  • To examine whether DGRF inhibits $cPLA_2$ activity in vitro, we purified a 100 kDa $cPLA_2$enzyme from porcine spleen and performed an inhibition study at two concentrations of 5.0 and 50.0 $\mu$M 1-stearoyl-2-[1-$^{l4C}$ ]arachidonoyl-sn -glycero-3-phosphocholine as a substrate to rule out an apparent inhibition due to "substrate depletion". Here we reported that DGRF inhibited $cPLA_2$activity with $ID_{50}$ of 3.2 $\mu$M and virtually complete inactivation of the enzyme occurred at 60 $\mu$M. Interaction experiment between enzyme protein and inhibitor by ultrafiltration method indicated that 1-desgalloylrugosin-F inactivates $cPLA_2$enzyme by an irreversible mechanism.

  • Activation of the T lymphocytes results in a variety of early biochemical events ultimately leading to cell proliferation and lymphokine production. Stimulation of the signal transduction cascade in T cells through the T cell receptor coincides with activation of the phosphatidylinositol-phospholipase C (PI-PLC) pathway. Therefore, we have established a model system to screen immune-simulator that can increase the hydrolysis of phosphoinositides in human T cell leukemia Jurkat cells. As a result of screening from herbal medicine extract, 4 extracts (O1ibanum, Ephedrae Herba, Real Gar, Saussureae Radix) were found 14 increase the production of inositol phosphates. All the active fraction from the four kinds of extract were fluted in a different retention time on C-18 HPLC and these active fraction also showed difference in cell specificity. And all the active fractions increased DNA synthesis in T cell. Therefore, it is suggested that the active fraction among 4 extracts might contain a compound having different properties one another.

  • Phospholipase A$_2$(PLA$_2$) catalyzes the hydrolysis of the sn-2 position of membrane glycerophospholipids to liberate arachidonic acid(AA), a precursor of eicosanoids including prostaglandins(PGs) and leukotrienes (LTs). The same reaction also produces lyso-phospholipids. So far, at least 19 enzymes that possess PLA2 activity have been identified, consists of low-molecular-weight, Ca$\^$2+/-requiring, secretory enzymes that have been implicated in a number of biological processes, such as modification of eicosanoid generation, inflammation, host defense, and atherosclerosis. • The present study was conducted to assess the effect of total saponins from Korean red ginseng on the biosynthesis of inositol phospholipids in vivo and also effects on the metabolic enzymes, such as phosphatidylinositol-specific phospholipase C(Pl-PLC) and PI-kinases. The administration of 0.1% saponin solution, 0.1 ml 3 times a day intraperitoneally to 5 mice for 30 days has increased a 23% of the body weight when it compared with a control group. The amounts of 32P-phoschorus radioactivity incorporated into the phosphoinositides from the liver and brain tissues have increased a 310% and 260%, respectively, in the saponin treated mice. The activities of PI-PLC from liver and brain were stimulated in the various amounts by the conditions treated with saponins. The PI-kinases from liver and brain were also activated by saponins, but its effect was lower than that of PI-PLC. From these results, it was confirmed that red ginseng saponins have affected positively not only on the biosynthesis of phosphoinositides but also on the PI-PLC and PI-kinases related to the cellular signal transduction.

  • Chryseobacterium mulctrae KACC 21234T is a novel species isolated from raw bovine milk. Psychrotrophic bacteria are considered contaminants and are hypothesized to originate from the environment. In this investigation, the C. mulctrae KACC 21234T genome was determined to be 4,868,651 bp long and assembled into four contigs with a G+C ratio of 33.8%. In silico genomic analyses revealed the presence of genes encoding proteases (endopeptidase Clp, oligopeptidase b, carboxypeptidase) and lipases (phospholipase A(2), phospholipase C, acylglycerol lipase) that can catalyze the degradation of the proteins and lipids in milk, causing its quality to deteriorate. Additionally, antimicrobial resistance and putative bacteriocin genes were detected, potentially intensifying the pathogenicity of the strain. The genomic evidence presented highlights the need for improved screening protocols to minimize the potential contamination of milk by proteolytic and lipolytic psychrotrophic bacteria.

  • To examine the localization pattern of phospholipase D2 (PLD2) in the pancreatic islet (the islet of Langerhans) depending on species, we conducted a morphological experiment in the rat and guinea pig. Since individual islets display a typical topography with a central core of B cell mass and a peripheral boundary of A, D, and PP cells, double immunofluorescent staining with a panel of antibodies was performed to identify PLD2-immunoreactive cells in the islets PLD2 immunoreactivity was mainly present in A and PP cells of the rat pancreatic islets. And yet, in the guinea pig, PLD2 immunoreactivity was exclusively localized in A cells, and not in PP cells. These findings suggest a possibility that PLD2 is mainly located in A cells of rodent pancreatic islets, and that the existence of PLD2 in PP cells is not universal in all species. Based on these results, it is suggested that PLD2 may play a significant role in the function of A and/or PP cells via a PLD-mediated signaling pathway.

  • Phospholipase A$_2$(PLA$_2$) comprise a family of enzymes that hydrolyze the acyl bond at the sn-2 position of phospholipids to generate free fatty acids including arachidonic acid and lysophospholipids. Distinct forms of PLA$_2$are involved in digestion, inflammation, and intercelluar-and intracellular signaling pathways. The released arachidonic acid, which is enriched at the sn-2 position, serves as the precursor for eicosanoids such as prostaglandins and leukotrienes. During oxygenation of arachidonic acid to hydroxy endoperoxide, reactive oxygen radicals are generated. On the other hand, lysophospholipids increase membrane fluidity and can be cytotoxic with its detergent-like action. Thus, the biochemical features of the products of PLA$_2$activity suggest that PLA$_2$may be implicated in many destructive cellular processes.

  • Because of the potent effects of lipid mediators such as prostaglandins (PGs), leukotriens (LTs) and platelet activating factor (PAF) on a variety of cells and tissues, they are considered as major contributors to the process leading to inflammation and allergy. To pursue the mechanism of anti-inflammatory activity of Lonicera japonica, we tested inhibitory effects of 7 flavonoids from Lonicera japonica on arachidonic acid cascade related enzymes, such as inflammatory phospholipase $A_2$, cyclooxygenase-1 and 2, 5-lipoxygenase, in bone marrow derived mast cell (BMMC), and lyso PAF-acetyltransferase in rat spleen microsomes. Anti-inflammatory activities of lonicera japonica are thought to be attributed at least in part to the inhibition of arachidonic acid cascade releated enzymes by flavonoids such as apigenin, luteolin quercetin.

  • A series of phosphodiesters of p-nitrophenyl phosphoryl derivatives were synthesized and used as a model substrate for phospholipase D (PLD). The phosphodiester substrates were synthesized from p-nitrophenyl phosphorodichloridate and corresponding alcohols with different chain lengths and polar groups. To measure the activity of PLD, either spectroscopic method for p-nitrophenol or pH-stat titration method was employed. For each substrate, effects of substrate concentration, pH, and $Ca^{2+}$ ion were examined. The kinetic parameters $V_{max}$ for the different substrates were varied depending on the chain lengths or charge of the alcohols. No calcium effect was observed in the hydrolysis of neutral and negatively charged alcohol derivatives, while positively charged choline derivative showed a strong $Ca^{2+}$ ion dependence.
