Three Unrecorded Fungal Species from Fecal and Freshwater Samples in Korea

  • While evaluating fungal diversity in fecal and freshwater samples in Korea, three fungal strains, CNUFC-GHD83-1, CNUFC-RD8126, and CNUFC-YR2-1, were isolated from specific habitats including grasshopper and rat feces, and freshwater samples in Korea. On the basis of the morphological characteristics and phylogenetic analysis of the internal transcribed spacer (ITS) and 28S rDNA, the isolates CNUFC-GHD83-1, CNUFC-RD8126, and CNUFC-YR2-1 were identified as Albifimbria terrestris, Cephaliophora tropica, and Mariannaea aquaticola, respectively. These species have not been previously described in Korea.

  • Echinoids were collected at depths of 5-10 m in Munseom, Jeju Island by SCUBA diving on November 23, 2008 and September 15, 2009. Two specimens were identified as Echinometra mathaei (Blainville, 1825) based on morphological characteristics and molecular analyses of mitochondrial cytochrome c oxidase subunit I partial sequences. Echinometra mathaei collected from Korea was redescribed with photographs and was compared with other species from GenBank based on molecular data. Phylogenetic analyses showed that no significant differences were between base sequences of E. mathaei from Korea and that from GenBank. To date, 13 echinoids including this species have been reported from Jeju Island, and 32 echinoids have been recorded in Korea.

  • In 2016, as part of a larger effort to discover indigenous prokaryotic species in Korea, we isolated the family Deinococcaceae and Planctomycetaceae as unrecorded bacterial species. From the high 16S rRNA gene sequence similarity (>98.5%) and formation of a robust phylogenetic clade with known species, it was determined that each strain was a distinct bacterial species. There are no official reports that these two species have been described in Korea; therefore, the bacterial strains of Deinococcus and Blastopirellula are described for the first time in Korea. Gram reaction, colony and cell morphology, basic biochemical characteristics, and isolation sources are also described in the species description section.

  • Bacterial community structure composing enhanced biological phosphorus removal (EBPR) activated sludge was analyzed phylogenetically by cloning 165 rDNA after direct DNA extraction. Then, this result was compared with 165 rDNA sequences of randomly isolated bacterial species. The results clearly showed that there are no coincidence between the sequences retrieved directly from activated sludge and those of isolated strains, suggesting that many important bacteria are hidden in activated sludge because of the difficulty in isolation and culture of them.

Complete Mitogenome of the Russian Sturgeon Acipenser gueldenstaedtii (Acipenseriformes; Acipenseridae)

  • Sturgeons and paddlefishes are frequently referred to as 'living fossils' among the actionpterygian lineage. They are increasingly facing threats to their existence because of various anthropogenic pressures. In this study, we present the complete mitogenome sequence of the Russian sturgeon Acipenser gueldenstaedtii (Acipenseriformes; Acipenseridae). The mitogenome showed highly homogeneous molecular features compared to previously known vertebrate mitogenomes. Phylogenetic tree inferred from concatenated protein-coding and tRNA genes unambiguously revealed the monophyly of A. gueldenstaedtii, Acipenser stellatus, and Huso huso. Genetic information of the endangered A. gueldenstaedtii will provide baseline data needed to develop molecular markers for stock identification and assessment of population diversity and also to develop future conservation strategies.

Characterization of a Zygomycete Fungus, Mortierella minutissima from Freshwater of Yeongsan River in Korea

  • During a survey of fungal diversity of the order Mortierellales, a zygomycete strain, EML-YS717-1, was isolated from a freshwater sample collected from the Yeongsan River in Gwangju, Korea. Based on its morphological characteristics and a phylogenetic analysis of the internal transcribed spacer (ITS1 and ITS2) and 5.8S rDNA sequences, the strain was identified as Mortierella minutissima. To the best of our knowledge, M. minutissima, has not previously been authentically reported in Korea.

Isolation and Characterization of Blakeslea trispora Isolated from Gut of Grasshopper and Soldier Fly Larva in Korea

  • During a survey of fungal diversity in insect guts in Korea, two fungal strains, EML-PGH2 and EML-PUKI88, were isolated from the gut of grasshopper and soldier fly larvae inhabiting the bulrush plants at a pond located in the Chonnam National University Arboretum, Gwangju, Korea. Based on their morphological characteristics and a phylogenetic analysis of the internal transcribed spacer (ITS1 and ITS2) and 5.8S rDNA sequences, the strains were identified as Blakeslea trispora. To our knowledge, the zygomycete species B. trispora has not been previously described in Korea.

  • Species of Phoma and its allies were isolated during a survey on the diversity of endophytic fungi associated with pine trees in Korea. Based on the phylogenetic analyses of internal transcribed spacer and ${\beta}$-tubulin gene sequences, two Phoma-like species from the needles of Pinus koraiensis were identified as Peyronellaea calorpreferens and P. glomerata. They were also morphologically identified based on the previous descriptions. Here, we report P. calorpreferens and P. glomerata being present in Korea as endophytic fungi in Pinus koraiensis.

New Records of Two Agarics: Galerina sideroides and Gymnopus luxurians in South Korea

  • Studies on the diversity of indigenous fungi were conducted in the central districts of South Korea from 2008 to 2013. During the studies, potentially unrecorded agarics were collected. All specimens collected in this study were examined morphologically and phylogenetic analysis was also performed. They were identified as Galerina sideroides and Gymnopus luxurians. These fungi have never been reported in South Korea. We report them here with detailed descriptions and figures.

A New Record on Umbelopsis vinacea and Mucor hiemalis f. corticola Isolated from Korea

  • In the screening of fungal diversity, two strains were collected from the soil of Yeongcheon and dissected guts from the bodies of Chinese rice grasshopper (Oxya chinensis), Chinese grasshopper (Acrida cinerea), and Far eastern devil grasshopper (Oedaleus infernalis) from Daejeon in Korea. They were identified as Umbelopsis vinacea (KNU-YC-1801B) and Mucor hiemalis f. corticola (KNU-20F7, KNU-20F8, KNU-20F9). Multigene phylogenetic analyses of the internal transcribed spacer (ITS) regions, and large subunit (LSU) sequence data confirmed two unreported taxa along with their morphology. The results of molecular phylogeny firmly supported the detailed description and illustration for each taxon. As far as we know, both Umbelopsis vinacea and Mucor hiemalis f. corticola are the first reported taxa in Korea.
