Caenorhabditis elegans as a Biological Model for Multilevel Biomarker Analysis in Environmental Toxicology and Risk Assessment

  • While in some instances, loss of diversity results from acute toxicity (e.g. major pollution incidents), in most cases it results from long-term sub-lethal effects that alter the relative competitive ability and fitness of certain organisms. In such cases the sub-lethal effects will cause a physiological response in the organism that ultimately leads to community level changes. Very sensitive tools are now available to study sub-lethal responses at the molecular level. However, relating such laboratory measurements to ecological effects represents a substantial challenge that can only be met by investigation at all scales (molecular, individual organism and community level) with an appropriate group of organisms. Among the various in vertebrates which can be used as model organisms in such a way, the soil nematode, Caenorhabditis elegans appear to be a promising biological model to diagnose environmental quality. This paper reviews the current status of multilevel biomarkers in environmental toxicology, and C. elegans as promising organisms for this approach.

Thermo Physiological responses of Far Infrared Ray Radiation Fabrics at outdoor in summer (옥외에서 원적외선 방사직물 착용시 온열생리반응)

  • This study was performed to define the effects of Far Infrared Ray Radiation Fabrics as summer garments during outdoor work by human trial. One healthy male subject was volunteered for this study. Experimental garments consisted of three kinds of trousers (Cotton, Cotton/linen blended, Far Infrared Ray Radiation Fabric/wool blended) and basic garments (panty, socks, shirts, and dress shirts). The measurements were rectal temp., skin temp., microclimate inside clothing, heart rate, subjective sensation etc. The results were as follows : 1. Rectal temperature showed the lowest in Far Infrared Ray Radiation Fabrics among 3 garments. 2. Skin temperature (forehead, chest abdomen temp.) and mean skin temperature were lower in Far Infrared Ray Radiation Fabrics than in others, especially during early stage of work. 3. Heart rate showed lower value in Far Infrared Ray Radiation Fabrics than in others but there was no significance among the garments. 4. Humidity inside clothing and total weight loss showed the highest value in Far Infrared Ray Radiation Fabrics.

  • N1 and N2 gross neural action potentials were measured from the round window of the guinea pig cochlea at the onset of the acoustic stimuli. N1-N2 audiograms were made by means of regulating stimulant intensities in order to produce constant N1-N2 potentials as criteria for different input tone pip frequencies. The lowest threshold was measured with an input tone pip I5 dB SPL in intensity and 12 KHz in frequency when the animal was in normal physiological condition. The procedure of experimental measurements is explained in detail. This experimental approach is very useful for the investigation of the Cochlear function. Both noN1inear and active functions of the Cochlea can be monitored by N1-N2 audiograms.

Nintendo Wii Fit-Based Sleepiness Testing is Not Impaired by Contagious Sleepiness

  • Sleep deprivation may cause accidents, and it has deteriorating effects on health. A measurement of postural steadiness by a portable and affordable Nintendo Wii Fit balance board can be used to quantify a person's alertness. At work, people are under the influence of their environment-often other peopl-dthat may affect their alertness. This work investigates whether sleep deprivation among people is "contagious," as quantified by sway measures. We measured 21 volunteers' postural steadiness while alert and sleep deprived. During the measurements, a screen placed in front of the participants showed a footage of either alert or sleep-deprived faces. We found a significant difference between the day time and night time steadiness, but found no effect resulting from watching footage of sleep-deprived people. This finding shows that a posturographic sleepiness tester quantifies physiological sleep deprivation, and is insensitive to the influence of social factors.

Physiological Studies on the Formation of Hairy Root by the Agrobacterium rhizogenes 1. Relationships between LAA content and morphological characteristics in carrot infected by A. rhizogenes (Agrobacterium rhizogenes에 의한 Hairy Root 형성에 대한 생리학적 연구)

  • Inoculation of carrot discs with Agrobacterium rhizogenes harboring Ri plasmid resulted in transformation of cells, as revealed by the tumors and hairy roots arising from them. Measurements of IAA content using HPLC indicate that it is higher in tumors and inoculated tissues than in uninoculated tissue. A lot of meristemoids and vessel elements formed I tumor tissue and the hairy roots differentiated from meristemoids. IAA content in tumor tissues is decreased with hairy root and vessel elements formation from them. Formation of wound callus in uninoculated tissues resulted on wound healing but no formation of vessel elements and hairy roots. Tumor tissues show continuous growing on hormone free medium.

  • Standark polysomnograms, and questionnaires were measured using 4 subjects during simulated 150m heliox saturation dives. These measurements were performed on 4 pre-dive nights, 8 bottom nights, 6 decompression nights and post decompression nights. Sleep disturbances caused by frequent awakening were found throughout the dive period, but the disturbances were more intense on the second and fourth nights at the bottom and on the third and fourth nights of decompression. Stage 4 of sleep decreased every night at the bottom and from the first through third nights of decompression. The subjective self-evaluation of sleep was in agreement with the physiological variables. Complaints of subjective feeling of fatigue slightly increased throughout the dive period. It could be surmised that sleep disturbance or fatigue was not as significant under the 150m deep hyperbalic environment of He-O/ sub 2/ mixture, and therefore should be able to be coped with partially, through there moght be individual differences in adaptation.

Investigation of Environmental Noise in Elementary School (국민학교의 환경소음실웅 조사연구)

  • Environmental noise raises serious problems in our ciries. Noise reduces the preformance task, and occasionially causes the physiological problems such as nervous strain, headache etc. This study is to investigate the environmental noise of elementary school classrooms in attempt to probide the basis of appropriate sound performance standards. Field interview survey with a total of 369 subjects in 3 elementary schools and simultaneously measurements of noise level have been made. The results of study show that the A-weighted noise level is in excess of the criteria recommended by I.S.O. and 69% of the subjects are annoyed by the environmental noises : in particular traffic ; and market noises.

Infrared Imaging for Screening Breast Cancer Metastasis Based on Abnormal Temperature Distribution

  • Medical infrared imaging is obtained by measuring the self-emitted infrared radiance from the human body. Infrared emission is related to surface temperature and temperature is one of the most important physiological parameters related to health. Though recent applications such as security identification and oriental medicine have provided new fields of biomedical applications, infrared thermography has had ups and downs in its usages in cancer detection. Some of the main difficulties include finding proper applications and efficient diagnostic algorithms. In this study, infrared thermal imaging was used to detect regional metastasis of breast cancer. Our measurements were done for 110 women. From 63 individuals of a Healthy Group and a Benign Breast Disease Group, we developed algorithms for differentiating malignant regional metastasis based on temperature difference and asymmetry of temperature distribution. Testing with 47 cancer patients, we achieved a positive predictive value of $87.5\%$ and a negative predictive value of $95.6\%$. The results were better than for mammogram examination. A proper analysis of infrared imaging proved to be a highly informative and sensitive method for differentiating regional cancer metastasis from normal regions.

  • In this study, when physician make a diagnosis of Sasang constitution of patients, anthropometric data are applied to seek the classification function into Sasang constitution. Data used in the analysis are the anthropometric data of 550 patients who had been treated in nine oriental medical hospital, and our data have no missing value in 12 anthropometric variables. In order to improve the accuracy of classification function into Sasang constitution, we consider one method of variable transformation of anthropometric data based on oriental medicine.
