Streptomycete isolator strain M0137 showed cytotoxic effect on THP-1 cells. One of the purified substances produced from the strain was identified as nonadecanoic acid. Morphological and physiological properties, phylogenetic analysis, and genomic fingerprinting of strain M0137 were determined. Strain M0137 showed a high similarity with Streptomyces scabiei, phenotypically and phylogenetically. In contrast, genomic fingerprinting and G+C content analysis revealed that strain M0137 could be distinguished from S. scabiei ATCC49173$\^$T/. We propose to name strain M0137 as Streptomyces scabiei Subsp.
Staphylococcus aureus cultures exposed to rotating magnetic field (RMF) were studied in order to analyse the possible induced changes in staphylococcal enterotoxin genes (se) expression. Liquid cultures of S. aureus strains carrying different se were exposed to the RMF of magnetic frequency 50 Hz and magnetic induction 34 mT for 10 h at $37^{\circ}C$. Three time points of bacterial growth cycle were considered for RNA extractions. Gene expression analyses were evaluated using real-time quantitative PCR method. The present study confirmed, that the RMF can stimulate the growth rate of S. aureus cultures in comparison to the unexposed controls, while the stimulation is not strain dependent. The studies have also shown, that the RMF, depending on the exposure time but regardless the bacterial strain, can influence on the expression of various se. In general, except for sea, as a result of bacterial exposure to the RMF through subsequent growth phases, the expression of se decreased, reaching the values below results recorded for unexposed controls. In the case of sea expression remained at a lower level as compared to the control, regardless the time of exposition.
A bacterial isolate (strain JS-2) characterized as Bacillus sp. was challenged with high concentrations of toxic selenite ions. The microbe was found to transform the toxic, soluble, colorless selenite (${SeO_3}^{2-}$) oxyions to nontoxic, insoluble, red elemental selenium ($Se^0$). This process of biotransformation was accompanied by cytoplasmic and surface accumulation of electron dense selenium ($Se^0$) granules, as revealed in electron micrographs. The cells grown in the presence of selenite oxyions secreted large quantities of extracellular polymeric substances (EPS). There were quantitative and qualitative differences in the cell wall fatty acids of the culture grown in the presence of selenite ions. The relative percentage of total saturated fatty acid and cyclic fatty acid increased significantly, whereas the amount of total unsaturated fatty acids decreased when the cells were exposed to selenite stress. All these physiological adaptive responses evidently indicate a potentially important role of cell wall fatty acids and extracellular polymeric substances in determining bacterial adaptation towards selenite-induced toxicity, which thereby explains the remarkable competitiveness and ability of this microbe to survive the environmental stress.
This study aimed to investigate the physiological characteristics and anti-obesity effects of milk fermented by L. plantarum KI134. The lipase, α-amylase, and α-glucosidase inhibitory activities of milk fermented by L. plantarum KI134 was 94.57±1.25%, 9.44±2.85%, and 2.74±1.24% (10 fold dilution), respectively. L. plantarum KI134 showed higher sensitivity to clindamycin and erythromycin in comparison to sixteen different antibiotics. It demonstrated the highest resistance toward ampicillin and vancomycin. The strain showed higher β-galactosidase, leucine arylamidase, valine arylamidase, acid phosphatase, β-glucosidase, and N-acetyl-β-glucosaminidase activities compared to other enzymes. It also did not produce carcinogenic enzymes, such as β-glucuronidase. The survival rate of L. plantarum KI134 in 0.3% bile was 96.90%. Moreover, the strain showed a 91.45% survival rate at a pH of 2.0. L. plantarum KI134 has resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus at the rates of 70.00%, 68.18%, 59.05%, and 40.63%, respectively. L. plantarum KI134 (23.01%) showed higher adhesion ability than the positive control (16.32%) L. rhamnosus GG. These results demonstrated that milk fermented by L. plantarum KI134 demonstrated an anti-obesity effect under in vitro conditions, with confirmed potential as a probiotic.
The Strain-Promoted Azide-Alkyne Cycloaddition (SPAAC) is a powerful method for synthesizing triazoles, even under physiological conditions, without a copper catalyst. This technique provides an efficient means for everyone to synthesize complex triazole derivatives rapidly. In order to investigate the configuration of triazole derivatives using bicyclo[6.1.0.]-nonyne (BCN) and chiral azide, it is necessary to employ the 2D NMR. Both 1D and 2D NMR (COSY, HSQC, 15N HMBC) are used to analyze the complex triazole product containing cyclooctyne, a diastereomeric product. The stereometric difference of the proton bonded to the same carbon is determined through the HSQC assignment. The intriguing splitting pattern of carbon resonances also reveals their diastereomeric configuration and will aid in further research based on physiological knowledge.
A bacterial strain J-59 was isolated from a humus soil, which produced simultaneously a thermostable glucose isomerase as well as xylanase. The morphological, cultural and physiological characteristics of the isoisomerase strain J-59 were detemined by the use of the media and methods described in International Streptomyces Project. The chemotaxonomic characteristics of the isolated strain J-59 were determined by the analysis of G+C molar % of DNA, diaminipimelic acid, composition of fatty acid and menaquinone. As the results of various examinations, the strain J-59 was identified to be Streptomyces chibaensis. This strain produced glucose isomerase intracellularly and xylanase extracellularly when grown in a medium containing xylan, but it was not able to utilize the xylose or xylan as a carbon source. The glucose isomerase of S. chibaensis J59 was highly thermostable, which retained more than 75% activity in the presence of Co$^{2+}$ at 80$\circ $C for 72 h.

The aim of the present research program was to develop a strain of actinomycetes producing antifungal substance. Soil samples were collected from various sites in Korea and a number of actinomycetes were isolated from the soil samples by applying selective agar for actinomycetes. Among over 440 isolates, a strain (NA-4803) producing antifungal substance against Trichophyton spp. Nannizzia otae and Pyricularia oryzae was selected. The strain NA-4803 was identified as strain similar to Streptoverticillium blastmyceticum with respect to morphological and physiological characteristics, lecithinase and lipolytic activity, degradation of organic compounds, resistance to antibiotics and utilization of carbon and nitrogen sources. But it showed some differences such as positive reaction of nitrate reduction, negative reaction of L-tyrosine degradation, resistance to cephaloridine, and utilization of I -rhamnose and inulin. The strain NA-4803 was named as Streptoverticillium sp. NA-4803.
Photosynthetic bacteria which can utilize ferulate as a sole carbon source for their metabolic activities were isolated from soils by liquid enrichment culture technique. The strain P-1 was selected by the highest capability of degrading ferulate in aerobic and anaerobic conditions. The strain P-1 was rod-shaped with its motility, strained gram negatively and could not utilize sulfur compounds. This strain has the bacteriochlorophyll a group I carotenoid and membrane structures like lamellae. As the results of physiological, morphological and cultural charactderistics, the isolate was identified as Rhodopseudomonas plaustris, one of the purple nonsulfer bacteria. The strain P-1 utilized 2mM/day in aerobic condition and 0.86 mM/day in anaerobic condition.
To clarify resistance mechanisms of lincosamide antibiotics, it was examined whether lincosamide antibiotics was able to induce high resistance to macrolide and lincosamide antibiotics against EMR-1 strain of Bacillus species. And it was also examined whether the inducible resistance was plasmid-mediated or chromosome-mediated. This strain was identified as Bacillus licheniformis by its morphological and physiological characteristics. The subinhibitory concentrations of lincomycin and clindamycin induced high resistance in the strain to lincosamide antibiotics, but not to macrolide antibiotics. The inducible resistance was not eliminated by treating the strain with ethidium bromide, and plasmid was not identified by the alkaline lysis method of plasmid preparation. These results indicate, therefore, that the inducible resistance to macrolide and lincosamide antibiotics in the strain may be chromosome-mediated, not plasmid-mediated.
