본 연구는 간암세포주인 HepG2 cell line에서 녹차씨박 추출물을 에탄올, 석유 에테르, 에틸 아세테이트, 부탄올의 순으로 분획 및 열수 추출물을 조제하여 암세포 증식 억제능을 확인한 후 에탄올 추출물을 선정하여 항종양 및 항염증효과를 생화학적, 분자적 방법으로 분석하였다. 녹차씨박 에탄올 추출물(DGTSE)의 polyphenol 함량을 HPLC로 분석한 결과 EGC($1039.1{\pm}15.26\;{\mu}g/g$)> tannic acid($683.5{\pm}17.61\;{\mu}g/g$)> EC($62.4{\pm}5.00\;{\mu}g/g$)> ECG($24.4{\pm}7.81\;{\mu}g/g$)> EGCG($20.9{\pm}0.96\;{\mu}g/g$)> gallic acid($2.4{\pm}0.68\;{\mu}g/g$)의 순이었으나 caffeic acid는 검출되지 않았다. DGTSE의 농도가 $10\;{\mu}g$/mL 이상에서는 84.13%의 세포 증식 억제능($IC_{50}$: $6.58\;{\mu}g$/mL)을 보여 간암세포의 증식을 효과적으로 억제하였고 제1상효소계인 CYP1A1와 CYP1A2의 발현을 농도 의존적으로 감소시키는 것으로 나타났다. QR 활성은 DGTSE을 $20\;{\mu}g$/mL 농도로 처리 시 대조군에 비해 2.6배 증가하였으며 reporter gene activity로 측정한 ARE 활성은 1.94배로 각각 증가하였다. DGTSE의 처리는 염증생성 인자인 PGE2의 생성과 TNF-${\alpha}$의 단백질 발현은 유의적으로 저해하였으며 cytokine 반응, 염증, 세포성장조절과 같은 다양한 단계에 참여하는 전사인자인 NF${\kappa}$B의 핵으로의 translocation을 억제하였다. 이상의 결과에서 DGTSE은 간암세포인 HepG2 세포계에서 암세포 증식 억제능을 가지며 해독효소인 QR, ARE의 활성을 증진시키고 NF${\kappa}$B의 translocation을 방해하고 TNF-${\alpha}$의 단백질 발현과 $PGE_2$의 생성을 억제하여 암세포의 증식을 억제하였다. 따라서 녹차씨박 추출물에 함유된 암세포 증식효과를 나타내는 생리활성 물질들에 대한 연구가 앞으로 진행되어야 할 것으로 사료된다.
Cancer is a complex disease and the genetic susceptibility to it could be an outcome of the inherited difference in the capacity of xenobiotic metabolizing enzymes. Glutathione-S-transferases (GSTs) are phase II metabolizing enzymes whose various genotypes have been associated with increased risk of different types of cancer. Null mutations caused by the deletion of the entire gene result in the absence of the enzymatic activity and increase in the risk of developing cancer including chronic myeloid leukaemia (CML). In the present case-control study we evaluated the effect of null mutations in GSTM1 and GSTT1 genes on the risk of developing CML. The study included 75 CML patients (43 males and 32 females; age (mean ${\pm}$ S.D) $42.3{\pm}13.4$ years) and unrelated non-malignant controls (76 male and 48 females; age (mean ${\pm}$ S.D) $41.5{\pm}12.9$). The distribution of GSTM1 and GSTT1 genotypes in CML patients and controls was assessed by multiplex-PCR method. Logistic regression was used to assess the relationship between GSTM1 and GSTT1 genotypes and risk of CML. Chi-square test was used to evaluate the trend in modulating the risk to CML by one or more potential high risk genotype. Although GSTM1 null genotype frequency was higher in CML patients (41%) than in the controls (35%), it did not reached a statistical significance (OD = 1.32, 95% CI: 0.73-2.40; P value = 0.4295). The frequency of GSTT1 null genotypes was higher in the CML patients (36%) than in the controls (21%) and the difference was found to be statistically significant (OD = 2.12, 95% CI: 1.12-4.02; P value = 0.0308). This suggests that the presence of GSTT1genotype may have protective role against the CML. We found a statistically significant (OD = 3.09, 95% CI: 1.122-8.528; P value = 0.0472) interaction between the GSTM1 and GSTT1 null genotypes and thus individuals carrying null genotypes of both GSTM1 and GSTT1 genes are at elevated risk of CML.
Nuclear factor erythroid 2-related factor 2 (Nrf2) has been recognized as a transcription factor that controls mechanisms of cellular defense response by regulation of three classes of genes, including endogenous antioxidants, phase II detoxifying enzymes and transporters. Previous studies have revealed roles of Nrf2 in resistance to chemotherapeutic agents and high level expression of Nrf2 has been found in many types of cancer. At physiological concentrations, luteolin as a flavonoid compound can inhibit Nrf2 and sensitize cancer cells to chemotherapeutic agents. We reported luteolin loaded in phytosomes as an advanced nanoparticle carrier sensitized MDA-MB 231 cells to doxorubicin. In this study, we prepared nano phytosomes of luteolin to enhance the bioavailability of luteolin and improve passive targeting in breast cancer cells. Our results showed that cotreatment of cells with nano particles containing luteolin and doxorubicin resulted in the highest percentage cell death in MDA-MB 231cells (p<0.05). Furthermore, luteolin-loaded nanoparticles reduced Nrf2 gene expression at the mRNA level in cells to a greater extent than luteolin alone (p<0.05). Similarly, expression of downstream genes for Nrf2 including Ho1 and MDR1 were reduced significantly (p<0.05). Inhibition of Nrf-2 expression caused a marked increase in cancer cell death (p<0.05). Taken together, these results suggest that phytosome technology can improve the efficacy of chemotherapy by overcoming resistance and enhancing permeability of cancer cells to chemical agents and may thus be considered as a potential delivery system to improve therapeutic protocols for cancer patients.
Glutathione S-transferase (GST) is a multigene family of phase II detoxifying enzymes that metabolize a wide range of exogenous and endogenous electrophilic compounds. GSTM1 and GSTT1 gene polymorphisms may account for inter-individual variability in coping with oxidative stress. We investigated the relationships between the level of lymphocyte DNA and antioxidative parameters and the effect on GST genotypes. GSTM1 and GSTT1 were characterized in 301 young healthy Korean adults and compared with oxidative stress parameters such as the level of lymphocyte DNA, plasma antioxidant vitamins, and erythrocyte antioxidant enzymes in smokers and non smokers. GST genotype, degree of DNA damage in lymphocytes, erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase (GSH-Px), and plasma concentrations of total radical-trapping antioxidant potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene, and cryptoxanthin were analyzed. Lymphocyte DNA damage assessed by the comet assay was higher in smokers than that in non-smokers, but the levels of plasma vitamin C, ${\beta}$-carotene, TRAP, erythrocyte catalase, and GSH-Px were lower than those of non-smokers (p < 0.05). Lymphocyte DNA damage was higher in subjects with the GSTM1- or GSTT1-present genotype than those with the GSTM1-present or GSTT1- genotype. No difference in erythrocyte antioxidant enzyme activities, plasma TRAP, or vitamin levels was observed in subjects with the GSTM1 or GSTT1 genotypes, except ${\beta}$-carotene. Significant negative correlations were observed between lymphocyte DNA damage and plasma levels of TRAP and erythrocyte activities of catalase and GSH-Px after adjusting for smoking pack-years. Negative correlations were observed between plasma vitamin C and lymphocyte DNA damage only in individuals with the GSTM1-present or GSTT1- genotype. The interesting finding was the significant positive correlations between lymphocyte DNA damage and plasma levels of ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. In conclusion, the GSTM1- and GSTT1-present genotypes as well as smoking aggravated antioxidant status through lymphocyte DNA damage. This finding confirms that GST polymorphisms could be important determinants of antioxidant status in young smoking and non-smoking adults. Consequently, the protective effect of supplemental antioxidants on DNA damage in individuals carrying the GSTM1- or GSTT1-present genotypes might show significantly higher values than expected.
브로콜리와 같은 십자화과 식물에서 glucoraphanin의 가수분해를 통해 생성되는 isothiocyanate의 일종인 sulforaphane은 강력한 항암효과를 가지며, 역학적 조사를 포함한 다양한 선행 연구에서 androgen 비 의존적으로 성장하는 전립선 암세포의 증식을 억제하는데 효과가 있었다. 최근 연구 결과에 따르면 sulforaphane은 다양한 인체암세포의 증식을 억제하고 apoptosis를 유발할 수 있는 것으로 알려지고 있으나, 정확한 분자생물학적 기전은 밝혀져 있지 않은 상태이다. 본 연구에서는 sulforaphane의 항암작용 기전을 조사하기 위하여 HeLa 인체자궁경부암세포의 증식에 미치는 sulforaphane의 영향을 조사하였다. Sulforaphane의 처리에 의한 HeLa 세포의 증식억제 및 형태적 변형은 세포주기 C2/M arrest 및 apoptosis 유발과 밀접한 관련이 있음을 알 수 있었다. RT-PCR 및 Western blot 분석 결과, sulforaphane 처리에 의하여 cyclin A 및 cyclin-dependent kinase (Cdk)4 단백질의 발현이 선택적으로 저하되었으며, Cdc2, Cdk inhibitor인 p16 및 p21의 발현은 증가되었다 그러나 sulforaphane은 cyclooxygenases의 발현이나 telomere 조절에 중요한 역할을 하는 인자들의 발현에는 큰 영향을 주지 못하였다. Sulforaphane의 항암 기전을 규명하기 위해서는 더 많은 연구가 부가적으로 필요하겠지만, 본 연구의 결과들에 의하면 sulforaphane은 강력한 인체암세포의 증식 억제 및 항암작용이 있을 것을 시사하여 준다고 할 수 있다.
본 연구의 목적은 항 methicillin-resistant Staphylococcus aureus (MRSA) 활성을 가지는 해양미생물을 분리, 동정하고 항MRSA 물질을 분리 정제하여 그 물질의 활성과 시너지 효과를 밝히는 것이다. 본 연구에서 분리한 해양미생물 중에서, YJ-1 분리 균주가 가장 강한 항MRSA 활성을 나타내었다. YJ-1 균주는 생화학적 특성과 16S rRNA 유전자 염기서열에 기초하여 분류 동정되었다. YJ-1 균주의 염기서열은 Pseudomonas stutzeri와 99.2%의 상동성을 나타내어, Pseudomonas sp. YJ-1이라 명명하였다. 이 균주의 최적 성장조건은 $25^{\circ}C$와 초기 pH 농도 7.0이었다. 항MRSA 물질들을 분리정제하기 위하여 YJ-1 균주를 PPES-II 배지에 배양하였으며, 배양 상등액을 ethyl acetate, hexane과 80% MeOH로 순차적으로 추출하였다. 활성을 보인 80% MeOH 분획을 $C_{18}$ ODS 칼럼 크로마토그래피, silica gel 크로마토그래피와 역상 HPLC법으로 순차적으로 정제하여 항MRSA 활성을 가지는 3개의 순수물질 MR1, MR2 및 MR3를 얻었다. $250{\mu}g\;mL^{-1}$ 농도의 MR1 물질을 MRSA 세포에 접종했을 때, MRSA 세포의 95%가 48시간 이내에 사멸하였다. 활성을 vancomycin 및 ampicillin과 비교해 보아도, MR1 물질은 보다 우수한 항MRSA 활성을 나타내었다. 또한 항MRSA 활성은 투여량과 시간에 비례하여 증가하였다. 더욱이 MR1 물질과 vancomycin을 조합하여 시너지 효과를 보았을 때, MR1 물질을 단독으로 투여했을 때보다 신속한 MRSA 세포의 감소를 관찰할 수 있었다. 이상의 결과를 종합해보면, MRSA 감염에 있어서 Pseudomonas sp. YJ-1과 이것이 생산하는 항MRSA 물질은 천연 항균제로서 기여할 수 있으리라 판단된다.
치주조직에 존재하는 주요한 세포의 한 형태인 인체 치은섬유아세포는 다양한 구강유해세균으로부터 염증이 유발되어지며, 그중 대표적으로 치주염 원인균인 P. gingivalis의 내독소인 LPS-PG로부터 염증성 자극에 반응하여 다양한 염증매개 물질을 분비한다. 본 연구에서는 치주염을 일으키는 주요한 원인균 중 하나인 P. gingivalis로 부터 분리한 LPS-PG를 이용하여 인체 치은섬유아세포주인 HGF-1 세포에 염증을 유도한 후 LRE에 대한 항염증 및 항산화 효과를 분석하였다. 실험 결과, LRE는 LPS-PG 유도에 따라 iNOS에 의한 NO 생성과 COX-2에 의한 PGE2와 같은 염증 매개 인자의 발현 및 생성 억제와 함께 염증성 싸이토카인(TNF-α, IL-1β및 IL-6)의 생성 또한 억제하였다. 신호전달계에서 염증성 전사인자의 발현 경로를 확인하기 위하여 TLR4/Myd88/NF-κB의 활성을 확인한 결과, LRE 처리에 따라 농도 의존적으로 억제되는 것을 확인하였다. 또한 산화 환원 효소로 항염증효과를 나타내는 것으로 알려진2상 효소 중 하나인 NQO-1과 이의 전사인자인 Nrf2를 분석 한 결과 LRE 처리에 의해 효소의 활성이 높아지는 것을 확인할 수 있었다. 결론적으로 LRE는 TLR4/Myd88/NF-κB 신호전달 경로를 억제하고 NQO1/Nrf2 활성을 유도함으로써 HGF-1 세포에서 LPS-PG에 의해 유도된 염증을 억제하는 것으로 사료되며, 향후 LRE는 식·의약품 소재 개발에서 치주질환 개선의 가능성이 있는 후보물질이 될 수 있을 것으로 사료된다.
