Effects of selected phytochemicals and fruit extracts on Poly(ADP-ribose) polymerase (PARP) activity induced by H2O2 in MCF-7 breast cancer cells (식물생리활성물질과 과일류 추출물이 MCF-7 유방암 세포에서 H2O2로 유도된 Poly(ADP-ribose) Polymerase (PARP) 활성도에 미치는 영향)

  • Poly(ADP-ribose) polymerase (PARP) is a nuclear enzyme which is activated in response to DNA damage, and which mediates DNA repair. PARP inhibitors can be used to reduce resistance of cancer cells to anticancer treatments. The objective of this study was to investigate the effects of selected phytochemicals and fruit extracts on PARP activation in MCF-7 breast cancer cells subjected to oxidative stress. Pre-incubation with epigallocatechin gallate (EGCG), apple extract (AE), cranberry extract (CE), or grape extract (GE) for 2 hours at test concentrations reduced PARP activity induced upon treatment with hydrogen peroxide in a dose-dependent manner (p<0.05). GE was found to be the most efficient PARP inhibitor among the fruit extracts examined. These results suggest that phytochemicals of fruit extracts might be used as PARP inhibitors in order to assist anticancer agents.

Phytochemical Characterization of Vitex negundo Leaves: a Potent Antiandrogenic and Antioxidant Agent

  • This study was conducted to characterise phytochemicals and to explore the biological activities of Vitex negundo leaves. The washed, course powder of V. negundo leaves were extracted with different solvents of increasing polarity. All the extracts were characterized and biological activities were compared. The results revealed that the ethanolic and cold water extracts showed the presence of all phytochemicals studied except protein compared to other extracts. Further, the quantitative estimation of phytochemicals showed that the ethanolic extract had highest yield and maximum amount of total polyphenols, flavonoids, and alkaloids with the least amount of tannins compared to other extracts studied. Furthermore, the highest total polyphenol content corresponds with the potent biological activities. Indeed, in vitro antioxidant and antisteroidogenic activities were highest in the ethanolic extract than others. To conclude, the present study is the first to report the characterization and antiandrogenic property of V. negundo leaf extracts. The ethanolic extract of V. negundo leaves can be used as an antioxidant and antiandrogenic agent. Hence, it can be considered for the treatment of hyperandrogenic conditions like polycystic ovary syndrome, etc.

Analysis of Phytochemicals in Popular Medicinal Herbs by HPLC and GC-MS (HPLC와 GC/MS를 활용한 약용식물 유래 phytochemicals 분석)

  • Oriental herbs are reported as having potent functions for preventing many types of diseases. They also appear to have positive effects and potential capabilities for skin care. Among the many oriental herbs that are available, we chose to analyze four medicinal herbs, Korean red ginseng, Artemisia capillaries Thunb, Schizonepeta tenuifolia Briq, and Foeniculum vulgare Mill, because all are popular and considered as favorite medicinal plants in Korea. Extracts of the herbs were obtained by various methods such as using distilled water, ethyl ether, methanol, ethanol, benzene, 1-butanol, and chloroform. Nine phytochemicals were detected in the extracts: maltol, adenosine, b-pinene, menthone, pulegone, limonene, anethole, estragole, and fenchone, which reportedly have multi-functionalities. All phytochemicals were analyzed quantitatively by various chromatographic techniques such as HPLC and gas chromatography-mass (GC-MS) spectrometry. This article also presents the optimum conditions for extracting these 9 targeted phytochemical compounds that were derived from 4 popular oriental herbs, which could be useful for the efficient preparation of each phytochemical.

Global Gene Expression Changes by Several Phytochemicals in Human Colorectal Cancer Cell (인간 대장암 세포주에서 파이토케미칼 처리에 의한 유전자 발현 변화)

  • Phytochemicals, non-nutrient chemicals derived from plants, have been shown to have anti-inflammation, anti-oxidation, and chemopreventive effects. In the current study, we investigated whether five different phytochemicals (resveratrol, genistein, epicatechin gallate, diallyl disulfide, and caffeic acid phenethyl ester) alter cell growth and gene expression in human colorectal cancer HCT116 cells. Using a cell viability assay, we found that each of the phytochemicals tested inhibited HCT116 cell growth in a dose-dependent manner. Additionally, using human oligo DNA microarray analysis, we found that only six genes were commonly up-regulated and seven genes were commonly down-regulated in response to each phytochemical treatment. For the commonly up-regulated genes, the microarray analysis was confirmed by reverse transcription.PCR using gene-specific primers. In addition, NAG-1 protein was up-regulated by all treated phytochemcials. The results of this study may help to enhance our understanding of the general molecular mechanisms of chemoprevention that are mediated by phytochemicals in human colorectal cancer.
