Hypocholesterolemic metabolism of dietary red pericarp glutinous rice rich in phenolic compounds in mice fed a high cholesterol diet

  • BACKGROUND/OBJECTIVES: The purpose of the current study was to investigate the effect of red pericarp glutinous rice rich in polyphenols (Jakwangchalbyeo, red rice) on serum and hepatic levels of cholesterol and hepatic protein expression linked to synthesis and degradation of cholesterol in a hypercholesterolemic mice diet as compared with brown rice. MATERIALS/METHODS: C57BL/6 male mice were randomly divided into four groups (n = 5 each), which were fed different diets for a period of 12 weeks: American Institute of Nutrition (AIN)-93G diet, AIN-93G diet with 2% cholesterol, brown rice with 2% cholesterol, or red rice with 2% cholesterol. RESULT: Consumption of red rice resulted in a significant decrease in serum level of low-density lipoprotein cholesterol and hepatic levels of triglyceride and total-cholesterol. Expression of acyl-coenzyme A cholesterol acyltransferase-2 (ACAT-2), sterol regulatory element binding protein-2 (SREBP-2), and 3-hydroxyl-3-methylglutaryl coenzyme A (HMG-CoA) reductase was decreased, while expression of phosphorylated adenosine monophosphate activated protein kinase (p-AMPK)/AMPK ratio, cholesterol 7-${\alpha}$-hydroxylase (CYP7a1), and sterol 12-${\alpha}$-hydroxylase (CYP8b1) was increased in mice fed red rice. Brown rice had similar effects on cholesterol metabolism, but the effect of red rice was significantly greater than that of brown rice. CONCLUSIONS: The current study suggested that red rice had a hypocholesterolemic effect by lowering hepatic cholesterol synthesis through ACAT-2, HMG-CoA reductase, and SREBP-2, and by enhancing hepatic cholesterol degradation through CYP7a1 and CYP8b1 in mice fed a hypercholesterolemic diet.

Colletotrichum fructicola, a Member of Colletotrichum gloeosporioides sensu lato, is the Causal Agent of Anthracnose and Soft Rot in Avocado Fruits cv. "Hass"

  • The filamentous Ascomycota Colletotrichum gloeosporioides sensu lato is a fungus that has been reported worldwide as a causal agent of anthracnose disease in avocado and other crops. In Mexico, this species affects fruits from an early stage of development in the orchard until the post-harvest stage. Although fungicides are continuously applied to control Colletotrichum species, pericarp cankers and soft rot mesocarp in fruits are still frequently observed. Considering the lack of a precise description of the causative agent, the aim of the current study was to determine the pathogens involved in this symptomatology. Twenty-four isolates were consistently obtained from the pericarp of avocado fruits cv. "Hass" collected in the central avocado-producing area of Mexico. Morphological features such as colony growth, conidia size, and mycelial appressorium were assessed. Bayesian multilocus phylogenetic analyses were performed using amplified sequences of the internal transcribed spacer region of the nuclear ribosomal DNA; actin, chitin synthase, glyceraldehyde-3-phosphate dehydrogenase partial genes; and APn2-Mat1-2 intergenic spacer and mating type Mat1-2 partial gene from the nine selected isolates. In addition, fruits were inoculated with a conidial suspension and reproducible symptoms confirmed the presence of Colletotrichum fructicola in this area. This pathogenic species can now be added to those previously reported in the country, such as C. acutatum, C. boninense, C. godetiae, C. gloeosporioides, and C. karstii. Disease management programs to reduce the incidence of anthracnose should include C. fructicola to determine its response to fungicides that are routinely applied, considering that the appearance of new species is affecting the commercial quality of the fruits and shifting the original population structure.

Histopathological Changes in Tissues of Bithynia siamensis goniomphalos Incubated in Crude Extracts of Camellia Seed and Mangosteen Pericarp

  • The present study was performed to observe histopathological changes in tissues of Bithynia siamensis goniomphalos (Gastropoda, Bithyniidae) incubated in crude extract solutions of camellia (Camellia oleifera) seed and mangosteen (Garcinia mangostana) pericarp, and furthermore to estimate the molluscicidal effects of 2 plant substances. Substantial numbers of bithyniid snails were incubated in various concentrations of 2 plant solution for 24 hr. As the positive control, snails incubated in various concentrations of niclosamide, a chemical molluscicide, were used. The histopathological findings were observed in sectioned snail specimens of each experimental and control groups. The results showed that both camellia and mangosteen extracts had molluscicidal effects at 24 hr with 50% lethal concentration ($LC_{50}$) at concentrations of 0.003 and 0.002 g/ml, respectively, while niclosamide had $LC_{50}$ at concentrations 0.599 ppm. B. siamensis goniomphalos snail tissues (foot, gill, and digestive system) showed disruption of columnar muscle fibers of the foot, reduction of the length and number of gill cilia, numerous mucous vacuoles, and irregularly shaped of epithelial cells. Irregular apical and calciferous cells, dilatation of the digestive gland tubule, and large hemolymphatic spaces, and irregular apical surfaces, detachment of cilia, and enlargement of lysosomal vacuoles of epidermis were also shown in all groups. By the present study, it is confirmed that 2 plants, camellia and mangosteen, are keeping some substance having molluscicidal effects, and histopathological findings obtained in this study will provide some clues in further studies on their action mechanisms to use them as natural molluscicides.

The Effect of Irrigation Concentration on the Growth and Fruit Quality of Sweet Pepper(Capsicum annuum L.) in Fertigation (관비재배에서 급액농도가 착색단고추의 생육과 품질에 미치는 영향)

  • Objective of this research was to investigate the effect of irrigation concentration on the growth and fruit quality of sweet pepper(Capsicum annuum L.) in fertigation. The sweet pepper was grown for 210 days with irrigation concentration of EC 0.5, 1.0, 1.5, 2.0, and 3.0 dSㆍ$m^{-1 }$ in fertigation nutrient solution developed by European Vegetable R & D Center, Belgium. The net $CO_2$ assimilation and transpiration rate were the highest in the treatment of 2.0 dSㆍ$m^{-1 }$. The pH in the soil was range of 5.63 ~6.03, the EC increased as the irrigation concentration was getting higher. The SPAD value also increased as the irrigation concentration was getting higher, N, P, K, Mg except Ca were highest in the treatment of EC 2.0 dSㆍ$m^{-1 }$. The growth was good in the treatment of EC 2.0 dSㆍm$m^{-1 }$. The fruit length, width, firmness, and pericarp thickness had no statistical differences among treatments, the fruit fresh weight and dry weight were good in the treatment of EC 2.0 dSㆍ$m^{-1 }$ the yield was good in the treatment of EC 1.5 dSㆍ$m^{-1 }$ and EC 2.0 dSㆍ$m^{-1 }$ The sugar contents was the highest in the treatment of EC 2.0 dSㆍ$m^{-1 }$ with 9.0$^{\circ}$Brix. In conclusion, the optimal irrigation concentration for sweet pepper fertigation was EC 2.0 dSㆍm$^{-1}$ .

Botanical Characteristics, Fresh Yield and Table Qualities of CNU Waxy Corn Hybrids

  • A total of CNU 28 hybrids were developed at the CNU Corn Breeding Lab. were evaluated to identify new cultivars in botanical characteristics, fresh yield per 10a and taste qualities. Most of these hybrids were stable in environmental stresses such as lodging, disease and insects. Stem height ranged from 115.0 to 239.3 cm, and ear height ranged from 30.7 to 107.0 cm. The ear height to stem height ratio was showed low than 50% of standard as a stable plant type to lodging. The range of ear length was 14.2 cm to 23.0 cm. Especially, CNU 13H-73 was very longest ear as a 23 cm. The fresh yield per 10a was high in purple of CNU13H-79 hybrid than control hybrid Miheuckchal, and CNU13H-73 in white hybrid was similar Yeonnong check hybrid. The 100-kernel weights in CNU13H-3 and CNU13H-9 hybrids were higher than that of the control hybrid. CNU13H-98 among hybrids had a 100-kernel weight of 20.32 g, which was heavier than that of the control hybrid Daehackchal Gold 1. The average pericarp thickness was $41.4{\mu}m$, CNU13H-46 among hybrids had a very thin pericarp as a $35.5{\mu}m$. The mean sugar content of the used hybrids was 14.95 brix%; CNU13H-73 and CNU13H-55 had higher than Mibak2 as a control hybrid.

Quality Changes of Fresh Green Pepper Paste during Storage (생청고추 페이스트의 저장 과정 중 품질 변화)

  • To maintain flavor and color of fresh green pepper, the fresh green pepper paste was directly prepared from fresh green pepper. The characteristic of fresh green pepper paste and processing properties were investigated, and the effect of salt, glucose, acid and heat on product quality during processing and storage were studied. After the processed fresh green pepper paste was stored at 3$0^{\circ}C$ and 5$^{\circ}C$ without light for 6 months. The weight and pH of pericarp were 86% of total and 4.5~5.0, respectively. Addition of 10% salt, 5% glucose, and 0.1 dl-malic acid to the fresh green pepper paste maintain flavor of fresh green pepper could be preserved for 6 months at 5$^{\circ}C$. Panel test showed fresh green pepper paste which was made of 10% salt, 5% glucose and 0.1% dl-malic acid to the fresh green pepper was quite acceptable.

Detecting Seed-Borne Fungi of Rice and Transmission of Helminthosporium oruzae in Germinating Seed (벼종자수반진균의 검정 및 종자발아에 따른 깨씨무늬병균의 전염경로)

  • From the sample seeds of rice cultivar Palkoeng with the brown spot symptom, Helminthosporium oryzae was detected in $86\~92\%$ of hulls including empty glumes, lemma, palea, and rachilla in five to seven days of incubation. When the seeds were dehulled, the percentage detection decreased to $74\~83\%$ of pericarps. The fungus sporulated on whole surface of the seed and was detected within about ten cell layers deep of endosperm but not detected in embryo. Fusarium moniliforme was found in embryo as well as pericarp and endosperm. The frequency of F. roseum, Trichoconis padwickii, Curvularia spp., Atternaria tenuis, Cladosporium cladosporoides and Phoma sp. was less than three percent of hulls and pericarps. In seven days of incubation, $60\~87\%$ of the germinating seeds showed the symptom of H. oryzae while $13\~40\%$ were apparently healthy. This fungus in hilum of infected rice transmitted through pericarp to plumule shoot and radicle of the germinating seed.
