A viral disease causing severe mosaic, necrotic, and yellow symptoms on Vigna angularis var. nipponensis was prevalent around Suwon area in Korea. The causal virus was characterized as Cucumber mosaic virus (CMV) on the basis of biological and nucleotide sequence properties of RNAs 1, 2 and 3 and named as CMV-wVa. CMV-wVa isolate caused mosaic symptoms on indicator plants, Nicotiana tabacum cv. Xanthi-nc, Petunia hybrida, and Cucumis sativus. Strikingly, CMV-wVa induced severe mosaic and malformation on Cucurbita pepo, and Solanum lycopersicum. Moreover, it caused necrotic or mosaic symptoms on V. angularis and V. radiate of Fabaceae. Symptoms of necrotic local or pin point were observed on inoculated leaves of V. unguiculata, Vicia fava, Pisum sativum and Phaseolus vulgaris. However, CMV-wVa isolate failed to infect in Glycine max cvs. 'Sorok', 'Sodam' and 'Somyeong'. To assess genetic variation between CMV-wVa and the other known CMV isolates, phylogenetic analysis using 16 complete nucleotide sequences of CMV RNA1, RNA2, and RNA3 including CMV-wVa was performed. CMV-wVa was more closely related to CMV isolates belonging to CMV subgroup I showing about 85.1-100% nucleotide sequences identity to those of subgroup I isolates. This is the first report of CMV as the causal virus infecting wild Vigna angularis var. nipponensis in Korea.

The development of reverse transcription-polymerase chain reaction using degenerate primers against conserved regions of most potyviral genomes enabled sampling of the potyvirome. However, these assays usually involve sampling potential host plants, but identifying infected plants when they are asymptomatic is challenging, and many plants, especially wild ones, contain inhibitors to DNA amplification. We used an alternative approach which utilized aphid vectors and indicator plants to identify potyviruses capable of infecting common bean (Phaseolus vulgaris). Aphids were collected from a range of asymptomatic leguminous weeds and trees in Iran, and transferred to bean seedlings under controlled conditions. Bean plants were tested serologically for potyvirus infections four-weeks postinoculation. The serological assay and symptomatology together indicated the presence of one potyvirus, and symptomology alone implied the presence of an unidentified virus. The partial genome of the potyvirus, encompassing the complete coat protein gene, was amplified using generic potyvirus primers. Sequence analysis of the amplicon confirmed the presence of an isolate of Wisteria vein mosaic virus (WVMV), a virus species not previously identified from Western Asia. Phylogenetic analyses of available WVMV sequences categorized them into five groups: East Asian-1 to 3, North American and World. The Iranian isolate clustered with those in the World group. Multiple sequence alignment indicated the presence of some genogroup-specific amino acid substitutions among the isolates studied. Chinese isolates were sister groups of other isolates and showed higher nucleotide distances as compared with the others, suggesting a possible Eastern-Asian origin of WVMV, the main region where Wisteria might have originated.
After a mixed carbohydrate diet, inhibition of ${\alpha}$-amylase and ${\alpha}$-glucosidase involved in the digestion and absorption of carbohydrates can significantly decrease the postprandial increase of blood glucose level. In the course of screening these useful enzyme inhibitors, we selected five kinds of bean, using an in-vitro enzyme inhibition assay method. To evaluate the effect of germination process on the functionality of the bean, we investigated the inhibitory activities of the water extracts of non-germinated bean and germinated bean against ${\alpha}$-amylase and ${\alpha}$-glucosidase, relevant to postprandial hyperglycemia. We also investigated the oxygen radical absorbance capacity(ORAC), total phenolics content, and postprandial blood glucose lowering effect in rats(Sprague-Dawley rat model). Most germinated beans showed significantly higher ${\alpha}$-glucosidase inhibitory activity, compared with non-germinated beans. Among germinated beans, Glycine max had the highest ${\alpha}$-glucosidase inhibitory activity(53.3%). The water extract of germinated Phaseolus vulgaris L. had the highest ${\alpha}$-amylase inhibitory activity(95.1%), followed by Glycine max(58.7%), and Glycine max L. Merr(54.1%). Furthermore, the five germinated beans also showed high antioxidant activities in ORAC assay. Results suggested that the germination process may improve and enhance the anti-hyperglycemia potential and antioxidant activity of the bean.
The ${\alpha}-Amylase$ inhibitor from black bean(Phaseolus vulgaris) was purified to homogeneity using 70% saturated ammonium sulfate, DEAF-cellulose, Concanavalin-A sepharose chromatography and gel filtration with Superose 6. The purified α-amylase inhibitor showed a single band of 25 KD in molecular weight on the SDS-PAGE. The specific activity of the inhibitor was 544.0 units/mg and the purity was enhanced about 18-fold. The amino acids of ${\alpha}-Amylase$ inhibitor from black bean was mainly glutamic acid, aspartic acid and lysine. The inhibitor was glycoproteins and its carbohydrate contents was 3.2%.

The seeds of soybean (Glycine max), adzuki bean (Vigna angularis), mung bean (Vigna radiata), and kidney bean (Phaseolus vulgaris L.) were examined the germination rate after 10 years of long-term storage ($-18^{\circ}C$) conservation. For soybean seeds, 2,313 accessions were examined and germination rate of 1,082 accessions was decreased with below 15% of initial germination rate. For 227 accessions of soybean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 589 accessions was increased and showed no change for 415 accessions after 10 years of long-term storage. For adzuki bean seeds, 2,058 accessions were examined and germination rate of 739 accessions was decreased with below 15% of initial germination rate. For 63 accessions of adzuki bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 535 accessions was increased and showed no change for 721 accessions after 10 years of long-term storage. For mung bean seeds, 438 accessions were examined and germination rate of 139 accessions was decreased with below 15% of initial germination rate. For 5 accessions of mung bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 155 accessions was increased and showed no change for 139 accessions after 10 years of long-term storage. For kdney bean seeds, 366 accessions were examined and germination rate of 7 accessions was decreased with below 15% of initial germination rate. For 65 accessions of kidney bean, germination rate was decreased with above 15% of initial germination rate after 10 years of long-term storage, which is needed to be rejuvenated. Germination rate of 201 accessions was increased and showed no change for 93 accessions after 10 years of long-term storage.
Stem rot symptoms of common bean occurred sporadically in Jinju, Korea. The typical symptom included water-soaking on the main stem, rotting, wilting, and blighting, which eventually leads to death of the plant. The sclerotia, white to brown, spherical with size of 1-3 mm, formed over lesions and surface soil line. The optimum temperature for mycelial growth and sclerotia formation was $30^{\circ}C$ on potato dextrose agar (PDA). The typical clamp connections were observed in the hyphae of the fungus grown on PDA. Mycological characteristics and pathogenicity to host plants indicated that the fungus was Sclerotium rolfsii Saccardo. This is the first report of stem rot on common bean caused by S. rolfsii in Korea.

Trout production is a growing activity in recent years but requires new alternative sources of feed to be sustainable over time. The objective of this research was to determine the apparent digestibility coefficient (ADC) of dry matter (DM), organic matter (OM), crude protein (CP) and digestible energy (DE) of $ka{\tilde{n}}iwa$ (Chenopodium pallidicaule Aellen), kiwicha (Amaranthus caudatus L), quinoa (Chenopodium quinoa Willd), beans (Phaseolus vulgaris L.), sacha inchi, (Plukenetia volubilis L) and jumbo squid (Dosidicus gigas) meal in juvenile rainbow trout. The experimental diets were composed of a 70% basal diet and 30% of any raw materials. The ADC was determined by the indirect method using insoluble ash as a non-digestible marker. Jumbo squid, sacha inchi and quinoa showed the highest values of ADC (%) of DM (84.5, 73.5 and 69.7), OM (89.1, 78.4 and 72.9), CP (93.2, 98.0 and 90.3), and DE (4.57, 4.15 and 2.95 Mcal/kg DM), respectively. The ADC values for $ka{\tilde{n}}iwa$, kiwicha and bean were significantly lower. In conclusion, quinoa meal and jumbo squid meal have an acceptable digestibility but sacha inchi meal is a potential alternative for rainbow trout feeding in the future.

Common bean (Phaseolus vulgaris L.) is one of the most important crops in human food production. The occurrence of diseases, such as white mold, caused by Sclerotinia sclerotiorum can limit the production of this legume. The use of Trichoderma has become an important strategy in the suppression of this disease. The aim of the present study was to evaluate the effect of volatile organic compounds (VOCs) emitted by Trichoderma azevedoi CEN1241 in five different growth periods on the severity of white mold in common bean. The in vitro assays were carried out in double-plate and split-plate, and the in vivo assays, through the exposure of the mycelia of S. sclerotiorum to the VOCs of T. azevedoi CEN1241 and subsequent inoculation in bean plants. Chemical analysis by gas chromatography coupled to mass spectrometry detected 37 VOCs produced by T. azevedoi CEN1241, covering six major chemical classes. The profile of VOCs produced by T. azevedoi CEN1241 varied according to colony age and was shown to be related to the ability of the biocontrol agent to suppress S. sclerotiorum. T. azevedoi CEN1241 VOCs reduced the size of S. sclerotiorum lesions on bean fragments in vitro and reduced disease severity in a greenhouse. This study demonstrated in a more applied way that the mechanism of antibiosis through the production of volatile compounds exerted by Trichoderma can complement other mechanisms, such as parasitism and competition, thus contributing to a better efficiency in the control of white mold in bean plants.

This study was conducted to obtain basic data on efficient and eco-friendly crop cultivation for urban residents who enjoy urban agriculture as a hobby or leisure activity. We planted strawberry(Fragaria x ananassa)(S) and kidney bean (Phaseolus vulgaris var. humilis)(K) in different ratios to analyze the differences in soil environment, growth, physiology, and productivity, and to investigate the effects. Strawberry growth was optimal with S1K2 ratio, whereas the S1K1 ratio treatment showed the highest levels of physiology and productivity. In terms of growth and physiology, kidney beans tended to perform best in S1K2 treatment. The average number of productive pods was two, with a highest average value of 2.3 being recorded in S2K1 treatment planted with a high percentage of strawberries. In terms of growth, physiology, and prodctivity our findings indicate that it would be desirable to plant starwberries and kidney bean in a 1:1 ratio. However, considering the environmental characteristics of walls and rooftops, it is necessary to effectively manage crops suitable for these conditions. Furthermore, additional studies should be conducted to analyze the quality of fruits and seeds producted, both qualitatively and quantitatively in the future.
The measurements of rubisco parameters are important in photosynthetic studies. In this experiment, we used photometric assay method to detect these major parameters, such as activity, carbamylation and amount of rubisco. The main advantages of this method are very simple and as sensitive as conventional methods which usually produce radioactive waste. In this study, with kidney bean (Phaseolus vulgatis L.) leaves grown at normal $CO_2$ (350ppm) and high $CO_2$ (650 ppm), we investigated the effect of $CO_2$ concentration on activation and carbamylation of rubisco by measuring the rubisco activity, carbamylation rate and amount of rubisco using a dual beam (334nm and 405nm) spectrophotometer, and analyzed the polypeptide profiles of rubisco by SDS-PAGE. When $CO_2$ concentration was raised from 350ppm to 650ppm, all parameters of rubisco were decreased : $41.2{\mu}M/m^2/s and 52.2{\mu}M/m^2/s$ to $27.4{\mu}M/m^2/s and 46.1{\mu}M/m^2/s$ for initial and total rubisco activity, respectively ; from 79% to 58.9% for carbamylation rate ; from $1.94 {\mu}M/m^2$ to 1.58{\mu}M/m^2$ for amount of rubisco. These results suggests that the decrease in rubisco activity at high $CO_2$ was caused by carbamylation. The analysis of the preparation by SDS-PAGE showed two major polypeptides at 50 and 14.5 kD which were identified as the large and the small subunits of rubisco. There were no differences in the intensity compared high $CO_2$ to normal $CO_2$ in both 50 kD and 14.5 kD bands. We also found that these inhibitory effects of $CO_2$ were reversible. When high $CO_2$ was switched to normal $CO_2$, the parameters of rubisco changed were almost the same as normal rubisco parameters. These data provide an evidence that activity of rubisco was recovered by $CO_2$ concentration of 350 ppm.
