  • In the presence of laccase, generation of monomeric aromatic acids from nonphenolic lignin model dimer veratrylglycerol-$\beta$-vanillate ether (VVE) was observed. The addition of acetovanillone (AV) or acetosyringone (AS) intensified this process, i.e. transformation was more extensive than in the experiments omitting mediators. Among the products isovanillic (IA) and vanillic (VA) acids were identified.

Determination of Phenolic Compounds Responsible for Allelopathy in Upland Weeds (밭 잡초(雜草)중에 존재(存在)하는 Allelopathy 관련(關聯) Phenol 화합물(化合物)의 검색(檢索))

  • Phenolic compounds present in upland weeds (Artemisia asiatica Nakai, Capsella bursa-pastoris (L.) Medik, Portulaca orleracea L. and Trifolium repens L.) which have shown allelopathic activity were determined using paper chromatography (PC) and high performance liquid chromatography (HPLC). Effect of the determined phenolic compounds on germination and post-germination growth of test plants was also investigated. Kinds of phenolic compounds determined by PC in the four weed species were greater in the aqueous extract than in the methanol extract. Ferulic acid was found in both extracts of the weeds studied, whereas benzoic acid was that the weeds commonly contained hydroquinone, p-hydroxybenzoic, ferulic and cinnamic acids. Out of the phenolic compounds determined by PC and HPLC p-hydroxybenzoic, cinnamic and ferulic acids inhibited germination and post-germination growth of radish and sesame. Inhibition of the phenolic compounds on the radicle growth was greater than on the germination of the test plants.

Component Profile Analysis of Irradiated Korean White Ginseng Powder (방사선 조사 인삼의 성분변화에 관한 분석)

  • Currently, some food materials are disinfected by $\gamma$-irradiation (using Co-60) or ethylene oxide treatment. These treatments were applied to ginseng powder and the ginseng components such as ginsenosides, polyacetylenes and phenolic acids were analyzed by HPLC to determine any compositional changes due to irradiation. No appreciable difference was observed in the HPLC pattern of ginsenosides, polyacetylenes of ginseng powder after 10 key irradiation or ethylene oxide treatment (EO $CO_2$= 3 : 7, w/wfb) from those of untreated fresh ginseng powder when they were analyzed soon after treatments. When the ginseng powders were stored at room temperature for three years after the same treatment, the HPLC patterns of polyacetylenes and phenolic acid fraction showed appreciable change from those of fresh ginseng powder, however, the HPLC patterns of three year old samples did not show any appreciable difference.

  • Korean cabbage, a member of the Brassicaceae family which also includes cauliflower, mustard, radish, and turnip plants, is a crucial leafy vegetable crop. Korean cabbage is harvested after completion of the leaf heading process and is often prepared for use in "baechu kimchi", a traditional Korean food. Many of the components in Korean cabbage are essential for proper human nutrition; these components can be divided into two groups: primary metabolites, which include carbohydrates, amino acids, fatty acids, and organic acids, and secondary metabolites such as flavonoids, carotenoids, sterols, phenolic acids, alkaloids, and glucosinolates (GSLs). Using gas chromatography-mass spectrometry, this study examined the variety of volatile compounds (including isothiocyanates) contained in Korean cabbage and its seed, which resulted in the identification of 16 and 12 volatile compounds, respectively. The primary volatile compound found in the cabbage was ethyl linoleolate (~23%), while 4,5-epithiovaleronitrile (~46%) was the primary volatile component in the seed.

  • Humic acids extracted from decomposing plant residues were characterized by infrared(IR) spectra. The IR spectra were further interpreted by chemical analyses for oxygen-containing functional groups such as carboxyl, phenolic, alcoholic, carbonyl, and quinionic groups. 1. The IR spectra obtained in this study were divied into three categories: spectra of humic acids from grain crop straws of rice, barley, wheat and rye produced Type I, while that from wild grass hay yielded Type II, and those from forest tree litter of the deciduous and conifers were led to give Type III. 2. There were no significant changes in the absorption bands observed among humic acids extracted at various stages of decomposition of a given Plant material. 3. The absorption band at about $3,430cm^{-1}$ represents the presence of hydrogen-bonded hydroxyl groups, phenolic-OH groups being the major component. 4. A close relationship was found between the total acidity and the content of phenolic-OH groups of humic acids. The content of carboxyl groups maintains a direct relationship with the content of total hydroxyl groups, and such a close relationship also exists between the content of alcoholic hydroxyls and that of total hydroxyl groups. 5. Overlapping of the absorption bands of carbonyl groups and quinones renders it difficult to make differentiation between the two. 6. A variety of non-armoatic cyclic hydrocarbons appears to be a structural component as evidenced by a sharp absorption peak near $995-1000cm^{-1}$.

Anti-hyperglycemic and Antioxidative Activities of Phenolic Acid Concentrates of Rice Bran and Hydroxycinnamic Acids in Cell Assays (미강 페놀산 농축물과 Hydroxycinnamic Acids의 세포내 항당뇨 및 항산화 활성)

  • Phenolic acid concentrates of rice bran(RB-ex) and hydroxycinnamic acids were investigated for their anti-hyperglycemic activities through glucose uptake and glucokinase activity using HepG2 cells and stimulatory effects on insulin secretion using HIT-T15 cells. RB-ex was prepared as an ethylacetate extract after alkaline hydrolysis and hydroxycinnamic acids, found as major compositions of RB-ex, such as ferulic acid(FA), sinapic acid(SA) and p-coumaric acid(p-CA) were investigated to compare with the properties of RB-ex. The properties of glucose uptake in HepG2 cells were examined in the absence of insulin and two different glucose concentrations(5.5 mM and 25 mM). RB-ex and FA showed anti-hyperglycemic activities through the increase of glucose uptake and the stimulation of glucokinase activity in HepG2 cells. RB-ex exhibited higher glucose uptakes with higher glucose concentrations, whereas FA exhibited the same increasing effects on both concentrations of glucose. RB-ex and FA exhibited doubled glucokinase activities relative to control. In the presence of insulin in the 25 mM glucose-containing medium, the levels of glucose uptake were increased in all treatments compared with control. As stimulatory effects of samples on insulin secretion were estimated, RB-ex and FA stimulated insulin secretion at a concentration of 25 ${\mu}g/m{\ell}$ and in particular, FA showed the highest amount of insulin-release in HIT-T15 cells. Antioxidative effects on HIT-T15 cells, RB-ex and hydroxycinnamic acids, excluding p-CA, showed inhibitory activities of 78% to 80% at a concentration of 100 ${\mu}g/m{\ell}$. On the basis of these results, we conclude that RB-ex and FA could help decrease blood glucose levels and prevent the cell damages via antioxidant activity.

Influence of Candide parapsilosis on the Changes in Various Components of Korea Red Ginseng Extract (Candida payapsilosis가 홍삼엑기스의 성분 변화에 미치는 영향)

  • The Quality characteristics of pH, ginsenosides, fatty acid, phenolic compounds were studied for their changes during growth of Candia parapsilosis. The yeast growth not only scarcely affected the total amount of saponins and ginsenosidvs of recd ginseng tail root extract, but also was not affected by the saponin C. pnrapsilosis did not utilize the ginsenosides as a carbon source. Glucose, fructose and free sugars were utilized in the initial phase of growth, whereas sucrose and maltose were used as the growth continued and completely redured after 43 hours of incubation. Unsaturated fatty acids were significantly reduced with cell growth, showing a relationship between unsaturated fatty acid content and the yeast growth, whereas the amount of saturate[1 tatty acids in red ginseng extract was not affected by the yeast growth. Generally, there were no changes in major organic acids and phenolic compounds (vanillie acid , m-coumaric acid) except the 50% reduction in maltol and ferulic acid in the ginseng extract. The amounts of amlno acids were gradually decreased, but that of arginine was remarkably reduced.

Compositional Analysis of Lentil (Lens culinaris) Cultivars Related to Colors and Their Antioxidative Activity

  • Metabolite profile is a powerful analytical technique to identify the functional characterization of plants. In this study, the phytochemicals and secondary metabolites of lentils (Lens culinaris) were analyzed to compare the anti-oxidative activities according to the different colors. The polar metabolites, fatty acids, carotenoids, flavonoids, anthocyanins, total phenolic acids, DPPH activity were analyzed. Three kind of lentils, French green whole lentil (FG), red whole lentil (LR), and green whole lentil (LG) (ASIA SEED Co., LTD), were used for this study. Fatty acids, phytochemicals, and antioxidative components from each lentil varieties were analyzed by official methods. The contents of lutein in carotenoids were 6-9 times higher than zeaxanthin in all lentils, but were not significantly different among three varieties. The content of carotenoids in FG was lower significantly than those in the LR and LG. Myricetin and luteolin were detected in the only FG. Kaempferol and delphinidin were significantly highest in the FG. Most of the phenolic acids except coumarate were higher in FG and LG than in LR. Also antioxidant effects ($EC_{50}$) were higher in FG and LG than in LR. The analyzed metabolites obtained from lentils showed distinct separation in the PCA results according to the varieties. Also, lentils showed different anti-oxidant profiles according to the colors. FG and LG showing higher contents of phytochemicals showed higher antioxidative activity than LG containing relative low contents of phytochemicals.

Structure-Guided Identification of Novel Phenolic and Phenolic Amide Allosides from the Rhizomes of Cimicifuga heracleifolia

  • Two phenolic allopyranosides and two phenolic amide allopyranosides, along with eight known phenolic compounds, including cimicifugic acids, shomaside B, fukiic acid, isoferulic acid, and piscidic acid, were isolated from the n-butanolic extract of rhizomes of Cimicifuga heracleifolia. On-line spectroscopic data for UV, NMR, and MS from a combination of LC-NMR and LC-MS techniques directly and rapidly provided sufficient structural information to identify and confirm all the structures of major phenolic compounds in the extract, in addition to their HPLC profiles. This combined analytic information was then used as a dereplication tool for structure-guided screening in order to isolate unknown phenolic compounds in the extract. Successive fractionation and purification using semi-preparative HPLC acquired four unknown allopyranosides, and their structures were identified as cis-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-ferulic acid 4-O-${\beta}$-D-allopyranoside, trans-feruloyltyramine 4-O-${\beta}$-D-allopyranoside, and trans-feruloyl-(3-O-methyl)dopamine 4-O-${\beta}$-D-allopyranoside, based on a subsequent spectroscopic interpretation.

Identification and Quantification of Phenolic Acid Derivatives from Three Korean White and Brown Rice Samples Based on High-Resolution Mass Spectrometry (쌀(백미, 현미) 시료로부터 고해상도 질량 분석 기반의 페놀산 유도체 동정 및 정량 평가)

  • Reported positive ion fragmentation of phenolic acid derivatives in rice (Oryza sativa L.) were summarized based on the literature. A total of eight phenolic acids (4 derivatives of ferulic acid, 3 derivatives of sinapic acid and p-coumaric acid) were isolated and identified from rice (raw and steamed) using UPLC-DAD-QToF/MS. Results revealed that 6-O-feruloylsurose was the major component with 3'-O-sinapoylsucorse being tentatively identified in Oryza sativa L. for the first time as a new hydroxycinnamoyl derivative in rice grains. In our study, raw brown rice had the highest phenolic acid contents with Samkwang showing higher phenolic acid content than Saeilmi and Sindongjin (12.41 vs. 7.89 and 3.10 mg/100 g dry weight, respectively). Of all varieties, brown rice had higher phenolic acid contents than white rice. These contents decreased considerably when rice was steamed whereas, p-coumaric acid and ferulic acid contents were increased. Additionally, contents of rice (raw and steamed) can be used as a fundamental report for new rice varieties.
