Effects on osteoporosis of Senescence Accelerated Mice (SAM) P6 by Injection of Placenta extracts ($Beuronel^{(R)}$) (SAM p6 mouse에서 자하거 주사제의 골다공증 개선에 대한 연구)

  • Objectives : The purpose of this study was to examine the effects of the Hominis placenta extracts on improvement of senile osteoporosis (Type 2) in SAM P6. In traditional Oriental medicine, Placenta (Jahageo) is indicated for insufficiency of blood and tidal fever due to steaming bone disorder. Methods : At 10 weeks after birth, Placenta ex. was given 100 mL, 200 ml/kg/day, i.p. 3 times a week for 4 weeks in SAM P6 mice. And then we measured complete blood cells, blood urea nitrogen, creatinine, inorganic phosphate and total iron. In addition, we tested bone mineral density (BMD) using the soft X-ray. Results : As a result, in SAM P6, injection of Placenta ex. (200 mL) increased in RBC, HB and PLT, in comparison with control group. It was also found that the inorganic phosphate levels increased significantly in the injection groups of the Placenta ex. from that of the control group, but blood urea nitrogen was no significant. Moreover, Placenta ex. were showed a trend of increase in bone mineral density (BMD) of the vertebrae lumbales and tibia in P6 mice. Conclusions : These findings suggest that Placenta ex. are effective in preventing bone loss in SAMP6.

Enhanced Production of Avermectin B1a with Streptomyces avermitilis by Optimization of Medium and Glucose Feeding (배지 및 유가식 회분배양 최적화에 의한 Streptomyces avermitilist 의 Avermectin B1a 생산성 향상)

  • The effect of phosphate on the production of avermectin B1a was studied. Response surface methodology was applied to optimize the concentration of organic nitrogen sources. The portion of B1b in total avermectins was decreased from 5.8% to 3.0% by the addition of 1.5 g/ι inorganic phosphate to the production medium. Among organic nitrogen sources, soybean meal was the most effective on avermectin biosynthesis. Results showed that B1a productivity was increased by 44.8% in a laboratory scale fermenter cultivation of Streptomyces avermitilis YA99-40 through fed-batch process. A maximal B1a productivity was obtained by repeated 30 and 20 g/ι of glucose feeding at 136 and 206 hour, respectively. The B1a productivity was increased by 86.3% and the proportion of B1a in the total avermectins was improved from 38% to 45% with respect to the control process. These results would be very useful for enhancing productivity of B1a in an up-scaled processes.

Effects of Mixed Organic Fertilizer Application with Rice Cultivation on Yield and Nitrogen Use Efficiency in Paddy Field (벼 재배시 혼합유기질비료 시용이 질소이용율과 수량에 미치는 영향)

  • This study was carried out to investigate optimum application rate with mixed organic fertilizer for chemical fertilizer alternative where the paddy rice (Chucheong) was cultivated in clay loam of paddy field. The mixed organic fertilizer were applied at 0, 50, 100 and 150% levels of recommended nitrogen by soil testing plot compared with plot of chemical fertilizer (nitrogen, phosphate and potash), respectively. The ammonium nitrogen content in paddy soil and surface water of mixed organic fertilizer 100% plot were higher than chemical fertilizer plot. The absorbed amount of nitrogen and nitrogen use efficiency by rice plant in mixed organic fertilizer 100% plot were higher than chemical fertilizer plot. The number of tillers on rice at 30 days and 60 days after transplanting mixed organic fertilizer 100% plot were many more than chemical fertilizer plot. The milled rice yield of mixed organic fertilizer 100% plot was increased by 4% than chemical fertilizer plot. The optimum application rate of mixed organic fertilizer was as follows ; $OAR_{MOF}(Mg\;ha^{-1})=[(NAR_{ST}{\div}NC_{MOF}/1,000{\times}0.93)/1,000]$ ($OAR_{MOF}$ : Optimum application rate of mixed organic fertilizer, NARST : Nitrogen application rate($kg\;ha^{-1}$) by soil testing, $NC_{MOF}$ : Nitrogen content($g\;kg^{-1}$) of mixed organic fertilizer, and $0.93:124.3kg\;ha^{-1}/133.0kg\;ha^{-1}$, respectively).

Effect of Nitrogen Level on Yield and Quality of Gyokuro Tea (질소비료 시용량에 따른 옥로차의 수량 및 품질)

  • The reasonable level of nitrogen fertilizer of Gyokuro tea which is producted fresh leaves plucked from shade tea plants is a key factor to increase crop yield and quality. Nitrogen were applied at the level to 60, 80, 100 and $120kg\;10a^{-1}$. Analytical results of yield and quality of Gyokuro tea are summarized as follows : The soil fertility of experimental field was higher in organic matter($59.8g\;kg^{-1}$) and available phosphate($1,285mg\;kg^{-1}$), but lower in pH(4.79) compared to the common field. As the amount of nitrogen fertilizer application was increased to $80kg\;10a^{-1}$, the yield of tea leaves, the content of total nitrogen, total amino acid, caffeine and chlorophyll were increased. However, the yield of tea leaves was not increased above $100kg\;10a^{-1}$ and the increased level of nitrogen fertilizer did not show any different in the content of tannin and vitamin C. When the amount of nitrogen fertilizer application was raised, the content of thiamine and arginine increased, but the content of aspartic acid, serine and glutamic acid decreased. The content of fatty acid was produced $2.850{\sim}3.012mg\;100g^{-1}$ Especially, the content of oleic acid, linoleic acid and linolenic acid was higher at the level of $80kg\;10a^{-1}$ application than other treatments. As nitrogen was applied at $80kg\;10a^{-1}$, sensory score was 0.2~4.6 point higher than other treatments. Consequently, $80kg\;10a^{-1}$ is considered to be the best level of nitrogen fertilizer both quality and crop yield.

Reducing Nitrogen Fertilization Level of Rice (Oryza sativa L.) by Silicate Application in Korean Paddy Soil (논토양에서 규산질 비료 시용에 의한 질소 시비 저감수준 평가)

  • Silicate (Si) fertilizers are well-known for soil amendment and to improve rice productivity as well as nitrogen efficiency. In this study, we investigated the possible reduction level of nitrogen fertilization for rice cultivation by amending Si fertilizer application. Field experiments were carried out to evaluate the productivity of rice (Oryza sativa L.) on a silt loam soil, where three levels of nitrogen (0, 110 and $165kg\;ha^{-1}$) were selected and Si fertilizer as a slag type was applied at 0, 1 and 2 times of the recommendation level (available $SiO_2\;130mg\;kg^{-1}$). Application of Si fertilizer increased significantly the rice yield and nitrogen efficiency. With increasing N uptake of rice, 1 and 2 times of recommended levels of Si fertilization could decrease nitrogen application level to about 76 and $102kg\;N\;ha^{-1}$ to produce the target yield, the maximum yield in the non-Si amended treatment. Silicate fertilizer improved soil pH and significantly increased available phosphate and Si contents. Conclusively, the Si fertilizer could be a good alternative source for soil amendment, restoring the soil nutrient balance and to reduce the nitrogen application level in rice cultivation.

Evaluation of Potentially Available Soil Nitrogen by Using Buffer Phosphote Solution of pH7 (답토양(畓土壤)의 인산완형액(燐酸緩衡液)에 의(依)한 지력질소(地力窒素) 평가(評價)에 관(關)한 연구(硏究))

  • Laboratory experiments were conducted to estimate extractable nitrogen by buffer phosphate solution of pH 7. A series of experiment soils were a) Sandy soil applied with compost, lime, and Wallastonite every years for 32 years. b) Sandy soils with and without waterlogging for 70 days before transplanting. c) Normal soils produced high and common yields. The results were summarized as follows: 1. Extractable organic nitrogen by pH 7 phosphate buffer solution was increased in order of NPK + compost > NPK > NPK + compost + lime + Wollast-onite > NPK + compost + lime > NPK + wollastonite > no fertilizer plot. 2. Extractable organic nitrogens at plots of NPK and NPK + compost were decreased as the growth stage processed regardless of tretments. 3. In case of normal soils having high and common yields the content of total N, organic matter and $NH_4-N$ were increased in high productive soil, while, only $NO_3-N$ content was increased in common productive soil. Especially, there was a highly positive correlation between extractable total nitrogen and $NH_4-N$ content submerged for 4 weeks under incubated condition. 4. Organic nitrogen content of soil was increased on the condition of non-waterlogging, however, nitrogen uptake by rice plant was increased in waterlogged paddy. 5. The content of extractable total nitrogen increased in the order of normal soil, sandy soil, unmatured soil, saline soil, and estimation of optimum nitrogen rates by extracted organic nitrogen was in order of saline soil, unmatured soil, sandy soil, normal soil.

Effect of Chemical Fertilizer and Compost on Soil Physicochemical Properties, Leaf Mineral Content, Yield and Fruit Quality of Red Pepper (Capsicum annuum L.) in Open Field

  • Nowadays, sustainable and environment-friendly agriculture has become an important issue all around the world, and repeated applications of mineral and/or organic fertilizer will probably affect mineral nutrient dynamics in soil in the long term but only a limited number of observations are available. This study was carried out to investigate whether there is any influence of different fertilizer management for red pepper (Capsicum annuum L.) cultivation on soil physicochemical properties, leaf mineral content, yield and fruit quality in the aspect of long-term practice in open field condition. NPK, NPK+compost, compost only, and unfertilized control plot were included in the treatments. The application of chemical fertilizer and/or compost repeated annually for 17 years from 1994 to 2011. Soil organic matter content was higher in compost treatments than in no-manure treatments. Available phosphate and the yield of red pepper were highest in NPK+compost treatment followed by NPK (chemical fertilizer), compost, and control. The results indicate that in the long term, nitrogen supply is still needed for increasing red pepper yield, but reduction in the use of chemical fertilizer could be also possible with the proper application of compost.

Transcriptional Induction of a Carbon Starvation Gene during Other Starvation and Stress Challenges in Pseudomonas putida MK1: A Role of a Carbon Starvation Gene in General Starvation and Stress Responses

  • Thirteen transcriptionally-fused carbon starvation mutants, derived from Pseudomonas putida ATCC 12633, were analyzed for their survivability and transcriptional induction profiles upon carbon starvation. One of these mutants, MK114, which exhibited the lowest survivability and the highest induction rate, was selected and further examined under different starvation (nitrogen and phosphate) and stress (osmolarity, H2O2, salts, alcohol, and heat) conditions. Under all tested conditions MK114 induced ${\beta}$-galactosidase activity, implying that the interrupted gene (cst114) is a general starvation and stress response gene. The rate of induction ranged from 2.6-fold for phosphate starvation to 3.7-fold for osmotic shock. The mini-Tn5 flanking DNA was cloned from the chromosome of MK114. The cloned DNA fragment exhibited carbon starvation activity, indicating that this fragment contains a carbon starvation-related promoter region. This region was partially sequenced. Possible physiological roles of Cst114 in a carbon sensing mechanism and in other stress responses are also discussed.

Emulsification Activity of Acinetobacter sp. 2-3A Isolated from Petroleum Oil-Contaminated Soil (유류오염 토양에서 분리한 Acinetobacter sp. 2-3A의 유화활성)

  • Fifty hydrocarbon-metabolizing microorganisms were isolated from soil samples polluted by the petroleum oils in Gamman-dong, Busan. Among them, strain 2-3A, showing strong emulsification activity, was selected by oil film-collapsing method. This bacterium was identified as Acinetobacter sp. and designated as Acinetobacter sp. 2-3A. The optimum temperature and pH on the growth of Acinetobacter sp. 2-3A were $25^{\circ}C$ and pH 7.0, respectively. The carbon and nitrogen sources for the most effective emulsification activity were 3.0% olive oil and 0.5% peptone, respectively. The 0.15% potassium phosphate was the most effective emulsification activity as a phosphate source. The optimum emulsification activity condition was $20^{\circ}C$, pH 7.0, and 2.0% NaCl. The optimum time for the best production of biosurfactant was 27 hrs. The emulsification stability was maintained at the temperature range from $4^{\circ}C$ to $100^{\circ}C$, pH range from 6.0 to 10.0, and NaCl range from 0% to 10%. For the oil resolvability of the biosurfactant, the residual oils were investigated by gas chromatography. As a result, it was verified that the biosurfactant decreased and decomposed crude oils from $_nC_{10}$ to $_nC_{32}$.

Effect of salinity on growth and nutrient uptake of Ulva pertusa (Chlorophyta) from an eelgrass bed

  • The effects of salinity on various ecophysiological parameters of Ulva pertusa such as growth, nutrient uptake, photosynthetic performance and internal nutrient composition were tested. U. pertusa was collected from an eelgrass bed in a semi-protected embayment on the southwest coast of Korea. Under salinity regimes from 5 to 40 psu, the specific growth rates $(\mu)$ of U. pertusa ranged from 0.019 to $0.032\;d^{-1}$. Maximum growth rate was observed at 20 psu, and minimum at 40 psu. This species showed various uptake rates for nitrate and phosphate. Nutrient uptake was noticeably higher at intermediate salinity levels, and lower at both extremes. Salinity significantly influenced chlorophyll-$\alpha$ content and effective quantum yield. Tissue nitrogen content ranged from 1.5 to 2.9% N (dry weight), whereas tissue phosphorus ranged from 0.1 to 0.14% P (dry weight). The N : P ratio in the tissue of U. pertusa was considerably higher, ranging from 30 to 50. Increased growth at lower salinity suggests that the initial growth rate of U. pertusa is greater during the rainy season (i.e., late spring and early summer) than any other season during the year. The appearance of an Ulva bloom in eelgrass beds may be triggered by salinity more than by other environmental factors such as light and temperature.
