Inhibition of HBV ]Replication by the Extract of Phyllanthus ussuriensis (여우주머니(Phyllanthus ussuriensis) 추출물의 B헝 간염바이러스 증식 억제)

  • Abstract -Phyllanthus ussuriensis which belongs to Euphorbiaceae has been used as a component of Korean traditional remedy against hepatitis and jaundice. Aqueous methanolic extract of P. ussuriensis was tested for antiviral activity of hepatitis B virus (HBV) in HepG2 2.2.15 cells which were derived through transfection of cloned HBV DNA into HepG2 human hrpatoblastoma cells. P. ussuriensis extract decreased the levels of extracellular HBV virion DNA and inhibited HBV replication at concentrations ranging from 50 to 500$\mu$g/ml. Partitioning of water suspension of the extract revealed that the activity mainly resides in the EtOAc fraction. Consecutive purification of the EtOAc fraction by silica-gel column chromatography resulted in the two active anti-HBV fractions. Southern blot analysis shows that the action mechanisms or active site of the two fractions seems to be different in terms of their inhibition of HBV replication steps.

Phenolic Compounds from Phyllanthus ussuriensis (여우주머니의 Phenol성 화합물)

  • The herbaceous species of phyllanthus in this genus are used as hyperglycemic, diuretic, and malaria agent in the world. For the phytochemical studies and the investigation of medicinal resources in the Phyllanthus species, Phyllanthus ussuriensis (Euphorbiaceae) were used and the studies of constituents in this plant were carried out. From the aqueous fraction of methanolic extract, one flavonoid (quercetin-3-O-rutinoside), two gallotannins (gallic acid, methyl gallate), and two ellagitannins (corilagin, geraniin) were isolated through fractionation and repeated column chromatography using Amberlite XAD-4, ODS gel, and sephadex LH-20. The structures of these compounds were identified on the basis of spectroscopic evidences.

  • For the investigation of medicinal resource in Phyllanthus ussuriensis, the studies were conducted to evaluate the pharmaco-constituents in Phyllanthus ussuriensis, which is used as folk medicine in China. From the hot water extract of leaves, three phenolic compounds were isolated and identified as phloroglucinol, gallic acid and rutin by physico-chemical properties and spectroscopic evidences$(IR,{\;}^1H-NMR,{\;}^{13}C-NMR{\;}and{\;}Mass,{\;}etc.)$.

  • Phyllanthus ussuriensis Rupr. et Maxim. (Euphorbiaceae) has long been used in folk medicine to treat kidney and urinary bladder disturbances, intestinal infections, diabetes, and hepatitis. Reported chemical constituents of this species are one flavonoid (rutin), two gallotannins (gallic acid, methyl gallate) and two ellagitannins.An investigation of the n-BuOH fraction of P. ussuriensis led to the isolation of two new megastigmane glycosides, 10-hydroxy-4,7-megastigmadiene-3-one-9-O-b-D-glucopyranoside (1), 10- hydroxy-4,6-megastigmadiene-3-one-9-O-b-D-glucopyranoside (2) and two known compounds roseoside (3) and 3-oxo-a-ionol-9-O-b-D-glucopyranoside (4).
