Effect of Arresting MCF-7 Human Breast Carcinoma Cell at G2/M Phase of Trichosanthes Kirilowii (천화분이 MCF-7 유방암 세포주의 G2/M 세포주기 억제에 미치는 영향)

  • The purpose of this study is to investigate the anti-proliferative mechanism by Trichosanthes kirilowii (TCK) in MCF-7 human breast carcinoma cell. In this study, we used human breast cancer cell line, Michigan cancer foundation-7 cells (MCF-7 cells). They were co-incubated with 30~200 ${\mu}g$/ml TCK for 48 hours, and cell viability was measured by Water-soluble tetrazolium salt-1 (WST-1) assay. After MCF-7 cells were exposed to 60 ${\mu}g$/ml of TCK for 0, 3, 6, 12, 24, 48 hours, We performed flow analysis cytometry sorting(FACS) and western blot analysis. We investigated the effect of dose-dependent cell growth inhibition by TCK, which could be proved by WST-1 assay. Also, flow cytometry analysis showed that TCK increased percentage of subG1 phase and G2/M phase cell cycle. In addition, TCK induced apoptosis through the expression of caspase-9, -3 and poly(ADP-ribose) polymerase(PARP) activation. Moreover, we showed that ATM-dependent G2/M phase arrest by DNA damage and phosphorylation of chk2, cdc25C, cdc2(Tyr15). Taken together, these results suggest that by G2/M phase arrest through DNA damage and inducing of apoptosis through intrinsic pathway, TCK may have potential tumor suppressor in breast cancer.

  • In this paper, the characteristics of frequency dependent dispersion are analyzed for the phase shifters in Ku-band phased array antenna system using full-wave method, SDA(Spectral Domain Approach).The design technique of asymmetric coupled-line phase shifter with dielectric overlay is proposed. The compared results of phase responses are shown with conventional symmetric coupled-line Schiffman phase shifter. The experimental results verify the improvement of broadband characteristics by the increasing bandwidths and the decreasing phase deviations.

  • A novel torque sharing function (TSF) is presented. To improve efficiency and to reduce torque ripple in commutation region, only a phase torque under commutation is regulated to produce a uniform torque. And the torque developed by the other phase remains with the previous state under a current limit of the motor and drive. If the minimum change of a phase torque reference can not satisfy the total reference torque, two-phase changing mode is used. Since a phase torque is constant and the other phase torque is changed at each rotor position, total torque error can be reduced within a phase torque error limit. And the total torque error is dependent on the change of phase torque. To consider non-linear torque characteristics and to suppress a tail current at the end of commutation region, the incoming phase current is changed to torque increasing direction, but the outgoing phase current is changed to torque decreasing direction. So, the torque sharing of the outgoing phase and incoming phase can be smoothly changed with a minimum current cross over. The proposed control scheme is verified by some computer simulations and experimental results.

Cell Cycle-Dependent Activity Change Of $Ca^{2+}/$Calmodulin-Dependent Protein Kinase II In NIH 3T3 Cells

  • Although the blockage of a cell cycle by specific inhibitors of $Ca^{2+}/$calmodulin-dependent protein kinase II (CaMK-II) is well known, the activity profile of CaMK-II during the cell cycle in the absence of any direct effectors of the enzyme is unclear. The activity of native CaMK-II in NIH 3T3 cells was examined by the use of cell cycle-specific arresting and synchronizing methods. The total catalytic activity of CaMK-II in arrested cells was decreased about 30% in the M phase, whereas the $Ca^{2+}$-independent autonomous activity increased about 1.5-fold in the M phase and decreased about 50% at the G1/S transition. The in vivo phosphorylation level of CaMK-II was lowest at G1/S and highest in M. The CaMK-II protein level was unchanged during the cell cycle. When the cells were synchronized, the autonomous activity was increased only in M. These results indicate that the physiologically relevant portion of CaMK-II is activated only in M, and that the net activation of CaMK-II is required in mitosis.

Dihydroartemisinine Enhances Dictamnine-induced Apoptosis via a Caspase Dependent Pathway in Human Lung Adenocarcinoma A549 Cells

  • Dictamnine (Dic) has the ability to exert cytotoxicity in human cervix, colon, and oral carcinoma cells and dihydroartemisinin (DHA) also has potent anticancer activity on various tumour cell lines. This report explores the molecular mechanisms by which Dic treatment and combination treatment with DHA and Dic cause apoptosis in human lung adenocarcinoma A549 cells. Dic treatment induced concentration- and time-dependent cell death. FCM analysis showed that Dic induced S phase cell cycle arrest at low concentration and cell apoptosis at high concentration in which loss of mitochondrial membrane potential (${\Delta}{\Psi}m$) was not involved. In addition, inhibition of caspase-3 using the specific inhibitor, z-DQMD-fmk, did not attenuate Dic-induced apoptosis, implying that Dic-induced caspase-3-independent apoptosis. Combination treatment with DHA and Dic dramatically increased the apoptotic cell death compared to Dic alone. Interestingly, pretreatment with z-DQMD-fmk significantly attenuated DHA and Dic co-induced apoptosis, implying that caspase-3 plays an important role in Dic and DHA co-induced cell apoptosis. Collectively, we found that Dic induced S phase cell cycle arrest at low concentration and cell apoptosis at high concentration in which mitochondria and caspase were not involved and DHA enhanced Dic induced A549 cell apoptosis via a caspase-dependent pathway.

NUMERICAL INVESTIGATION OF THE SPREADING AND HEAT TRANSFER CHARACTERISTICS OF EX-VESSEL CORE MELT

  • The flow and heat transfer characteristics of the ex-vessel core melt (corium) were investigated using a commercial CFD code along with the experimental data on the spreading of corium available in the literature (VULCANO VE-U7 test). In the numerical simulation of the unsteady two-phase flow, the volume-of-fluid model was applied for the spreading and interfacial surface formation of corium with the surrounding air. The effects of the key parameters were evaluated for the corium spreading, including the radiation, decay heat, temperature-dependent viscosity and initial temperature of corium. The results showed a reasonable trend of corium progression influenced by the changes in the radiation, decay heat, temperature-dependent viscosity and initial temperature of corium. The modeling of the viscosity appropriate for corium and the radiative heat transfer was critical, since the front progression and temperature profiles were strongly dependent on the models. Further development is required for the code to consider the formation of crust on the surfaces of corium and the interaction with the substrate.

  • Ti2O3 is known as a typical Mott insulator with a transition temperature of near $200^{\circ}C$. Unlike VO2, Ti2O3 does not have a structural phase transition near the metal-insulator-transition (MIT) temperature. We investigated the temperature-dependent thermal vibration change using temperature-dependent x-ray absorption fine structure (XAFS) at Ti K-edge in the temperature range of 300~600 K. Ti2O3 powder and films were synthesized using thermal chemical vapor deposition (CVD) at $800{\sim}900^{\circ}C$. X-ray diffraction measurements show a single phased Ti2O3 at room temperature. XAFS confirmed no structural phase transition in the temperature of 300~600 K. A small but distinguishable structural disorder change was observed near the transition temperature. We will discuss the MIT behavior with the change of structural disorder.

Tannic acid-induced apoptosis in FaDu hypopharyngeal squamous cell carcinoma

  • Tannic acid (TA) is a water-soluble polyphenol compound found in various herbal plants. We investigated the chemopreventive effects of TA on FaDu hypopharyngeal squamous carcinoma cells. In an 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, TA showed dose-dependent cytotoxicity with a half maximal inhibitory concentration (IC50) of 50 ?M. Cell cycle analysis and immunofluorescence imaging demonstrated that under low-dose ($25{\mu}M$) treatment, FaDu cells were arrested in G2/M phase, and as the dose of TA was increased, apoptosis was induced with the increase of cell population at sub-G1 phase. The expressions of various cyclins, including cyclin D1 and cyclin-dependent kinases (CDK-1 and CDK-2), were down-regulated at low doses of TA, whereas apoptotic effectors such as cleaved caspase 3, cleaved caspase 7, and poly (ADP-ribose) polymerase (PARP) were expressed in a dose-dependent manner in Western blotting. In addition, TA-induced apoptosis of FaDu cells might be mediated by the extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinase pathway, with the upregulation of p-AKT/p-PKB (phosphorylated protein kinase B) and p-ERK. Overall, our data support the hypothesis that TA is a potential candidate agent for the treatment of hypopharyngeal cancer.

Thermoelastic interaction in functionally graded nanobeams subjected to time-dependent heat flux

  • This paper investigates the vibration phenomenon of a nanobeam subjected to a time-dependent heat flux. Material properties of the nanobeam are assumed to be graded in the thickness direction according to a novel exponential distribution law in terms of the volume fractions of the metal and ceramic constituents. The upper surface of the functionally graded (FG) nanobeam is pure ceramic whereas the lower surface is pure metal. A nonlocal generalized thermoelasticity theory with dual-phase-lag (DPL) model is used to solve this problem. The theories of coupled thermoelasticity, generalized thermoelasticity with one relaxation time, and without energy dissipation can extracted as limited and special cases of the present model. An analytical technique based on Laplace transform is used to calculate the variation of deflection and temperature. The inverse of Laplace transforms are computed numerically using Fourier expansion techniques. The effects of the phase-lags (PLs), nonlocal parameter and the angular frequency of oscillation of the heat flux on the lateral vibration, the temperature, and the axial displacement of the nanobeam are studied.
