Antitumor Activities of Polysaccharudes fractuibuzed from Zoogloea sp. Against Meth A Cells (Zoogloea sp.의 다당체가 Meth A 세포에 의한 종양형성 억제 효과)

  • The antitumor activities of the cell bound polysaccharide(CBP), water soluble polysaccharide(WSP) and sulfated polysaccharide(SP) of Zoogloea sp. were observe. The results obtained were as follows : 1) The CBP, WSP, and SP showed cytotoxic effect on the Meth A cells in vitro, however, the effect of CBP and WSP was more ten-fold greater than that pf SP. 2) When CBP, WSP, and SP was inoculated into the peritoneal cavity of the Meth A cells transplanted mice, the average survival days tended to prolonged slightly as compared with the control. 3) When Meth A cells were transplanted subcutaneously into the back side of mice, and then CBP, WSP, and Sp was inoculated into the peritoneal cavity of mice, the tumor growth inhibition ratio was 46.9% for WSP, 40.4% for CBP, and 16.2% for SP. 4) The phagocytic activity of peritoneal macrophages elicited with CBP, WSP, and SP was significantly increased than that of control. 5) The production of nitric oxide in the peritoneal macrophages stimulated with CBP, WSP, SP, and LPS aloneo was not increased than that of control. The production of nitric oxide in the peritoneal macrophages stimulated with IFN-r and CBP, IFN-r and WSP and IFN-r and SP was significantly increased than that of control, but in the case of stimulated with IFN-r and WSP was increased 50% for CBP and SP. These results suggest that the CBP, WSP and SP of Zoogloea sp. showed direct cytotoxic effect and tumor growth inhibition on Meth A cells in vitro and in vivo, and induced nitric oxide production of activated macrophages.

  • Recombinant murine stem cell factor (rmSCF) or recombinant murine nerve growth factor (rmNGF) induced the morphological change of large numbers of rat peritoneal mast cells (RPMC). We investigated the role of phosphatidylinositol $3^{l}-kinase$ (PI3-kinase) in receptors-mediated morphological change in RPMC. Exposure of RPMC to PI3-kinase inhibitor, wortmannin, before the addition of rmSCF and rmNGF antagonized those factors-induced morphological change. These results suggest that the PI3-kinase is involved in the signal transduction pathway responsible for morphological change following stimulation of rmSCF and rmNGF and that wortmannin blocks these responses.

  • Objective : This Study was to investigate effects of Takliikgi-tang on the anti-cancer and proliferation of immunocytes, nitric oxide (NO) production of peritoneal macrophages. Methods : We used Takliikgi-tang extract (TLT) with freeze-dried, 8weeks-old male mice and cancer cell lines (L1210, Sarcoma-180) for this Study. The cytotoxicity and proliferation of cells were tested using a colorimetric tetrazoliun assay (MTT assay). Results : 1. TLT was significantly showed cytotoxicity on the L1210, 5-180 cell lines. 2. TLT was significantly increased proliferation of thymocytes in vitro. 3. TLT was significantly increased proliferation of thymocytes and splenocytes in normal mice. 4. TLT was significantly increased NO production from peritoneal macrophages in normal mice. 5. TLT was significantly decreased proliferation of L1210 cells in L1210 cells transplanted mice. 6. TLT was significantly increased proliferation of thymocytes and splenocytes by all-dosage in L1210 cells transplanted mice. 7. TLT was significantly increased NO production from peritoneal macrophages in L1210 cells transplanted mice. Conclusions : The present author thought that TLT had action of anti-cancer by becoming immunocytes activity (NO production, proliferation of thymocytes).

  • Yolk sac tumors are rare malignant germ cell neoplasms that usually arise from the gonads. Extragonadal yolk sac tumors (EGYSTs) frequently occur in the mediastinum in post-pubertal females. EGYSTs in the pelvis are extremely rare, and to date, only thirteen cases have been reported in the English literature. Among them, the primary EGYST of the pelvic peritoneum in post-pubertal females has only been reported in ten cases. The present case describes a 26-year-old female diagnosed with primary peritoneal yolk sac tumor located in the rectouterine pouch. We report clinical and tumor imaging features, including ultrasound, computed tomography (CT), magnetic resonance images (MRI), positron emission tomography-computed tomography (PET-CT), and present a review of the literature.

Effects of the Administration of water extract of Juglandis Semen without Inner cortex and with Inner cortex on Activity of Splenocytes and Macrophages in Mice (호두 속껍질 없는 것과 있는 것의 물 추출물 투여가 생쥐의 비장세포 및 대식세포의 활성에 미치는 영향)

  • The purpose of this research was to investigate the effects of the administration of Juglandis Semen without inner cortex (JE) or with inner cortex (JEIC) on activity of splenocytes and peritoneal macrophages in BALB/C mice. JE (300 mg/kg, p.o.) did not affect the cell viability of T- and B-lymphocytes in murine splenocytes, but JEIC (300mg/kg, p.o.) increased the cell viability of T- and B-lymphocytes. Furthermore, JE decreased the population of B220$^+$ cells in splenocytes, but JEIC enhanced the population of Thyl$^+$ cells. Also, JEIC enhanced the population of splenic CD4$^+$ cells. JE decreased the production of nitric oxide and the phagocytic activity of peritoneal macrophages, but JEIC increased the production of nitric oxide and the phagocytic activity of peritoneal macrophages. These results suggest that JEIC is more potent than JE against the immune response induced by splenocytes and macrophages.

Effect of Bojung-Ikgi-Tang-Gami(BITG) on Immune Response in the Young Mice (보중익기탕가미(補中益氣湯加味)가 새끼 생쥐의 면역능(免疫能)에 미치는 영향(影響))

  • The purpose of this research was to investigate the effects of BITG on immune response in the young mice. BITG(500mg/kg) was administerd per orally once a day for 7 days to the young BALB/C(4 wks old)mice. The administrationo of BITG enhanced the cell viability of splenocytes, but did not affect that of thymocytes. In vitro system, BITG decreased the cell viability of thymocytes and splenocytes at the concentration of $100{\mu}g/m{\ell}$. The administration of BITG did not affect DNA fragmentation of thymocytes, but decreased that of splenocytes. The administration of BITG increased the population of Thy1+ cell and CD4+CD8- cell in splenocytes. In addition, BITG decreased the production of NO and increased the lucigenin chemiluminescence from peritoneal macrophages. These results suggest that BITG enhances the specific-immune and nonspecific-immune response via increase of cell viability in splenocytes and of phagocytic activity in peritoneal macrophages.

  • Objectives : Yukmi-Jihwang-Tang(YJT) has been used in Oriental Medicine as a drug for tonifying and nourishing yin. So, the purpose of this Study was to investigate effects of Yukmi-Jihwang-Tang extract(YJTE) on the T-lymphocytes and peritoneal macrophages in mice. Methods : YJTE consists of the following components; Rehmaniae Radix Preparata(熟地黃), Dioscoreae Rhizoma(山藥), Corni-Fructus(山茱萸), Hoelen alba(白茯), Moutan Cortex Radicis(牧丹皮), Alismatis Rhizoma(澤瀉). Result : The results of this Study were obtained as follow; The administration of YJTE did not affect T-lymphocytes apoptosis. YJTE decreased sub-population of TH and TC cells, and proliferation of T-lymphocytes too. But YJTE accelerated phagocytic activity and Nitric Oxide(NO) production from peritoneal macrophages in mice. Conclusions : These results suggest that the administeration of YJTE suppresses T-cell mediated immunity, but activates peritoneal macrophages.

Macrophage activation by glycoprotein isolated from Dioscorea batatas

  • We demonstrate that glycoprotein isolated from Dioscorea batatas (GDB) activates macrophage function. Analysis of the infiltration of macrophages into peritoneal cavity showed GDB treatment significantly increased the recruitment of macrophages into the peritoneal cavity. In order to further confirm and investigate the mechanism of GDB on macrophage activation, we analyzed the effects of GDB on the cytokine expression including IL-$1{\beta}$, TNF-${\alpha}$, and IL-6 in mouse peritoneal macrophages. GDB increased the expression of IL-$1{\beta}$, TNF-${\alpha}$, and IL-6. Cytokine induction by GDB was further confirmed by RT-PCR and ELISA in mouse macrophage cell line, RAW264.7 cells. Treatment of RAW264.7 cells with GDB produced strong induction of NF-${\kappa}B$ DNA binding and MAPK phosphorylation, markers for macrophage activation and important factors for cytokine gene expression. Collectively, this series of experiments indicates that GDB stimulates macrophage activation.

Effects of Calendula Officinalis L. Extract on the Activation of Murine Lymphocytes and Macrophages (금잔화 추출물이 생쥐의 임파구 및 대식세포의 활성에 미치는 영향)

  • The purpose of this research was to investigate the effects of Calendula officinalis extract (CE) on the activation of murine lymphocytes and macrophages. CE was administered p.o. once a day for 7 days at the concentration of 500 mg/kg. The administration of CE increased the viability of thymocytes, but decreased the viability of splenocytes. Also, CE increased the viability of thymocytes and splenocytes at the concentration of $5{\mu}g$/ml in vitro. The administration of CE did not affect the population of thymic $CD4^+$/$CD8^+$ cells and splenic $CD4^+$/$CD8^+$ cells. Furthermore, the administration of CE enhanced the phagocytic activity of peritoneal macrophages, but decreased the phagocytic activity in vitro. CE decreased the production of nitric oxide from peritoneal macrophages in vivo and in vitro system. These results suggest that CE enhance of cell viability by a direct influence on thymocytes and phagocytic activity by an indirect influence on peritoneal macrophages.
