Electron Donating Ability and Contents of Phenolic Compounds, Tocopherols and Carotenoids in Waxy Corn (Zea mays L.) (찰옥수수의 전자공여작용과 페놀성화합물, Tocopherols 및 Carotenoids의 함량)

  • The antioxidative activity measured by electron donating ability was investigated for the breeding of the highest antioxidative waxy corn (Zea mays L.) and the research for the most effective antioxidant in waxy corn. The electron donating ability was $15.5{\sim}65.0%$. The contents of phenolic compounds and tocopherols and the absorbance at 450 nm were $102.3{\sim}323.5\;{\mu}g/mL$, $15.6{\sim}144.2\;{\mu}g/mL$ and $0.047{\sim}0.206$, respectively. The mean values of electron donating ability and contents of phenolic compounds and tocopherols of four black waxy corn were comparatively high, that is, 48.7%, $267.0\;{\mu}g/mL$ and $87.0\;{\mu}g/mL$, respectively. The electron donating ability was significantly correlated with the level of phenolic compounds and tocopherols but not with the content of carotenoids.

Phenolic compounds in domestic and imported grape cultivars in Korea (국내산 포도와 수입산 포도의 페놀계화합물 함량)

  • In this study, fruit phenolics were investigated with using LC/MS and HPLC analysis in order to compare the differences between domestic ('Campbell Early', 'Kyoho', 'Heukbosuk', and 'Hongju SDS') and imported ('Crimson SDS' and 'Thomson SDS') grapes. In the case of fruit characteristics, imported grape 'Crimson SDS' and 'Thompson SDS', had lighter skin weight (300-350 mg/berry) and hard flesh (5.2-5.6 kg·f) than domestic grape cultivar. The phenolic compound contents of 'Crimson SDS' skin was higher, but resveratrol (25-29 mg/kg), quercetin (350-380 mg/kg), and myricetin (31-32 mg/kg) contents were similar in to those of 'Hongju SDS'. The anthocyanin content was different from differed between grape cultivars. 'Hongju SDS' grape was showed higher in Delphinidin-3-glucose (D3G) levels, and 'Crimson SDS' was showed higher in Peonidin-3-glucoside (P3G) levels. The contents of phenolic compounds were investigated differently for each grape berry part. Catechin, epicatechin, procyanidin B1, and B2 were found in grape seeds.

  • In this study, Tongue fern (Pyrrosia lingua) plants that have been used traditionally as medicines. Their traditional medicinal uses, regions where indigenous people use the plants, parts of the plants used as medicines. This study was designed to assess the antioxidant and inhibition activities of extracts from P. lingua. In the P. lingua extracts was measured ethanol activity, 80.0% ethanol was high activity. The antioxidant activity was measured in 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-Azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS), assays. DPPH and ABTS radical in this experiment, solid and phenolic of extract were tested, but only an average concentration of 100 ㎍/mL was used. However, the phenolic extract is shown phenolic activity reached a peak. Also, phenolic extracts ware reached peak water and ethanol extracts. As a result, using the phenolic extracts did other antioxidant assays such as DPPH, ABTS, protection factor, and thiobarbituric acid reactive substances at 50-200 ㎍/mL concentrations. The activity of elastase and collagenase, inhibiting their activities may retard skin aging. α-Glucosidase and α-amylase, inhibitors need to be explored for the benefit of postprandial hyperglycemia in diabetic patients. Activities of tyrosinase, hyaluronidase and xanthine oxidase inhibitors of these enzymes are increasingly important ingredients in cosmetics and medications to protect the skin against hyperpigmentation and skin aging. Inhibition effects were investigated using the P. lingua extracts at 50-200 ㎍/mL concentrations. The expression levels of enzyme inhibitions activities were decrease in dependent-concentrations manner when P. lingua extracts were treated.

Occurrence and Concentrations of Estrogenic Phenolic Compounds in Surface Waters of Rivers Flowing into Masan Bay, Korea

  • The estrogenic phenolic compounds, nonylphenol (NP), octylphenol(OP), bisphenol A (BPA) and nonylphenol mono- and diethoxylate ($NP_{1-2}EO$) were analyzed in 24 surface water samples from six rivers flowing into Masan Bay. All of the phenolic compounds were detected in all six rivers in high concentrations. The most abundant compound was $NP_{1-2}EO$ (86.0%), followed by NP ($10.1 \%$), BPA ($3.6\%$) and OP ($0.3\%$). The levels of phenolic compounds were 1.42-22.70 ${\mu}g$/L for $NP_{1-2}EO$, 0.15-1.68 ${\mu}g$/L for NP, 0.024-0.610 ${\mu}g$/L for BPA and 0.003-0.067 ${\mu}g$/L for OP. Especially, high concentrations were recorded in the rivers that pass through industrial complexes. The concentrations of phenolic compounds observed in these river waters were 1-2 orders of magnitude lower than the reported acute toxicity levels (hundreds of micrograms per liter). However, they were only slightly lower than the chronic toxicity levels. Most of the water samples also exceeded the Canadian nonylphenolic compounds water quality guideline, 1 ${\mu}g$/L, for the protection of aquatic life and the maximum permissible concentrations (MPC), 0.33 ${\mu}g$/L for NP and 0.12 ${\mu}g$/L for $NP_{1-2}EO$.

Fluctuations in Phenolic Content and Antioxidant Capacity of Green Vegetable Juices during Refrigerated Storage

  • Shinseoncho and kale were made into green vegetable juices by building block [shinsenocho branch (SB), shinsenocho leaf (SL), kale branch (KB), and kale leaf (KL)]. Fluctuations in their phenolic contents and antioxidant capacities were analyzed during refrigerated storage at $4^{\circ}C$ for 28 days. Total polyphenolic contents of leaf parts showed a decreasing tendency after 4 days (SL) or 7 days (KL), whereas branch parts showed fluctuating values during the entire storage period. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging capacity was rapidly decreased in SB and in SL at 28 days (P<0.001), whereas KL showed a slightly increasing tendency after 14 days. For the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging capacity, SL showed a sharp fall at 28 days (P<0.001), and KL showed a decreasing tendency after 14 days (P<0.001). SB showed a steady decrease during the entire storage period and KB indicated a nearly zero (0.97%) at 28 days. Pearson's coefficients for the correlation between antioxidant capacities measured by the ABTS and DPPH assays, and the total polyphenolic contents were determined. The results showed that the ABTS assay (r=0.934, P<0.001) was more strongly positively correlated with the total phenolic contents than the DPPH assay (r=0.630, P<0.001). In conclusion, when considering all building blocks, green vegetable juices, including kale and shinseoncho may have kept antioxidant capacities for up to 14 days under refrigeration, and the ABTS assay better reflects a positive correlation with the total phenolic contents when compared to the DPPH assay.

Phenolic Acid Contents and ROS Scavenging Activity of Dandelion(Taraxacum officinale) (민들레(Taraxacum officinale) 잎과 뿌리의 페놀산 조성 및 활성산소 소거활성)

  • The propose of this study was to investigate the antioxidant activity of 80% ethanol extracts and various solvent fractions of dandelion (Taraxacmn officinale) leaves and roots, Total phenolics and phenolic acid contents were also examined. The total phenol content of leaves and roots were $7.9{\pm}0.4%$ and $9.4{\pm}0.3%$ respectively. Eight phenolic acids were separated by GC, among which caffeic acid (113.7 mg%)and m-coumaric acid (152.6 mg) were the dominant phenolic acids in leaves and roots, respectively. Amongst solvent functions of leaves and roots, the ethyl acetate fraction showed the strongest radical scavenging activity. A strong correlation was found between total phenol content and electron-donating ability, and ABTS radical scavenging activity showed a similar trend as electron-donating ability. Hydroxyl-radical-scavenging activity and lipid peroxidation were significantly higher in the ethyl acetate fraction than other factions. In particular, the SOD-like activity was highest (43.6%) in the ethyl acetate fraction of dandelion leaves, and was higher than that of trolox. Thus, the ethyl acetate fraction of dandelion leaves exhibited significant phenol content, antioxidant activity, and free-radical-scavenging effects.

Immobilization of Recombinant Bacterial Biosensors: a Simple Approach for the On-Site Detection of Phenolic Compounds (재조합 박테리아 바이오센서의 고정화: 페놀계 화합물의 현장 검출을 위한 간단한 접근 방법)

  • We herein report the development of an agarose-gel-immobilized recombinant bacterial biosensor simple system for the field monitoring of phenolic compounds. Escherichia coli cells harboring the pLZCapR plasmid, which was previously designed to express the ${\beta}$-galactosidase reporter gene in the presence of phenolic compounds, were co-immobilized with a substrate [chlorophenol red ${\beta}$-galactopyranoside (CPRG) in agarose gel, and dispensed to the wells of a 96-well plate. Field samples were added to the wells and color development was monitored. In the presence of 5 ${\mu}M$ to 10 mM of phenol, the biosensor developed a red (representing hydrolysis of CPRG) color. Other phenolic compounds were also detected by this immobilized system, with the pattern resembling that previously reported for the corresponding non-immobilized biosensor. The immobilized cells showed optimum activity when the gel was simultaneously supplemented with 6% dimethyl formamide (DMF), 0.1% SDS and 10 mM $CaCl_2$. The immobilized biosensor described herein does not require the addition of a substrate or the use of unwieldy instruments or sample pretreatments that could complicate field studies.

Changes in Physicochemical Properties and Antioxidant Activities of Brown Rice (Oryza sativa L.) throughout Germination

  • The objective of this research was to investigate the changes in the contents of physicochemical properties including $\gamma$-aminobutyric acid (GABA), total dietary fiber (TDF), amylose, protein, and fat content in brown rice through germination for 2 different years. Total phenolic contents and antioxidant activities of DPPH and ABTS radical scavenging capacities were also determined in different solvent extracts. For the physicochemical properties, GABA, TDF, protein, and fat content increased, whereas amylose levels decreased. Specially, GABA and TDF levels showed the greatest variations among cultivars and harvest years. Total phenolic content and antioxidant activity significantly increased. The average total phenolic content at a concentration of 0.5 mg/mL in different extract solvents occurred in this order: methanol>ethylacetate>chloroform>hexane extracts. Additionally, 'Keunnun' exhibited the highest GABA levels, highest total phenolic content, and highest antioxidant activity after germination, with increases of approximately 3.7, 2.0, and 1.9 times, respectively, compared to levels before germination. These results suggest that, because of its high physicochemical contents and strong radical scavenging activities, germinated brown rice can be used as beneficial supplement.

Changes of Phenol Compounds according to Storing Years in Soybean

  • The objective of this study was to determine the role of storing years with the variation of total phenol and individual phenolic compounds in soybean (Glycine max L.) seeds. The total phenol content varied from 0.36 to 0.42% over four years, with the highest value (0.42%) found at storage for two and three years. Among the nine soybean varieties examined, Daweonkong had the highest total average phenol content (0.58%). The total content of 11 phenolics varied from 730.0 to 1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$ over storage for four years, and the highest concentration (1812.8 $\mu\textrm{g}\;\textrm{g}^{-1}$) was found at storage for two years. Myeongjunamulkong (1465.4 $\mu\textrm{g}\;\textrm{g}^{-1}$) had the highest mean content among the nine soybean varieties. The total content of 11 phenolic compounds measured in this study occupied from 20.96 to 47.73% of the total phenol contents. The highest total phenol contents were in seeds with black coats (5279.4 $\mu\textrm{g}\;\textrm{g}^{-1}$), while the highest concentration of individual phenolic compounds were in seeds with green coats (1419.5 $\mu\textrm{g}\;\textrm{g}^{-1}$). Our study suggests that it may be feasible to improve soybean varieties with high functional substances such as phenolic compounds.

Antioxidant Activity and Total Phenolic Compounds in Grain Extracts of Wheat, Barley, and Oat

  • Cereal grains are rich in phenolic compounds that give beneficial effect in human health. Although several research works have been reported on the effects of phytochemicals of plant origin, such as fruits, vegetables, few studies have examined the antioxidative effects of whole cereal grains. The objective of this study was to determine total antioxidant capacity of 80% ethanolic extracts of cereal grains by testing the ability of the extracts to inhibit UV -induced lipid peroxidation in vitro using linoleic acid in comparison to well-known antioxidant such as ascorbic acid and tannic acid. The total phenolic content of the cereal grain (80% ethanolic extracts) investigated in this study varied from 2.1 mg/g (wheat cv. Olgeurumil) to 10.4 mg/g (barely cv. Seodunchalbori). Highly positive relationship between total phenol compounds and antioxidant activity was found. When the antioxidant activities of all investigated extracts were measured with application of same quantity of phenol compounds, oat grain extracts showed similar antioxidant activity of barely cultivars. However, barely extract appeared as the most potent antioxidant activity of inhibition of UV -induced lipid peroxidation. This indicated that factors such as phenolic compound composition and their individual antioxidant activity could playa crucial role in the total antioxidant activity of cereal grains.
