In order to investigate biochemical activities of harvested tobacco leaves, photosynthetic rate, soluble protein contents, and peroxidale activities were analysed during different maturing period. Physiological activities of harvested leaves during maturing period were higher in topped than those of non-topped plants. Chlorophyll content and photosynthetic rate in both topped and non-topped plants decreased at 4 days and 3 days before harvest, respectively. The chloroplast numbers in topped and non-topped plants decreased at 3 days and 5 days before harvest, respectively. Changes of soluble protein and total RNA contents showed similar patterns during maturing period. Soluble protein contents were slightly decreased from 5 days before harvest in topped plants, but decreased drastically from 3 days before harvest in non-topped plants. Not much changes were found in total RNA contents in topped plants until 2 days before harvest, and it was largely decreased after 5 days before harvest in non-topped plants. The peroxidase activities drastically decreased in topped plants and increased in non-topped plants after 3 days before harvest during maturing period. The largest change of biochemical activities in tobacco leaves during maturing periods were observed at 3 days before harvest.
To investigate the influence of the mesophyll cells on stomatal opening in response to white light, the segments of isolated epidermis were transferred on partly exposed mesophyll cells of a leaf and stomatal apertures were measured. Transferring the isolated epidermis on partly exposed mesophyll cells of a leaf caused a marked increase on stomatal apertures while stomata in isolated epidermis incubated in MES buffer hardly opened. Mesophyll infiltration with photosynthetic inhibitors (DCMU, DCCD, $NaN_3$) was performed to elucidate the correlation between stomatal apertures and the degree of photosynthetic activity. It was found that transferring the isolated epidermis on partly exposed mesophyll cells of a leaf caused an increase of stomatal apertures depending on the degree of photosynthetic activities. In $NaN_3$ infiltrated leaf discs, transferring the fresh isolated epidermis on partly exposed mesophyll cells of a leaf showed no significant effect, but a slight increase on stomatal apertures. Isolated epidermis alone did not respond to the light properly, but if it was closely contacted with mesophyil cells, the stomata regained the ability of the light response. Therefore, it could be suggested that stomatai apertures were related with the degree of photosynthetic activity in the mesophyll cells.
In order to characterize the chloroplasts of Korean ginseng as a semi-shade plant and radish as a sun plant, effects of growth light intensity on photosynthetic electron transport (PS) activity in chloroplasts and superoxide (O2.-) production in thylakoid membrane by irradiation were investigated. High-light chloroplasts of both plants showed higher PS activities than those grown under ow growth light intensity. High PS II and low PS I activities in ginseng chloroplasts (ratio of PS II/PS I : 1.1) were observed, but radish chloroplasts showed low PS II and high PS I activities (ratio of PS II/PS I : 0.3). PS II activity of both plants was little affected by temperature in range of 15-35$^{\circ}C$. Activities of whole -chain (PS II+I) in ginseng and PS I in radish were increased at high temperature (4$0^{\circ}C$). Preincubation of chloroplasts at 4$0^{\circ}C$ during 30 min, as a mild heat stress, caused rapid decrease in PS II and PS II+I activities of both plants. However PS I activity was not decreased in ginseng and rather increased in radish. O2.- production (NBT reduction) in Mehler reaction in the thylakoid membrane was inhibited by DCMU in both plants. DMBIB inhibited O2.- production in ginseng, but radish was insensitive to DMBIB. Electron flow system in ginseng thylakoid membrane was more susceptible to damage of photooxidation than that of radish.
덩굴식물 개머루, 계요등, 까마귀머루, 담쟁이덩굴, 댕댕이덩굴, 인동덩굴, 하늘타리 등 7수종과 덩굴식물이 아닌 쇠뜨기, 쑥 등 2수종을 이용하여 광색소 함량과 SOD활성에 대하여 분석한 결과는 다음과 같다. 100 ppb 처리구에서 볼 때 엽록소 a, 엽록소 b, 총 엽록소 함량, 카로테노이드에서 담쟁이덩굴이 가장 많이 증가하였으며, 엽록소 b와 a의 비에서는 쇠뜨기, 카로테노이드와 총 엽록소의 비에서는 계요 등에서 가장 많이 증가하였다. 덩굴식물과 덩굴식물이 아닌 쇠뜨기, 쑥의 비교에서는 엽록소 b와 엽록소 a의 비에서 확실한 차이를 보였다. 엽록소 a와 b의 비에서보면 엽록소 b보다 엽록소 a가 더 민감한 차이를 보였으며, 수종에서는 담쟁이덩굴, 덩굴식물과 덩굴식물이 아닌 쇠뜨기, 쑥의 비교에서는 덩굴식물이 민감한 것으로 나타났다. SOD 활성에서는 100 ppb 처리구에서 덩굴식물류 7수종과 쇠뜨기, 쑥의 처리전과 처리후의 차이는 쑥에서 3535.7 unit/g로 매우 크게 증가하였으며, 계요 등이 109.3 unit/g로 가장 적게 증가하였고, 개머루 한 수종에서 131.7 unit/g로 감소했다.
본 연구는 오존에 노출된 회화나무 8 가계에서 측정한 생리적 지표들을 대상으로, 오존에 대한 민감성을 평가하고, 이 결과들을 종합하여 가계 간 차이를 구명하고자 실시하였다. 회화나무(Sophora japonica L.) 8가계를 대상으로 오존 처리 후, 광합성 특성, 엽록소 함량, 항산화효소 활성 및 MDA 함량을 측정하였고, 각 측정 지표들의 표준화 지수를 이용하여 내성 순위를 결정하였다. 탄소고정효율은 6번 가계를 제외한 모든 가계에서 오존 처리 후 뚜렷하게 감소하였으며, 탄소고정효율의 감소율은 -38%에서 -67% 범위를 보였다. 또한 순양자수율은 7번 가계를 제외한 모든 가계에서 오존 처리 후 뚜렷한 감소를 보였으며, 감소율은 -27%에서 -61%를 나타냈다. 엽록소 a를 비롯한 광색소 함량의 가계 간 차이는 컸으나, 오존 처리 효과는 나타나지 않았다. 오존 처리 후, SOD 활성의 증가율은 7%에서 64%로, 가계 간 차이가 뚜렷하였다. APX 활성은 모든 가계에서 오존 처리 후 증가하였으며, 7번 가계가 가장 큰 활성 증가율(218%)을 보였다. 표준화 지수를 기준으로 할 때, 6번 가계의 내성 수준이 가장 낮았으며, 7번 가계의 내성 수준이 가장 높았다. 6번 가계는 오존 처리 후 탄소고정효율을 비롯한 광합성 특성의 감소율과 광색소 함량의 감소율이 높게 나타난 반면, 내성을 나타내는 항산화효소의 활성 증가가 매우 적었다. 반대로 내성 수준이 높은 7번 가계는 오존 처리 후 광합성 특성의 감소율이 다른 가계에 비해 적었으며, 광색소 함량은 오히려 오존처리 후 증가하였고, 항산화효소의 증가율도 다른 가계에 비해 높게 나타났다.
As a mean to recover photosynthetic bacterial(PSB) cells and its practical uses in food industrial wastewater treatment, various biodegradable polyelectrolytes were first investigated for flocculation of suspended colloids in the PSB treatment process of soybean curd wastewater. Anionic polyelectrolytes such as sodium alginate and carrageenan were not effective but a cationic polyelectrolyte chitosan isolated from Portunus trituberclatus showed very effective flocculation activity. The concentration of chitosan, pH and temperature of wastewater for maximal flocculation were 40 mg/l, pH 7 and room temperature, respectively. Test using deacetylated chitosan to various degree showed higher flocculating activities in samples deacetylated over 75% and time for maximum flocculation was 40 min by stirring slowly under the above optimal conditions. Chitosan was not only effective to flocculate cells but also removed COD and MLSS of the wastewater. COD of 42% and MLSS of 87% were removed by addition of chitosan to the soybean curd wastewater treated with PSB.
Correlative changes between photosynthetic O2 exchange rates and room temperature Chl fluorescence were investigated in wheat (Triticum aestivum L.) chloroplasts treated with high temperature for 5 min. With increasing treatment temperature, photosynthetic O2 evolution rate mediated by PSII was decreased, showing 50% inhibition at 38$^{\circ}C$ (I50). But PSI activity measured by O2 uptake rates was stimulated as a function of increasing temperature. Dark level fluorescence (Fo)-temperature (T) analysis showed that fluorescence rising temperature (Tr), critical temperature (Tc), and peak temperature (Tp) was 38, 43, and 52$^{\circ}C$, respectively. Quenching analysis of Chl fluorescence showed that both the oxidized fraction of plastoquinone (qQ) and degree of thylakoid membrane energization (qNP) increased up to 4$0^{\circ}C$ and then declined dramatically. These results suggest that Tr is correlated with temperature showing a 50% of inhibition of photosynthesis and under mild high temperature stress, qNP is worth regarding as indicator for heat-induced damage of photosynthesis.
To study the effect of IAA on the growth of Chlorella, the alage wre cultured on the media for six days by bubbling $_{2}$ enriched air under 10K lux at 20-$25^{\circ}C$. The culture media were made by adding a concentration of $10^{-3}$M, $10^{-4}$M, and 0M(as a control) IAA to the standard media. During the period of culture, Chlorella was smapled for the given time of interval and photosynthetic and respiratory activities were measured by Warburg manometer and change of chemical components of Chlorella was determined by spectrophotometry after the Chlorella cell was fractionated by Schmidt-Thannhauser method. 1) Photosynthetic and respiratory activities were enhanced by IAA ; especially the enhancement of respiratory activity was so remarkable. 2) As to the chemical components of Chlorella, carbohydrates and amino acids were reduced a little but phosphate, RNA, DNA, and protein were increased by $10^{-3}$M IAA ; the increase of RNA, in particular, was noticable. 3) The above results suggest that the enhancement of growth of Chlorella, by IAA and ATP induced by respiratory activity accelerated with IAA enhanced RNA synthesis, resulting in an increase of protein synthesis.
Edible canna is a productive starch source in some tropical and semitropical regions. In these regions, water deficit stress is one of factors that limit the crop yield. In the present study, we investigated seven physiological indexes and photosynthetic responses of three edible canna (Canna edulis Ker.) cultivars ('PLRF', 'Xingyu-1', and 'Xingyu-2') under 35 days drought stress. Our results indicated that drought treatment caused visible wilting symptoms in all cultivars, especially in 'Xingyu-1'. Coupled with the increase of wilting symptoms, relative water content (RWC) and chlorophyll content decreased progressively, malondialdehyde (MDA) content gradually increased, and key antioxidant enzymes such as superoxide dismutase (SOD), peroxidase (POD), catalase (CAT) activities increased first and then decreased in all three cultivars. The effect of water stress was more pronounced in 'Xingyu-1' than in 'PLRF' and 'Xingyu-2', and in lower leaves than in upper leaves. In addition, 35 days drought stress also significantly reduced the photosynthetic capacity. Consistent with antioxidant parameters, photosynthetic changes of 'Xingyu-2' were less than those of the other cultivars under water deficit stress. Drought stress caused a significant increase of water use efficiency (WUE) in 'Xingyu-2', but little in 'PLRF', and obvious decrease in 'Xingyu-1'. These results indicated that 'Xingyu-2' was more tolerant to drought stress than 'PLRF' and 'Xingyu-1' by maintaining lower lipid peroxidation and higher antioxidant enzyme activities.
