Effect of CRATAEGII FRUCTUS(山査) on Antioxidant Capacity in D-galactose induced Aging Rats (산사(山査)가 노화유발(老化誘發) 흰쥐의 항산화능(抗酸化能)에 미치는 영향(影響))

  • Objectives : CRATAEGII FRUCTUS(山査) is known as the substance which delays aging by the antioxidant effects. The purpose of this study is to investigate the effects of CRATAEGII FRUCTUS(山査) on antioxidant enzyme activities such as Thiobarbituric acid reactive substance(TBARS), Superoxide dismutase(SOD), Catalase(CAT), Glutathione peroxidase (GSH-px) in rat erythrocytes and blood plasma. Methods : Sprague-Dawley rats were divided into 3 groups, Normal group (supplied enough water and feeds only, Normal Group), D-galatose administered group(injected D-galatose 50mg/kg, 1time/day for 6 weeks, Control Group) and CRATAEGII FRUCTUS(山査) administered group (D-galactose 50mg/kg and CRATAEGII FRUCTUS(山査) extracts 85.0mg/200g 1time/day for 6 weeks, SS Group). Rats were sacrificed and TBARS, SOD, CAT, GSH-px, neutral fat and cholesterol were measured in rat erythrocytes and blood plasma. Results : There was no significant difference in the level of TBARS in the blood plasma between each experimental group. Red blood cell SOD activities were significantly different in each group. They were significantly decreased in the Control group compared to those of Normal and there was an increasing phenomenon in the SS group compared to the Control group. There was a significant difference in the activities of the red blood cell - glutathione peroxides in each group. There was a significant increase in the SS group compared to the Control group. Red blood cell catalase activities was no significant difference in each group. Plasma total lipid concentration was significantly different in each group. It was significantly increased in the Control group compared with the Normal group and it was decreasing in the SS group compared to the control group. Plasma triglyceride was not significantly different in each group. Plasma total cholesterol and Plasma HDL -cholesterol concentrations in the blood plasma were not significantly different in each group. Conclusions : According to the above results, it is considered that CRATAEGII FRUCTUS(山査) is effective in inhibiting lipid peroxides and increasing the activities of antioxidative(anti aging) enzyme in D-galactose induced aging rat.

Protection Effects of Allylmercaptan, Metabolite of Garlic on Endothelial Cell Injury Induced by Oxidized Low Density Lipoprotein (산화된 low density lipoprotein (LDL)에 의해 유도된 내피세포의 손상에 대한 마늘 대사산물인 allylmercaptan의 보호 효과)

  • Oxidation of low density lipoprotein (LDL) has been recognized as an important role in the initiation and progression of atherosclerosis. In this study, effects of allylmercaptan, a major metabolite compound of garlic, was studied on endothelial cell injury induced by oxidized low density lipoprotein (ox-LDL). The antioxidative activity of allylmercaptan was investigated by monitoring a thiobarbituric acid substance (TBARS). Allylmercaptan inhibited LDL oxidation induced by $Cu^{2+}$ at concentrations of 0.1, 1 and 10 mM in a dose dependent manner. Lactate dehydrogenase (LDH) release, as an index of cell injury, and intracellular glutathione levels were determined. Pulmonary artery endothelial cells were preincubated with allylmercaptan at $37^{\circ}C$ and 5% $CO_2$ for 24 hr, washed, and then exposed to 0.1 mg/ml oxidized LDL for 24 hr. Preincubation of endothelial cells with allylmercaptan significantly prevented the LDH release and depletion of GSH. Peroxides were measured directly in 24 well plates using a fluorometric assay. Allylmercaptan inhibited release of peroxides induced by ox-LDL in pulmonary artery endothelial cells. In a free system, allylmercaptan was shown to scavenge hydrogen peroxide. The data indicate that allylmercaptan can protect pulmonary artery endothelial cells from injury caused by oxidized LDL, and suggest that allylmercaptan may be useful for the prevention of atherosclerosis.

Effect of Solvent Extracts from Sargassum hemiphyllum on Inhibition of Growth of Human Cancer Cell Lines and Antioxidant Activity (짝잎모자반(Sargassum hemiphyllum)의 암세포주 증식 억제 및 항산화 효과)

  • This study was carried out to determine the inhibitory effects of solvent extracts from Sargassum hemiphyllum on growth of cancer cell lines (AGS human gastric adenocarcinoma and HT-29 human colon cancer cells) and production of lipid peroxides. Inhibitory effects of acetone with methylene chloride extract from S. hemiphyllum on the growth of AGS and HT-29 cancer cells were increased as dose dependent patterns (p<0.05). The methanol extract was more effective on inhibition of growth of AGS. The treatments of hexane, 85% aq. methanol, butanol and water fractions significantly inhibited the growth of cancer cells (p<0.05) and the inhibitory effect was stronger in HT-29. In DCFH-DA (dichlorodihydrofluorescin diacetate) assay, acetone with methylene chloride and methanol extracts showed a stronger inhibitory effect on the production of cellular lipid peroxides (p<0.05) compared with the butanol and hexane fractions. These results indicate that the consumption of S. hemiphyllum may be recommended as a potent functional food for preventing cellular oxidation and cancer.

Pigmentation and Delayed Oxidation of Broiler Chickens by the Red Carotenoid, Astaxanthin, from Chemical Synthesis and the Yeast, Xanthophyllomyces dendrorhous

  • The red carotenoid, astaxanthin was studied to improve the meat quality of broiler chickens. Astaxanthin pigmented chickens and delayed oxidation of lipid in them. Two sources of astaxanthin were used to pigment broiler chickens in a five-wk feeding trial: biological astaxanthin (BA) from the red yeast, Xanthophyllomyces dendrorhous, and chemical astaxanthin (CA) from chemical synthesis. The concentrations of CA (45 mg/kg feed) and BA (22.5 mg/kg feed) were set to give similar levels of pigmentation. The colorimetric values (a and b) of breast muscles were significantly changed by astaxanthin (p${\leq}$0.01). Absorption and accumulation of BA were higher than those of CA, probably due to the high contents of lipids in the yeast (17%). Lipid peroxide formation in skin was significantly decreased by astaxanthin (p${\leq}$0.05). This result indicated that the production of lipid peroxides in the carcasses of broiler chickens during storage could be delayed by astaxanthin. Therefore, astaxanthin could be used as an antioxidant as well as a colorant for broiler chickens.

Effects of Polyacetylene Compounds from Panax Ginseng C.A. Meyer on $CCl_4$-Induced Lipid Peroxidation in Mouse Liver

  • The inhibitory effect of three polyacetylene compounds, panaxydol, panaxynol and panaxytriol isolated from Panax ginseng C.A. Meyer on $CCl_4$induced lipid peroxidation in vivo and in vitro hepatic microsomal lipid peroxidation induced by ADP-$Fe^{3+}$, NADPH and NADPH-cytochrome P-450 reductase were investigated. Their effects on lowering the lipid peroxide levels both in serum and liver and lowering the serum enzyme (GOT, GPT, LDH) activities without the $CCl_4$-induction were also determined. Male ICR mice were pretreated i.p. with polyacetylene compounds or DL-${\alpha}$-tocopherol before administration of $CCl_4$ i.p. and 20 hr after the administration of $CCl_4,$ serum and liver were analyzed. Hepatic microsome was isolated and used for the in vitro NADPH-dependent lipid peroxidation system. Except for panaxynol, treatment with polyacetylenes to control mice did not reduce the levels of lipid peroxides and serum enzyme activities. Panaxynol itself inhibited lipid peroxidation in the liver of normal mice. Polyacetylene compounds protected from the $CCl_4$-induced hepatic lipid peroxidation and lowered serum lipid peroxide levels. Polyacetylenes also inhibited the in virto hepatic microsomal lipid peroxidation in a dose-dependent manner. The results suggest that panaxydol, panaxynol and panaxytriol seem to be the antioxidant components which contribute the anti-aging activities of Panax ginseng C.A. Meyer.

Effects of Onion Juice on Ethanol-Induced Hepatic Lipid Persoxidation in Rats (양파즙이 에탄올에 의한 백서의 지질산화물 생성에 미치는 영향)

  • The effect of onion juice on ethanol -induced lipid peroxidation were studied were studied in rats. The contents of thiobarbituric acid (TBA) -reactants increased significantly in liver thanol(4ml/kg/day) administered -rats. The activities of serum alanine aminotransferase and alkaline phosphatase increased by ethanol administration compared with control group, but alterations of antioxidant enzymes activities in liver of ethanol administered rats were not significant vs control group. The glutathione contents in liver decreased by ethanol , whereas the glutathione level increased in ethanol and onion juice group compared with ethanol group. The contents of hepatic TBA-reactants and serum aminotrasnferase activity in ethanol group were reduced by onion juice administration. In these results, increased hepatic TBA-reactants of liver in ethanol group might be due to decreased glutathione contents in liver. Reduced glutathione (GSH) plays an important roles in the liver in several detoxification and the reduction of lipid peroxides. So the protective effects of onion juice on ethanol-induced increment of TBA-reactants may be due to the increament of lgutathions content. The glutathione depletion by ethanol was an important factor of ethanol-induced cell damage, and the prevention of onion juice to the glutathione depletion reduced by ethanol may be an important factor on the protection from ethanol-induced lipid perpxidation in rats.

Anti-effects of Photodynamic Therapy in Peroxiredoxin IV-induced AMC-HN3 Cell Lines

  • Photodynamic therapy (PDT) is a treatment utilizing the generation of singlet oxygen and other reactive oxygen species (ROS), which selectively accumulate in target cells. Peroxiredoxin (prx) plays an important role in eliminating peroxides generated during metabolism. Prx exert protective antioxidant role in cells though peroxidase activity. The aim of present work is to investigate the cytotoxicity of photofrin-mediated PDT in prx IV-transfectant AMC-HN3 cell lines. We confirmed that PDT has an effect on ROS generation in prx IV-induced cell lines. Treatment of PDT in prx IV-HN3 cell lines inhibits cytotoxic effects. Prx IV-induced HN3 cell lines resists in cell death during PDT. Also, prx IV-HN3 cell lines treated PDT inhibited ROS generation in contrast with vector control. We indicated that prx IV-induced AMC-HN3 cell lines have a function as inhibitors during PDT.

The Effect of Lipid Concentration in Culture Medium on Senescence and Lipid Peroxides Production of Fibroblast from Neonate Rats (배양액내 지방함량의 변화가 신생흰쥐 피부섬유아세포의 노화와 지질과산화물 생성에 미치는 영향)

  • This study was performed to investigate the effects of lipid on cellular senescence, lipid peroxide production, and morphological changes. For this study we used primary skin fibroblasts from neonate rats grown in media various lipid contents. Fibroblasts were cultured until they lost their proliferation potential either in control medium (Dulbecco's modified Eagle's medium supplement with 10% fetal bovine serum) or in media supplemented with various concentrations of lipid-cholesterol rice component from bovine serum. Cumulative population doublings(CPD, as an index of cellular life span), and cellular thiobarbituric acid reactive substances (TBARS, as an index of lipid peroxide) concentrations were measured and morphological changes were observed. CPD were shortened with increasing lipid concentration in media ; 28.12 for cells grown in control medium and 13.42, 11.42, and 6.19 for those grown in 0.1%, 1% and 5% lipid rich components containing media, respectively. Cellular proliferation ratios were those grown in 5% lipid rich components containing media were delayed and they were degenerated soon. TBARS concentrations were increased with increasing concentration of lipid in media. Morphological changes were observed in cells grown in control medium by cellular senescence. Especially lipid droplets were observed in cells grown in 5% lipid rich components containing media. Therefore it seems that lipid contents in media had an effect on cellular proliferation and cellular life span, possibly via lipid peroxide production.

Effectiveness of Nutrition Counseling on Dietary Behavior and Serum Levels of Lipids and Antioxidants in Patients with Cardiovascular Disease (영양상담에 의한 심혈관질환자의 식행동, 혈청 지질 및 항산화영양소 상태 변화)

  • The purpose of this study was to investigate whether nutrition counseling could lead to a beneficial outcomes in patients with cardiovascular disease(CVD). Fifty CVD patients(23 men and 27 women) participated in a dietary counseling program which was based on serum lipid management. Various markers of disease risk including lifestyles, anthropometric indices, eating behaviors, serum lipids, antioxidants, lipid peroxides were measured before and after the program. The program lasted 3-11 months depending on individuals. Waist/hip ratio decreased after nutrition counseling only in women. Smoking and alcohol drinking status did not change significantly after counseling. Food habit score increased significantly in both men and women(p<0.05), and patients consumed mixed cereal rice, fruits, seaweeds, legumes more frequently and snacks less frequently after nutrition counseling (p<0.05). Although intakes of energy, carbohydrate and vitamin B1 were reduced significantly after nutrition counseling, nutrient densities of calcium and iron of diets have increased significantly after nutrition counseling, and those of other micronutrients were not different Serum total cholesterol decreased significantly in men and LDL cholesterol decreased significantly in women. In women lipid peroxide level decreased and $\alpha$-tocopherol level increased significantly, while there was no significantly change in men. We conclude that well-planned nutrition counseling would reduce risks of cardiovascular disease through improving dietary behavior, lipid profile and antioxidant status.

Activation Energy Measurement of the $^{60}Co$ $\gamma$-ray Irradiated Epoxy Resin ($^{60}Co$ $\gamma$-ray 조사에 따른 에폭시 수지의 활성화 에너지 측정)

  • In this paper, we measured the activation energy of the Bisphenol A type Epoxy resin (DGEBA)-Jeffamine system according to an irradiation. We put the mixed solutions to the silicon mold after mixing both DGEBA(216g) and Jeffamine(93.9g), curing agent of amine system. The mixed solutions were cured in the atmosphere during 24hours after finishing the first curing during 4hours in the vacuum oven under $80^{\circ}C$. The mixed solutions were cured in the atmosphere during 24hours after finishing the second curing during 12hours in the vacuum oven under $60^{\circ}C$ again. Prepared samples were irradiated to the dose rate of 8kGy/hr with 500kGy, 700kGy, 1000kGy, 1500kGy, 2000kGy, 2500kGy. Experimental results indicated activation energy of the samples reduced as the irradiation dose increased because of the peroxides of the Epoxy created by the oxidation and the radiation.
