biocontrol activities against the phytophthora blight of chili pepper caused by Phytophthora capsici. MA and MB contained the microbe Paenibacillus polymyxa and Bacillus subtilis, respectively, with concentrations over those posted on the microbial products. In comparison of the isolated microbes (termed as MAP from MA and MBB from MB) in the antagonistic activities against P. capsici was effective, prominently against zoospore germination, while MBB only significantly inhibited the mycelia growth of the pathogen. Some effectiveness of MAP and MBB was noted in the inhibition of zoosporangium formation and zoospore release from zoosporangia; however, no such large difference between MAP and MBB was noted. In a pot experiment, MA reduced the severity of the phytophthora blight more than MB, suggesting that the disease control efficacy would be more attributable to the inhibition of zoospore germination than mycelia growth of P. capsici. These results also suggest that the similar microbes MA and MB targeting different points in the life cycle of the pathogen differ in the disease control efficacies. Therefore, to develop microbial fungicides it is required to examine the targeting points in the pathogen's life cycle as well as the action mode of antagonistic microorganisms.

Efficacy of Suppression of Phytophthora Blight of Red Pepper Caused by Phytophthora capsici by Treatment with Antagonistic Trichoderma harzianum DYMC in Greenhouse (온실에서 길항미생물 Trichoderma hazianum DYMC 처리에 의한 고추 역병 억제 효과)

  • To control Phytophthora blight of red pepper biologically caused by Phytophthora capsici, we developed Trichoderma harzianum DYMC for commercial product. DYMC was storage at room temperature and was investigated their population every 3 months for 1 year. For investigating the dynamic population of T. harzianum in the pot soils, we applied powder and suspension applications with DYMC, and then investigated for 95 days. The efficacy of powder and suspension applications of DYMC for control of Phytophthora blight of red pepper and plant growth were investigated for 50 days in greenhouse experiment. The population of T. harzianum was decreased at the room temperature for 1 year but there was not statistically significance. After soil treated in the pot with DYMC, the population of Trichoderma spp. was the highest when DYMC powder at 5 g was applied to mix with pot soil, and the population was deceased significantly among treatment means as time goes by ($R^2=0.76$, F=10.5960, P=<.0001). Incidence of Phytophthora blight of, red pepper was significantly reduced among treatment means on 50th day after treated with DYMC ($R^2=0.82$, P=16.4758, P=<.0001). Disease control value was the highest at 62.5% when DYMC powder at 5 g was applied to mix with pot soil. No significant difference (P=0.05) of effects of plant and root growth showed by treated with DYMC on 60th day, except stem. Mixing the application of DYMC powder with soil to control Phytophthora blight of red pepper was greater than suspension application to dilute with water. DYMC could be used as an effective biocontrol agent to control Phythophthora blight of red pepper.

Development of the Microbial Consortium for the Environmental Friendly Agriculture by the Antagonistic Rhizobacteria (다기능 PGPR 균주들의 기작별 상호보완형 컨소시엄 구성을 통한 고추역병 방제 및 고추생장촉진)

  • We found out the new method of the consortium for the environmental friendly agriculture by 8 kinds of the selected antagonistic rhizobacteria. This research involved composition of mutual complementary consortium by each antagonistic function such as production of antibiotic, siderophore, antifungal cellulase and insoluble phosphate solubilization. The consortium No.11 among composed consortium candidates showed the most pepper growth promoting activity and Phytophthora blight suppression on the in vivo pot test of red-pepper plant. The consortium No. 11 is combination of PGPR Bacillus subtilis AH18 and Bacillus licheniformis K11. B. subtilis AH18 and B. licheniformis K11 both could produce the auxin, antifungal ${\beta}$-glucannase and siderophore. Also, they had mechanism for solubilization of insoluble phosphate. But, B. licheniformis K11 could produce the antibiotic of iturin which was able to inhibit Phytophthora capsici. We confirmed complementary noncompetitive mutualism between B. subtilis AH18 and B. licheniformis K11 of the consortium No.11. The results came out through treatment of two strains co-culture, treatment of individual culture and co-treatment of two individual cultures for the growth and Phytophthora blight suppression of red-pepper. The treatment of two strains co-culture didn't show a synergic effect in comparing sole treatment on the pepper growth promotion and Phytophthora blight suppression. But, when the pots were treated simultaneously with co-treatment of two individual cultures, an synergic effect was seen in the growth promotion of roots, stem, leaves and suppressed Phytophthora blight on red-pepper in vivo pot test.

Damage Analysis and Establishment of Control Threshold for Phytophthora Blight of Hot Pepper (Capsicum annuum) (고추 역병 피해 해석과 방제가 필요한 발병수준의 설정)

  • Incidence of Phytophthora blight of hot pepper (Capsicum annuum) and yield (fresh weight) of pepper fruits were investigated at four separate fields located in Cheongwon, Boeun, Eumsung, and Goesan, which are major pepper production areas in Chungcheongbuk-Do. In all of the experimental fields except the Goesan field, increased incidence of Phytophthora blight led to decreased yield of pepper fruits. The harvest time in which the yield of red pepper fruits was highly correlated with the incidence of Phytophthora blight was different between areas: it was highly correlated in the third harvest in Cheongwon (y=-11.0x+435.2, $r^2$=0.99), but in the second harvest in Boeun (y=-15.0x+944.6, $r^2$=0.76). In contrast, there was a very low correlation between the pepper yield and the disease incidence in Goesan in which pepper seedlings grafted on resistant stocks were planted. The final disease incidence in the Cheongwon experimental field reached 100% more than 40 days later in 2007 compared with that in 2006. The control threshold of Phytophthora blight in the pepper fields where disease incidence had been lower than 5% was set as 0.8% disease incidence, which caused less than 5% yield loss.

Control Effects of Indole Isolated from Xenorhabdus nematophila K1 on the Diseases of Red Pepper (Xenorhabdus nematophila K1 유래물질 인돌의 고추 병해 방제 효과)

  • Indole compound is a bacterial metabolite synthesized and released by an entomopathogenic bacterium, Xenorhabdus nematophila K1. The antibiotic activity was evaluated against plant pathogens, such as Phytophthora blight and anthracnose of red pepper. Indole significantly suppressed mycelial growth of Phytophthora blight and anthracnose pathogens. Under natural sunlight conditions, indole maintained the antifungal activity for at least sixty days. The activity was not affected under the condition of soil-water. When the indole suspension was applied to surface soil before transplanting of red pepper seedlings and was then regularly sprayed to the foliage of the plants with ten days interval, it resulted in significant reduction of the disease occurrences (Phytophthora blight, anthracnose, soft rot, and black mold) by about 30%. These results suggest that indole can be used to control Phytophthora blight and anthracnose of red pepper.
