Effect of Amino acids on the Oxidation of Linoleic acid (일부 아미노산이 Linoleic acid의 산화반응에 미치는 영향)

  • A study on the oxidation effect of some amino acids(glutamic acid, phenylalanine, alanine) addition to the linoleic acid emulsion was conducted. Amino acid solutions in concentrations of $10^{-2}$, $10^{-3}$ and $10^{-4}M$ were mixed with equal volume of linoleic acid emulsions. The prepared samples were incubated at $60{\pm}1^{\circ}C$ and the extent of diene conjugation and TBA values were measured by using the UV visible spectrophotometer. The results were as follows: 1) From the extent of diene conjugation, we found that the addition of phenylalanine and alanine prolonged the induction period and the addition of glutamic acid shortened. There was an optimum concentration for each amino acid to act as an antioxidant during the induction period. The optimum concentration of alanine was $10^{-3}M$ and that of phenylalanine was $10^{-2}M$. 2) The results of TBA values showed that three amino acids possesed antioxidant activity after the induction period. There was also an optimum concentration to act as antioxidant after the induction period. The optimum concentrations of glutamic acid, phenylalanine, and alanine were $10^{-2}$, $10^{-3}$, and $10^{-3}M$, respectively.

Phenylketonuria: Current Treatments and Future Developments (페닐케톤뇨증의 치료: 현재와 미래)

  • Phenylketonuria is the most prevalent disorder caused by an inborn error in aminoacid metabolism. It results from mutations in the phenylalanine hydroxylase (PAH) gene. If untreated or late treated, results in profound and irreversible mental disability. Newborn screening test identify patients with phenylketouria. The early initiation of a phenylalanine restricted diet very soon prevents most of the neuropsychiatric complications. However, the diet therapy is difficult to maintain and compliance is poor, especially in adolescents and adulthood. Since 2015, American Medical College of Medical Genetics and Genomics (ACMG) recommended more strong restrictive diet therapy for target blood level of phenylalanine (<360 umol/L). For over four decades the only treatment was a very restrictive low phenylalanine diet. This changed in 2007 with the approval of cofactor therapy (Tetrahydrobiopterin, BH4) which is effective in up to 30% of patients. Data from controlled clinical trials with sapropterin dihydrochloride indicate a similar occurrence of all-cause adverse events with this treatment and placebo. Large neutral aminoacids (LNAA) competes with phenylalanine for transport across the blood-brain-barrier and have a beneficial effect on executive functioning. A new therapy has just been approved that can be effective in most patients with PAH deficiency regardless of their degree of enzyme deficiency or the severity of their phenotype. Phenylalanine ammonia lyase (PAL-PEG) was approved in the USA by FDA in May of 2018 for adult patients with uncontrolled blood phenylalanine concentrations on current treatment. Nucleic acid therapy (therapeutic mRNA or gene therapy) is likely to provide longer term solutions with few side effects.

Enhanced Activity of Phenylalanine Ammonia Lyase in Permeabilised Recombinant E. coli by Response Surface Method

  • To improve phenylalanine ammonia lyase (E.C.4.3.1.5-PAL) activity in recombinant Escherichia coli, Some approaches for improving phenylalanine ammonia lyase (PAL) activity in recombinant E. coli were developed following preliminary studies by means of response surface method. The results shown that permeabilization with combination of Triton X-100, cetyl trimethyl ammonium bromide (CTAB), and acetone enriched cellular recombinant PAL activity significantly, which improved over 10-fold as compared with the control (untreat cell), as high as 181.37 U/g. The optimum values for the tested variables were Triton X-100 0.108 g/L, CTAB 0.15 g/L, and acetone 45.2%(v/v). Furthermore, a second-order model equation was suggested and then validated experimentally. It was indicated that addition of surfactants and organic solvents made the cells more permeable and therefore allowed easier access of the substrate to the enzyme and excretion of the product, which increased the rate of transport of L-phenylalanine and trans-cinnamic acids. These improved methods of PAL activity enrichment could serve as a rich enzyme source, especially in the biosynthesis of L-phenylalanine.

  • Three new peptide analogs 5a-c were obtained through coupling of 5-Amino benzimidazoles 2a-c with L-phenylalanine. For the purpose ${\alpha}$-amino group was blocked with phthalic anhydride and activation of ${\alpha}$-carboxy group of phenylalanine was carried out by preparing phthaloyl-L-phenylalanyl chloride 4. After developing a successful peptide linkage, the phthaloyl group was removed by treating 5a-c with hydrazine hydrate to get free peptides 6a-c, purified through a column of Amberlite (IR-4B). All of these compounds 2a-c and 5,6a-c have been characterized on the basis of their IR, 1H NMR and EIMS analyses. Antibacterial activity of these compounds is also been reported.

  • The activity of PAL, an initial and key regulatory enzyme in biosynthesis of phenolics, peaked during ripening stage. The enzyme activities were 5.60 and 4.25 units/100g fr. wt. in ripening and overripening stages. The increase in PAL activity in ripening strawberry fruits is due to de novo synthesis of the enzyme. PAL extracts from ripe strawberry fruits have a native molecular weight of about 260, 000 Da.

Kinetics of L-Phenylalanine Production by Corynebacterium glutamicum (Corynebacterium glutamicum에 의한 L-Phenylalanine 생산의 동역학적 특성)

  • Microbial production of L-phenylalanine using Corynebacterium glutamicum ATCC 21674, a tyrosine auxotroph resistant to aromatic amino acid analogues, has been studied and kinetic analysis was performed. Even though the strain was reported as a tyrosine auxotroph, it produced tyrosine and was able to grow on the minimal medium where no tyrosine was present. The average specific growth rate at the exponential growth phase was 0.087 hr-1. There was a dissociation of growth from the formation of the product. Linear correlation between biomass production and total CO2 production was obtained. The relationship between CO2 evolution rate and sugar consumption rate was also found to be linear.

양자계산을 이용한 Phenyl Alanine Layase (PAL)의 메커니즘 연구

  • Phenylalanine Ammonia Lyase (PAL)는 phenylalanine의 산성도가 더 큰 ammonium hydrogen 과는 반응하지 않고 그것의 nonacidic ${\beta}$ proton을 제거하는 역할을 한다. Phenylalanine은 electrophilic 그룹을 갖고 있으며 지난 30 여년 동안 phenylalanine의 electrophilic 그룹이 PAL의 반응 기작에 중요한 역할을 한다고 믿어왔다. 그러나 최근 X-ray와 UV spectroscopy를 통하여 상당히 electrophilic 한 5-methylene-3,5-dihydroimidazol-4-one (MIO) 그룹이 발견되었다. 이전의 연구들은 실험을 통하여 MIO와 관련된 electrophile에 의한 Friedel-Crafts Attack 메커니즘과 electrophile에 대한 nucleophilic addition 메커니즘을 제시하였으며, 본 연구진은 양자계산을 통하여 두 가지 메커니즘의 에너지 차이를 살펴봄으로써 더욱 합리적인 메커니즘을 제시, 규명하고자 한다. 본 연구에서는 특히 electrophile에 대한 nucleophilic addition 메커니즘에 대하여 양자계산을 이용하여 반응물, 생성물, 전이상태의 분자 구조를 제시하고 반응이 일어나는데 필요한 에너지를 계산하고자 한다.

Raman Spectroscopy of L-Phenylalanine, L-Tyrosine, and their Peptides Adsorbed on Silver Surface

  • The surface-enhanced Raman scattering of L-phenylalanyl-glycine (L-Phe-Gly), L-phenylalanyl-glycyl-glycine (L-Phe-Gly-Gly), glycyl-glycyl-L-phenylalanine (Gly-Gly-L-Phe), L-tyrosyl-glycine (L-Tyr-Gly), and L-tyrosyl-glycyl-glycine (L-Tyr-Gly-Gly) adsorbed on silver colloidal particles have been investigated. More detailed investigations on the surface-enhanced Raman scattering from L-phenylalanine (L-Phe), glycyl-L-phenylalanine (Gly-L-Phe), L-tyrosine (L-Tyr), and glycyl-L-tyrosine (Gly-L-Tyr) than in ref. 17 have also been made. It has been found that the above molecules adsorb on the surface via both the carboxylate ($COO^-$) and amino ($NH_2$) groups.

Effect of Functional Monomer on Retention Factor of Chiral Racemate (기능성 단량체가 키랄 물질의 체류인자에 미치는 영향)

  • In this work, molecular imprinted polymers (MIPs) using the template of the N-CBZ (carbobenzyloxy)-L-phenylalanine, MAA and 4-VPY as a monomer, EGDMA as a crosslinker and AIBN as an initiator were considered. The prepared polymer particles $(Ca.\;25-35\;{\mu}m)$ were packed into a chromatographic column $(3.9\;\times\;150\;mm)$. The chromatographic characteristics of the retention on the MIP were experimented with acetonitrile as a mobile phase at the flow rate of mobile phase, 0.5 ml/min. The retention factors and resolutions of chiral racemate of the N-CBZ-D, L-phenylalanine were measured. The results showed that the retention factor and resolution by the two co-monomer imprinting polymer were higher than the single monomer imprinting polymers, which indicated an increase in the affinity of the MIP with the sample as a result of the cooperation effect of the binding sites.
