Antioxidant Activity of Royal Jelly Hydrolysates Obtained by Enzymatic Treatment

  • Recently, research on the processing of raw functional materials with the aim of improving various physiological activities has been conducted. In this study, we investigated the antioxidant activity of royal jelly (RJ) hydrolysates obtained from three commercial proteases. Enzyme-treated royal jelly (ERJ), in which the RJ hydrolysates were converted into easy-to-absorb shorter chain monomers through the removal of two known allergen proteins, showed no difference in the content of (E)-10-hydroxydec-2-enoicacid (10-HDA) or the freshness parameter and showed a significant increase in total free amino acid content. The antioxidant activity of ERJ was determined by 1,1-diphenyl-2-picrylhydrazyl (DPPH) and chemical assays. The ERJ showed about 80% DPPH-radical scavenging activity at same concentration of ascorbic acid. The antioxidant effect of ERJ was confirmed to be due to reduction of intracellular reactive oxidative species (ROS) and nitric oxide (NO) production in LPS-treated macrophages. Moreover, ERJ significantly increased the activity of the antioxidant enzyme superoxide dismutase (SOD) and the level of the antioxidant glutathione (GSH) in a dose-dependent manner. Interestingly, these antioxidant activities of ERJ were stronger than those of non-treated RJ. These findings indicate that ERJ has high potential as an antioxidant agent for use in human and animal diets.

한국 중년 여성의 건강증진 행위 예측 모형 구축 (Determinants of Health Promoting Behavior of Middle Aged Women in Korea)

  • Health promoting behaviors of an individual are affected by various variables. Recently, there has been a growing concern over important health problems of the middle aged women. Physiological changes in the middle aged women and their responsibility for family care can result in physical and psychological burden experienced by middle aged women. This study was designed to test Pender's model and thus purpose a model that explains health promoting behaviors among middle-aged women in Korea. The hypothetical model was developed based on the Pender's health promoting model and the findings from past studies on women's health. Data were collected by self-reported questionnaires from 863 women living in Seoul, between 20th, April and 15th, July 1995. Data were analyzed using descriptive statistics and correlation analysis. The Linear Structural Relationship(LISREL) modeling process was used to find the best fit model which assumes causal relationships among variables. The results are as follows : 1. The Overall fit of the hypothetical model to the data was good expect chi-square value(GFI=.96, AGFI=.91, RMR=.04). 2. Paths of the model were modified by considering both its theoretical implication and statistical significance of the parameter estimates. Compared to the hypothetical model, the revised model has become parsimonious and had a better fit to the data expect chi-square value(GFI=.95, AFGI= .92. RMR=.04). 3. Some of modifying factors, especially age, occupation, educational levels and body mass index (BMI) are revealed significant effects on health promoting behaviors. 4. Some of cognitive-perceptual factors, especially internal health locus of control, self-efficacy and perceptive health status are revealed significant effects on health promoting behaviors. 5. All predictive variables of health promoting behaviors, especially age, occupation, educational levels, body mass index(BMI), internal health locus of control, self-efficacy & perceptive health status are explained 20.0% of the total variance in the model.

회복 시 심박수의 역할: 심장질환과 운동프로그램 적용 가능성 (The Role of Heart Rate Recovery: Possibility of Heart Disease and Exercise Program Application)

  • Heart rate is a relatively simple and non-invasive method that is used as an important physiological indicator in many studies and has a close relationship with heart structure and function, cardiovascular disease and sudden cardiac death. In general, appropriately low heart rate during resting means effective heart function and cardiovascular fitness; heart rate at recovery is an important indicator of health and disease condition. We found a beagle dog (Dog_1) with a high heart rate in the previous preliminary experiment. Therefore, purpose of this study was to compare the heart rate response of the Dog_1 with the control group during 12 weeks of interval exercise, to evaluate the structural and functional abnormalities of the heart and to verify the applicability of exercise program. Heart rate was checked during 12 weeks of interval exercise, and after the exercise was over, imaging examination and hematological and serum biochemistry were performed. As a result, Dog_1 (165.6 ± 1.5) showed significantly higher heart rate in low intensity session of interval exercise than control group (133.3 ± 0.5, p < 0.01). In addition, Dog_1 (181.2 ± 1.4) showed significantly higher heart rate than control group (155.1 ± 0.9) in high intensity session (p < 0.01). The heart rate (30 sec, 60 sec) during recovery state was higher in Dog_1 (30 sec: 156.8 ± 4.0, 60 sec: 166.8 ± 5.8) than in the control group (30 sec: 111.2 ± 2.5, 60 sec: 104.0 ± 5.1, p < 0.01). The results of the imaging examination of Dog_1 with high heart rate confirmed that the heart had no functional and structural abnormalities. All beagles with the interval exercise program did not show maladjustment, and in the hematological and serum biochemistry results, all the parameter were within the reference range. If the interval exercise program of this study is used in the future, it is expected to be used as an important basic data to achieve the purpose of health, welfare, and physical fitness improvement of dogs.

  • Moxa-combustion therapy make use of heat stimulus and chemicals result form when cauterize the skin with moxa cones to medical cares. However, recently moxibustion have been utilized less than acupuncture for treatment of disease in a practicing oriental physicians. To determine variable parameters (the numbers, the times, the quantities, the locations) affected moxibustion's effects, the gastrin serum level in rats were observed. In experiment, male Spraque-Dawley rats (body wt. 140-160g) were selected. Anything is performed to normal group. Control group were only anesthetized with inhalation in normal group. Experimental group were anesthetized and cauterized with moxa at BL21 by way of direct moxibustion. The size of moxa cone is 1.6±0.2㎎. The moxibustion as 5 times a day for five days has shown most significant effects and the moxibustion as a aquantity of 1, 5, 10 times moxa united one respectly, inverse to quantity in effects. The moxibustion for five days has also shown an most significant effects. The moxibustion at unilateral acupoints BL21 have less effects than bilateral one and these effects had no difference between control group after vagotomy. This results indicate that moxibustion's effects are not direct proportion to moxa cone size and frequency but imply that there is adequate value of moxibustion.

  • This study was carried out to investigate the comparison of immunomodualtory effects of water-extracted Ginseng Radix(GR), Pilose Asia-bell(PA), Astragali Radix(AR), Astractylodes Rhizoma alba(AA) and Dioscoreae Rhizoma (DR). The parameter examined to assess apparent immunomodulatory effect of the water-extracted GR, PA, AR, AA and DR included the regulation of Nitric oxide (NO), the expression of Th1/Th2 type cytokine, the change of B cell phenotype. The water-extracted GR, PA, AR, AA and DR inhibited NO production and iNOS protein expression in LPS stimulated RAW 264.7 macrophage cells. In the Th1 and Th2 cytokine expression, the water-extracted GR, PA, AR, AA and DR induced IL-2 and IFNr mRNA gene expression. Therefore, it seems that the water-extracted GR, PA, AR, AA and DR have a inducing effect of Th1 type cytokines. In the Flow cytometry analysis, the water-extracted GR, PA, AR, AA and DR did not change B cell phenotype (CD45R/B220). The water-extracted GR, PA, AR, AA and DR have a reducing effect of immune suppression cause by Methotrexate (MTX), an agent of immune suppression. These results suggest that the immunomodulatory effects of the water-extracted GR, PA, AR, AA and DR may be, in part, associated with the inducing IL-2 and IFNr mRNA gene expression in and regulation of NO production in macrophage cells.

BIOFIT - Smart, Portable, Wearable and Wireless Digital Exercise Trainer Device with Biofeedback Capability

  • Today Human Personal Trainers are becoming very famous in this health conscious world. They teach user to achieve fitness goals in managed way. Due to their high fee and tight schedule they are unavailable to mass number of people. Another solution to this problem is to develop digital personal trainer portable instrument that may replace human personal trainers. We developed a portable digital exercise trainer device - BIOFIT that manages, monitors and records the user's physical status and workout during exercise session. It guides the user to exercise efficiently for specific fitness goal. It keeps the full exercise program i.e. exercises start date and time, duration, mode, control parameter, intensity in its memory which helps the user in managing his exercise. Exercise program can be downloaded from the internet. During exercise it continuously monitors the user's physiological parameters: heart rate, number of steps walked, and energy consumed. If these parameters do not range within prescribed target zone, the BIOFIT will alarm the user as a feedback to control exercise. The BIOFIT displays these parameters on graphic LCD. During exercise it continuously records the heart rate and number of steps walked every 10 seconds along with exercise date and time. This stored information can be used as treatment for the user by an exercise expert. Real-time ECG monitoring can be viewed wirelessly (RF Communication) on a remote PC.

광용적맥파 신호를 이용한 수면 중 호흡 추정 (Estimation of Respiration Using Photoplethysmograph During Sleep)

  • Respiratory signal is one of the important physiological information indicating the status and function of the body. Recent studies have provided the possibility of being able to estimate the respiratory signal by using a change of PWV(pulse width variability), PRV(pulse rate variability) and PAV(pulse amplitude variability) in the PPG (photoplethysmography) signal during daily life. But, it is not clear whether the respiratory monitoring is possible even during sleep. Therefore, in this paper, we estimated the respiration from PWV, PRV and PAV of PPG signals during sleep. In addition, respiration rates of the estimated respiration signal were calculated through a time-frequency analysis, and errors between respiration rates calculated from each parameter and from reference signal were evaluated in terms of 1 sec, 10 sec and 1 min. As a result, it showed the errors in PWV(1s: $36.38{\pm}37.69$ mHz, 10s: $36.53{\pm}38.16$ mHz, 60s: $30.35{\pm}38.72$ mHz), in PRV(1s: $1.45{\pm}1.38$ mHz, 10s: $1.44{\pm}1.37$ mHz, 60s: $0.45{\pm}0.56$ mHz), and in PAV(1s: $1.05{\pm}0.81$ mHz, 10s: $1.05{\pm}0.79$ mHz, 60s: $0.56{\pm}0.93$ mHz). The errors in PRV and PAV are lower than that of PWV. Finally, we concluded that PRV and PAV are more effective than PWV in monitoring the respiration in daily life as well as during sleep.

Force Field Parameters for 3-Nitrotyrosine and 6-Nitrotryptophan

  • Nitration of tyrosine and tryptophan residues is common in cells under nitrative stress. However, physiological consequences of protein nitration are not well characterized on a molecular level due to limited availability of the 3D structures of nitrated proteins. Molecular dynamics (MD) simulation can be an alternative tool to probe the structural perturbations induced by nitration. In this study we developed molecular mechanics parameters for 3-nitrotyrosine (NIY) and 6-nitrotryptophan (NIW) that are compatible with the AMBER-99 force field. Partial atomic charges were derived by using a multi-conformational restrained electrostatic potential (RESP) methodology that included the geometry optimized structures of both $\alpha$- and $\beta$-conformers of a capped tripeptide ACE-NIY-NME or ACE-NIW-NME. Force constants for bonds and angles were adopted from the generalized AMBER force field. Torsional force constants for the proper dihedral C-C-N-O and improper dihedral C-O-N-O of the nitro group in NIY were determined by fitting the torsional energy profiles obtained from quantum mechanical (QM) geometry optimization with those from molecular mechanical (MM) energy minimization. Force field parameters obtained for NIY were transferable to NIW so that they reproduced the QM torsional energy profiles of ACE-NIW-NME accurately. Moreover, the QM optimized structures of the tripeptides containing NIY and NIW were almost identical to the corresponding structures obtained from MM energy minimization, attesting the validity of the current parameter set. Molecular dynamics simulations of thioredoxin nitrated at the single tyrosine and tryptophan yielded well-behaved trajectories suggesting that the parameters are suitable for molecular dynamics simulations of a nitrated protein.

Effects of Clove Oil and Lidocaine-HCl Anesthesia on Water Parameter during Simulated Transportation in the Marine Medaka, Oryzias dancena

  • Optimum concentrations of anesthetic clove oil and anesthetic lidocaine-HCl were determined for a species of adult marine medaka, Oryzias dancena, over a range of salinity conditions, and investigated in a transport simulation experiment by analyzing various water and physiological parameters. Research indicated that the higher the concentration of anesthetic at each salinity, the shorter the anesthesia time at each salinity. At each concentration, fish were anesthetized slower at water salinities over 10 ppt (P<0.05). Anesthesia time at 10 ppt was faster than any other salinity. In 10 ppt salinity, the dissolved oxygen (DO) concentrations and respiratory frequencies of the clove-oil-administered groups decreased until 48 hours (P<0.05), whereas the $NH_4{^+}$ and $CO_2$ concentrations increased until 48 hours (P<0.05). In same period, the DO, $NH_4{^+}$, and $CO_2$ concentrations and respiratory frequencies all decreased as the clove oil concentration increased (P<0.05). The trends in the DO, $NH_4{^+}$, and $CO_2$ concentrations and respiratory frequencies in the lidocaine-HCl-administered groups were similar to those in the clove-oil-administered groups. In conclusion, clove oil and lidocaine-HCl are effective anesthetics, improving the transportation of the marine medaka. The results from this study will contribute to safe laboratory handling of the marine medaka, which are commonly required by many research studies and experiments.

말의 운동 스트레스에 관한 새로운 생물지표 (A novel biomarker of exercise-induced stress in horses)

  • Exercise is one of the most common stressors in horses. Although various physiological parameters such as cortisol respond to exercise, there is no reliable parameter for the measurement of exercise-induced stress in sport horses. This study was performed to discover a new biomarker with high sensitivity for exercise-induced stress. The expression of fos mRNA was increased more than 10-fold in horse blood samples collected after an hour of exercise, as compared with before the exercise. The plasma cortisol levels were also increased after the exercise, but only by about two-fold. The fos mRNA levels were well-correlated with plasma cortisol concentrations. These findings suggest that fos mRNA expression in blood may be useful for the measurement of exercise-induced stress in horses.
