On-line Monitoring of IPTG Induction for Recombinant Protein Production Using an Automatic pH Control Signal

  • The response of IPTG induction was investigated through the monitoring of the alkali consumption rate and buffer capacity during the cultivation of recombinant E. coli BL21 (DE3) harboring the plasmid pRSET-LacZ under the control of lac promoter. The rate of alkali consumption increased along with cell growth, but declined suddenly after approximately 0.2 h of IPTG induction. The buffer capacity also declined after 0.9 h of IPTG induction. The profile of buffer capacity seems to correlate with the level of acetate production. The IPTG response was monitored only when introduced into the mid-exponential phase of bacterial cell growth. The minimum concentration of IPTG for induction, which was found out to be 0.1 mM, can also be monitored on-line and in-situ. Therefore, the on-line monitoring of alkali consumption rate and buffer capacity can be an indicator of the metabolic shift initiated by IPTG supplement, as well as for the physiological state of cell growth.

  • Apoptosis is now widely recognized as a common form of cell death and represents mechanism of cell clearance in many physiological situations where deletion of cells is required. In vivo administration of bacterial lipopolysaccharide (LPS) to Balb/c mice induced DNA fragmentation in the thymus. DNA fragmentation in the thymus was roughly dependent on the dose of LPS injected and reached the peak 18 hours after injection. This apoptosis in the thymus might be mediated due to LPS stimulant. DEN (diethylnitrosamine) has been shown to cause liver cancer in experimental animals and humans. The hepatic tumorigenesis was induced by ad libitum feeding of DEN only. It was suggested that DEN induced hepatic tumorgenesis in rat is a good reproducible model for studying biochemical and pathophysiological changes associated with the development of hepatic tumorigenesis and apoptosis.

Sleep-Promoting Effect of Ecklonia cava: Ethanol Extract Promotes Non-rapid Eye Movement Sleep in C57BL/6N Mice

  • We investigated the effects of Ecklonia cava ethanol extract (ECE) on sleep architecture and sleep profiles. ECE was orally administered at a dose of 100, 250, or 500 mg/kg to C57BL/6N mice and its effects were measured by recording electroencephalogram (EEG) and electromyogram. Administration of ECE (250 and 500 mg/kg) significantly induced non-rapid eye movement sleep (NREMS) without affecting rapid eye movement sleep. The increase in NREMS by ECE (500 mg/kg) was significant (P < 0.05) during the first 2 h after administration. In addition, ECE had no effect on EEG power density (an indicator of sleep quality) in NREMS. These results suggest that ECE induces NREMS in a manner similar to physiological sleep.

  • Review is made on the sweat in 'Donguibogam' from the view point of Hyungsang medicine. The conclusions are as follows: Clinically sweet is an important indicator for the condition and constitution of the patients. Sweat should be taken into consideration in diagnosis and treatment. Sweat is one of the symptoms that appears when there is a disharmony among Jung(精), Ki(氣) and Shin(神). The cause and phase of sweat varies with every patient. Dam type persons (膽體) are inclined to have night sweat, while Bangkwang type persons(膀胱體) are apt to have day sweat.

  • During the period of 1975-76, a survey was carried out to find out zinc indicators in the natural vegetation in Korea. The symptoms fo chlorosis were observed in flowering plants in the areas of zinc outcrop of Wolgok-A, Seokgok-9, and Sowolgok. Although 28 species were found to be chlorotic, the total quantity of chlorotic foliage observed was small. Reasons for chlorosis in the areas of zinc are deposits is considered as effects of zinc, lead, copper and calcium ions. Sedum sp. and Dianthus sinensis were confined to soil containing more than exchangeable zinc of 30 ppm and to accumulation in the plants contained at least 1,300-14,000 ppm of zinc. Therefore Sedum sp. and Dianthus sinensis might be used as zinc indicators in Korea.

Systematic Review on the Effect of Forest Healing Activities on the Elderly (산림치유 활동이 노인에게 미치는 효과에 대한 체계적 문헌고찰)

  • Life expectancy is increasing due to the aging of the population, which is in turn exacerbating problems such as the prevalence of various geriatric diseases. This study was established to provide basic data for the expansion of forest healing activities for the elderly by systematically analyzing the literature on how such activities affect this age group. For the collection of studies, the Korean databases RISS, KISS, Korea Med, and Science On were used, while PubMed, Cochrane Central, MDPI, and Google Scholar were used to identify reports published elsewhere. To assess the quality of the methodology used in the collected studies, the risk of bias was analyzed using Cochrane's RoB2 and RoBANS. Among 1,856 reports initially identified, 21 were finally selected for analysis in this study, which were limited to research papers on forest healing activities for the elderly published between 2000 and January 2022. In this review, the subjects were those aged 60 or older, with a total of 750 participants, ranging from at least 7 to a maximum of 88 per study. The analysis showed that the most frequently performed tests in each category were on depression as a psychological indicator in 7 studies, MMSE(Mini Mental State Examination) as a cognitive indicator in 2 studies, on blood pressure as a physiological indicator in 4 studies, on melatonin as a biochemical indicator in 2 studies, and on body fat and muscle strength as physical indicators in 3 studies. Of the 21 studies, 19 used two or more test items, with psychological indicators being most commonly measured. For the future application of forest healing activities for the elderly, various forest healing programs to prevent cognitive function decline should be developed and distributed, and follow-up studies should be continuously presented to provide the basis for forest healing activities.

The Role of Heart Rate Recovery: Possibility of Heart Disease and Exercise Program Application (회복 시 심박수의 역할: 심장질환과 운동프로그램 적용 가능성)

  • Heart rate is a relatively simple and non-invasive method that is used as an important physiological indicator in many studies and has a close relationship with heart structure and function, cardiovascular disease and sudden cardiac death. In general, appropriately low heart rate during resting means effective heart function and cardiovascular fitness; heart rate at recovery is an important indicator of health and disease condition. We found a beagle dog (Dog_1) with a high heart rate in the previous preliminary experiment. Therefore, purpose of this study was to compare the heart rate response of the Dog_1 with the control group during 12 weeks of interval exercise, to evaluate the structural and functional abnormalities of the heart and to verify the applicability of exercise program. Heart rate was checked during 12 weeks of interval exercise, and after the exercise was over, imaging examination and hematological and serum biochemistry were performed. As a result, Dog_1 (165.6 ± 1.5) showed significantly higher heart rate in low intensity session of interval exercise than control group (133.3 ± 0.5, p < 0.01). In addition, Dog_1 (181.2 ± 1.4) showed significantly higher heart rate than control group (155.1 ± 0.9) in high intensity session (p < 0.01). The heart rate (30 sec, 60 sec) during recovery state was higher in Dog_1 (30 sec: 156.8 ± 4.0, 60 sec: 166.8 ± 5.8) than in the control group (30 sec: 111.2 ± 2.5, 60 sec: 104.0 ± 5.1, p < 0.01). The results of the imaging examination of Dog_1 with high heart rate confirmed that the heart had no functional and structural abnormalities. All beagles with the interval exercise program did not show maladjustment, and in the hematological and serum biochemistry results, all the parameter were within the reference range. If the interval exercise program of this study is used in the future, it is expected to be used as an important basic data to achieve the purpose of health, welfare, and physical fitness improvement of dogs.

Condition index and hemocyte apoptosis as a health indicator for the Pacific oysters, Crassostrea gigas cultured in the western coastal waters of Korea (서해안 굴, Crassostrea gigas의 건강도 평가를 위한 Condition index와 혈구 apoptosis 분석)

  • A significant production decrease has been witnessed for the Pacific oysters, Crassostrea gigas farmed in the western coastal waters of Korea, presumably by the loss of physiological viability. We evaluated the viability in terms of health indicators, the condition indices and hemocyte apoptosis rates of the oysters inhabiting two representative farming sites, Incheon and Taean each with different environmental variables. In our monthly measurements for the whole year 2013, the indicators were location specific. The condition indices of Incheon were highly variable, 1.67-8.58%, while those of Taean were less, 2.28-5.57%. The condition indices decreased during the spawning seasons, July and September in common. The two oysters exhibited also differed in the apoptotic activities of hemocyte, highly active, 4.03-30.15% for Incheon oysters and less active, 2.87-17.48% for Taean oysters. One thing we could identify was the two measurements were adverse during the critical seasons of spawning, reminiscent of being a useful tool for a health indicator for the oysters. Similar trend was also observed in the time when change in temperature was extreme. The findings in this study are highly indicative of health indicators for the oyster aquaculture.

Study on the Basic Pulse Indicators for Pattern Identifications in Stroke (중풍환자의 변증에 사용되는 맥진 지표에 관한 연구)

  • The purpose of this study is to select the major pulse indicators and evaluate their significance in discriminating the subtypes of Pattern Identifications (PI) from stroke patients. Decision tree analysis was carried out using clinical data collected from 835 stroke patients with the same subtypes diagnosed identically by two experts with more than 3 year clinical experiences. Among the 10 pulse indicators, 6 major pulse indicators (slow, rapid, strong, weak, slippery, and fine pulse) were selected by decision tree analysis. The accumulated distributions of six pulse indicators in each PI showed that strong was major pulse indicator in Fire-Heat pattern, slippery in Dampness Phlegm pattern, weak in Qi Deficiency pattern. But there were two major combinations in Yin deficiency pattern, weak or fine with rapid pulse and weak or fine without rapid pulse. Therefore, it is suggested that 6 pulse indicators can be used for discrimination of PI in stroke patients, though the combination studies between these pulse indicators and the other PI indicators are left for further study.

Relationship between Personality Type and Academic Achievement of Korean Medical Students

  • The purpose of this study is to examine the relationship between Myers-Briggs Type Indicator(MBTI) personality type and academic achievement of Korean medical students. A total of 97 (57 men, 40 women; ages 24 to 36) fourth-year Korean medical students participated in this study. The MBTI questionnaire was administered to all the students for identification of their personality type and academic performance. The results are as follows. First, the proportions of Personality type of Korean medical students were Extroversion (E) 33.0% - Introversion (I) 67.0%, Sensing (S) 70.1%- Intuition (N) 29.9%, Thinking (T) 58.8%- Feeling (F) 41.2%, and Judging (J) 54.6% - Perceiving (P) 45.3%. Second, the most common personality type was ISTJ (22.7%), followed by ISTP (13.4%), ISFJ (12.3%). Third, according to the analysis of this study, academic achievement was significantly related with their personality type in the preference : Sensing (S) - Intuition (N) and Judging (J)-Perceiving (P). In analysis of Sensing (S) - Intuition (N) and Judging (J)-Perceiving (P) index, Sensing (S) and Judging (J) type students had higher academic achievement than Intuition (N) and Perceiving (P) type students. This is the study to identify the characteristics of MBTI in Korean Medical students. The findings indicate that academic achievement was significantly related to their personality type in the preference. Using the results of MBTI in Korean medical students, is helpful in selection of appropriate teaching and learning strategies to provide better education.
