Effects of resveratrol on laminar shear stress-induced mitochondrial biogenesis in human vascular endothelial cells

  • [Purpose] The purpose of the study was to determine the combined effects of resveratrol supplementation with high-flow LSS on mitochondrial biogenesis in human vascular endothelial cells. [Methods] Cultured human umbilical vein endothelial cells were treated with 20 μM of RSV. For the shear experiments, cells grown to a >90% confluence were exposed to physiological levels of LSS (5 to 20 dyne/cm2) for 12 to 36 hours using a cone and plate shear apparatus. Gene expressions were analyzed by western blotting. [Results] Depletion of mitochondrial integrity was directly associated with increase in endothelial activation/dysfunction. The expressions of mitochondrial biogenesis regulator genes, such as SIRT1, PGC-1α, and TFAM, and the mitochondrial contents were significantly increased after treatment with both resveratrol and high-flow LSS for 12 hours. However, supplementation of resveratrol to high-flow LSS for a prolonged duration had no synergistic effect on the levels of mitochondrial biogenesis regulator gene expressions and mitochondrial content compared to the LSS treatment alone. [Conclusion] The present study demonstrated that the supplementation of resveratrol to high-flow LSS has no synergistic effects on enhancing mitochondrial integrity in human vascular endothelial cells.

Role of the Cervical Sympathetics on the Submaxillary Reflex Secretion Evoked by Stimulation of the Afferent Glossopharyngeal Nerve of the Cat (가묘(家猫)에 있어서 설인신경중추단자극(舌咽神經中樞端刺戟)에 의한 반사성(反射性) 악하선분필(顎下腺分泌)에 미치는 경부교감신경(頸部交感神經)의 영향(影響))

  • To investigate whether the cervical sympathetics contains specific secretory fibers for the salivary glands, reflex salivation was evoked and the role of the sympathetics or the reflex was examined in ketamine-anesthetized cat. Stimulation of the central end of the glossopharyngeal nerve produced a copious secretion from the submaxillary gland and the response was not affected by the section of the cervical sympathetics or by the administration of phenoxybenzamine, whereas the response was abolished by severing the chorda tympani or by the administration of atropine. The salivary response was always associated with an increase in glandular blood flow. Both salivary and blood flow responses were decreased markedly by the superimposed stimulation of the cervical sympathetics or by the administration of norepinephrine. The decreased submaxillary blood flow always preceded the decrease in salivary flow on stimulation of the cervical sympathetics and the decreased blood flow recovered prior to the salivary flow upon cessation of the sympathetic stimulation. The inhibitory effects of the sympathetics and norepinephrine were completely abolished by the pretreatment with phenoxybenzamine. These results indicate that the glossopharyngeal nerve is one of the afferent limbs of the submaxillary salivary reflex and the chorda tympani is the only efferent limb of the reflex pathway. Thus, it is suggested that the cervical sympathetics does not contain the specific secretory fibers for the gland, but plays a role in inhibiting the reflex secretion by decreasing the blood flow to the gland.

  • Ethylene gas classified as one of five major plant hormones plays an important role in various plant metabolism. The precise analysis of ethylene production of plants or plant parts is a valuable research procedure because knowledge of ethylene production facilitates measures of the physiological activity within the tissue. This paper describes procedures for analyzing ethylene from plant tissues by gas chromatography and discusses problems associated with extracting gas samples either by introducing a vacuum to plant samples or by using a hypodermic syringe. Introduced are a continuous flow system for efficient analysis and an automated system for sampling, analyzing, calculating and recording ethylene production data.

The common physiological effects of electric current on the body, stated in order of increasing current magnitude, are threshold perception, muscular contraction, unconsciousness, fabrillation of the heart. In this study, determined the tolerable voltage about earth-layered structure in the grounding system.

  • Aging effect on the cardiovascular circulation is simulated by lumped parameter model. Aging phenomena can be hemodynamically explained as (1) the increase of flow resistance induced by remodeling of artery vessels and increased viscosity of blood and (2) the reduction of the vessel capacitance caused by arteriosclerosis. Appropriate physiological parameters are evaluated from the clinical data of adults and old men. Simulation results well explain the hypertension with aging of cardiovascular system.

Study on the Relationship Between 12Meridians Flow and Facial Expressions by Emotion (감정에 따른 얼굴 표정변화와 12경락(經絡) 흐름의 상관성 연구)

  • Facial expression was an important communication methods. In oriental medicine, according to the emotion the face has changed shape and difference occurs in physiology and pathology. To verify such a theory, we studied the correlation between emotional facial expressions and meridian and collateral flow. The facial region divided by meridian, outer brow was Gallbladder meridian, inner brow was Bladder meridian, medial canthus was Bladder meridian, lateral canthus was Gallbladder meridian, upper eyelid was Bladder meridian, lower eyelid was Stomach meridian, central cheeks was Stomach meridian, lateral cheeks was Small intestine meridian, upper and lower lips, lip corner, chin were Small and Large intestine meridian. Meridian and collateral associated with happiness was six. This proves happiness is a high importance on facial expression. Meridian and collateral associated with anger was five. Meridian and Collateral associated with fear and sadness was four. This shows fear and sadness are a low importance on facial expression than different emotion. Based on yang meridian which originally descending flow in the body, the ratio of anterograde and retrograde were happiness 3:4, angry 2:5, sadness 5:3, fear 4:1. Based on face of the meridian flow, the ratio of anterograde and retrograde were happiness 5:2, angry 3:4, sadness 3:5, fear 4:1. We found out that practical meridian and collateral flow change by emotion does not correspond to the expected meridian and collateral flow change by emotion.

Changes of Facial Temperature and Blood Flow Rates by Treatment of Miso Facial Rejuvenation Acupuncture (미소안면침이 안면 피부 온도와 혈류량에 미치는 영향)

  • To investigate the effects of facial temperature and blood flow rates generated by Miso Facial Rejuvenation Acupuncture treatment. Ten women in their twenties to fifties with no skin diseases were recruited. Miso Facial Rejuvenation Acupuncture(MFRA) was performed on the both sides of their face. We measured their facial temperature using Digital Infrared Thermal Imaging(DITI) and blood flow rates using Laser Doppler Perfusion Imaging(LDPI) at pre-treatment, immediately, twenty and sixty minutes after treatment. We analyzed data using student's t-test(p<0.05). After MFRA treatment, facial temperature on the measurement area increased immediately from $30.5{\pm}1.0^{\circ}C$ to $31.5{\pm}1.0^{\circ}C$, a statistically significant increase. Sixty minutes after treatment, facial temperature on the measurement area decreased a little bit($30.2{\pm}0.6^{\circ}C$), but there was no statistical significance. After MFRA treatment, facial blood flow rates on the measurement area increased immediately from $165.1{\pm}52.3$ PU to $342.7{\pm}51.3$ PU, a statistically significant increase. Sixty minutes after treatment, facial blood flow rates measurement area were recovered almost at the same level as before treatment. MFRA treatment could increase facial temperature and blood flow rates.

Effect of Mori Folium Extract on Improvement of Blood Flow in Ferric Chloride-Induced Carotid Artery Damage Rat Model (염화제2철이 유도하는 경동맥 손상 렛트 모델에서 상엽 추출물의 혈행개선 효과)

  • In a previous study, we have shown that mulberry leaves (Mori Folium) extract MFE) and its compounds have the antioxidant effect in human red blood cells. However, the possible effect of MFE and its compounds on improvement of blood flow were not reported. Therefore, the aim of this study is to investigate the effects of MFE and its compounds on improvement of blood flow in a rat model of topical ferric chloride ($FeCl_3$)-induced carotid artery damage. The $FeCl_3$ treatment seriously damaged the carotid artery: the walls of the artery, blood flow rate, blood vessel diameter, blood vessel area and blood flow amount. However, administration of MFE or its compound has ameliorated the blood flow and suppressed thrombus in blood vessels. Moreover, the concentrations of serum total cholesterol, triglycerides, and LDL cholesterol in the MET and its compound groups were remarkably reduced in comparison to the control group, and HDL cholesterol concentration was higher in the MET and its compound groups than in the control group. These results suggest that MFE and its compounds ameliorate the thrombosis against blood vessel damage.

A study on determinants of flow status in smoking simulation game (흡연시뮬레이션 게임에서 몰입 결정요인에 대한 연구)

  • This research was conducted to find out determinant variables which can influence the degree of flow in smoking simulation game. We set up flow experience as a dependent variable. Several factors including empathy, health concern, and brainwave(${\alpha}$, ${\beta}$) EEG(Electroencephalogram) as a physiological indicator, which was expected to affect flow, were set up as independent variables to perform regression analysis. As a result, empathy, health consciousness, and ${\beta}$ wave had positive influences on flow in smoking simulation game but ${\alpha}$ wave had negative effect. This research empirically analyzed users' experience and neurophysiological changes which can influence on flow in serious game and contributed to theoretical and methodological development in health-related game research.

A study on anti-thrombotic activity of Hwao-tang

  • The thrombosis is the category of blood stasis. Blood stasis is a pathologicial state resulting from the reverse or impeded flow of blood in the body or the stagnation of blood flow in local parts as well as abnormal blood outside of the vessles which remains in the body and fails to disperse. Hwao-tang has been reported to have a hypolipidemic effect in patients with hypercholesterolemia, and in highcholesterol-induced experimental models. The present paper reports the effects of HOT on atherosclerosis using a spontaneous experimental model, Kurosawa and Kusanagi-hypercholesterolemic (KHC) rabbits. We have also investigated the pharmacological effect of extracts obtained from HOT on collagen-and ADP-induced blood platelet aggregation, thrombin-induced conversion of fibrinogen and fibrinolysis in in vitro experiments. In conclusion, the protection of extracts of Korean herbs' HOT on the ischemic infarction induced artificially might be involved to their inhibition of thrombotic action.
