Antioxidative Effectiveness of Terminalia chebula Rets Extracts (가자(Terminalia chebula Retz)추출물의 항산화 효과)

  • In this study, antioxidative effectiveness of BHA, BHT at 0.02%(w/w) was compared with those of separated free phenolic acid, ester form and insoluble bound phenolic acid which were extracted from 50 g of Terminalia chebula Retz by MeOH/aceton solvents. Antioxidative effectiveness was measured by peroxide values and TBA values for 7 days, storaging respective substrates and contrast tube at $45{\pm}1^{\circ}C$ for 35days. Laboratory tubes was added by BHA, BHT, separated free, soluble and insoluble phenolic acid extracts and peroxide value of contrast tube after 21 day storage were 60, 30, 14, 11, 100. On the other hand, at the same conditions, TBA values of each antioxidants were 0.150, 0.108, 0.105, 0.073, 0.078, 0.185. This results remarkably appeared antioxidative effectiveness in meal soybean oil substrates. Phenolic acid separated and identificated were p-coumaric acid, Ferulic acid, Phloroglucinol, Pyrogallol, Vanillic acid and Caffeic acid.

도정도를 달리한 백미 및 미강에 함유된 phenolic acids와 총 폴리페놀 함량

  • 최근 쌀 소비량이 감소되고 쌀중심 식생활의 영양적 우수성을 인식하지 못하여 쌀은 주식으로서 위치가 흔들리고 있다. 그동안 쌀의 영양특성에 관한 연구로서 주로 탄수화물 급원으로서의 당질에 관한 연구와 미강을 중심으로한 식이섬유, 미강유등에 관한 연구결과들이 발표되었다. 특히 미곡 부산물인 미강으로부터 다양한 유효성분들이 확인되어 그 효능이 보고되고 있으며 일부가 기능성 소재로서 제품화되기도 하였다. 그러나 실제 우리의 식생활에서 섭취되고 있는 백미 또는 현미상태에서의 유효성분의 분포 및 함량에 대한 연구는 미진한 상태이다. 본 연구에서는 쌀의 영양적우수성을 규명하고자 유효성분 중 phenolic acid와 폴리페놀 함량을 품종 및 도정도에 따라 조사하였다. 조생종인 오대와 만생종인 남평 품종으로부터 현미와 7분도, 10분도 및 12분도로 도정도를 달리한 백미, 도정도별 미강을 각각 제조하였다. Ferulic acid 등 쌀과 미강에 존재하는 phenolic acid 는 알칼리로 추출한 후 pH를 조정하고 에틸아세테이트로 반복 추출하여 HPLC로 분석하였다. 현미에 함유된 phenolic acid는 ferulic acid가 가장 많아 50% 내외를 차지하였으며 p-coumaric acid, benzoic acid, sinapinic acid 순으로 검출되었다. 오대 현미의 총 phenolic acid 함량은 65.9 mg%로서 남평 현미의 57.2 mg%보다 높았으며 도정도가 증가할수록 백미 중의 총 phenolic acid 함량은 감소되었다. 미강 중에는 백미의 10배 량에 해당하는 phenolic acid가 검출되었고 benzoic acid와 m-hydroxy benzoic acid는 미강 시료에서만 검출되었다. 도정한 백미 중의 phenolic acid는 28.8∼51.7 mg%, 미강에서 321.4∼438.4 mg% 범위로 나타났다. 현미, 백미 및 미강에 함유된 총 폴리페놀의 함량을 표준 페놀화합물로 카테친을 사용하고 비색법에 의하여 측정하였을 때 오대 현미의 폴리페놀 함량은 78.4 mg%, 남평 현미 88.8 mg% 였다. 도정한 백미 중의 총 폴리페놀 함량은 30.3∼56.9 mg%, 미강이 541.5∼472.6 mg%의 범위였다. 이상과 같이 쌀에는 phenolic acid 및 총 폴리페놀이 상당량 함유되어 있으며 특히 배유보다는 강층에 많이 존재하므로 이들 성분의 효율적인 이용을 위한 쌀의 섭취방안이 필요한 것으로 나타났다.

Comparative phenolic compound profiles and antioxidative activity of the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) according to cultivation years

  • Background: The study of phenolic compounds profiles and antioxidative activity in ginseng fruit, leaves, and roots with respect to cultivation years, and has been little reported to date. Hence, this study examined the phenolic compounds profiles and 2, 2-diphenyl-1-picrylhydrazyl (DPPH) free-radical-scavenging activities in the fruit, leaves, and roots of Korean ginseng (Panax ginseng Meyer) as a function of cultivation year. Methods: Profiling of 23 phenolic compounds in ginseng fruit, leaves, and roots was investigated using ultra-high performance liquid chromatography with the external calibration method. Antioxidative activity of ginseng fruit, leaves, and roots were evaluated using the method of DPPH free-radical-scavenging activity. Results: The total phenol content in ginseng fruit and leaves was higher than in ginseng roots (p < 0.05), and the phenol content in the ginseng samples was significantly correlated to the DPPH free-radical-scavenging activity ($r=0.928^{****}$). In particular, p-coumaric acid ($r=0.847^{****}$) and ferulic acid ($r=0.742^{****}$) greatly affected the DPPH activity. Among the 23 phenolic compounds studied, phenolic acids were more abundant in ginseng fruit, leaves, and roots than the flavonoids and other compounds (p < 0.05). In particular, chlorogenic acid, gentisic acid, p- and m-coumaric acid, and rutin were the major phenolic compounds in 3e6-yr-old ginseng fruit, leaves, and roots. Conclusion: This study provides basic information about the antioxidative activity and phenolic compounds profiles in fruit, leaves, and roots of Korean ginseng with cultivation years. This information is potentially useful to ginseng growers and industries involved in the production of high-quality and nutritional ginseng products.

Changes in element accumulation, phenolic metabolism, and antioxidative enzyme activities in the red-skin roots of Panax ginseng

  • Background: Red-skin root disease has seriously decreased the quality and production of Panax ginseng (ginseng). Methods: To explore the disease's origin, comparative analysis was performed in different parts of the plant, particularly the epidermis, cortex, and/or fibrous roots of 5-yr-old healthy and diseased red-skin ginseng. The inorganic element composition, phenolic compound concentration, reactive oxidation system, antioxidant concentrations such as ascorbate and glutathione, activities of enzymes related to phenolic metabolism and oxidation, and antioxidative system particularly the ascorbate-glutathione cycle were examined using conventional methods. Results: Aluminum (Al), iron (Fe), magnesium, and phosphorus were increased, whereas manganese was unchanged and calcium was decreased in the epidermis and fibrous root of red-skin ginseng, which also contained higher levels of phenolic compounds, higher activities of the phenolic compound-synthesizing enzyme phenylalanine ammonia-lyase and the phenolic compound oxidation-related enzymes guaiacol peroxidase and polyphenoloxidase. As the substrate of guaiacol peroxidase, higher levels of $H_2O_2$ and correspondingly higher activities of superoxide dismutase and catalase were found in red-skin ginseng. Increased levels of ascorbate and glutathione; increased activities of $\text\tiny L$-galactose 1-dehydrogenase, ascorbate peroxidase, ascorbic acid oxidase, and glutathione reductase; and lower activities of dehydroascorbate reductase, monodehydroascorbate reductase, and glutathione peroxidase were found in red-skin ginseng. Glutathione-S-transferase activity remained constant. Conclusion: Hence, higher element accumulation, particularly Al and Fe, activated multiple enzymes related to accumulation of phenolic compounds and their oxidation. This might contribute to red-skin symptoms in ginseng. It is proposed that antioxidant and antioxidative enzymes, especially those involved in ascorbate-glutathione cycles, are activated to protect against phenolic compound oxidation.

Antioxidative Materials in Domestic Meju and Doenjang 4. Separation of Phenolic Compounds and Their Antioxidative Activity (재래식 메주 및 된장 중의 항산화성 물질에 관한 연구 4. 페놀화합물의 함량과 항산화력)

  • In order to investigate the antioxidant activity of phenolic compounds contained in domestic Meju and Doenjang , the methanolic extract from defatted Meju and Doenjang was fractionated into the phenolic acid and the isoflavone fractions by alumina column and polyamide-6 column chromatography, respectively. Both phenolic acid and isoflavone fractions exhibited an identical antioxidative effect against the oxidation of linoleic acid. GC analysis revealed that the phenolic acid fractio contained vanillic , chlorogenic ,p-coumalic , ferulic, and caffeine acid. The content of caffic acid was greater than 70% in the phenolic acid fraction and the content of vanillic and chlorogenic acids were disappeared during fermentation, and thus, the facts are thought to be reasons for the low antioxidant activity of the phenolic aicd fraction from Doenjang. Similarly, the isoflavone fraction cotained daidzin, glycitin-65-0 glucosider and genistin and their aglycons such as daidzein, glyciten and genistein. The content of daidzin and genistin in Meju dramatically decreased at the early stage of fermentation ,whereas the content of daidzein an dgenistein rather increased ; however, these change in isoflavone contents did not affect the antioxidant activity of isoflavone fractions.

Effect of Phenolic Resin According to Relative Humidity on Submerged Entry Nozzle with ZrO2-C System in Fabrication Process (ZrO2-C계 침지노즐 제조시 상대습도에 따른 바인더용 페놀수지의 영향)

  • The thermodynamic behavior of phenolic resin was investigated to verify the relation between the properties of porous ceramics with $ZrO_2$-C system for submerged entry nozzle and the characteristics of phenolic resin with various relative humidity. The green and the sintered density were decreased between 25% and 50% relative humidity, whereas they were gradually enhanced above 50% relative humidity. The highest value of apparent porosity was 20.1% and the minimum compressive strength was 69MPa in the specimen using the powder exposed to 50% relative humidity. As a result of thermal analysis for phenolic resin, the shift of endothermic peak to low temperature and the reduction of exothermic peak were observed, and the peaks corresponded to melting and curing of phenolic resin, respectively. The melting and the curing of phenolic resin generate the change of green density, and it can affect the properties of submerged entry nozzle.

  • Phenolic compounds play a major role in the interaction of plants with their environment. They are thought to have been a feature of higher plants since early colonization of the land. Phenolics are crucial for many important aspects of plant life. They can play structural roles in different supporting or protective tissues, for example in cell walls, they can be involved in defence strategies, and signalling properties particularly in the interactions between plants and their environment. In brown algae, phenolic compounds are contained within membrane bound vesicles known as physodes, and their roles in algae are thought to be similar to those of higher plant phenolics. They can be stained using various histochemical stains, however, none of these stains are phenolic specific so care must be taken during interpretation of such results. Many, but not all phenolics are also autofluorescent under UV or violet light. Physodes are involved in cell wall construction, both in primary and secondary walls in brown algae. They bind together with other wall components to make a tough wall. They have also been found to play a role at fertilization, in blocking polyspermy in some species. Sperm are very quickly rendered immobile after phenolic release from newly fertilized zygotes seconds after fertilization. Phenolic compounds are thought to be important herbivore deterrents in some species due to their astringent nature. Phenolic compounds also offer effective UV protection in the early life stages and also the adults of many algal species. In the future, this factor may also make them an important player in the pharmaceutical and skincare industries.

Determination of Total Phenolic Compounds from Korean Red Ginseng, and Their Extraction Conditions (고려홍삼의 총 페놀성환의 정량 및 그 추출조건)

  • This study was carried out to investigate the possible determination of total phenolic compounds by Folin-Denis method from Panax ginseng C.A. Meyer and their extraction conditions. It was possible to apply the method for determination of total phenolic compounds from Korean red ginseng. But 3 kinds of amino acids such as tyrosine, cystein and tryptophan, and 3 kinds of vitamins such as ascorbic acid, pyridoxin HCI and thiamine HCI affected strongly the colorization by the method. Effective alcoholic solvent for the extraction was 60% ethanol, appropriate extraction temperature, time and times were 40-80。C, 1-2 hours and 3 times, respectively.

The Stimultaneous Determination of Phenolic Compounds by GC and GC/MS

  • To develop a simple, rapid and simultaneous analytical method of phenolic compounds using gas chromatography (GC) and gas chromatography/mass spectrophometer (GC/Ms), this experiment was carried out to search the retention times of capillary columns and the characteristics of fragment ions in electron impact mass spectra. Most of trimethylsilyl derivatives and underivatized phenolic compounds were separated very well on three kinds of capillary columns(HP-1), Ultra-2 and HP-35). Quantitiative determination of phenolic compounds was achieved by internal standards (p-hydroxybenzoic acid iopropyl ester, p-hydroxybenzoic acid ethyl ester). Calibration plts were linear in the investigated range, and the limits of detection were about 5 ng at split mode method. When analyzed by three columns, theseparation times were fairly constant on two nonpolar columns, but a few compounds showed slightly different separation order by the itnermediate polar HP-35 column. The important characteristic patterns of TMS derivatives of phenolic compounds on the EI/MS spectrra appeared at the base peak of [M-15]+ ion and presented at high abundance in most TMS derivatives of phenoloc compounds. [M]+, [M-CH3-COO]+, [M-Si(CH3)4]+ and [M-Si(CH3)4 -CH3]+ also observed in mass spectra of these compounds . Although several compounds have the same retention times on GC column, it might be possible to identify these compounds by the different patternsof mass frgement ions. The TMS derivatives, thus , provide additional information for identification of phenolic compounds in biological systems.

Relationship of Phenolic Compounds and Free-radical Scavenging Activity in Black and Red Rice Extract

  • Total phenolic content, total anthocyanin content and antioxidant activity were analyzed from rice samples collected in Korea, Japan and China. The results showed that the total phenolic content and free-radical scavenging activity differed significantly in these rice lines. The correlation between content and activity was subsequently investigated. The results showed that in black rice, anthocyanin was the major phenolic component and that both phenolic content and anthocyanin content were closely correlated with free-radical scavenging activity. Reverse Phase High Performance Liquid Chromatography (RP-HPLC) data showed that cyanidin-3-O-glucoside and peonidin-3-O-glucoside composed about 90% of the total anthocyanin content in black rice and in red rice. In the red rice extract, the total phenolic content produced a high correlation coefficient with antioxidant activity but correlated very poorly with the total anthocyanin content. The $OD_{458}$ and the $OD_{500}$ values which represent the proanthocyanidin content of the rice extract, produced high correlation coefficients with antioxidant activity and total phenolic content. These results suggest that the $OD_{458}$ and the $OD_{500}$ values can be used to evaluate the quality of red rice. In addition, based on the data obtained, a competitive accumulation model of anthocyanin and proanthocyanidin in black and red rice was proposed.
