무마취가토 신동맥내 투여에 의한 Trifluoperazine의 신장기능에 미치는 영향 (Effects of Renal Arterial Trifluoperazine on the Renal Function in Conscious Rabbit)

  • Renal arterial infusion of renotropic agents has been a very useful technique in the renal function studies. This type of experiments have usually been conducted in the large animals such as dogs and sheep. In these animals a catheter can be placed in the site without much disturbances of renal blood flow. Rabbits as an experimental model, however, caused a disturbances of renal blood flow by a catheterization of renal artery by its properties. Therefore we have developed a new technique that allows a simple and selective access to one side of renal arteries and the other as a control, without any disturbances of renal function. The distance between the both bifurcations of renal arteries on abdominal aorta is about 7 mm. To locate the tip of catheter on one side renal artery, ascending cannulation performed via femoral artery was done. We did an experiment with the technique to clarify the effect of calmodulin inhibitor on the renal function. One of the phenothiazine derivatives, trifluoperazine known as a powerful calmodulin inhibitor. Trifluoperazine, actual dose ranges of $2.76-5.20\;ug\;{\cdot}\;kg^{-1}\;{\cdot}\;min^{-1}$, increased urine volume and glomerular filtration rate significantly. Significant increases in urinary excretion of sodium, chloride and potassium were found. Fractional excretion of sodium and free water clearance increased significantly. These data suggest that this new technique is very useful in field of renal physiology and that striking effect of trifluoperazine on the renal function may be caused by increasing the renal hemodynamics, and by the inhibition of sodium, chloride and potassium reabsorption in the renal tubules.

Cholinergic Role on Insulin Action in Exocrine Secretion of the Isolated Rat Pancreas

  • In order to investigate intra-pancreatic cholinergic roles on insulin action in exocrine secretion, the pancreas was isolated from rats and continuously perfused with modified Krebs-Henseleit solution. Intra-arterial infusion of insulin (100 nM) or cholecystokinin (CCK, 14 pM) alone resulted in stimulation of the volume flow and amylase output. Also insulin potentiated the action of CCK in the exocrine secretion. Tetrodotoxin and atropine completely abolished the potentiating action of insulin and CCK as well as the action of insulin alone, but did not change the action of CCK alone. In order to see an effect of intra-pancreatic neural activation on the insulin action, electrical field stimulation (EFS) with parameters of 20 V, 2 msec and 8 Hz was applied to the isolated pancreas for 10 min under 2.5 or 18 mM glucose background. The EFS voltage-dependently elevated the flow rate and amylase output, and potentiated exocrine secretion in 18 mM glucose infusion compared with 2.5 mM glucose. The potentiating effects of EFS and 18 mM glucose were not observed in the streptozotocin-treated pancreas although it was perfused with 18 mM glucose. However, it was restored when the diabetic pancreas was perfused with porcine insulin(100 nM). Tetrodotoxin and atropine inhibited the pancreatic secretion induced by EFS with the background of 18 mM glucose. The results of present investigation indicate that the intra-pancreatic cholinergic tone exerts a stimulatory influence on the action of insulin in pancreatic exocrine secretion of rats.

Estimation of Suitable Methodology for Determining Weibull Parameters for the Vortex Shedding Analysis of Synovial Fluid

  • Weibull distribution with two parameters, shape (k) and scale (s) parameters are used to model the fatigue failure analysis due to periodic vortex shedding of the synovial fluid in knee joints. In order to determine the later parameter, a suitable statistical model is required for velocity distribution of synovial fluid flow. Hence, wide applicability of Weibull distribution in life testing and reliability analysis can be applied to describe the probability distribution of synovial fluid flow velocity. In this work, comparisons of three most widely used methods for estimating Weibull parameters are carried out; i.e. the least square estimation method (LSEM), maximum likelihood estimator (MLE) and the method of moment (MOM), to study fatigue failure of bone joint due to periodic vortex shedding of synovial fluid. The performances of these methods are compared through the analysis of computer generated synovial fluidflow velocity distribution in the physiological range. Significant values for the (k) and (s) parameters are obtained by comparing these methods. The criterions such as root mean square error (RMSE), coefficient of determination ($R^2$), maximum error between the cumulative distribution functions (CDFs) or Kolmogorov-Smirnov (K-S) and the chi square tests are used for the comparison of the suitability of these methods. The results show that maximum likelihood method performs well for most of the cases studied and hence recommended.

Induction of Apoptosis by Gagamhwanglyeonhaedog-tang through Activation of Caspase-3 in Human Leukemia Cell Line HL-60 Cells

  • Gagamhwanglyeonhaedog-tang(GHH), a Korean genuine medicine, is a newly designed herbal drug formula based on the traditional oriental pharmacological knowledge for the purpose of treating tumorous diseases. Apoptosis is an evolutionarily conserved suicide program residing in cells. It leads to cell death through a tightly regulated process resulting in the removal of damaged or unwanted tissue. In the present study, the apoptosis inducing activities of the decocted water extract of GHH were studied. Results of the 3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay showed that GHH had a strong cytotoxic effect on HL-60 cells. The number of live cells was less than 20% after exposure to 1㎎/㎖ GHH for 48 hr. GHH increased cytotoxicity of HL-60 cells in a dose- and time­dependent manner. Cell apoptosis by GHH was confirmed by flow cytometric analysis of the DNA-stained cells. The percentage of apoptotic cells increased to 28%, 31% and 37% 24 hr and 37%, 44% and 81% 48 hr after treatment with 0.01, 0.1 and 1㎎/㎖ GHH, respectively. Flow cytometric analysis of GHH treated HL-60 cells showed increase of hypodiploid apoptotic cells in a dose- and time- dependent manner. DNA fragmentation also occurred in apoptosis and was characterized by a ladder pattern on agarose gel. In addition, GHH (0.01 and 0.1㎎/㎖) increased the secretion of tumor necrosis factor-alpha in 24 and 48 hr. The author showed that GHH-induced apoptosis was accompanied by activation of caspase-3. These results suggest that GHH induces activation of caspase-3 and eventually leads to apoptosis.

천마반하탕이 뇌허혈에 미치는 기전 연구 (Mechanism Study of Cheonmabanhwa-Tang on the Cerebral Ischemia in Rats - Focusing arround Improvement in Changes of Cerebral Hemodynamics -)

  • Cheonmabanhwa-Tang(CBT) has been used in the Oriental Medicine for many centuries as a therapeutic agent for dizziness due to Poong-Dam. This Study was designed to investigate the mechanism of Prescription on cerebral hemodynamics [regional cerebral blood flow(rCBF) and pial arterial diameter(PAD)J in cerebral ischemia rats, The results in cerebral ischemic rats were as follows: Both rCBF and PAD were significantly and stably increased by CBT (10 ㎎/㎏, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. Pretreatment with indomethacin(1 ㎎/㎏, i.p.), an inhibitor of cyclooxygenase significantly but unstably increased the CBT-induced increases in PAD as well as rCBF during the period of cerebral reperfusion. Pretreatment with methylene blue(10 (.1.㎍/㎏, i.p.), an inhibitor of guanylate cyclase significantly but unstably increased the CBT-induced increases in PAD as well as rCBF during 150 minutes of cerebral reperfusion, but decreased stably the CBT-induced increases in rCBF and PAD after 180 minutes of cerebral reperfusion. In conclusion, the present authors thought that CBT caused effect on cerebral hemodynamics via mediation of cyclooxygenase.

목향조기산(木香調氣散) 추출물이 뇌허혈 흰쥐의 뇌혈류개선에 미치는 작용기전 (Mechanism of Mokhyangjoki-san Extract on the Improvement in Cerebral Blood Flow in Ischemic Rats)

  • This study was designed to investigate the effects of Mokhyangjoki-san extract (MJS) on the improvement in cerebral blood flow (rCBF) in cerebral ischemic rats, and further to determine cytokines production (IL-1${\beta}$, TNF-${\alpha}$, IL-10, TGF-${\beta}$) of MJS. The results in cerebral ischemic rats were as follows ; The rCBF was significantly and stably increased by MJS (10 mg/kg, i.p.) during the period of cerebral reperfusion, which contrasted with the findings of rapid and marked increase in control group. In cytokine production in serum from femoral arterial blood 1 hr after middle cerebral arterial occlusion, MJS significantly decreased IL-1${\beta}$ and TNF-${\alpha}$ production, and increased IL-10 production compared with control group. In cytokine production in serum from femoral arterial blood 1 hr after reperfusion, MJS significantly decreased IL-1${\beta}$ and TNF-${\alpha}$ production compared with control group. IL-10 and TGF-${\beta}$1 production in MJS group were significantly increased compared with control group. These results suggested that MJS significantly and stably increased rCBF by inhibiting the production IL-1${\beta}$ and TNF-${\alpha}$, and accelerating that of IL-10 and TGF-${\beta}$. The result in nerve cells was as follows ; MJS significantly inhibited lactate dehydrogenase activity in vitro in a dose-dependent manner. This result suggested that MJS prevented the neuronal death.

천화분이 MCF-7 유방암 세포주의 G2/M 세포주기 억제에 미치는 영향 (Effect of Arresting MCF-7 Human Breast Carcinoma Cell at G2/M Phase of Trichosanthes Kirilowii)

  • The purpose of this study is to investigate the anti-proliferative mechanism by Trichosanthes kirilowii (TCK) in MCF-7 human breast carcinoma cell. In this study, we used human breast cancer cell line, Michigan cancer foundation-7 cells (MCF-7 cells). They were co-incubated with 30~200 ${\mu}g$/ml TCK for 48 hours, and cell viability was measured by Water-soluble tetrazolium salt-1 (WST-1) assay. After MCF-7 cells were exposed to 60 ${\mu}g$/ml of TCK for 0, 3, 6, 12, 24, 48 hours, We performed flow analysis cytometry sorting(FACS) and western blot analysis. We investigated the effect of dose-dependent cell growth inhibition by TCK, which could be proved by WST-1 assay. Also, flow cytometry analysis showed that TCK increased percentage of subG1 phase and G2/M phase cell cycle. In addition, TCK induced apoptosis through the expression of caspase-9, -3 and poly(ADP-ribose) polymerase(PARP) activation. Moreover, we showed that ATM-dependent G2/M phase arrest by DNA damage and phosphorylation of chk2, cdc25C, cdc2(Tyr15). Taken together, these results suggest that by G2/M phase arrest through DNA damage and inducing of apoptosis through intrinsic pathway, TCK may have potential tumor suppressor in breast cancer.

유체의 성질에 따른 자계용적맥파의 검출 특성에 관한 연구 (A Study on the Detection Characteristics of the Magneto-Plethysmography According to Fluid Properties)

  • Photo-plethysmography (PPG), which measures changes in the peripheral blood flow of a human body using difference in absorption rate of light, is a measurement method that is studied and used in clinical and various applications due to its simple circuit configuration and measurement convenience. Magneto-plethysmography (MPG), which is newly developed by our team, is a method of measuring changes in the conductivity of biological tissues by using a eddy current induced by a time-varying magnetic field, and is not subject to optical interference. In this study, we investigated the detection characteristics of MPG according to the change of the conductivity of the object and fluid to be measured by simultaneously measuring PPG and MPG. In order to control the speed of fluid known in advance, a blood flow simulator was implemented and used. The fluid used in the experiment was general mineral water and physiological saline (0.9% NaCl) solution. Experimental results show that the amplitude change of the measured PPG was 0.3% in normal water and saline solution, and that of MPG was 77.3%. Therefore, it is considered that the magneto-plethysmography (MPG) has a strong correlation with the conductivity of the fluid.

소청용탕이 Helper T Cell의 활성에 미치는 영향 (Effect of Herbal Extract on Helper T Cell activity)

  • SCRT (Sochungyong-tang) has been used for immune disease in human. The purpose of this study was effect of Helper T cell, major regulator of immune system. Spleen cell from 8 week BALB/c mice were cultured in SCRT containing medium without activation for 48 h. The MTS assay and flow cytometry revealed that SCRT treated Iympocyte were non-effect in percentage of CD4+ T cell. Subsequently CD4+ T cell were isolated and cultured in SCRT containing medium. SCRT were non-effective on CD4+ T cell without any involvement of APC. In order to evaluate the direct effect of SCRT on Helper T cell, CD4+ T cell isolated after 48 h of culture in SCRT containing medium and activated with and without anti-CD3/anti-CD28 activation for 48 h. A lower level of CD69 was observed in SCRT treated cells in flow cytometry analysis. Subsequently Using RT-PCR analysis the expression of mRNA for IL-2, INF-γ are upregulated and, IL-4 is downregulated in CD4 T cell. The result suggests that SCRT makes Th1 significantly increased and Th2 relatively inhibited. The results suggest that SCRT potentiate Th1 cell and decrease Th2 development at the same time, which is believed to be bemeficial for IgE-mediated responses.

내상병에 대한 형상의학적 치료 -위질환을 중심으로 (Treatment on Internal Damage Disease in Hyungsang Medicine)

  • Review on internal damage disease(disease of the spleen and stomach) in Hyungsang medicine is as follows; In case of woman, Disease of the spleen and stomach is caused mostly a excessive thought. In case of man, Disease of the spleen and stomach is related to the genitals's disease for man is based on the kidney and body. In shapes of the Jung(精), Ki(氣). Shin(神) and Hyul(血), the seven feelings(emotions) in the Ki(氣) and Shin(神) types and deficiency of essence and blood in the Jung(精) and Hyul(血) types induce mostly diseases of the spleen and stomach. Treatments are used frequently to promote the normal flow of Ki(行氣), promote digestion and remove food stagnancy(消導), resolve phlem(化痰) and check upward perverted flow of fire(降火) in the Ki(氣) and Shin(神) types and tonity the essence(補精), tonity the blood(補血) and replenish Ki(補氣) in the Jung(精) and Hyul(血) types. The main parts to appear outside diseases of the spleen and stomach are face color, depression condition of eyes, lips' condition, LU10(魚際)'s color and the rise and fall of flesh etc. Shapes of the six meridians have to treat to divid shapes and symtoms of the six meridians according to 'deviation of Ki and blood' and 'the expression part of symtoms'.
