상이한 광질하에서 활착된 수박 접목묘 결합 부위의 식물체온 변화 (Variation of Plant Temperature at Joining Parts of Grafted Watermelon Seedlings Graft-taken under Different Light Quality)

  • Effect of light quality on plant temperatures at joining parts of grafted watermelon seedlings was investigated using a thermal imaging system in this study. Plant temperatures at joining parts lowered than those at stem region during graft-taking. However, difference in plant temperatures at joining parts and at stem region decreased by days after graft-taking. Plant temperatures of grafted seedlings graft-taken under white, blue, red, and blue + red fluorescent lamps repeatedly fluctuated high at photoperiod and low at dark period. Considering the variation of plant temperatures at joining pa퍼5, the illumination of blue and red light as well as white light will be effective for grafting of grafted watermelon seedlings. It is recommended that air temperature around grafted seedlings should be maintained at 27 to $29^{\circ}C$ for enhancing the physiological reaction of callus and for the smooth joining of scion and rootstock.

  • The present study was conducted to investigate the anti-allergic activity of Saingheylyunbooem(SHU)). We investigated the anti-allergic effects of SHU in RBL-2H3 basophilic leukemia cells by compound 48/80, a mast cell degranulator in mice. SHU inhibited ${\beta}$-hexosaminidase and histamine release from compound 48/80 stimulated RBL-2H3 cells. The in vitro anti-inflammatory activities of SHU in LPS-stimulated RAW 264.7 cells were investigated. SHU inhibited NO production effectively dowregulated the expression of iNOS mRNA and iNOS protein expression in LPS-stimulated RAW 264.7 cells. These result provide evidences that SHU may be beneficial in the treatment of allergic inflammtory disease.

  • Many pathovars of the species Pseudomonas syringae are known to produce different phytotoxins as secondary metabolites. Although phytotoxins generally enhance the virulence of P. syringae, they are not required for pathogenesis. Among the phytotoxins produced by P. syringae, lipodepsipeptides, coronatine, phaseolotoxin, and tabtoxin are the most well-known toxins which have been intensively studied for their structure, mode of action, biosynthesis, and regulation. A polymerase chain reaction (PCR) technique that amplifies a segment of the phytotoxin gene cluster using a primer set has been developed in recent years. This method offers the advantages of speed and sensitivity compared to the approaches based on physiological and biochemical methods. PCR detection of genes involved in the production of toxins could be exploited for early diagnosis of plant diseases caused by P. syringae pathovars.

  • The effect of levanheptaose produced by levanase from Streptomyces sp. 366L on principle intestinal microflora was investigated. The reaction product, levanheptaose, was used as a carbon source for various intestinal microflora. As a results, Bifidobacterium adolescentis, Lactobacillus acidophilus, and Eubacterium limosum grew effectively in the in vitro experiment, whereas Clostridium perfringens, E. coli, and Staphylococcus aureus did not. Therefore levanheptaose seems to promote selectively the growth of B. adolescentis and L. acidophilus. In the in vivo experiment, the effect of levanheptaose on the growth of intestinal microflora, $\beta$-fructosidase activity, pH, and butyrate concentration were examined in rats. Apparently, the number of fecal Bifidobacteria, the amount of butyrate, and $\beta$-fructosidase activity were increased, whereas total aerobes and pH were reduced in rats fed by levanheptaose diets, compared with those of control diets. We concluded that those effects may be beneficial in improving gastrointestinal health.

인삼사포닌이 비둘기 가슴근육으로부터 분리된 Malate Dehydrogenase의 조절기능에 미치는 영향 (Effects of Ginseng Saponin on the Regu lately Properties of Malate Dehydrogenase from Pigeon Breast Muscle)

  • In an endeavour to elucidate effects of ginseng on some characteristics of enzymes, malate dehydrogenase (EC 1.1.1.37) was chosen as a model enzyme and effects of ginseng saponin on the enzyme such as optimum pH, product inhibition, optimum temperature and the activity was investigated. The product inhibition by NADH-a reaction product of the enzyme-was increased 33% by 0.3% ginseng saponin. And the optimum pH of the enzyme was 8.3 but in the presence of 0.3% ginseng saponin it increased to 8.5. The enzyme activity and the optimum temperature was not affected by ginseng saponin in the concentration of 1.0% and 0.3%, respectively. In this work, the possibility of contribution of ginseng saponin to the adaptogen activity is suggested; Potentiation of the regulatory activity of an enzyme may contribute to the normalization of the physiological state and consequently may increase the nonspecific resistance of an organism.

야생 Killer 효모 Candida dattila의 분리 및 동정

  • 본 연구는 야생 killer 효모에 의한 포도주의 이상발효를 방지하고 오염균주에 내성이 강한 killer 특성을 이용하는 전 단계로서 국내 포도에서 생육하는 야생 killer 효모를 분리하고자 수행되었다. 그 결과 17개의 killer 효모 균주를 분리하였고 그 중 K109와 K112의 killer 활성이 가장 강하였으며, K117 균주의 killer 활성이 가장 약하였다. 분리된 17개의 균주는 서로의 상호작용에 의하여 생육을 억제하는 특성을 보이지 않았으며, 생리학적 및 배양학적 특성을 조사한 결과 지금까지 killer 효모로서 보고된 적이 없는 Candida dattila로 동정되었다. 이에 각 균주를 Candida dattila K101-K117로 명명하였다.

고온성 Bacillus amyloliquefaciens NS 15-4가 생산하는 내열성 Protease의 특성 (Characterization of a Thermostable Protease from Thermophilic Bacillus amyloliquefaciens NS 15-4)

  • A thermophilic bacteria showing proteolytic activity against defatted soybean was isolated from soil. It was identified as Bacillus amyloliquefaciens based on its morphological and physiological characteristics. The Bacillus amyloliquefaciens NS 15-4 was cultivated at 50$\circ$C by rotary shaking in a medium containing defatted soybean. An extracellular protease from this strain was purified to homogeneity by ammonium sulfate precipitation, ion exchange, and hydrophobic interaction chromatographies. The molecular weight of the enzyme was estimated to be approximately 30,000 by SDS-PAGE and the N-terminal amino acid sequence of the enzyme was turned out to be AQSVPYGISQIKAPA. The optimum temperature and pH for the enzyme reaction were 60$\circ$C and 11, respectively, and its thermostability was increased by the addition of calcium ion. The enzyme was inactivated by phenylmethylsulfonylfluoride, suggesting it be a serine protease. Comparing with other commercial proteases, the enzyme showed relatively high proteolytic activity against defatted soybean, a water-insoluble protein substrate.

Kinetic Approaches to Measuring Peroxiredoxin Reactivity

  • Peroxiredoxins are ubiquitous thiol proteins that catalyse the breakdown of peroxides and regulate redox activity in the cell. Kinetic analysis of their reactions is required in order to identify substrate preferences, to understand how molecular structure affects activity and to establish their physiological functions. Various approaches can be taken, including the measurement of rates of individual steps in the reaction pathway by stopped flow or competitive kinetics, classical enzymatic analysis and measurement of peroxidase activity. Each methodology has its strengths and they can often give complementary information. However, it is important to understand the experimental conditions of the assay so as to interpret correctly what parameter is being measured. This brief review discusses different kinetic approaches and the information that can be obtained from them.

  • Taurine is present in a variety of tissue and exhibits many important physiological functions in the cell. Although it is known that many tissues mediate taurine transport, its functions of taurine transport in bone have not been identified yet. In the present study, we investigated the expression of taurine transporter (TauT) and taurine uptake using mouse stromal ST2 cells and osteoblast-like MC3T3-E1 cells, which is bone related cells. Detection of TauT MRNA expression in these cells were performed by reverse transcription polymerase chain reaction (RT-PCR). (omitted)
