Ozone sensitivity of physiological indicators and the difference of ozone tolerance on 4 families of Quercus aliena seedlings were investigated on the basis of the standardized physiological indicators. Photosynthetic parameters, photosynthetic pigment and malondialdehyde (MDA) content, and antioxidative enzyme activities were measured or analyzed from the leaves of Q. aliena seedlings at the end of ozone fumigation, and ozone tolerance indices among 4 families were calculated with the standardized physiological parameters. After ozone treatment, the reduction of carboxylation efficiency was observed in the leaves of four families, and their reduction were ranged from -24.1% to -56.9% of control seedlings. Photosynthetic pigment content differed significantly among 4 families and treatments. The reduction of total chlorophyll content showed the highest in family SU4 (-40.6%) and the lowest family US2 (-18.8%). Ascorbate-peroxidase (APX) activity showed significant difference among families and treatments, and increased as compared with control in three families, except for family US2. On the basis of the physiological indices, ozone tolerance of four families was ranked in the order of US1 > SU4 > US2 > SU1. In conclusion, photosynthetic parameters, pigment content and APX activity were recommended as appropriate indicators to assess the tolerance against ozone stress of Q. aliena.
American sycamore seedlings were grown in chambers with two different ozone concentrations ($O_3$-free air and air with additional $O_3$) for 45 days. Both the control and the $O_3$ chambers included non-fertilized and fertilized plants. After 18 days of $O_3$ fumigation, seedlings were placed in a clean chamber for 27 days. Seedlings under ozone fumigation showed a significant decrease in pigment contents and photosynthetic activity, and a significant increase in lipid peroxidation. Fertilization enhanced physiological damage such as the inhibition of photosynthetic activity and the increase of lipid peroxidation under ozone fumigation. During the recovery phase, the physiological damage level of seedlings increased with ozone fumigation. In addition, physiological damage was observed in the fertilized seedlings. Superoxide dismutase (SOD) and glutathione reductase (GR) activities of $O_3$-treated seedlings increased up to 33.8% and 16.3% in the fertilized plants. The increase of SOD activity was higher in the fertilized plants than in the non-fertilized plants. Negative effects of ozone treatment were observed in the biomass of the leaves and the total dry weight of the fertilized sycamore seedlings. The $O_3$-treated seedlings decreased in stem, root and total dry weight, and the loss of biomass was statistically significant in the fertilized plants. In conclusion, physiological disturbance under normal nutrient conditions has an effect on growth response. In contrast, in conditions of energy shortage, although stress represents a physiological inhibition, it does not seem to affect the growth response.
The deteriorative effect of ultraviolet-B(UV-B) radiation on photosynthesis was assessed by the simultaneous measurement of O$_2$ evolution and chlorophyll(Chl) fluorescence in green pepper. UV-B was given at the intensity of 1 W$.$m$\^$-2/, a dosage often encountered in urban area of Seoul in Korea, to detached leaves. Both Pmax and quantum yield of O$_2$ evolution was rapidly decreased, in a parallel phase, with increasing time of UV-B treatment. Chl fluorescence parameters were also significantly affected. Fo was increased while both Fm and Fv were decreased. Photochemical efficiency of PSII(Fv/Fm) was also declined, although to a lesser extent than Pmax. Both qP and NPQ were decreased similarly with increasing time of UV-B treatment. However, PS I remained stable. The addition of lincomycin prior to UV-B treatment accelerated the decline in Fv/Fm to some extent, suggesting that D1 protein turnover may play a role in overcoming the harmful effect of UV-B. The amount of photosynthetic pigments was less affected than photosynthetic response in showing decline in Chl a and carotenoids after 24 h-treatment. Presumptive flavonoid contents, measured by changes in absorbance at 270 nm , 300 nm and 330nm, were all increased by roughly 50% after 8 h-treatment. Among antioxidant enzymes, activities of catalase and peroxidase were steadily increased until 12h of UV-B treatment whereas ascorbate perxidase, dehydroascorvate reductase and glutathione reductase did not show any significant change. The results indicate that deteriorative effect of UV-B on photosynthesis precedes the protection exerted by pigment synthesis and antioxidant enzymes.
The response of Spirulina (Arthrospira) platensis to high salt stress was investigated by incubating the cells in light of moderate intensity in the presence of 0.8 M NaCl. NaCl caused a decrease in photosystem II (PSII) mediated oxygen evolution activity and increase in photosystem I (PSI) activity and the amount of P700. Similarly maximal efficiency of PSII (Fv/Fm) and variable fluorescence (Fv/Fo) were also declined in salt-stressed cells. Western blot analysis reveal that the inhibition in PSII activity is due to a 40% loss of a thylakoid membrane protein, known as D1, which is located in PSII reaction center. NaCl treatment of cells also resulted in the alterations of other thylakoid membrane proteins: most prominently, a dramatic diminishment of the 47-kDa chlorophyll protein (CP) and 94-kDa protein, and accumulation of a 17-kDa protein band were observed in SDS-PAGE. The changes in 47-kDa and 94-kDa proteins lead to the decreased energy transfer from light harvesting antenna to PSII, which was accompanied by alterations in the chlorophyll fluorescence emission spectra of whole cells and isolated thylakoids. Therefore we conclude that salt stress has various effects on photosynthetic electron transport activities due to the marked alterations in the composition of thylakoid membrane proteins.

This study was performed to investigate the physiological responses of Oplopanax elatus by water condition. Drought stress was induced by withholding water for 26 days. The results show that $P_{N\;max}$, SPAD, gs, E and Ci were significantly decreased with decreasing of soil moisture contents. However, AQY and WUE were decreased slightly only at 26 day. This implies that photosynthetic rate is reduced due to an inability to regulate water and $CO_2$ exchange through the stomatal. According to JIP analysis, ${\Phi}_{PO}$, ${\Psi}_O$, ${\Phi}_{EO}$ and $PI_{ABS}$ were dramatically decreased at 21 day and 26 day, which reflects the relative reduction state of the photosystem II. On the other hand, the relative activities per reaction center such as ABS/RC, TRo/RC were significantly increased at 26 day. Particularly, Dio/RC and DIo/CS increased substantially under drought stress, indicating that excessive energy was consumed by heat dissipation. These results of chlorophyll a fluorescence show that the sensitivity changes photosystem II activity. Thus, according to the results, O. elatus was exhibited a strong reduction of photosynthetic activity to approximately 10% soil moisture contents, and JIP parameters could be useful indicator to monitor the physiological states of O. elatus under drought stress.
Heavy metal pollution in soil, such as lead contamination, has become an area of interest in Korea because of urbanization and atmospheric deposition from neighboring countries. Therefore, in this research, eco-physiological responses such as chlorophyll contents, antioxidant enzyme activity, photosynthetic rate, biomass and phytoaccumulation abilities were investigated for 4 commonly used native roadside tree species to suggest suitable tree species to cope with lead contamination. The target species, Ginkgo biloba, Prunus yedoensis, Zelkova serrata and Chionanthus retusus showed lead toxicity by significant changes of chlorophyll contents and antioxidant enzyme activities on treatments over 200 mg Pb/kg. However, biomass and photosynthetic rates only showed significant responses of plants in the highest level (5,000 mg/kg) treatment. Especially, G. biloba did not show any significant changes of antioxidant enzyme activity, photosynthetic rate, and biomass even in the highest level treatment. In low level - environmentally realistic treatments, G. biloba and P. yedoensis showed the highest phytoaccumulation rate of lead from soil. Selecting and planting species like G. biloba which have good phytoaccumulation abilities and resistance to lead contamination by further research will be required to deal with emerging lead contamination.
Diurnal acid fluctuation, stomatal resistance, and solar radiation with regard to the cladode orientation were investigated in Opuntia lanceolata Haw. growing at WPeolryeong-ri, Hallim-eup, Chejudo, Korea. Diurnal changes of titratable acidity showed the typical CAM pattern in all investigated cladodes. Water tissue in the cladode had the same pattern of acid fluctuation as mesophyll tissue. Stomatal resistance was low during the night, increased rapidly to be a peak right after sunrise and decreased again thereafter. The southern side of the cladode showed higher stomatal resistance than the northern side during the day time. It suggests that the stomata of the northern side opens under diffuse radiation. The amount of solar radiation varied depending upon the cladode orientation. It is thought that C4 acids move inter and intra mesophyll tissues in the cladode through the unknown pathways. RuBP carboxyulase activity in the cladode was very high at 14:00, but was not significant at 01:00. PEP-carboxylase had high activities both at 14:00 and at 01:00. The results of this study showed the possibility that O. lanceolata Haw. had the C3, C4 and CAM photosynthetic patterns under the environmental conditions at Weolryeong-ri.
Highly stable $O_2$-evolving cells and thylakoid membranes have been obtained from the cyanobacterium, Synechococcus PCC7002, by immobilization with polyvinylalcohol(PVA). The absorption peak showed the blue-shift of about 3 nm after immobilization of intact cells and thylakoid membranes as well as isolation of thylakoid membranes. Photosynthetic electron transport activities, especially PS II activity showed greater stability in the PVA-immobilized cells and thylakoid membranes when stored at $4^{\circ}C$ than in those at $25^{\circ}C$. When the cells were threated at higher temperature, the level of Fo and Fv increased. After imobilization, however, Fo showed no change. This suggests that the immobilization can protect against the damages of PS II complex, especially a water-spiliting system, by heat treatment.

Seedlings of Pterocarpus indicus were grown in both well-watered and drought stress conditions in phytotron. Seedlings grown under well-watered and drought stress conditions were exposed to either combined or without ozone of 200 ppb for one month. First, the physiological responses to elevated ozone levels indicated a decreased biomass. The seedlings grown in arid soil and exposed to ozone showed less biomass than those grown in arid soil but not exposed to ozone. Moreover, all the seedlings except the well-watered and unexposed ones showed a significantly lower photosynthetic rate ($P_N$) over time. However, with the accumulation of ozone injuries, the antioxidant enzyme activities increased overall. In the study results, when exposed to ozone, the well-watered seedlings exhibited more antioxidative enzyme activity than did the seedlings grown in arid soil. Generally, P. indicus in arid soil suffered less damage from elevated ozone than did the well-watered plants.

Influence of gamma rays on the cyanobacterium Synechocystis sp. PCC 6803 cells was investigated in terms of a bidirectional hydrogenase, which is encoded by hoxEFUYH genes and responsible for biohydrogen production. Irradiated cells revealed a substantial change in stoichiometry of photosystems at one day after gamma irradiation at different doses. However, as evaluated by the maximal rate of photosynthetic oxygen evolution, maximal photochemical efficiency of photosystem II, and chlorophyll content, net photosynthesis or photosynthetic capacity was not significantly different between the control and irradiated cells. Instead, transcription of hoxE, hoxH, or lexA, which encodes a subunit of bidirectional hydrogenase or the only transcriptional activator, LexA, for hox genes, was commonly enhanced in the irradiated cells. This transcriptional enhancement was more conspicuously observed immediately after gamma irradiation. In contrast, hydrogenase activities were found to somewhat lower in the irradiated cells. Therefore, we propose that transcription of hox genes should be enhanced by gamma irradiation in a LexA-mediated and possibly photosynthesis-independent manner and that this enhancement might not induce a subsequent increase in hydrogenase activities, probably due to the presence of post-transcriptional and/or post-translational regulatory mechanisms.
