Incorporation of phosphate into protein and other nitrogenous compounds in Chlorella cells (Chlorella 세포에서의 $^{32}P$-인산의 단백질 및 다른 질소화합물로의 전환)

  • In the process of the incorporation of orthophsphate into protein and other cell constituents, the role of inorganic polyphosphate and RNA-polyphosphate complex and the correlation between them were pursued by analyzing the contents of $^{32}P$ and total P in various fractions of Chlorella cells, which had been uniformly labeled with $^{32}P$ before the inoculation in a normal "cold" medium or P-free medium during the culture. The effects of ionizing radiation and various micronutritional-element deficiencies on the phosphate incorporation into, and biosynthesis of, protein and other introgenus compounds in the cells were also observed. When the uniformly $^{32}P$-labeled algae were grown in a normal "cold" medium the contents of $^{32}$ P in the fractions of protein, DNA and RNA-polyphosphate complex increased, but those in the fraction of acid-insoluble polyphosphate decreased. On the other hand, amount of $^{32}P$in the fraction of RNA was almost unchanged in spite of rapid increase of the total P. In the growing period of $^{32}P$-labeled algae in a P-free medium, amounts of $^{32}P$ in the fractions of DNA, protein and lipid increased, while those in the fractions of RNA-polyphosphate and inorganic polyphosphates decreased. When the algal cells were irradiated with about 70, 000r of gamma-rays before the inoculation in the medium, amounts of phosphate in the fractions of DNA, RNA, nucleotides and protein decreased during the culture, compared with those of the control. However, the phosphate content in the fraction of acid-insoluble polyphosphate of the irradiated cells increased than those of the control. In the growing period of the algae in a Mo-free, medium, amounts of acid-soluble total phosphate and nucleotides of the cells increased, while the amounts of residual protein and RNA decresed compared with those of the normal cells. Amounts of alkali-labile protein and phospholipid of the cells grown in a B-free medium decreased, whereas amount of phosphate in acid-soluble fraction increased compared with the control. In general, the contents of protein and RNA in each microelement deficient cells decreased more or less, compared with those in the normal cells.in the normal cells.

  • In order to interpret the effect of IAA on the phosphate metabolism and biosynthesis of organic compounds, Saccharomyces uvarum were cultured in the media treated with various concnetration of IAA $(10^{-3}M,\;10^{-5}M,\;10^{-7}M)$. Sampling at the beginning and intervals of culture, yeast cells fractionated were traced the contents of inorganic phosphate and organic compounds of various fractions. 1. Growth of Saccharomyces uvarum were enhanced by IAA $(10^{-3}M,\;10^{-5}M)$ and phosphate contents in DNA and RNA fractions treated with IAA were accelerated 2.3 times and 2 times in comparison with those of control. 2. Amounts of poly-P"A" and poly-p"B" were increased but poly-P"C" decreased during the culture. Therefore, it is considered that poly-P"C" play on most important role as a phosphate pool. 3. It is suggested that because phosphate contents in DNA, protein and lipid fractions increased, inorganic phosphates required phosphates required RNA were transferred from phosphates in cytoplasm, because these increased slowly during the culture. 4. Alkali-labile protein were accelerated by IAA and alkali stable protein only were inhibiction were enhanced by IAA while, ethanol : ether soluble fraction was induced by $10^{-7}M$ IAA in comparison with those control.X> IAA in comparison with those control.

Turnover of Phosphate Compounds in Chlorella cells in a P-free medium (인산결핍배지에 있어서의 Chlorella 세포내의 인산화합물의 전환)

  • Using the Chlorella cells which had been uniformly labeled with $^{32}P$, the distribution of phosphorus in various fractions of cell material was investigated. Uniformly $^{32}P$-labeled Chlorella cells were further grown in a P-free medium, and some protions of the cells were taken out at intervals during the culture, and subjected to analyze the contents of $^{32}P$ in various fractins of the cell constituents. 2. Analysis of the $^{32}P$-labeled Chlorella cells showed that the highest in P-content was the fraction of RNA followed by those of lipid, RNA-polyphosphate complex, acid-insoluble polyphosphate, acid-soluble polyphosphate, DNA and protein. 3. During the culture of $^{32}P$-labeled Chlorella cells in a P-free medium, amounts of phosphate in DNA, protein and lipid fractions increased, while the P-contents in the fraction of RNA-polyphosphate complex decreased as well as those of acid-insoluble polyphosphate and acid-soluble polyphosphate fractions. 4. It was inferred that phosphorus used in the syntheses of DNA and protein was taken from polyphosphates of the cells, and RNA-polyphosphate complex would play an important role as a phosphate pool.

Effect of Triol and Diol Fractions of Ginseng Saponin on Glutamine Transport into Rat Renal Cortical Mitochondria (인삼의 Triol 및 Diol계 사포닌이 쥐의 신피질 미토콘드리아 의 Glutamine 이동에 미치는 영향)

  • Attempts were made if diol and triol fractions of ginseng saponin affect on glutamine transport into rat renal cortical mitochondria, swelling, phosphate dependent glutaminase activity, and consumption of oxygen. The following results were obtained. When mitochondrial preparation from rat renal cortex was incubated in medium containing 14C-glutamine and either triol or diol fractions, radioactivity was shown to increase at both 10-6% and 10-5% triol fractions of ginseng saponin, but reduce in case of diol fraction. The remarkable acceleration of the rate of swelling of renal cortical mitochondria was observed in the presence of 10-1% trios and diol fractions but no accerelation at lower concentrations. The activity of phosphate dependent glutaminase from renal cortical mitochondria was slightly activated at 10-2% of triol fraction. However, there was no effect in case of diol fraction. Oxygen consumption by mitochondria from renal cortex was remarkably increased at concentrations of 10-5% and 10-6% triol fractions, but reduced in the case of diol fractions. On the basis of these observations it was concluded that triol fraction of ginseng saponin might increase the transport of glutamine into mitochondria by accelerating the respiratory chain and supplying additional energy to mitochondria, and physiological role of triol fraction was entirely different from that of diol fraction of ginseng saponin.

Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate (Chlorella의 인산대사에 관한 연구)

  • Yung Nok Lee (Dept. of Biology, Korea University) : Studies on the phosphate metabolism in Chlorella, with special reference to polyphosphate. Kor. J. Microbiol., Vol.2, No.1, p1-11 (1964). 1. Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. According to these experimental results, it was inferred that polyphosphate "B" could transform into polyphosphate "C" in normal growing Chlorella cells.sults, it was inferred that polyphosphate "B" could transform into polyphosphate "C" in normal growing Chlorella cells.ing Chlorella cells.

Antidiabetic Effect of Lonicerae Japonica Thunb Extract (인동초(Lonicerae Japonica Thunb) 추출물의 혈당강하 효과)

  • This study was carried out to investigate the antidiabetic effect of Lonicerae japonica Thunb(LJ) in the streptozotocin(STZ)-induced diabetic rats. The effective fractions were prepared as a form of organic solvents of hexane, chloroform, ethylacetate, butanol, water fractions prepared from the ethanol extract of LJ. The content of serum glucose and the activities of glucose-6-phosphatase(G-6-Pase) in the hexane and water fractions treated rats were significantly decreased compared to those of the STZ control group. Whereas the activity of hepatic glucose-6-phosphate dehydrogenase(G-6-PDH) was significantly increased in the hexane and water fractions treated rats. In conclusion, these results indicated that the hexane and water fractions of LJ were effective for the antidiabetes in the STZ-induced diabetic rats.

Effect of magnesium-deficiency on the biosynthesis of nucleic acid, phosphoprotein, and phospholipid in Chlorella cells (Chlorella 세포의 핵산 인단백질 및 인지질의 생합성에 미치는 마그네슘의 결핍 효과)

  • Chlorella ellipsoidea were grown in a Mg-free medium. Aliquots of the algal cell were taken out at the beginning and predetermined time intervals during the culture and were analyzed the contents of phosphate in various fractions of the cell constituents. The results obtained were compared with those of the control. When Chlorella cells were grown in a Mg-free medium, the contents of phosphate in the DNA protein, RNA-olyphosphate complex, nucleotidic-lbileP, and PCA-soluble, fractions decreased compared with those of the control, while the content of acid insoluble polyphosphate increased significantly. On the otherhand, RNA-P and lipid-P showed the tendency of decrease at the early stage of the culture, but they were increased more than those in the control as culture proceeds. It is showed that phosphate turnover from acid-insoluble polyphosphate into DNA, protein, and RNA-polyphosphate complex was inhibited by magnesium-deficiency of the cells.

  • Weanling female Sprague Dawley rats were fed d diets containing 22mg pyridoxine. BCI/kg diet (control diet) and l.2mg pyridoxine. BCI/kg diet (deficient diet). One control group and one defi­c dent group were fed their diet throughout growth, g gestation and lactation. After the pups were born and weaned, the deficient group was divided into two groups. One switched to control diet(supple­I mented group) and the other continued the same d deficient diet( deficient group) until 10 week -old. The liver mitochondrial and cytosolic asparate a aminotransferase activity and pyridoxal phosphate content were determined in offspring rats. The aspartate aminotransferase activities in both liver mito$\phi$ondrial and cytosolic fractions of den­d cient group were significantly lower than those of controls, but there were no significant differences between two groups after addition of 1O^{-4}M pyri­d do뼈I phosphate to the medium. By pyridoxine s supplementation after weaning, the reduced aspar­a tate aminotnmsferase activities were only partialy I restored to control levels. The pyridoxal phospha­t te content of deficient group in Iiver mitochondr­ial and cytosoIic fractions were alo significantly different from those of controls, but readily restored by dietary supplementation. These results suggest that there is a quantitative and a qualitative changes of aspartate amino trans­f ferase and pyridoxal phosphate in liver mitochon­d drial and cytosolic fraction by long-term pyrido­x xine deficiency and these reductions can partially recovered by dietary pyridoxine supplementation after weaning.

Studies on the Phosphate Accumulation in Tobacco Production Area (연초경작지 토양의 인산 축적에 관한 연구)

  • The experiment was conducted to investigate accumulation o( phosphate in tobacco production area. The results are as follows: 1) The content of available P in burley production area was about 200ppm higher than that in flue-cured tobacco production area, the available P content of 20% tobacco fields surveyed was above 1000ppm P. 2) Tobacco fields having 200ppm of available P which were grouped as medium category for phosphate recommendation were more than 90% out of tobacco fields surveyed. 3) Total phosphate content was above 4000ppm in 42.5% burley tobacco fields before transplanting, and 2000~3000ppm in about 40% flue-cured tobacco fields. 4) Phosphate fractions in soil increased in order of Al-P > Fe-P > Ca-p, and their content was about 50% of total phosphate in tobacco fields surveyed. 5) pH value in most of tobacco fields surveyed ranged from 4.5 to 5.0 and, field soils with pH value below 5.5 were 64 %.

Characteristics of Bacillus sphaericus PSB-13 as Phosphate Solublizing Bacterium Isolated from Citrus Orchard Soil (감귤원 토양에서 분리한 인산염 가용화 미생물 Bacillus sphaericus PSB-13의 특성)

  • This study was carried out to measure insoluble phosphorus fractions content fixed in different soil type and isolate a superior phosphate solublizing bacteria(PSB) producing free phosphate in citrus orchard soil. Distribution of insoluble phosphate fraction ordered Al-P>Ca-P>Fe-P in the investigated citrus orchards. Insoluble phosphate fraction such as Al-P, Ca-P, Fe-P were higher in volcanic ash than in non-volcanic ash soil. A PSB with high holo zone in PDA-P medium isolated from citrus orchard soil. This strain identificated by MIDI system as Bacillus sphaericus. The optimum growth of pH and temperature were at 4~5, $30^{\circ}C$, respectively. When Bacillus sphaericus cultured at $25^{\circ}C$, 150 rpm condition in LB broth medium included different phosphate. Bacillus sphaericus produced free phosphate in the culture broth medium from tricalcium-phosphate(207.0 ppm), aluminium phosphate(324.5 ppm) and hydroxyapatite(334.8 ppm) and Phosphatase activity of Bacillus sphaericus was higher at $35^{\circ}C$ culture condition than that of $25^{\circ}C$. Two type preparation inoculated Bacillus sphaericus made with carrier materials such as Bentonite, $CaCO_3$, Sodium alginate. Density of PSB in this preparation conserved at $10^5c.f.u.\;g^{-1}$ level during storage in different temperature condition for 7 month. It also showed that free phosphate produced at PDA-P medium.
