Morphological Study on the Epithelial Cells in the Gall Blader of Vertebrates (각급 척추동물의 담낭 상피세포에 대한 형태학적 연구)

  • These experiments were performed in order to study histologically and histochemically on the epithelial cells of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus. The results of the observation were as follows: 1. There were different cell types in the epithelium of gall bladder in each animal and it could not be supported histochemically that the epithelia cells of gall bladder were divided into two cell types of the rod-shaped and barrel-shaped ones. 2. The epithelium of gall bladder in Carassius carassius, Bufo bufo gargarizans, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus was simple columnar epithelium. 3. The eosinophilities of cytoplasm in the epithelial cells of gall bladder were in uniform stronger in the upper portion of nucleus in Carassius carassius, Bufo bufo gargarizans and Natrix tigrina lateralis than its other portions, and in Urloncha striata var. domesticus and Lepus cuniculus var. domesticus existed uniformly in all portions, but there were many non-eosinophilic cells in Bufo bufo gargarizans and many cells that weakly eosinophilic around nucleus in Bos taurus var. domesticus. 4. The periodic acid Schiff's reactivities in the epithelial cells of gall bladder were different in each other and the epithelial cells in PAS reaction were divided into two cell types of the dark and light ones. There presented the light cells of 6.4%, 4.3% and 3.7% of epithelial cell of gall bladder in Carassius carassius, Bufo bufo gargarizans and Urloncha striata var. domesticus for each other, but were not presented in Natrix tigrina lateralis and Bos taurus var. domesticus. 5. The ninhydrin-Schiff-active proteins were much in the epithelial cells of gall bladder in Bos taurus var. domesticus, Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis in order and were much in epithelial cells in the upper portion of mucosal folds in Carassius carassius, Urloncha striata var. domesticus and Natrix tigrina lateralis, and the ninhydrin-Schiff-active protein of the epithelium of gall bladder in Bufo bufo gargarizans was uniformly distributed. 6. The epithelial cells of gall bladder in Carassius carassius, Natrix tigrina lateralis, Urloncha striata var. domesticus and Bos taurus var. domesticus had no stain reactivity or weak stain reactivity to neutral fat and all epithelial cells in Bufo bufo gargarizans had strong stain reactivity, though they were different in quantity of epithelial cell portion. 7. The stain reactivities to RNA and DNA were stronger in the epithelial cells of the upper portion of mucosal fold than in those of other portions.

Characteristics of Growth, Yield, and Physiological Responses of Small-Sized Watermelons to Different Soil Moisture Contents Affected by Irrigation Starting Point in a Plastic Greenhouse (소형 수박 시설 재배 시 관수개시점에 따른 토양수분 함량별 생육, 수량 및 생리적 반응 특성 구명)

  • Watermelon yield mainly depends on soil water content controlled by irrigation in a plastic greenhouse. In this study, we investigated the effect of different soil moisture contents affected by irrigation starting point on growth, yield, and physiological responses of small-sized watermelons. Irrigation was initiated at 5 different levels of soil water content as a starting point with soil moisture detecting sensor after 14 days of transplanting, and stopped at 7 ~ 10 days before harvest. These treatments were compared with the conventional periodic irrigation as control. When soil had the lowest moisture content (-50 kPa), the overall shoot growth was retarded, but the root length and root dry weight increased. The photosynthetic parameters (photosynthetic rate, stomatal conductance, and transpiration rate) of watermelon leaves decreased significantly in the lowest soil moisture content (-50 kPa). On the other hand, the photosynthetic rates of watermelon leaves grown with irrigation starting point between -20 and -40 kPa were observed to be higher than those of other treatments. Fruit set rate and marketable fruit yield increased significantly at -30 kPa and -40 kPa. Proline, abscisic acid (ABA), total phenol and citrulline, which are known to contribute to stress tolerance under drought condition, increased as soil water content decreased, particularly, the largest increases were recorded at -50 kPa. From these results, it was found that an appropriate water supply adjusted with an irrigation starting point between -30 and -40 kPa could help to keep favorable soil water content during the cultivation of small-sized watermelons, promoting the marketable fruit production as well as inducing the vigorous plant growth and reproductive development.

Effects of Sulfur Dioxide Exposure in Histological Structure and Mucosubstances of the Nasal Respiratory Mucosa of Rat ($SO_2$ 흰쥐 비강 호흡부 점막의 조직학적 구조 및 점액질에 미치는 영향에 관한 연구)

  • This experiment was performed to investigate the effects of sulfur dioxide on the histological changes, properties of mucosubstances and glycoconjugates of the nasal respiratory mucosa in the rat. Sprague-Dawley male rats weighing about 200~250g were divided into a control group and SO$_2$ exposed groups. Again SO$_2$ exposed groups were divided into 10 ppm, 25 ppm, 50 ppm, 100 ppm, and 200 ppm subgroups, according to concentrations of SO$_2$ and each SO$_2$exposed groups were divided into 1, 3 and 6 hours groups. For the histological changes, hematoxylin-eosin(H-E) and periodic acid Schiff's(PAS) stainings were used, and for the properties of mucosubstances, PAS, alcian blue (AB) pH 2.5, pH 2.5-PAS, AB pH 1.0 and aldehyde fuchsin (AF) pH1.7-AB pH 2.5 were used. In all the SO$_2$ exposed groups, loss of cilia and detachment of epithelial cells, vacuolation of goblet cells were observed in the respiratory epithelium while epithelial squamous metaplasia and intraepthelial mucous cells were observed in the higher concentration of SO$_2$ and the degree of the loss cilia was higher according as concentration was higher and exposed time was longer. The intraepitheial mucous cells appeared most remarkable in the 50 ppm SO$_2$ exposed group. The numbers of goblet cells and acini of nasal septal gland were varied according to concentration of SO$_2$ and exposed time, but the numbers in the 25 ppm and 50 ppm, SO$_2$ exposed increased remarkably. However, the numbers in the 100 ppm and 200 ppm SO$_2$ exposed group had a tendency to decrease noticeably, or disappeared.

  • Background : Goblet cell hyperplasia is a critical pathological feature in hypersecretory diseases of the airways. A bacterial infection of the lung is also known to induce inflammatory responses, which can lead to the overproduction of mucus. Recently, mucin synthesis in the airways has been reported to be regulated by neutrophilic inflammation-induced epidermal growth factor receptor (EGFR) expression and activation. In addition, it was reported that migration of the activated neutrophils is dependent on the matrix metalloproteinases (MMPs), especially MMP-9. In this study, bacterial lipopolysaccharide (LPS)-induced goblet cell hyperplasia and mucus hypersecretion by EGFR cascade, resulting from the MMPs-dependent neutrophilic inflammation were investigated in the rat airways. Methods : Pathogen-free Sprague-Dawley rats were studied in vivo. Various concentrations of LPS were instilled into the trachea in $300{\mu}{\ell}$ PBS (LPS group). Sterile PBS ($300{\mu}{\ell}$) was instilled into the trachea of the control animals (control group). The airways were examined on different days after instilling LPS. For an examination of the relationship between the LPS-induced goblet cell hyperplasia and MMPs, the animals were pretreated 3 days prior to the LPS instillation and daily thereafter with the matrix metalloproteinase inhibitor (MMPI; 20 mg/Kg/day of CMT-3; Collagenex Pharmaceuticals, USA). The neutrophilic infiltration was quantified as a number in five high power fields (HPF). The alcian blue/periodic acid-Schiff (AB/PAS) stain were performed for the mucus glycoconjugates and the immunohistochemical stains were performed for MUC5AC, EGFR and MMP-9. Their expressions were quantified by an image analysis program and were expressed by the percentage of the total bronchial epithelial area. Results : The instillation of LPS induced AB/PAS and MUC5AC staining in the airway epithelium in a time- and dose-dependent manner. Treatment with the MMPI prevented the LPS-induced goblet cell hyperplasia significantly. The instillation of LPS into the trachea induced also EGFR expression in the airway epithelium. The control airway epithelium contained few leukocytes, but the intratracheal instillation of LPS resulted in a neutrophilic recruitment. A pretreatment with MMPI prevented neutrophilic recruitment, EGFR expression, and goblet cell hyperplasia in the LPS-instilled airway epithelium. Conclusion : Matrix metalloproteinase is involved in LPS-induced mucus hypersecretion, resulting from a neutrophilic inflammation and EGFR cascade. These results suggest a potential therapeutic role of MMPI in the treatment of mucus hypersecretion that were associated with a bacterial infection of the airways.
