Bacillus subtilis dgk (diacylglycerol kinase) 유전자의 생리적 자극에 의한 유도발현 (Expression Patterns of Bacillus subtilis Diacylglycerol Kinase Gene Induced by Physiological Stimuli)

  • Diacylglycerol Kinase (DGK)는 E. coli 및 진핵세포에서의 신호전달에 관여하며, 또한 미생물에서 생리적 자극에 따라 다른 발현 양상을 보이는 것으로 밝혀져 있다. Bacillus subtilis에서 이 유전자에 대한 환경에서의 자극 신호들, 즉 pH 변화, 삼투압의 변화 및 온도의 변화에 따른 발현양상을 연구하였다. 이미 동정된 dgk locus의 KpnI-HindIII의 0.45 kb의 DNA fragment를 probe로 하여 Dot blot, Northern blot analysis를 통해 발현량을 조사해 본 결과 dgk 유전자는 pH변화, 삼투압의 변화 및 온도의 변화에 대응하여 발현되는 유전자임을 알 수 있었다. 특히 낮은 pH, 고 삼투압, 및 저온에서 dgk 유전자의 발현량이 많아짐을 확인 할 수 있었다. Northern hybridization에서 약 2.5kb의 mRNA가 관찰되었다. dgk gene의 ORF size는 약 0.4 kb로 관찰된 transcript size와는 일치하지 않았다. 따라서 Streptococcus mutans의 dgk gene과 마찬가지로 B. subtilis의 dgk 유전자도 polycistronic mRNA로 발현되는 것을 추정할 수 있었으며, 염색체상의 dgk gene에서 상류의 ORF2까지의 크기가 약 2.5 kb로 관찰된 mRNA size와 동일하였다. dgk gene 상류의 ORF2영역의 0.6 kb의 DNA fragment를 probe로 하여 northern blot hybridization을 수행한 결과, 2.5 kb의 mRNA가 관찰되었으며 발현되는 형태도 dgk probe를 이용한 결과와 동일하였다. 따라서 dgk gene은 상류부위의 ORF2 gene과 operon을 형성하여 polycistronic mRNA로 전사되는 것으로 판단된다.

Inferring Pedestrians' Emotional States through Physiological Responses to Measure Subjective Walkability Indices

  • Walkability is an indicator of how much pedestrians are willing to walk and how well a walking environment is created. As walking can promote pedestrians' mental and physical health, there has been increasing focus on improving walkability in different ways. Thus, plenty of research has been undertaken to measure walkability. When measuring walkability, there are many objective and subjective variables. Subjective variables include a feeling of safety, pleasure, or comfort, which can significantly affect perceived walkability. However, these subjective factors are difficult to measure by making the walkability index more reliant on objective and physical factors. Because many subjective variables are associated with human emotional states, understanding pedestrians' emotional states provides an opportunity to measure the subjective walkability variables more quantitatively. Pedestrians' emotions can be examined through surveys, but there are social and economic difficulties involved when conducting surveys. Recently, an increasing number of studies have employed physiological data to measure pedestrians' stress responses when navigating unpleasant environmental barriers on their walking paths. However, studies investigating the emotional states of pedestrians in the walking environment, including assessing their positive emotions felt, such as pleasure, have rarely been conducted. Using wearable devices, this study examined the various emotional states of pedestrians affected by the walking environment. Specifically, this study aimed to demonstrate the feasibility of monitoring biometric data, such as electrodermal activity (EDA) and heart rate variability (HRV), using wearable devices as an indicator of pedestrians' emotional states-both pleasant-unpleasant and aroused-relaxed states. To this end, various walking environments with different characteristics were set up to collect and analyze the pedestrians' biometric data. Subsequently, the subjects wearing the wearable devices were allowed to walk on the experimental paths as usual. After the experiment, the valence (i.e., pleasant or unpleasant) and arousal (i.e., activated or relaxed) scale of the pedestrians was identified through a bipolar dimension survey. The survey results were compared with many potentially relevant EDA and HRV signal features. The research results revealed the potential for physiological responses to indicate the pedestrians' emotional states, but further investigation is warranted. The research results were expected to provide a method to measure the subjective factors of walkability by measuring emotions and monitoring pedestrians' positive or negative feelings when walking to improve the walking environment. However, due to the lack of samples and other internal and external factors influencing emotions (which need to be studied further), it cannot be comprehensively concluded that the pedestrians' emotional states were affected by the walking environment.

EIV와 MLP를 이용한 뇌파 기반 운전자의 졸음 감지 시스템 (Electroencephalogram-Based Driver Drowsiness Detection System Using Errors-In-Variables(EIV) and Multilayer Perceptron(MLP))

  • 졸음운전은 전체 교통사고 원인 중 큰 비중을 차지하며 그 위험성이 음주운전보다도 크다고 알려져 있다. 따라서 운전자의 졸음을 판단하고 경고하는 시스템 개발에 대한 관심이 높아지고 있으며, 뇌파를 분석하는 것이 운전자의 피로와 졸음을 감지하는데 효과적이라는 연구결과들이 발표되었다. 본 논문은 짧은 시간에 높은 해상도를 가지는 auto-regressive 모델 기법 중 잡음에 강인한 errors-in-variables(EIV) 방법을 이용하여 특징벡터를 추출하고, 다층신경망(multilayer perceptron; MLP)에 적용하여 운전자의 상태를 각성, 천이, 졸음의 세 가지 상태로 분류하는 졸음 감지 시스템을 제안한다. 생체신호의 측정 환경에 따른 성능을 평가하기 위해 높은 진단률을 갖도록 하는 EIV차수를 결정하고, 잡음에 대한 강인성을 확인하기 위해 신호대 잡음비(signal-to-noise ratio; SNR)에 따른 성능을 선형 예측 부호화(linear predictive coding; LPC) 방법과 비교하였다. 이 결과로부터 제안한 EIV와 MLP를 결합한 졸음 감지 시스템은 기존의 LPC와 MLP를 이용한 시스템에 대해 우수한 성능을 얻을 수 있음을 확인하였다.

HRV를 이용한 자기공명영상 소음의 시간 변화에 따른 스트레스 평가 (Evaluation on the stress using HRV according to elapsed time of MRI noise)

  • 자기공명영상 촬영시 발생되는 소음은 100 dB정도의 큰 소리로 인체에 심리적 생리적으로 많은 영향을 미친다. 따라서 본 연구는 자기공명영상 소음에 따른 성인 남녀의 심박동변이률 신호의 주파수 분석을 통해 스트레스 정도를 분석하였다. 실험 방법은 귀마개를 착용하여 안정상태에서 15분간 심전도 신호를 측정하였다. 그 후 자기공명영상 장비에서 측정한 약 100dB의 음원을 30분동안 들으면서 동시에 심전도 신호를 측정하였다. 약 100 dB정도에서의 자기공명영상 소음은 HRV 분석 결과 교감신경과 부교감신경간의 불균형을 초래하여 스트레스 상태를 유발하였다. 안정 상태에서 10분까지 최대의 스트레스 상태를 나타내고, 15분부터는 스트레스 정도가 줄어들어 30분에는 안정 상태 보다 높은 스트레스 상태를 나타내었다. 본 연구를 기초 자료로 하여 자기공명영상 촬영 종사자들은 환자의 청각보호에 관심을 가지고, 자기공명영상 소음에 관한 객관적인 지표연구가 필요할 것으로 사료된다.

우슬이 산화적 스트레스와 관련한 세포내 신호전달계에 미치는 영향 (Effect of Achyrantis Radixs on Rat Chondrocyte Oxidative Stress and Its Signal Transduction)

  • Archyranthes radix has had extensive therapeutic application, and there has been increasing interest in its biological effects. However, the biochemical effects of Archyranthes radix on chondrocyte oxidative stress have never been systematically investigated. Therefore, we investigated the effects of Acyranthes radix on role of MAPK signal transduction pathway on oxidative stress induced by hydrogen peroxide in rat articular chondrocytes. The statistically significant inhibitory action of Archyranthes radix on cell proliferation was observed at above $5{\mu}g/m{\ell}$. Next, we examined the time-dependent effect of $5{\mu}g/m{\ell}$ Archyranthes radix on cell proliferaion. Archyranthes radix significantly inhibited cell proliferation from 12 hr after treatment (P<0.05). $H_2O_2$, resulted in a time- and dose-dependent cell proliferation, which was largely attributed to oxidative damage. Acyranthes radix and $H_2O_2$ treatment caused marked sustained activation of phosphorylation of ERK1/2. Moreover, the synergistic phosphorylation of p44/42 MAPK by $H_2O_2$ and Archyranthes radix was selectively inhibited by PD 98059, a p44/42 MAPK inhibitor. In conclusion, these results are consistent with the hypothesis that under conditions of oxidative stress, the $H_2O_2$-induced inhibition of cell proliferation in the rat chondrocyte is mediated through a modulation of the Archyranthes radix signaling pathway, promoting further phosphorylation of p44/42 MAPK, indicating a potentially important role in cartilage repair and in the treatment of osteoarthritic cartilage.

  • 모바일 컴퓨팅은 무선이동체 통신과 휴대 정보 터미널, 인터넷을 이용하여, 컴퓨터와 인체의 정보 기술을 효과적으로 연계시켜, when, where, who, 이동하면서 사용이 가능한 현대 기술의 중추 적인 역할을 제공하며 새로운 기술을 생리학적 측정으로 재고하고, 창조적으로 재건 할 수 있다. 특히, 고령화 사회에서 병적 징후들이 질병으로 발전되기 이전에 생체 변화를 유도하는 과정에 개입할 수 있는 가능성을 제공해 줄 수가 있다. 그러나 많은 파라미터가 데이터처리, 데이터의 기준화의 모호함, 데이터의 동시수집이 어려움 등을 들 수 있다. 따라서 본 연구에서는 모바일 컴퓨팅을 활용하여 시간적 제한요소를 배제하고, 정확한 분석이 되는 접근으로 분석이 타당하고 생체전기 신호를 바탕으로 하는 모바일 신경역학적 코드화 방법을 채택하여 시스템을 구현하여 실험한 결과, 생체 신호 모바일 분석 장치의 모델이 될 수가 있었다.

생체정보를 이용한 유비쿼터스 심리상태 인식 모델 연구 (A Study on Ubiquitous Psychological State Recognition Model Using Bio-Signals)

  • 본 논문에서는 다양한 생체신호들을 이용하여 심리상태와 생체정보를 판별하고, 외부환경 정보와 함께 이용자의 현재 상황을 인식하여 그에 맞는 적절한 서비스를 제공하는 생체정보기반 상황인식시스템(Bio-Signal Context aware system :BSC)을 설계하고 구현한다. 본 논문에서 구현한 생체정보기반 상황인식시스템은 센서를 통하여 측정된 뇌파(EEG), 심전도(ECG), 피부전도도(GSR) 등의 생체신호들로부터 특징들을 추출하고 분석하였으며, 분석된 결과를 입력받아 평온, 집중, 긴장, 우울의 네 가지 심리상태를 판별하였다. 판별된 심리상태의 결과와 함께 심박변이도(HRV), 피부전도도, 체온 등의 생체신호로부터 분석된 생체 상황정보, 그리고 외부 환경의 상황정보로부터 이용자의 현재 상황을 추론하고 인식하여 현재 생체 상황에 맞는 적절한 서비스를 제공하였다.

Ginsenosides attenuate the 3-nitropropioic acid-induced rat striatal degeneration in an age-dependent manner

  • The number of reporting the effects on ginseng's physiological, pharmacological, and behavioral effects has been increased every year. Major active components of Panax ginseng, are the ginsenosides, which are mainly triterpenoid dammarane derivatives. 3-Nitropropionic acid (3-NP) is blown to induce cellular energy deficit and oxidative stress related neurotoxicity via an irreversible inhibition of the mitochondrial enzyme succinate dehydrogenase (SDH). Intraperitoneal injection of 3-NP produces striatal degeneration. Aged animals was more vulnerable to 3-NP than young animal. We used three different ages of 5-, 8-, and 26-week-old rats. 3-NP alone treatment induced striatal lesion and increased lesion volume with age-dependent manner in 5-, 8-, and 26-week-old rats by $30.2{\pm}5.8$, $v$, and $51.3{\pm}8.4mm^3$, respectively. However, pretreatment of GTS (100 mg/kg/day) before 3-NP reduced striatal lesion in 5-,8-, and 26-week-old rats by $3.15{\pm}6.1$, $8.89{\pm}1.9$, and $27.3{\pm}5.6mm^3$, respectively. Pretreatment of GTS also significantly increased survival rate in 5-week-old rats (3-NP alone: GTS +3-NP = $40.4{\pm}6.3$: $72.5{\pm}9.5\%$) than 8-week-old rats (3-NP alone: GTS + 3-NP : $13.5{\pm}5.2\%$ : $45.1{\pm}3.1\%$). In 26-week-old rats, 3-NP alone treated group died on day 18, whereas GTS +3-NP-treated group prolonged lifespan to 30 days. Thus, pretreatment of GTS before administration of 3-NP extended lifespan in all ages. The present results indicate that aged animals are more vulnerable to 3-NP and GTS pretreatment protected 3-NP-induced striatal damage in different ages of animals.

The Effect of Epidermal Growth Factor on Cell Proliferation and Its Related Signal Pathways in Pig Hepatocytes

  • It has been reported that liver is a very important organ to xenotransplantation. Pig is known to be a most suitable species in transplantation of human organs. However, the physiological function of pig hepatocytes is not clear elucidated. Epidermal growth factor (EGF) is known to be a mitogen in various cell systems. Thus, we examined the effect of EGF on cell proliferation and its related signal cascades in primary cultured pig hepatocytes. EGF stimulates cell proliferation in a dose (>1ng/ml) dependent manner. EGF-induced increase of $[^3H]-thymidine$ incorporation was blocked by AG 1478 ($10^{-6}M$, an EGF receptor antagonist) genistein and herbymycin A (tyrosine kinase inhibitors, $10^{-6}M$), suggesting the role of activation and tyrosine phosphorylation of EGF receptor. In addition, EGF-induced increase of $[^3H]-thymidine$ incorporation was prevented by neomycin $(10^{-4}M)$, U73122 $(10^{-5}M)$ (phospholipase C [PLC] inhibitors), staurosporine ($(10^{-8}M)$, or bisindolylmaleimide I $(10^{-6}M)$ (protein kinase C [PKC] inhibitors), suggesting the role of PLC and PKC. Moreover, EGF-induced increase of $[^3H]-thymidine$ incorporation was blocked by PD 98059 (a p44/42 mitogen activated protein kinase [MAPK] inhibitor), SB 203580 (a p38 MAPK inhibitor), and SP 600125 (a JNK inhibitor). EGF increased the translocation of PKC from cytosol to membrane fraction and activated p42/44 MAPK, p38 MAPK and JNK. In conclusion, EGF stimulates cell proliferation via PKC and MAPK in cultured pig hepatocytes.

Toll-Like Receptor 2 매개 Dual-Specificity Phosphatase 4 발현에서 Extracellular Signal-Regulated Kinase 1/2와 활성산소의 역할 (Role of Extracellular Signal-Regulated Kinase 1/2 and Reactive Oxygen Species in Toll-Like Receptor 2-Mediated Dual-Specificity Phosphatase 4 Expression)

  • Background: Toll-like receptors (TLRs) are well-known pattern recognition receptors. Among the 13 TLRs, TLR2 is the most known receptor for immune response. It activates mitogen-activated protein kinases (MAPKs), which are counterbalanced by MAPK phosphatases [MKPs or dual-specificity phosphatases (DUSPs)]. However, the regulatory mechanism of DUSPs is still unclear. In this study, the effect of a TLR2 ligand (TLR2L, Pam3CSK4) on DUSP4 expression in Raw264.7 cells was demonstrated. Methods: A Raw264.7 mouse macrophage cell line was cultured in Dulbecco's modified Eagle's medium supplemented with 10% fetal bovine serum and 1% antibiotics (100 U/mL penicillin and 100 g/mL streptomycin) at $37^{\circ}C$ in 5% $CO_2$. TLR2L (Pam3CSK4)-mediated DUSP4 expressions were confirmed with RT-PCR and western blot analysis. In addition, the detection of reactive oxygen species (ROS) was measured with lucigenin assay. Results: Pam3CSK4 induced the expression of DUSP1, 2, 4, 5 and 16. The DUSP4 expression was also increased by TLR4 and 9 agonists (lipopolysaccharide and CpG ODN, respectively). Pam3CSK4 also induced ERK1/2 phosphorylation and ROS production, and the Pam3CSK4-induced DUSP4 expression was decreased by ERK1/2 (U0126) and ROS (DPI) inhibitors. U0126 suppressed the ROS production by Pam3CSK4. Conclusion: Pam3CSK4-mediated DUSP4 expression is regulated by ERK1/2 and ROS. This finding suggests the physiological importance of DUSP4 in TLR2-mediated immune response.
